Mechanism of Brucea javanica against multiple myeloma via TNF-NF-κB signaling.

This study aims to explore the pharmacological mechanism of Brucea javanica (BJ) in treating multiple myeloma (MM) through network pharmacology and validate this mechanism via in vitro experiments. Active pharmaceutical ingredients (APIs) of BJ and their potential targets were identified, along with MM-related targets. By plotting protein-protein interaction (PPI) networks, hub genes responsible for BJ in treating MM were identified and subjected to molecular docking. In MM cell lines H929 and U266, regulatory effects of BJ and luteolin (the major compound of BJ) on the behaviors of MM cells w

Intraperitoneal administration of doxorubicin-encapsulated Brucea javanica oil nanoemulsion against malignant ascites.

Malignant ascites is a common complication of advanced cancers, which reduces survival rates and diminishes patients' quality of life. Intraperitoneal chemotherapy is a conventional method for treating cancer-related ascites, but the poor drug retention of conventional drugs requires frequent administration to maintain sustained anti-tumor effects. In this study, we encapsulated doxorubicin (DOX) into Brucea javanica oil (BJO) to develop a water-in-oil (W/O) nanoemulsion called BJO@DOX for the treatment of malignant ascites through in-situ intraperitoneal administration. BJO significantly indu

In vitro and in vivo antiplasmodial activity of the root extracts of Brucea mollis Wall. ex Kurz.

Malaria control is compromised worldwide by continuously evolving drug-resistant strains of the parasite demanding exploration of natural resources for developing newer antimalarials. The northeastern region of India is endemic for malaria characterized by high prevalence of drug-resistant Plasmodium falciparum strains. Many plants are used by the indigenous communities living in the northeast India in their traditional system of medicine for the treatment of malarial fever. Folklore claim of antimalarial property of one such plant Brucea mollis was evaluated in vitro and in vivo for antiplasm

Therapeutic Effect of Brucea Javanica Oil Emulsion in Mice with Irinotecan-Induced Delayed Diarrhea.

BACKGROUND: Chemotherapy-induced diarrhea (CID), particularly delayed diarrhea, often limits clinical use. Brucea javanica oil emulsion (BJOE), an adjuvant chemotherapy agent, has been shown to reduce irinotecan-related gastrointestinal side effects. However, its underlying molecular mechanism remains unclear. The cGAS-STING pathway, composed of the cytosolic DNA sensor cyclic GMP-AMP synthase (cGAS) and the adaptor protein stimulator of interferon genes (STING), plays an essential role in delayed diarrhea. This work aimed to investigate the therapeutic potential and underlying mechanism of BJ

Bruceine E, a natural quassinoid from Brucea javanica, inhibits PARthanatos via targeting PARP1 in ischemic stroke.

Ischemic stroke currently lacks evidence-based neuroprotective agents, primarily due to the challenge of timely intervention, which often occurs after the onset of irreversible neuronal damage. To address this, this study investigates the PARthanatos pathway, a form of regulated cell death triggered by DNA damage. Utilizing MNNG-induced cellular PARthanatos models, we screened a library of 2,939 traditional Chinese medicine monomers and identified Bruceine E, a natural product derived from Brucea javanica (bitterwood), as a potent inhibitor of PARthanatos at nanomolar concentrations, acting vi

Brucea javanica oil inhibits tongue squamous cell invasion and metastasis by regulating miR-138-EZH2 pathway.

Tongue squamous cell carcinoma (TSCC) is one of the most common malignant tumors of head and neck. Its incidence is on the rise, and the proportion of young patients is gradually increasing, which is prone to tumor recurrence and metastasis. At present, there is no effective method to completely treat TSCC. Studies have shown that brucea javanica oil (BJO) has good antitumor activity against lung cancer and gastrointestinal tumors, but its therapeutic effect on TSCC is not clear. We have previously confirmed that oleic acid, the main component of BJO, can induce apoptosis of TSCC and reduce it
