Insight into the Genome of Brochothrix thermosphacta, a Problematic Meat Spoilage Bacterium.

Brochothrix thermosphacta is a dominant but poorly studied meat spoilage organism. It is a close relative of the foodborne pathogen Listeria monocytogenes, and Brochothrix constitutes the second genus in the Listeriaceae family. Here, the genomes of 12 B. thermosphacta strains were sequenced, assembled into draft genomes, characterized, and compared with the genomes of Brochothrix campestris and L. monocytogenes Phenotypic properties including biogenic amine production and antibiotic and heavy metal susceptibilities were tested. Comparative genomic analyses revealed a high degree of similarity

Genomic Relationship Between Brochothrix campestris and Its Phages: A Cross-Replicon and Interspecies Perspective.

Background/Objectives: The bacterium Brochothrix campestris is closely related to Brochothrix thermosphacta, a known food spoilage agent, and Listeria monocytogenes, the causative agent of listeriosis. B. campestris garnered attention several years ago because it produces brochocin-C, a bacteriocin capable of inhibiting the growth of certain pathogens. It has been recently suggested that phages play a significant role in the evolution of B. thermosphacta, similar to the role they play for L. monocytogenes. However, understanding the role of phages in the evolution of B. campestris has been cha

Synergistic antibacterial effects of zinc oxide with cinnamaldehyde or carvacrol against Brochothrix thermosphacta and their application in chicken breast preservation.

Brochothrix thermosphacta (B. thermosphacta) is a significant spoilage organism in meat products, posing a major challenge to food safety. The research investigated the individual and combined antibacterial effects of zinc oxide nanoparticles (ZnO), cinnamaldehyde (Cin) and carvacrol (Car) against B. thermosphacta. The findings showed that ZnO, Cin and Car had considerable antibacterial activity, with minimum inhibitory concentrations of 4, 0.5 and 2 mg/mL respectively. Combination of ZnO-Cin and ZnO-Car assays demonstrated synergistic interactions, and the fractional inhibitory concentration

Modeling the growth and volatile metabolite production of spoilage-causing Brochothrix thermosphacta on solid meat substrates under modified atmospheres.

Spoilage microorganisms including Brochothrix thermosphacta are associated with various volatile organic compounds (VOCs) and off-odors in meat. Modified atmosphere packaging (MAP) limits microbial growth and affects metabolic activities. However, the exact impact of gas compositions on B. thermosphacta still remains unclear, especially regarding the direct relationship between its growth and VOC accumulation. This study thus aimed to investigate and model these behaviors by growing B. thermosphacta on solid pork simulation media under different packaging atmospheres. Five O2/CO2/N2 ratios (v/

The off-odor identification and CO2-inhibition for high‑oxygen packaged beef spoilage associated with Brochothrix thermosphacta growth and metabolism.

Controlling bacterial spoilage off-odor is a major challenge for meat preservation, in which the influences of packaging systems on the microbial-induced off-odor characteristics and formation need further in-depth exploration. This study examined the CO2 effect on high‑oxygen packaged beef spoilage (specifically for off-odor) caused by Brochothrix thermosphacta growth and targeted diacetyl/acetoin biosynthetic pathway during chilled storage, and it will provide a new insight into understanding the odor preservation mechanism of CO2 for packaged beef. Compared with CMAP (50% O2/50% N2), TMAP (

Multi-target prediction in volatolomics with deep neural networks: Modeling volatile organic compounds produced by Brochothrix thermosphacta under modified atmospheres.

The microbial spoilage of meat is associated with the generation of volatile organic compounds (VOCs) and off-odors. While predictive modeling benefits shelf-life determination, predicting the volatolome is very complex as the related microbial metabolism is easily affected by the packaging O2/CO2 composition. Whereas traditional supervised learning mostly focuses on predicting one single target, this study introduces the concept of multi-target prediction (MTP) by presenting a study on predicting multiple VOCs produced by Brochothrix thermosphacta under different packaging atmospheres. The cytochrome composition of the meat spoilage bacterium Brochothrix thermosphacta: identification of cytochrome a3-and d-type terminal oxidases under various conditions.

Brochothrix thermosphacta, grown in batch culture in a yeast-dextrose broth, at temperatures from 30 degrees C to 10 degrees C, contained diverse membrane-bound respiratory cytochromes. Under conditions of moderate aeration, cytochromes of the a-, b- and d-type were detected at all growth temperatures, but the proportions changed as a function of temperature, with the spectra of cells grown at 10 or 15 degrees C being dominated by a-type cytochrome(s). Cytochrome a3 was detected by its reactions with CO and cyanide in cells from all growth conditions. An additional cytochrome a, which was not

Changes in volatile organic compounds of tray-packed fresh beef sausages under refrigeration: The roles of dominant spoilage bacteria.

This study aimed to investigate the change of volatile organic compounds (VOCs) in fresh beef sausages during refrigerated storage and assess the relationship between VOC levels and microbial population. Forty-one VOCs were detected using headspace solid-phase microextraction coupled with gas chromatography-olfactometry-mass spectrometry (GC-O-MS). The results of the 16S rDNA amplicon sequencing indicated that the spoilage bacteria predominated were Brochothrix and Serratia. Acetoin, hexanal and 1-octen-3-ol were significantly positively related to Brochothrix (Mantel's r = 0.95, p = 0.017) an

Microbiota encompassing putative spoilage bacteria in retail packaged broiler meat and commercial broiler abattoir.

It is well established, that certain bacteria within the Brochothrix, Carnobacterium, Lactobacillus, Lactococcus, and Leuconostoc genera have an important role in the spoilage of chill stored poultry meat packaged in modified atmosphere. However, little is known about the role of microorganisms that are difficult to culture and the microbiota during poultry spoilage. We combined traditional cultivation and culture-independent 16S rRNA amplicon sequencing to investigate the microbiota encompassing putative bacteria of whole broiler meat, packaged in modified atmosphere, during and exceeding she

Psychrotolerant spoilage bacteria enhanced Campylobacter jejuni culturability on refrigerated chicken meat.

Campylobacter jejuni is a leading cause of bacterial foodborne illness, with poultry as a major transmission vehicle. The bacterium can enter a viable but non-culturable (VBNC) state, which complicates detection using culture-based methods. While cold and oxidative stress are known VBNC inducers, the role of poultry-associated microbiota in influencing C. jejuni culturability and viability during refrigeration is poorly understood. This study characterized the effect of retail chicken microbiota on C. jejuni over 10 days of cold storage in aerobic and vacuum conditions. We profiled microbiota

Synthesis and antibacterial properties of Houttuynia cordata-derived carbon dots regulated by carbonization temperature for prolonging shelf life of chilled beef.

Houttuynia cordata carbon dots (HCO-CDs) were synthesized via the hydrothermal method and incorporated into polyvinyl alcohol/pectin films (PVA/Pec-CD), followed by exploring their applications to extend the shelf life of chilled beef. HCO-CDs transformed from flake-like structures to quasi-spherical nanoparticles at 160 °C, with aggregation occurring at 240 °C. HCO-CDs-160 displayed the strongest antibacterial activity against Pseudomonas aeruginosa and Brochothrix thermosphacta, due to their smaller particle size, higher membrane permeability, AKP leakage induction, ATP depletion, and ROS ge

Meat spoilage by bacteria: Influencing factors, volatile compounds, and organoleptic alterations.

This review provides an understanding of spoilage factors of meat, the spoilage bacteria, the spoilage mode of major spoilage bacteria, and alterations due to spoilage. pH and water activity of meat, packaging atmosphere, and storage temperature affect the growth and activity of spoilage bacteria. The spoilage bacteria cause the production of various compounds. Volatile compounds such as volatile basic nitrogen [ammonia (NH3) and amines (RNH2)], alcohols (ROH), aldehydes (RCHO), ketones (RCOR'), and sulfur (S) compounds are generated by decomposition from glycogen, proteins and lipids in meat

Microbial succession and interaction in vacuum-packed beef: a longitudinal study of bacterial and fungal dynamics.

The microbial dynamics of vacuum-packed (VP) beef are shaped by interactions between bacterial and fungal communities, influencing spoilage and meat quality during storage. While bacterial succession is well studied, fungal roles remain underexplored. We examined microbial communities in VP beef over 85 days using spike-in, qPCR, 16S/18S rRNA gene amplicon sequencing, culture-based methods, whole genome sequencing, and co-culture experiments. Initially dominated by Pseudomonas and Brochothrix, the bacterial community shifted toward lactic acid bacteria (LAB) by day 15. Fungal communities remai

Unraveling the improvement of (-)-epigallocatechin-3-O-gallate marination against salt reduction-induced quality deterioration of dry-cured grass carp: Mechanistic links among antioxidation, bacterial community rebalance, and flavor evolution control.

The improving effects and underlying mechanism of EGCG marination against salt reduction-induced quality deterioration of dry-cured grass carp (DCGC) were unraveled. Results showed that ≥500 mg/L EGCG marination contributed to water activity decline, inhibiting spoilage bacterial growth (Streptococcus parauberis, Carnobacterium maltaromaticum, Brochothrix, unclassified Enterobacteriaceae, Vibrio, etc.) and increasing flavor-contributing bacterial richness (mainly some lactic acid bacteria, Staphylococcus, and Psychrobacter species). The bacterial community structural rebalance and antioxidatio

Suitability of lactic acid bacteria and deriving antibacterial preparations to enhance shelf-life and consumer safety of emulsion type sausages.

Ready-to-eat (RTE) sliced emulsion type sausages are sensitive to recontamination with Listeria (L.) monocytogenes during processing and packaging steps. Since Listeria spp. are able to grow on those products under cold storage conditions, taking steps to reduce the recontamination risk and implementing antibacterial hurdles contribute to consumer safety and increase the product quality. With this study data about the suitability of culture broth, cell-free supernatant (CFS) or concentrated bacteriocin preparations (CFSconc) of bacteriocin-producing lactic acid bacteria (LAB) obtained from fer
