Aphidius ervi venom regulates Buchnera contribution to host nutritional suitability.

The association between the pea aphid, Acyrthosiphon pisum (Harris) (Homoptera: Aphididae), and the endophagous parasitoid wasp Aphidius ervi Haliday (Hymenoptera: Braconidae) offers a unique model system for studying the molecular mechanisms underlying the complex interactions between the parasitoid, its host and the associated primary symbiont. Here, we investigate in vivo the functional role of the most abundant component of A. ervi venom, Ae-γ-glutamyl transpeptidase (Ae-γ-GT), which is known to induce host castration. Microinjections of double-stranded RNA into A. ervi pupae stably knocke

Lipid metabolism in the aphid-Buchnera symbiosis: an underexplored dimension of insect physiology.

Lipids are essential biomolecules that function as energy reserves, signaling mediators, and structural components of cellular membranes, yet their roles in insect-microbe symbiosis remain poorly understood. In contrast to the well-established contributions of microbial symbionts to amino acid and vitamin metabolism, symbiotic lipid metabolism has received comparatively little attention. Here, we review current knowledge of lipid metabolism in insect symbiosis, focusing on the aphid-Buchnera association as a model of obligate mutualism. Physiological evidence indicates that aphids rely primari

Plasmid-encoded anthranilate synthase (TrpEG) in Buchnera aphidicola from aphids of the family pemphigidae.

Buchnera aphidicola is an obligate intracellular symbiont of aphids. One of its proposed functions is the synthesis of essential amino acids, nutrients required by aphids but deficient in their diet of plant phloem sap. The genetic organization of the tryptophan pathway in Buchnera from proliferous aphids of the family Aphididae has previously been shown to reflect a capacity to overproduce this essential amino acid (C.-Y. Lai, L. Baumann, and P. Baumann, Proc. Natl. Acad. Sci. USA 91:3819-3823, 1994). This involved amplification of the genes for the first enzyme in the pathway, anthranilate s

Evolutionary relationships of Pemphigus and allied genera (Hemiptera: Aphididae: Eriosomatinae) and their primary endosymbiont, Buchnera aphidicola.

Aphids harbor primary endosymbionts, Buchnera aphidicola, in specialized cells within their body cavities. Aphids and Buchnera have strict mutualistic relationships in nutrition exchange. This ancient association has received much attention from researchers who are interested in endosymbiotic evolution. Previous studies have found parallel phylogenetic relationships between non-galling aphids and Buchnera at lower taxonomic levels (genus, species). To understand whether relatively isolated habitats such as galls have effect on the parallel relationships between aphids and Buchnera, the present

Molecular characterization of the leucine cluster in Buchnera sp. strain PSY, a primary endosymbiont of the aphid Pemphigus spyrothecae.

Buchnera strains from most aphid subfamilies studied to date have been found to carry the leucine gene cluster (leuA, -B, -C, and -D) on a plasmid, an organization unique among bacteria. Here, however, we demonstrate a classical chromosomal location of the cluster in Buchnera sp. strain PSY from the aphid Pemphigus spyrothecae (subfamily Pemphiginae). The genes that flank leuABCD in Buchnera sp. strain PSY appear to be adjacent in the genome of Buchnera sp. strain APS, a strain carrying a leucine plasmid. We propose that the presence of a leucine plasmid predates the diversification of symbiot

Integrating Host Phylogeny and Bacterial Detection Patterns Reveals Contrasting Associations of Buchnera aphidicola and Other Aphid-Associated Bacteria.

Aphids are important agricultural pests that interact with diverse bacterial taxa living within or associated with their bodies. Some bacteria, such as the obligate symbiont Buchnera aphidicola, provide essential nutritional functions and are predominantly inherited vertically, whereas other aphid-associated bacteria may have more variable distributions and ecological roles. In this study, we investigated patterns of association between aphid evolutionary relationships and selected bacterial taxa. Aphids were identified using morphological characteristics and mitochondrial cytochrome c oxidase

A genomic perspective on nutrient provisioning by bacterial symbionts of insects.

Many animals show intimate interactions with bacterial symbionts that provision hosts with limiting nutrients. The best studied such association is that between aphids and Buchnera aphidicola, which produces essential amino acids that are rare in the phloem sap diet. Genomic studies of Buchnera have provided a new means for inferring metabolic capabilities of the symbionts and their likely contributions to hosts. Despite evolutionary reduction of genome size, involving loss of most ancestral genes, Buchnera retains capabilities for biosynthesis of all essential amino acids. In contrast, most g

Genome sequence of the pea aphid Acyrthosiphon pisum.

Aphids are important agricultural pests and also biological models for studies of insect-plant interactions, symbiosis, virus vectoring, and the developmental causes of extreme phenotypic plasticity. Here we present the 464 Mb draft genome assembly of the pea aphid Acyrthosiphon pisum. This first published whole genome sequence of a basal hemimetabolous insect provides an outgroup to the multiple published genomes of holometabolous insects. Pea aphids are host-plant specialists, they can reproduce both sexually and asexually, and they have coevolved with an obligate bacterial symbiont. Here we

Metabolic relay gene of aphid and primary symbiont as RNAi target loci for aphid control.

INTRODUCTION: Aphids form a stable and mutually beneficial relationship with their primary symbiont Buchnera aphidicola, which play an important role in providing the missing nutrients to the host aphid. Based on the genome sequence of wheat aphid Siotobion miscanthi and its primary symbiont Buchnera that we obtained in our previously study, we identified a metabolic relay gene, ilvA, involved in the isoleucine synthesis pathway between aphids and Buchnera. METHOD: In this study, we identified the location and sequence structure of ilvA gene in aphid genome, the expression level in different

Symbiont dominance and microbiome dysbiosis in wheat-aphid revealed through 16 S rRNA gene amplicon sequencing analysis.

Insect herbivory represents a major biotic stress that leads to significant losses in crop yield and quality worldwide. Aphids, in particular, damage wheat (Triticum aestivum L.), a staple crop that provides nearly 20% of the global protein and caloric intake, and also harbor diverse bacterial symbionts that can influence plant health. In this study, 16 S rRNA gene amplicon sequencing was used to characterize the bacterial communities associated with healthy and aphid-infested wheat plants. Alpha-diversity indices, including Simpson, Shannon, Chao1, and ACE, revealed statistically significant

Yellow protein co-opted to sustain obligate symbiosis in leaf beetles.

Yellow proteins are best known for their roles in pigmentation, behavior, and development across insects. Here, we uncover their striking evolutionary co-option for a wholly distinct function: sustaining a Paleocene-aged digestive symbiosis in tortoise beetles. We show that a female-specific Yellow forms the gelatinous spheres that encapsulate the bacterium Stammera during vertical transmission, allowing it to subsist extracellularly despite its drastically reduced genome (0.24 Mb) and limited metabolic capacity. Yellow expression is highly localized to symbiont-harboring glands in the ovaries

Acetone preservation: a practical technique for molecular analysis.

In attempts to establish a convenient and reliable method for field collection and archival preservation of insects and their endosymbiotic microorganisms for molecular analysis, acetone, ethanol, and other organic solvents were tested for DNA preservability of the pea aphid Acyrthosiphon pisum and its intracellular symbiotic bacterium Buchnera sp. After 6 months' storage, not only the band of high-molecular-size DNA but also the bands of rRNA were well preserved in acetone, ethanol, 2-propanol, diethyl ether and ethyl acetate. Polymerase chain reaction (PCR) assays confirmed that the DNA of b

Multiple origins of endosymbiosis within the Enterobacteriaceae (γ-Proteobacteria): convergence of complex phylogenetic approaches.

BACKGROUND: The bacterial family Enterobacteriaceae gave rise to a variety of symbiotic forms, from the loosely associated commensals, often designated as secondary (S) symbionts, to obligate mutualists, called primary (P) symbionts. Determination of the evolutionary processes behind this phenomenon has long been hampered by the unreliability of phylogenetic reconstructions within this group of bacteria. The main reasons have been the absence of sufficient data, the highly derived nature of the symbiont genomes and lack of appropriate phylogenetic methods. Due to the extremely aberrant nature

Antibiotics, primary symbionts and wing polyphenism in three aphid species.

The possible role of the primary Buchnera symbionts in wing polyphenism is examined in three aphid species. Presumptive winged aphids were fed on antibiotic-treated beans to destroy these symbionts. As previously reported, this leads to inhibited growth and low/zero fecundity. When such treatment is applied to the short-day-induced gynoparae (the winged autumn migrant) of the black bean aphid, Aphis fabae, it also causes many insects to develop as wingless or winged/wingless intermediate adult forms (apterisation). However, whilst antibiotic treatment of crowd-induced, long-day winged forms of

A Member of an Ancient Family of Bacterial Amino Acids Transporters Contributes to Chlamydia Nutritional Virulence and Immune Evasion.

Many obligate intracellular bacteria, including members of the genus Chlamydia, cannot synthesize a variety of amino acids de novo and acquire these from host cells via largely unknown mechanisms. Previously, we determined that a missense mutation in ctl0225, a conserved Chlamydia open reading frame of unknown function, mediated sensitivity to interferon gamma. Here, we show evidence that CTL0225 is a member of the SnatA family of neutral amino acid transporters that contributes to the import of several amino acids into Chlamydia cells. Further, we show that CTL0225 orthologs from two other di

Peptidoglycan in obligate intracellular bacteria.

Peptidoglycan is the predominant stress-bearing structure in the cell envelope of most bacteria, and also a potent stimulator of the eukaryotic immune system. Obligate intracellular bacteria replicate exclusively within the interior of living cells, an osmotically protected niche. Under these conditions peptidoglycan is not necessarily needed to maintain the integrity of the bacterial cell. Moreover, the presence of peptidoglycan puts bacteria at risk of detection and destruction by host peptidoglycan recognition factors and downstream effectors. This has resulted in a selective pressure and o

Implications of environmentally shaped microbial communities for insecticide resistance in Sitobion miscanthi.

Insect-associated bacteria play an important role in the resistance to pesticides, yet bacterial community compositions in wild insect host populations and the environmental factors that shape them are mostly elusive. In this study, Sitobion miscanthi (Takahashi) populations were collected from major wheat growing regions in China. Following high-throughput sequencing of 16S rRNA gene fragments, association analyses were performed within the bacterial community associated with S. miscanthi, as well as with population resistance levels to four commonly used pesticides and different environmenta

Evolutionary relationships of "Candidatus Riesia spp.," endosymbiotic enterobacteriaceae living within hematophagous primate lice.

The primary endosymbiotic bacteria from three species of parasitic primate lice were characterized molecularly. We have confirmed the characterization of the primary endosymbiont (P-endosymbiont) of the human head/body louse Pediculus humanus and provide new characterizations of the P-endosymbionts from Pediculus schaeffi from chimpanzees and Pthirus pubis, the pubic louse of humans. The endosymbionts show an average percent sequence divergence of 11 to 15% from the most closely related known bacterium "Candidatus Arsenophonus insecticola." Parasitoids as vectors of facultative bacterial endosymbionts in aphids.

Heritable bacterial endosymbionts play an important role in aphid ecology. Sequence-based evidence suggests that facultative symbionts such as Hamiltonella defensa or Regiella insecticola also undergo horizontal transmission. Other than through male-to-female transfer during the sexual generation in autumn, the routes by which this occurs remain largely unknown. Here, we tested if parasitoids or ectoparasitic mites can act as vectors for horizontal transfer of facultative symbionts. Using symbiont-specific primers for diagnostic PCR, we demonstrate for the first time, to our knowledge, that pa

Plasmids derived from Gifsy-1/Gifsy-2, lambdoid prophages contributing to the virulence of Salmonella enterica serovar Typhimurium: implications for the evolution of replication initiation proteins of lambdoid phages and enterobacteria.

Gifsy-1 and Gifsy-2 are lambdoid prophages which contribute to the virulence of Salmonella enterica serovar Typhimurium. The nucleotide sequence of the replication region of both prophages is identical, and similar in organization to the replication region of bacteriophage lambda. To investigate the replication of the Gifsy phages and the relationship between Gifsy and host chromosome replication, a plasmid which contained all the genes and regulatory sequences required for autonomous replication in bacterial cells was constructed. This plasmid, pGifsy, was stably maintained in Escherichia col
