A single large dose of butylated hydroxytoluene (BHT, 1000 mg/kg) in male Fischer 344 rats produced some renal damage, reduced accumulation of p-aminohippuric acid in renal slices, proteinuria and enzymuria, in addition to hepatic damage. Further, prior administration of phenobarbital (80 mg/kg, i.p., daily for 4 days) in the high-dose BHT-treated male rats produced renal damage accompanied by slight tubular necrosis. The renal damage was confirmed by biochemical and histological changes. These changes were dose dependent, with a maximum at 24 h after BHT administration, but had returned to th

Two patients with chronic idiopathic urticaria in whom remissions were achieved with dye- and preservative-elimination diet had exacerbations of their urticaria when they were challenged under double-blind, placebo-controlled conditions with butylated hydroxyanisole and butylated hydroxytoluene. After elimination of butylated hydroxyanisole and butylated hydroxytoluene from their diets, there was marked abatement of the frequency, severity, and duration of their urticaria. These antioxidants appear capable of aggravating symptoms in certain patients with chronic urticaria.

Butylated hydroxytoluene (BHT), one of the most widely used synthetic phenolic antioxidants, is a popular food additive. Previous studies have reported the possible health hazards of BHT. However, BHT effects on female reproduction, especially on endometrial decidualization, are still unknown. During early pregnancy, decidualization plays important roles for embryo implantation and pregnancy establishment. This study aimed to explore the effects of BHT on endometrial decidualization in pregnant mice. The pregnant mice received BHT via intraperitoneal injection at doses of 0, 200, and 400 mg/kg

Senile cataractogenesis advanced by diabetes, thought mainly to be due to accumulation of polyols, is one of the major causes of blindness throughout the world. Rats fed a 50% galactose diet developed mature cataracts in 23 days, whereas rats fed a 50% galactose diet with 0.4% butylated hydroxytoluene (BHT) had clear lenses. On day 10, both groups had comparable galactitol levels in the lens, but hydration in the former group, as determined by the lens weight, was approximately 10% more than in the latter. On day 23, galactitol levels in the latter group were 92% higher (approximately 85 mM) t

The present study was to determine the effect of butylated hydroxytoluene (BHT) on quality and fertilizing ability of frozen-thawed boar semen. In the first experiment, five crossbreds of Polish Landrace and Large White boars (five ejaculates per boar) were frozen in 0.5 mL straws after dilution with lactose-egg yolk-glycerol extender supplemented with 0 (control), 0.5, 1.0, and 2.0 mM BHT. The sperm quality was verified based on the motility (computer-assisted sperm analysis; total motility, %; progressive motility, %), membrane integrity (YO-PRO-1/propidium iodide [PI] assay), acrosome integ

Butylated hydroxytoluene (BHT) is a synthetic phenolic antioxidant utilized as a preservative in many products from foods to cosmetics. Population-based studies have detected BHT in the vast majority (>90%) of human specimens (serum, urine, fingernails) as well as home dust samples. Thus, BHT constitutes a ubiquitous environmental contaminant without obvious adverse health effects to humans. In contrast, exposure of mice to a single intraperitoneal dose of BHT triggers distal epithelial damage including loss of gas-exchanging alveolar type 1 epithelial cells, thus providing an invaluable tool

Chemoprevention of toxicoses and/or cancer through the use of nutrients or pharmacologic compounds is the subject of intense study. Among the many compounds examined, food additives such as antioxidants are being considered due to their ability to reduce disease formation by either induction or inhibition of key enzyme systems. One such compound, butylated hydroxytoluene (BHT), has been found to protect against cancer formation caused by exposure to aflatoxin B1 (AFB1) in rodents. We have shown that dietary BHT protects against clinical signs of aflatoxicosis in turkeys, a species that is very

The effect of butylated hydroxytoluene (BHT), which is used widely as an antioxidant, on IgE-dependent allergic responses in vivo and in vitro was investigated. For in vivo study, passive cutaneous anaphylaxis (PCA) was elicited in rats by i.d. injection of anti-DNP IgE and 48 h later by i.v. injection of DNP-HSA. BHT was i.p. given immediately after anti-DNP IgE injection. For in vitro studies, the rat mast cell line RBL2H3 sensitized with monoclonal anti-dinitrophenol (DNP) IgE was challenged with the multivalent antigen DNP-human serum albumin (DNP-HSA) in the presence or absence of BHT. be

We previously reported on the induction by vitamin A of gallstones, rich in calcium and phosphate, in hamsters. On the other hand, it has been reported that the phenolic antioxidant butylated hydroxytoluene (BHT) potentiates the hepatotoxicity of vitamin A. In the present work we have tested the effect of BHT on the lithogenicity of vitamin A and on bile composition. The urinary excretion of calcium and phosphate was determined to assess a possible asymptomatic bone resorption due to vitamin A toxicity, and/or an effect of BHT on the homeostasis of calcium and phosphate. Three groups of 18 mal

We have shown previously that the extreme sensitivity of turkeys to aflatoxin B(1) (AFB(1)) is due to a combination of efficient AFB(1) activation by cytochrome P450s (CYPs) 1A and deficient detoxification by glutathione S-transferases (GSTs). Phenolic antioxidants such as butylated hydroxytoluene (BHT) have been shown to be chemoprotective in some animal models due, in part, to modulation of AFB(1)-relevant phase I and/or phase II activities, and we wished to determine whether BHT has a similar effect in turkeys. Ten-day-old male turkeys were maintained on diets amended with 1000 or 4000 ppm

Butylated hydroxytoluene (2,6-di-tert-butyl-4-methylphenol, BHT) has been reported to be a lung toxicant. Mice treated with BHT (200-800 mg/kg, po) in combination with an inhibitor of glutathione (GSH) synthesis, buthionine sulfoximine (BOS; 1 hr before and 2 hr after BHT, 4 mmol/kg per dose, ip) developed hepatotoxicity characterized by an increase in serum glutamic pyruvic transaminase (GPT) activity and centrilobular necrosis of hepatocytes. The hepatotoxic response was both time- and dose-dependent. BHT (up to 800 mg/kg) alone produced no evidence of liver injury. As judged by the observat

This study was designed to characterize the biochemical, cellular, and morphologic events produced in mice by butylated hydroxytoluene (BHT) and to relate these events to changes in extracellular angiotensin-converting enzyme (ACE) activity. On Day 1 after the administration of BHT, bronchoalveolar lavage (BAL) ACE activity increased 4-fold (p less than 0.001), its specific activity relative to BAL protein increased 3-fold (p less than 0.001), and both type 1 cell damage and endothelial cell damage were detected by electron microscopy. The early increase in BAL ACE activity preceded changes in

The aim of the present study was to test the effects of butylated hydroxytoluene (BHT) on the cryopreservation of common carp spermatozoa. BHT is widely used in the cryopreservation of the spermatozoa of different animal species and successfully sustains the characteristics of spermatozoa during freezing and thawing, but it has not previously been used with fish. After sampling, common carp spermatozoa were diluted with an extender composed of modified Kurokura's extender, 10% DMSO, and 10% egg yolk containing 0.0001, 0.001, 0.01, 0.1, 1, 2.5, 5, or 10mM BHT and subsequently frozen in liquid n

Diethyldithiocarbamate (DTC) and carbon disulfide (CS2), at nearly equimolar doses (po), prevented mice from lung injury induced by butylated hydroxytoluene (BHT), as evidenced by suppression of increased lung weight and total DNA content as well as by histopathologic observations. CS2 pretreatment dose dependently decreased the amount of covalently bound [ring-14C]BHT to lung macromolecules in vivo. A slight, but significant, loss of lung GSH observed early after BHT administration was also prevented. The lung microsomal fraction exhibited NADPH-dependent covalent binding of BHT in vitro; thi

Butylated hydroxytoluene (BHT) is a synthetic phenolic antioxidant extensively used in food. Given the increasing biosafety concerns regarding BHT and its metabolites (BHT-OH and BHT-COOH), their interactions with pepsin were systematically investigated using multispectral techniques, molecular docking, and simulation methods. Fluorescence analysis indicated static quenching and stable complex formation, with binding affinities following the order BHT-OH > BHT-COOH > BHT. Hydrogen bonding and van der Waals forces dominated the interactions, and the carboxyl group of BHT-COOH contributed additi

Arsenic is a global environmental contaminant. Sodium arsenite (NaAsO2), one of the arsenic compounds, is toxic. Prolonged exposure to NaAsO2 causes various detrimental effects on body organs. Butylated hydroxytoluene (BHT), a synthetic antioxidant may retard the deleterious effects of free-radical-induced damage. We hereby investigated the chemo-protective role of BHT on sodium arsenite-induced hepatorenal toxicity in rats. Rats (n = 49, 110 ± 10 g) were grouped into seven with 7 animals each: groups 1 and 2 served as the negative controls and were administered corn oil and distilled water re

Butylated hydroxytoluene (BHT) is widely employed as an antioxidant in various industries. However, concerns persist regarding its safety and environmental impact, as its toxicological mechanisms remain poorly understood. Zebrafish embryos were exposed to BHT at 3, 5, and 7 mg/L to assess developmental toxicity. BHT exposure dose-dependently induced spinal cord malformations concomitant with reduced larval motility. Mechanistically, BHT suppressed Hedgehog signaling, evidenced by dysregulation of Shha, Gli1, and Smo. Notably, pharmacological activation of the Hh pathway by SAG rescued both str

BACKGROUND: Type 2 diabetes mellitus (T2DM) is a global health concern associated with systemic as well as oral complications. The preventive antioxidants found in saliva naturally reduce the damaging effects of reactive oxygen molecules. Any disruption to the regular functioning of these antioxidants may lead to oxidative stress, which could boost an individual's vulnerability to oral diseases. Diabetes patients are vulnerable to various dental complications, such as oral mucosal disorders, dental caries, dry mouth, and periodontal disease.
AIM: This study aimed to assess the salivary butyla
