Relation of the CD11/CD18 family of leukocyte antigens to the transient neutropenia caused by chemoattractants.

Adherence of leukocytes to the endothelium is a prerequisite for infiltration and accumulation of cells at an inflammatory site. Recent studies suggest that the CD11/CD18 family of adhesion-promoting receptors plays a crucial role in the initial adherence of polymorphonuclear leukocytes (PMNs) to endothelium. We have studied the effect of the anti-CD18 monoclonal antibody (MoAb) IB4, on movement of PMN in rabbits. Accumulation of PMNs in the skin induced by a local injection of the chemoattractant, zymosan-activated serum (ZAS), was strongly inhibited, in a dose-dependent fashion, by intraveno

The role of Mac-1 (CD11b/CD18) in antigen-induced airway eosinophilia in mice.

Mac-1 (CD11b/CD18) is an important adhesion molecule involved in the migration of leukocytes, cell signaling, and subsequent secretory responses. Its precise role in eosinophil recruitment and activation in vivo is not entirely clear. We wished to directly examine the role of Mac-1 in eosinophil migration in a murine model of allergic pulmonary inflammation. Briefly, wild-type (C57Bl/6) and Mac-1-deficient/knockout (Mac-1 KO) mice were intraperitoneally sensitized with ovalbumin (OVA) and alum (AlOH) on Days 0 and 14, and intranasally challenged with OVA either once on Day 14 or five times on

The effects of two anti-CD18 antibodies on antigen-induced airway hyperresponsiveness and leukocyte accumulation in the guinea pig.

Guinea pigs sensitized to ovalbumin develop airway eosinophilia and bronchial hyperresponsiveness to acetylcholine and histamine 24 h after ovalbumin challenge. We have used two different antibodies to the leukocyte integrin CD18 (R15.7 and 6.5E) to study the link between eosinophilia and the development of bronchial hyperresponsiveness. In this model, both these antibodies reduced, to a similar degree, the number of eosinophils recovered from bronchoalveolar lavage fluid, although the eosinophil numbers in lung tissue were not reduced. However, only R15.7 reduced bronchial hyperresponsiveness

[The characteristics of CD18, CD11b, CD4, CD8 and CD3 antigen expression detectable by ICO monoclonal antibodies in the blood of hemoblastosis patients].

CD18-positive cells (beta-chain of antigen LFA-1), cells CD11b+, CD4+, CD8+ and CD3+ were quantified in the blood of 32 acute leukemia patients, 38 chronic lymphoid leukemia patients and 15 mature-cell lymphosarcoma patients. A standard reaction of indirect immunofluorescence was employed using ICO monoclonal antibodies. Alterations in the content and in the proportions of the main effector populations in the peripheral blood of hemoblastosis patients were observed. These were more apparent in the unfavourable course of acute leukemia and in chronic lymphoid leukemia. The defect was more marke

A novel CD18 genomic deletion in a patient with severe leucocyte adhesion deficiency: a possible CD2/lymphocyte function-associated antigen-1 functional association in humans.

Leucocyte adhesion deficiency (LAD) is an autosomal-recessive genetic disease that is characterized clinically by severe bacterial infections and caused by mutations in the CD18 gene that codes for the beta2 integrin subunit. A patient with a severe LAD phenotype was studied and the molecular basis of the disease was identified as a single homozygous defect in a Herpes virus saimiri (HVS)-transformed T-cell line. The defect identified involves a deletion of 171 bp in the cDNA that encodes part of the proteic extracellular domain. This genetic abnormality was further studied at the genomic DNA

Transfection of cells from patients with leukocyte adhesion deficiency with an integrin beta subunit (CD18) restores lymphocyte function-associated antigen-1 expression and function.

Leukocyte adhesion deficiency (LAD) is an inherited immunodeficiency disease that is characterized by the deficient expression of the leukocyte adhesion glycoproteins lymphocyte function-associated antigen-1 (LFA-1), Mac-1, and p150,95. This loss of expression is attributed to heterogeneous defects in the common beta subunit shared by these glycoproteins. Here we demonstrate that expression of the LFA-1 alpha beta heterodimer in EBV-transformed B lymphoblastoid cells from LAD patients can be recovered after transfection with the beta subunit cDNA contained in an EBV-based vector. Four patients

Flow cytometric immunophenotypic characteristics of plasma cell leukemia.

The aim of this prospective study was to define the flow cytometric characteristics of simultaneously investigated bone marrow and peripheral blood plasma cells antigens expression in 36 plasma cell leukemia (PCL) patients. The immunophenotypic profile of plasma cells was determined with a panel of monoclonal antibodies. The antigen expression intensity was calculated as relative fluorescence intensity (RFI). Bone marrow plasma cells showed expression of particular antigens in the following proportion of cases: CD49d 100%, CD29 94%, CD54 93%, CD44 83%, CD56 60%, CD18 26%, CD11b 29%, CD11a 19%,

Epidermal dendritic cells in psoriasis possess a phenotype associated with antigen presentation: in situ expression of beta 2-integrins.

BACKGROUND: Epidermal dendritic cells (DCs) isolated from psoriasis possess greatly enhanced T lymphocyte-activating properties compared with DCs from normal skin, suggesting that DCs in psoriasis express surface antigens crucial for antigen presentation. These include beta 2-integrins and intercellular adhesion molecule (ICAM)-1. OBJECTIVE: Our purpose was to determine DC phenotype in psoriatic compared with normal epidermis with respect to these molecules. METHODS: Tissue sections were single labeled with a peroxidase antiperoxidase (PAP) immunohistochemical technique and double labeled wh

Adhesion molecule CD11a/CD18-deficient Burkitt's lymphoma cells lack the transcript for the beta, but not the alpha, integrin subunit.

Adhesion to cells and matrices participates in the regulation of lymphocyte proliferation, maturation and tissue localization. Consequently, abnormal patterns of adhesion molecule expression may contribute to the pathophysiology of lymphoproliferative disorders. Integrins are major cell-surface adhesive proteins composed by alpha and beta subunits. In contrast to normal lymphocytes, Burkitt's lymphoma (BL) cells lack the beta2 integrin CD11a/CD18. To study the molecular mechanism underlying this deficiency, presence of the transcript for each subunit was analysed by Northern blotting in group

Phosphorylation of the β2 integrin subunit CD18 contributes to Actinobacillus pleuropneumoniae ApxI-induced cytotoxicity through the downregulation of Akt activity.

Actinobacillus pleuropneumoniae, the causative agent of porcine pleuropneumonia, produces various virulence factors, including the exotoxin ApxI, which contributes to disease pathogenesis. ApxI induces cytotoxicity through porcine lymphocyte function-associated antigen-1 (LFA-1; CD11a/CD18) while concurrently attenuating Akt activity. This study aimed to investigate the regulatory role of porcine CD18, focusing on the tyrosine 735 residue in its cytoplasmic tail, and to examine the involvement of Akt in ApxI-induced cytotoxicity. Through transfection experiments, we demonstrated that the expre

Amelioration of antigen-induced arthritis in rabbits treated with monoclonal antibodies to leukocyte adhesion molecules.

OBJECTIVE: To examine the effects of treatment of antigen-induced arthritis in rabbits with a monoclonal antibody against CD18, the common beta chain of the leukocyte adhesion molecules. Intraarticular injection of antigen into primed rabbits elicits an acute inflammatory response followed by chronic arthritis in this model. METHODS: Anti-CD18 was given at the time of intraarticular antigen administration, and effects on the acute and chronic arthritis were investigated. Twenty-four rabbits were examined (11 controls, 3 receiving normal mouse IgG, and 10 receiving anti-CD18). RESULTS: Flow c

Canine leukocyte integrins: characterization of a CD18 homologue.

The major characteristics of the canine CD18 leukocyte antigen family, defined by a murine monoclonal antibody CA1,4E9, are described. The overall molecular organization of this family of leukocyte integrins was similar to the human counterpart, and consisted of a common beta subunit of 95 kD (CD18) and non-covalently linked alpha subunits of 150 kD (CD11C), 165 kD (CD11b) and 180 kD (Cd11a). Conservation of CD18 epitopes between dog, human, feline, equine and bovine was observed. CD18 expression in lymphoid tissue had a characteristic pattern which was based on differences in the intensity of

Neutrophil alloantibodies react with cytoplasmic antigens: a possible cause of false-positive indirect immunofluorescence assays for antibodies to neutrophil cytoplasmic antigens.

Antibodies to neutrophil cytoplasmic antigens (ANCA) can be detected in patients with Wegener's granulomatosis and systemic vasculitis. During pregnancy or following transfusion, subjects sometimes produce alloantibodies to neutrophil antigens. If patient sera being tested for ANCA contain alloantibodies directed at neutrophil antigens that residue in the cytoplasm, the results may be difficult to interpret. At least one neutrophil antigen, NB1, is expressed on both neutrophil plasma membranes and secondary granules. We tested alloantibodies specific for neutrophil antigens NA1, NA2, NB1, NB2,

Occupancy of CD11b/CD18 (Mac-1) divalent ion binding site(s) induces leukocyte adhesion.

The group of leukocyte integrins CD11a-c/CD18 coordinate disparate adhesion reactions in the immune system through a regulated process of ligand recognition. The participation of the receptor divalent ion binding site(s) in this mechanism of ligand binding has been investigated. As compared with other divalent cations, Mn2+ ions have the unique property to dramatically stimulate the adhesive functions of the leukocyte integrin CD11b/CD18 (Mac-1), expressed on myelo-monocytic cells. This is reflected in a three- to fivefold increased early monocyte adhesion (less than 20 min) to resting, unpert

Coronin 1A, a novel player in integrin biology, controls neutrophil trafficking in innate immunity.

Trafficking of polymorphonuclear neutrophils (PMNs) during inflammation critically depends on the β2 integrins lymphocyte function-associated antigen 1 (LFA-1) (CD11a/CD18) and macrophage-1 antigen (CD11b/CD18). Here, we identify coronin 1A (Coro1A) as a novel regulator of β2 integrins that interacts with the cytoplasmic tail of CD18 and is crucial for induction of PMN adhesion and postadhesion events, including adhesion strengthening, spreading, and migration under flow conditions. Transition of PMN rolling to firm adhesion critically depends on Coro1A by regulating the accumulation of high-a

Adhesion molecules on MHC-nonrestricted lymphocytes: high density expression and role in oncolysis.

We have shown that interleukin-2 (IL-2) -activated adherent lymphocytes (A-LAK) display superior oncolytic activity against acute myelogenous leukemia (AML) blasts when compared to conventionally prepared lymphocytes with lymphokine-activated killing (LAK) activity. The A-LAK activity was generated promptly and from donors whose lymphocytes did not display any LAK activity. In comparison to LAK, a higher percentage of A-LAK expressed the CD25 and HLA class II (HLA-DR) activation-associated structures and high density of HLA class I antigens. Most striking, however, was the observation that lym

Immunophenotyping of human dendriform cells from the conjunctival epithelium.

PURPOSE: Conjunctival Langerhans cells are bone marrow-derived, antigen-presenting cells that play a major role in the immune response of the ocular surface, but they have as yet been little investigated, either functionally or phenotypically. This study was undertaken in impression cytology to provide an extended immunophenotype of human conjunctival dendriform cells. METHODS: Immunostaining procedures were used to seek for the expression of the following 30 membrane antigens related to the immune system, using dendriform cells obtained in conjunctival specimens from 80 normal subjects and 1

The role of higher-order protein structure in supporting binding by heteroclitic monoclonal antibodies: the monoclonal antibody KIM185 to CD18 also binds C4-binding protein.

Heteroclitic monoclonal antibodies are characterized by the ability to bind multiple epitopes with little or no similarity. Such antibodies have been reported earlier, but insight into to the molecular basis of this propensity is limited. Here we report that the KIM185 antibody to human CD18 reacts with the plasma protein C4b-binding protein (C4BP). This was revealed during affinity purification procedures where human serum was incubated with surfaces coated with monoclonal antibodies to CD18. Other monoclonal antibodies to CD18 (KIM127 and TS1/18) showed no such interaction with C4BP. We cons

Adhesion molecules mediating recruitment of monocytes to inflamed tissue.

Three families of cell-surface proteins are largely responsible for the adherence of leukocytes to cells and matrices: integrins, immunoglobulin (Ig)-related molecules and selectins. Blood monocytes express beta 1 integrins VLA-4, -5 and -6 and beta 2 integrins CD11a/CD18, CD11b/CD18 and CD11c/CD18. These cells also express the Ig-related molecules ICAM-1, -2 and -3, ligands for the beta 2 integrins. In addition, monocytes express L-selectin and the oligosaccharides Lex and sialyl Lex, ligands for the endothelial selectins E- and P-. In vitro studies with blocking antibodies have identified ad
