Brompheniramine, terfenadine, and placebo in allergic rhinitis.

BACKGROUND: Second-generation antihistamines, reported to lack central nervous system depressant activity, may be considered to have a clinical advantage over traditional antihistamines. OBJECTIVE: To compare the effectiveness, at recommended doses, of an extended-release formulation of nonprescription brompheniramine and prescription terfenadine in the treatment of allergic rhinitis. METHODS: This was a double-blind, randomized, placebo-controlled, multicenter, parallel study. Subjects with symptoms of allergic rhinitis received brompheniramine 12 mg (n = 96), terfenadine 60 mg (n = 96), or

Efficacy of brompheniramine maleate for the treatment of rhinovirus colds.

We tested the efficacy of brompheniramine maleate in a large randomized, controlled trial of volunteers with experimental rhinovirus colds. Brompheniramine (12 mg) or placebo was administered at 8:00 A.M. and 8:00 P.M. for < or = 4 days after the onset of symptoms (24, 36, or 48 hours after virus challenge). During the first 3 days of treatment (the first 4 days after virus challenge), nasal secretion weights were lower for infected evaluable subjects receiving brompheniramine (n = 113) than for controls (day 1: 4.3 g vs. 6.8 g; day 2: 4.8 g vs. 7.7 g; and day 3: 3.3 g vs. 5.3 g) (P < or = .03

Pregnancy after brompheniramine treatment of a diabetic with incomplete emission failure.

Ejaculation disorders in diabetic men are most frequently caused by retrograde ejaculation resulting from a damage of the sympathetic nerves at the level of the bladder neck. This results in incomplete or complete emission failure because of insufficient sperm transport. Diabetic ejaculatory sterility can be treated with anticholinergic drugs. The reported case shows that as a result of its antihistaminic and anticholinergic properties, brompheniramine can be successfully used in the treatment of men with diabetic ejaculatory sterility, particularly in patients with incomplete emission failure

Longterm treatment with brompheniramine and phenylpropanolamine in recurrent otitis media--a double-blind study.

A double-blind study has been performed in 44 children with recurrent episodes of otitis media and/or secretory otitis media using a combined preparation of brompheniraminemaleate and phenylpropanolaminehydrocloride (Lunerin mite). No significant prophylactic effect was found on symptoms of respiratory tract infections or middle ear diseases during the trial period of nine months. The result suggests that further critical studies should be performed with such preparations which for the time being are used abundantly for the prevention of middle ear infections or effusions.

Alkylamine antihistamine toxicity and review of Pediatric Toxicology Registry of the National Association of Medical Examiners. Report 4: Alkylamines.

Antihistamines are popular nonprescription medications for the treatment of allergy and cold symptoms. Accidental exposures to these preparations are common with > 14,000 occurring annually in children under the age of 6 years (1). Despite this, there is limited information about toxic and lethal concentrations of these drugs in children. We present a case of a pediatric fatality due to a common brompheniramine and phenylpropanolamine preparation and review available Pediatric Toxicology Registry data on alkylamine antihistamines. The data collected by the Pediatric Toxicology Registry on the

Clinical pharmacokinetics of H1-receptor antagonists (the antihistamines).

This article reviews clinical pharmacokinetic data on the H1-receptor antagonists, commonly referred to as the antihistamines. Despite their widespread use over an extended period, relatively little pharmacokinetic data are available for many of these drugs. A number of H1-receptor antagonists have been assayed mainly using radioimmunoassay methods. These have also generally measured metabolites to greater or lesser extents. Thus, the interpretation of such data is complex. After oral administration of H1-receptor antagonists as syrup or tablet formulations, peak plasma concentrations are usua

Negative middle ear pressure and hearing thresholds in secretory otitis media. A double-blind crossover study with Lunerin.

No significant effect of an oral decongestant (Lunerin: brompheniramine maleate and phenylpropanolamine hydrochloride) can be shown in this study. Negative middle ear pressure, effusions in the middle ear and hearing threshold levels were evaluated in 26 cleft palate children over a 3 month period, applying a double-blind crossover method. Variations in middle ear pressure occurred in all patients during the trial. The shape of the tympanograms gave no information additional to the clinical evaluation.

S-carboxymethylcysteine in otitis media with effusion. (A double-blind study).

S-carboxymethylcysteine syrup (SCMC); brompheniramine, phenylephrine with phenylpropanolamine elixir (BPP); and a placebo were compared in a double-blind trial in 58 children with otitis media with effusion (OME). All patients underwent myringotomies after an initial period of medical treatment. SCMC, but not BPP, was significantly better than the placebo in speeding up resolution of the effusion after surgery, as judged by pure tone audiometry, showing this drug to be a useful adjunct to the surgical management of OME. It should also be considered as an alternative to surgery.

Quantitative determinations of two decongestants and an antihistamine in combination using paired ion high-pressure liquid chromatography.

A single method for the quantitative determinations of three active ingredients, phenylephrine hydrochloride, phenylpropanolamine hydrochloride, and brompheniramine maleate, and one inactive ingredient (sodium benzoate) in a commercial product for colds is reported. The method is based on paired ion high-pressure liquid chromatography using 1-heptanesulfonic acid as the counterion. It is accurate and precise. The relative standard deviations based on six readings are reported. This method is sensitive; less than 1 microgram of each ingredient can be assayed. The peak area of each ingredient is

A comparative evaluation of oral decongestants in the treatment of vasomotor rhinitis.

In a seven-week double-blind crossover study, Disofrol (dexbrompheniramine maleate +d-isoephedrine sulphate) was compared with Lunerin (brompheniramine + 2-amino-1-phenyl-propanol-(1)-hydrochloride) and with placebo in the treatment of vasomotor rhinitis in thirty-one patients. From the patients' evaluations on diary cards Disofrol reduced the symptom running nose to a higher degree than Lunerin, an effect possibly ascribable to a greater anticholinergic activity. Both preparations had equal ability to relieve nasal blockage and both combinations were found to be more effective than placebo.
