Phosphonates as Modulators of Brain Chemistry.

The present review describes the current knowledge on phosphonates designed for brain disease therapeutics, a less explored aspect of the studies on their medicinal chemistry. Because of their structural similarity with phosphates and of the ubiquitous presence of phosphate-dependent processes in the brain, phosphonate compounds are able to influence brain chemistry, interfering with neurotransmission and phosphorous-dependent bioprocesses. One of the main applications for phosphonates in the brain is Alzheimer's disease, for which they can be new drug candidates and/or afford innovative strat

Targeting receptor-ligand chemistry for drug delivery across blood-brain barrier in brain diseases.

The blood-brain barrier (BBB) is composed of a layer of endothelial cells that is interspersed with a series of tight junctions and characterized by the absence of fenestrations. The permeability of this barrier is controlled by junctions such as tight junctions and adherent junctions as well as several cells such as astrocytes, pericytes, vascular endothelial cells, neurons, microglia, and efflux transporters with relatively enhanced expression. It plays a major role in maintaining homeostasis in the brain and exerts a protective regulatory control on the influx and efflux of molecules. Howev

park+/+ and park-/- Drosophila have sexually dimorphic brain redox chemistry.

Sexual dimorphism in Parkinson's disease (PD) pathophysiology is poorly understood. Elucidating consequences of disease-causing mutations on brain redox chemistry may reveal therapeutic targets for all people with PD. We report that male Drosophila had increased hydrogen peroxide and glutathione (G-SH) redox disequilibrium in vulnerable dopaminergic neuron mitochondria. Levels of cysteine and oxidized cystine were decreased, with cysteine/cystine ratios (indicating less oxidative stress) and G-SH levels being elevated in parkin-null (park-/-) Drosophila brains, and more so in males. We report

Prenatal undernutrition: effects on behavior, brain chemistry and neuroanatomy in rats.

The behavior and cerebral effects of prenatal protein-calorie undernutrition were investigated in newborn and postweanling rat pups. At birth, prenatally undernourished (PU) animals showed a deficit in body weight; however, by day 15 the difference was diminished and not significant. On days 13-18, PU pups required more trials for reversal learning of a water-escape response in a T-maze than control (C) pups; however, on days 31-35, there were no difference between groups when trained to learn an active-avoidance response. Assessment of brain development showed that at birth, DNA and protein c

Effects of dietary choline on memory and brain chemistry in aged mice.

The purpose of this study was to investigate in more detail the characteristics of the age-related extension of the retrograde amnesia gradient previously demonstrated in a passive avoidance task [6]. In Experiment 1, it was found that while 2-3 month old mice were susceptible to the amnesic effects of anisomycin (ANI) only when given prior to 15 min post-training, memory of 14-16 month old mice was susceptible to disruption when ANI was given as late as 20 min post-training, and retention of 17-20 month old mice was impaired when ANI was injected even as late as 30 min after training. Glutathione in brain oxidative stress: from redox chemistry to clinical imaging.

Glutathione (GSH), a tripeptide composed of glutamate, cysteine, and glycine, is the most abundant intracellular low-molecular-weight thiol and serves as a central regulator of cellular redox homeostasis. In addition to its well-established roles in antioxidant defense and detoxification, GSH is essential for maintaining mitochondrial function and cellular viability. Since its discovery more than a century ago, extensive research has progressively revealed the diverse biological functions of GSH. More recently, growing evidence from experimental and clinical studies has demonstrated that disru

Citalopram and fluoxetine affects blood chemistry, hematology and brain serotonin in rats.

BACKGROUND: Depression is caused as a result of combination of genetic, biochemical, environmental, and psychological factors. Citalopram and fluoxetine are antidepressants which are considered the current standard for depression treatment. There are little or no reports as to whether these antidepressants affect blood chemistry and haematological parameters in humans. OBJECTIVE: The effects of citalopram and fluoxetine on blood chemistry, hematology and brain serotonin in rats were investigated. METHODS: Forty-five Sprague Dawley male albino rats (140.69 +/- 5.86g) were divided into 3 equal

Explore the Functional Connectivity between Brain Regions during a Chemistry Working Memory Task.

Previous studies have rarely examined how temporal dynamic patterns, event-related coherence, and phase-locking are related to each other. This study assessed reaction-time-sorted spectral perturbation and event-related spectral perturbation in order to examine the temporal dynamic patterns in the frontal midline (F), central parietal (CP), and occipital (O) regions during a chemistry working memory task at theta, alpha, and beta frequencies. Furthermore, the functional connectivity between F-CP, CP-O, and F-O were assessed by component event-related coherence (ERCoh) and component phase-locki

Pregabalin rectifies aberrant brain chemistry, connectivity, and functional response in chronic pain patients.

BACKGROUND: Chronic pain remains a significant challenge for modern health care as its pathologic mechanisms are largely unknown and preclinical animal models suffer from limitations in assessing this complex subjective experience. However, human brain neuroimaging techniques enable the assessment of functional and neurochemical alterations in patients experiencing chronic pain and how these factors may dynamically change with pharmacologic treatment. METHODS: To identify the clinical action of pregabalin, a proven analgesic, the authors performed three complementary brain neuroimaging proced

Imaging and recording of synaptic zinc in the brain by covalent chemistry-based semisynthetic biosensors.

Labile zinc ions (Zn2+) are stored in glutamatergic vesicles at specific excitatory synapses in the central nervous system and released into synaptic clefts in an activity-dependent manner. However, the precise roles of synaptic Zn2+ remain unclear due to a lack of tools capable of analyzing Zn2+ with synapse-level spatial resolution in vivo. Here, we developed neuroreceptor-based semisynthetic biosensors that function as integrating dosimeters, recording synaptic Zn2+ by covalent chemistry in the living mouse brain. Using a chemical knock-in strategy, we introduced activity-based Zn2+ probes

Distinct sensory atypicalities bridge the gap between brain chemistry and motor dysfunction in autism.

Sensory and motor difficulties are common in autism. Altered excitation-inhibition (E-I) balance is a putative framework for understanding atypical sensory and motor function. We investigated whether sensory differences of autism mediate motor difficulties of autism via differences in E-I balance. 106 children were included in the study (Autism n = 44, Typical development children (TDC) n = 62, age 10.32 ± 1.49). E-I balance was assessed through magnetic resonance spectroscopy (MRS), quantifying Glutamate and Glutamine (Glx) and Gamma-Aminobutyric Acid (GABA) in primary sensorimotor cortex (SM

Using novel click chemistry algorithm to design D3R inhibitors as blood-brain barrier permeants.

Dopamine receptor D3 (D3R) has gained attention as a promising therapeutic target for neurological disorders. In this study, an innovative in silico click reaction strategy was employed to identify potential D3R binders. The ligand template, 1-phenyl-4-[4-(1H-1,2,3-triazol-5-yl)butyl]piperazine, with substitution at the 1,2,3-triazole ring, served as the starting point. Generated compounds underwent filtration based on their brain-to-blood concentration ratio (logBB), leading to the identification of 1-{4-[1-(decahydronaphthalen-1-yl)-1H-1,2,3-triazol-5-yl]butyl}-4-phenylpiperazine as the most

Impact of Conjugation Chemistry on the Pharmacokinetics of Peptide-Polymer Conjugates in a Model of Traumatic Brain Injury.

Traumatic brain injury (TBI) remains a leading cause of long-term disability and mortality; however, there are no effective therapies to mitigate secondary injury and long-term neurological impairments. After the initial mechanical insult, there is a secondary injury that leads to neuroinflammation and blood-brain barrier (BBB) disruption, both of which are linked to changes in the extracellular matrix (ECM). A short peptide sequence, CAQK (Cys-Ala-Gln-Lys), targets upregulated ECM proteoglycans after TBI and has exhibited therapeutic properties in preclinical TBI studies. However, like many p

Click-Chemistry-Enhanced Dynamic Light Scattering Immunoassay for Ultrasensitive GFAP Detection in Traumatic Brain Injury.

Traumatic brain injury (TBI) requires a rapid and objective assessment; however, current diagnostic methodologies are constrained by subjectivity, reliance on imaging techniques, and insufficient sensitivity for the early detection of biomarkers. In this study, we present a click-chemistry-enhanced dynamic light scattering (DLS) immunoassay for the ultrasensitive detection of glial fibrillary acidic protein (GFAP), a clinically significant blood biomarker for brain injury. Unlike conventional DLS immunoassays, which rely on weak antigen-antibody interactions to drive DLS probe aggregation, the

Abnormal brain chemistry in chronic back pain: an in vivo proton magnetic resonance spectroscopy study.

The neurobiology of chronic pain, including chronic back pain, is unknown. Structural imaging studies of the spine cannot explain all cases of chronic back pain. Functional brain imaging studies indicate that the brain activation patterns are different between chronic pain patients and normal subjects, and the thalamus, and prefrontal and cingulate cortices are involved in some types of chronic pain. Animal models of chronic pain suggest abnormal spinal cord chemistry. Does chronic pain cause brain chemistry changes? We examined brain chemistry changes in patients with chronic back pain using

Functional Connectivity is Associated With Altered Brain Chemistry in Women With Endometriosis-Associated Chronic Pelvic Pain.

UNLABELLED: In contrast to women with relatively asymptomatic endometriosis, women with endometriosis-associated chronic pelvic pain (CPP) exhibit nonpelvic hyperalgesia and decreased gray matter volume in key neural pain processing regions. Although these findings suggest central pain amplification in endometriosis-associated CPP, the underlying changes in brain chemistry and function associated with central pain amplification remain unknown. We performed proton spectroscopy and seed-based resting functional connectivity magnetic resonance imaging to determine whether women with endometriosis

Effects of oral administration of energy drinks on blood chemistry, tissue histology and brain acetylcholine in rabbits.

BACKGROUND: Energy drinks are canned or bottled carbonated beverages that contain large amounts of caffeine and sugar with additional ingredients, such as B-Vitamins, amino acids and herbal stimulants. Previous reports have shown that consumption of large amounts of these energy drinks may result in adverse health consequences. OBJECTIVE: The present study is to ascertain if oral administration of energy drinks, such as "power horse" and "red bull", may affect blood chemistry, tissue histology and acetyl choline levels in rabbits. METHODS: Five ml of power horse and red bull energy drinks, c
