The recently conceptualized Cardiovascular-Kidney-Metabolic (CKM) syndrome represents a pressing global health burden, characterized by a vicious cycle of dysfunction among the cardiac, renal, and metabolic systems. Growing evidence suggests that gut microbiota dysbiosis, specifically, a loss of butyrate-producing bacteria (BPB) and the resulting systemic butyrate deficiency, may be an important but previously overlooked driver of CKM progression. In this review, we synthesize available evidence linking butyrate to the integrated, multi-organ pathophysiology of CKM and propose a conceptual fra

BACKGROUND: Dietary imbalance, such as a lower proportion of complex carbohydrates and a higher protein diet, may contribute to gestational diabetes mellitus (GDM) risks through their metabolisms. However, there is a lack of knowledge regarding the association between butyrate, iso-butyrate, and GDM, which are metabolisms of the two primary nutrients above. This study aimed to clarify the association of butyrate and iso-butyrate with GDM.
METHODS: A nested case-control study was conducted based on the Beijing Birth Cohort Study (BBCS) from 2017 to 2018. Totally, 99 singleton women were involv

Supplementation with Bifidobacterium bifidum G9-1 (BBG9-1) has been established to enhance the production of butyrate, a short-chain fatty acid (SCFA) known for its beneficial effects in alleviating constipation. We hypothesized that BBG9-1 alters gut microbiota such that bacteria that produce butyric acid from lactate and acetate become more abundant. In this study, we sought to determine whether BBG9-1 promotes the growth of butyrate-producing bacteria and thereby enhances butyrate production. BBG9-1 was cocultured with different butyrate-producing bacteria to compare differences in the SCFA

Spinal muscular atrophy (SMA) is an early-onset motor neuron disease that leads to loss of muscle function. Butyrate (BA)-based compounds markedly improve the survival and motor phenotype of SMA mice. In this study, we examine the protective effects of the BA prodrug pivaloyloxymethyl butyrate (AN9) on the survival of SMNΔ7 SMA mice. Oral administration of AN9 beginning at PND04 almost doubled the average lifespan of SMNΔ7 SMA mice. AN9 treatment also increased the growth rate of SMNΔ7 SMA mice when compared to vehicle-treated SMNΔ7 SMA mice. In conclusion, BA prodrugs like AN9 have ameliorati

BACKGROUND: Short-chain fatty acids (SCFAs), fermentation products of undigested fibers, are considered beneficial for colonic health. High plasma concentrations are potentially harmful; therefore, information about systemic SCFA clearance is needed before therapeutic use of prebiotics or colonic SCFA administration.
OBJECTIVE: The aim of this study was to investigate the effect of rectal butyrate administration on SCFA interorgan exchange.

Gamma Hydroxybutirate (GHB) is unfamiliar to many physicians in Israel. Recently GHB has emerged as an illicit drug of misuse for its purported euphoric and aphrodisiac effect. The clinical effects can progress rapidly to respiratory arrest and death. We provide a description of a case of GHB poisoning, with a comprehensive review of the literature emphasizing the pharmacodynamics, clinical effect, and suggestions for management. Physicians are urged to become familiar with GHB.

Butyrate has a dramatic effect on transformed cells in culture. This effect disappears as soon as butyrate is removed from the medium. The other short chain fatty acids are much less effective. Butyrate produces an arrest of cell proliferation at the early G1 phase of the cell cycle. The effect is very general and may be used for cell growth synchronization. This compound increases the expression of the c-fos oncogene and inhibits the expression of c-myc in all phases of the cell cycle. Butyrate modulates the expression of several genes. In general it induces the expression of markers of cell

In addition to being a part of the metabolic fatty acid fuel cycle, butyrate is also capable of inducing growth arrest in a variety of normal cell types and senescence-like phenotypes in gynecological cancer cells, inhibiting DNA synthesis and cell growth in colonic tumor cell lines, suppressing hTERT mRNA expression and telomerase activity in human prostate cancer cells, and inducing stem cell differentiation and apoptosis by DNA fragmentation. It regulates gene expression by inhibiting histone deacetylases (HDACs), enhances memory recovery and formation in mice, stimulates neurogenesis in th

Two cows each received intraruminal applications of 1.0 g/kg B.W. of butyric acid or sodium butyrate, for three weeks running, before daily applications were increased up to 2.0 kg/B.W., within one week. Resulting alterations primarily included increase in acidosis or alkalosis, whereas the typical symptoms of ketosis were lacking. Signs of impaired liver function were to some extent recordable in response to sodium butyrate.

Fibrosis is a progressive pathological condition characterized by excessive extracellular matrix deposition (ECM) deposition, which disrupts organ structure and function and can ultimately lead to organ failure. It can affect nearly all organs, and the associated morbidity and mortality increase with age. Nevertheless, few effective treatments are currently available for fibrosis. Targeting gut microbiota-derived metabolites has recently emerged as an active area of research. These metabolites participate in disease pathophysiology and profoundly affect host health. Butyrate, a major gut micro

Butyrate, a short-chain fatty acid (SCFA) produced by gut microbiota, plays crucial roles in maintaining intestinal homeostasis and modulating the gut-brain axis. Dysbiosis and SCFA imbalances are increasingly recognized as contributors to disease pathogenesis. A decrease in butyrate-producing bacteria leads to reduced butyrate levels, which have been linked to increased intestinal permeability, systemic inflammation, and neuroinflammation. Emerging evidence highlights a potential therapeutic role for butyrate in Parkinson's Disease (PD). This review examines butyrate's origins, functions, and

PURPOSE: Butyrate is a short-chain fatty acid produced in the intestine. It is controversial whether butyrate is protective or destructive for the intestinal epithelium in the development of diseases like necrotizing enterocolitis (NEC), and its mechanism of action remains unclear. We aimed to determine the effect of butyrate on the intestinal epithelium by studying its effects on intestinal epithelial cells (IEC-18) exposed to injury and in vivo by investigating the effects on the intestine in an experimental model of NEC.
METHODS: A) In vitro study: Butyrate was given to normal IEC-18 to de

A key event featured in the early stage of chronic gut inflammatory diseases is the disordered recruitment and excess accumulation of immune cells in the gut lamina propria. This process is followed by the over-secretion of pro-inflammatory factors and the prolonged overactive inflammatory responses. Growing evidence has suggested that gut inflammatory diseases may be mitigated by butyric acid (BA) or butyrate sodium (NaB). Laboratory studies show that BA and NaB can enhance gut innate immune function through G-protein-mediated signaling pathways while mitigating the overactive inflammatory re

This review investigates the therapeutic potential of butyrate, a short-chain fatty acid (SCFA) produced by gut microbiota, in the prevention and treatment of various gynecological diseases, including polycystic ovary syndrome (PCOS), endometriosis, and gynecologic cancers like cervical and ovarian cancer. These conditions often pose treatment challenges, with conventional therapies offering limited and temporary relief, significant side effects, and a risk of recurrence. Emerging evidence highlights butyrate's unique biological activities, particularly its role as a histone deacetylase (HDAC)

BACKGROUND: It is well established that the colonic mucosa is highly dependent upon the luminal presentation of nutrients. This dependence is most marked in the distal colon. The major luminal nutrients are short chain fatty acids (SCFA) that are produced as a by-product of colonic fermentation of carbohydrates. Butyrate appears to be the SCFA that is most avidly metabolized by the colonic mucosa. It has been suggested that ulcerative colitis (UC) is, at least in part, related to an energy deficiency state of the colonic mucosa which may be secondary to impaired SCFA production, uptake, or uti

It is widely known that the short chain fatty acid (SFA) butyrate, generated mainly through fermentation of dietary fibre by anaerobic bacteria, has beneficial effects on health. Less widely known is that butyrate, at the concentration (c20 mM) typically found in the healthy colonic lumen, is toxic to colon epithelial cells. It is probably the presence of the continuous adherent mucus layer in the healthy colon that ensures that butyrate concentrations in contact with the epithelium fall below the levels (c > 3 mM) that are consistently toxic in in vitro or ex vivo studies. In active ulcerativ

Maintaining an optimum microbial community within the gastrointestinal tract is intricately linked to human metabolic, immune and brain health. Disturbance to these microbial populations perturbs the production of vital bioactive compounds synthesised by the gut microbiome, such as short-chain fatty acids (SCFAs). Of the SCFAs, butyrate is known to be a major source of energy for colonocytes and has valuable effects on the maintenance of intestinal epithelium and blood brain barrier integrity, gut motility and transit, anti-inflammatory effects, and autophagy induction.
