CD5 B lymphocyte antigen in monoclonal gammopathy.

Because B lymphocytes bearing the CD5 antigen have been involved in many B-cell malignancies, we have investigated the presence of the CD5 B-cell antigen on B and plasma cells in monoclonal gammopathy. Quantification of CD5 B cells was made in the peripheral blood of seven individuals with monoclonal gammopathy of undetermined significance (MGUS) and in that of 21 patients with multiple myeloma (MM). The bone marrow of ten patients with MM was also studied. Patients with progressive MM presented a significant reduction in both B and CD5 B lymphocytes (i.e., percentages and absolute numbers), w

Comparative expression of major histocompatibility complex (MHC) antigens on CD5+ and CD5- B cells in patients with chronic lymphocytic leukaemia (CLL).

The aim of this study was to investigate the expression of major histocompatibility complex (MHC) antigens on CD5+ and CD5- B cells of 13 patients with chronic lymphocytic leukaemia (CLL). This was carried out using a series of monoclonal antibodies (MAbs) against polymorphic and monomorphic class I and class II antigens, as well as to the transferrin receptor and assessed by flow cytometry and direct and indirect immunofluorescence. The expression of these molecules was assessed as mean fluorescent intensity (MFI). The results showed that cells from all 13 individuals expressed monomorphic cl

CD5 is associated with the human B cell antigen receptor complex.

On human B cells the antigen receptor complex is composed of the membrane form of the immunoglobulin molecule and the non-covalently associated Ig alpha/beta heterodimer. A small subpopulation of normal B cells and chronic lymphocytic leukemia B cells express (analogous to T cells) the transmembrane molecule CD5, a counterstructure of B cell-specific CD72. Numbers of CD5+ B cells are increased in several physiological and pathological conditions. Moreover, CD5+ B cells are being held responsible for the production of autoreactive antibodies and seem to have signaling characteristics distinct f

Selective response of CD5+ B cell malignancies to activation of the CD72 antigen.

The function of the simultaneous expression of CD5 and its ligand CD72 on B cell malignancies like chronic lymphocytic leukemia and mantle cell lymphoma was assessed. It is unknown if reciprocal interactions between CD72 and CD5 exert an autocrine growth-promoting or -inhibiting effect. CD5+ (n = 13) and CD5- (n = 9) B cell malignancies were cultured with the anti-CD72 mAb WL225. For comparison, five other anti-CD72 mAbs were tested. Only CD5+ B cell malignancies proliferated upon CD72 activation (9 out of 13 cases). A strong suppressive effect of IL-4 on the anti-CD72-induced [3H]thymidine in

Evidence for a trans-acting activator function regulating the expression of the human CD5 antigen.

Interspecies somatic cell hybrids were generated by fusing the mouse T-lymphoma cell line, BW5147, with normal human T lymphocytes at different stages of differentiation. Thymocytes, activated peripheral T lymphocytes, or an activated T-cell clone were used as human partners, respectively, in three independent fusions. Irrespective of the human cell partner used for fusion, a certain number of hybrids lost CD5 surface expression over a period of time in culture. Analysis at the phenotype and genetic level showed that lack of CD5 expression was due neither to segregation of human autosome 11, o

CD5 is phosphorylated on tyrosine after stimulation of the T-cell antigen receptor complex.

When T cells are activated by the T-cell antigen receptor, a number of cellular proteins are phosphorylated on tyrosine. We investigated whether any of these proteins were present on the surface of activated T cells. Using the human leukemic T-cell line Jurkat and normal peripheral blood lymphocytes, we identified a 67-kDa cell surface glycoprotein in anti-phosphotyrosine immunoprecipitates, after treatment of the cells with CD3 antibody. When cell lysates were depleted of CD5 by sequential immunoprecipitation, the 67-kDa phosphotyrosyl polypeptide was no longer precipitated by the phosphotyro

Intravascular large B-cell lymphoma: the CD5 antigen is expressed by a subset of cases.

The CD5 antigen is a T-cell associated marker that is also usually expressed by two B-cell neoplasms, chronic lymphocytic leukemia/small lymphocytic lymphoma and mantle cell lymphoma. We observed CD5 antigen expression in a subset of cases of intravascular large B-cell lymphoma (IVLBL), and we report here five cases. The patients, two men and three women, ranged in age from 59 to 81 years. Biopsy specimens were obtained from kidney, lung, bone marrow, abdominal wall, and neck, the latter involving a lymphangioma. All of the cases had histologic features typical of IVLBL, with large and atypica

Interactome analysis of CD5 and CD6 ectodomains with tegumental antigens from the helminth parasite Echinococcus granulosus sensu lato.

Echinococcus granulosus sensu lato (s.l.) is a cestode parasite affecting both human and livestock health. Recombinant ectodomains of human scavenger receptors CD5 (rshCD5) and CD6 (rshCD6) were previously reported to bind its tegumental antigens and to exert prophylactic effects in a murine model of infection. Although the properties of mammalian scavenger receptors include the binding to diverse pathogen-derived structures, their interaction with helminth parasites has been scarcely explored. Therefore, we report here a search for CD5 and CD6 interactors within E. granulosus s.l. antigens. M

CD5+ B lymphocytes are the main source of antibodies reactive with non-parasite antigens in Trypanosoma congolense-infected cattle.

Mice infected with African trypanosomes produce exceptionally large amounts of serum IgM, a major part of which binds to non-trypanosome antigens such as trinitrophenol and single-strand DNA. In this paper, we describe that in cattle infected with Trypanosoma congolense and T. vivax, similar antibodies are found, although they bind mainly to protein antigens, such as beta-galactosidase, ovalbumin and ferritin. The parasite non-specific IgM antibodies appear around the same time as the parasite-specific antibodies, but their origin and function are not clear. We tested the hypothesis that CD5+

[Diminished expression of CD5 and/or CD7 surface antigens as the first clue of diagnosis for monoclonal T lymphocytosis].

The significance of precursor status for mature T-cell neoplasms has not been elucidated. Diagnosis of T-cell neoplasms is often challenging and sometimes overlooked when leukocytosis is not evident and neoplastic cell morphology is hardly distinguishable. Here, we report two cases of monoclonal T lymphocytosis (MTL) without evident lymphocytosis and lymphadenopathies that show slightly diminished CD5 and/or CD7 surface antigens as the first clue of diagnosis. Recently, some reports have demonstrated the possibility that monoclonal lymphocytosis precedes leukemia/lymphoma. Although its clinica

Antigen-binding affinity is a key determinant of the durable antitumor activity of CD5 CAR-T cells.

T cell fratricide in T cell antigen-targeted chimeric antigen receptor (CAR)-T cell therapies remains a critical barrier to achieving optimal antitumor efficacy. To address this challenge, we explored modulation of antigen-binding affinity as a simple yet effective strategy to mitigate fratricide. To this end, we aimed to develop low-affinity CD5-specific CAR-T cells and to test the hypothesis that low-affinity CD5 CAR-T cells can evade T cell fratricide, thereby alleviating T cell exhaustion and enhancing sustained antitumor activity. Our results demonstrate that CD5 CAR-T cells engineered wi

Preclinical targeting of aggressive T-cell malignancies using anti-CD5 chimeric antigen receptor.

The outlook for T-cell malignancies remain poor due to the lack of effective therapeutic options. Chimeric antigen receptor (CAR) immunotherapy has recently shown promise in clinical trials for B-cell malignancies, however, designing CARs for T-cell based disease remain a challenge due to the shared surface antigen pool between normal and malignant T-cells. Normal T-cells express CD5 but NK (natural killer) cells do not, positioning NK cells as attractive cytotoxicity cells for CD5CAR design. Additionally, CD5 is highly expressed in T-cell acute lymphoblastic leukemia (T-ALL) and peripheral T-

Discrimination between two subsets of porcine CD8+ cytolytic T lymphocytes by the expression of CD5 antigen.

Previous work has revealed striking differences in the peripheral T-lymphocyte compartments of swine compared to other species. The key difference is the existence of four T-lymphocyte subpopulations defined by their CD4/CD8 expression. Besides this difference in the CD4/CD8 antigen expression, we report here another unusual antigen distribution on porcine extrathymic T lymphocytes: the expression of the 63,000 MW CD5 antigen. In porcine thymus the CD5 antigen shows a biphasic antigen density, whereas the majority of thymocytes are characterized by an intermediate CD5 expression. In the extrat

The Class I Scavenger Receptors CD5 and CD6 Play a Role in the Early Peritoneal Immune Response to Echinococcus granulosus Tegumental Antigens.

Scavenger Receptors (SRs) comprise a structurally diverse group of pattern recognition receptors (PRRs) involved in sensing non-self (microbial-associated molecular patterns) or altered-self ligands. CD5 and CD6 are class I SRs (SR-I) preferentially expressed by lymphoid cells and characterized by the presence of several tandem scavenger receptor cysteine-rich (SRCR) domain repeats. Both receptors interact with diverse microbial structures, including tegumental antigens from Echinococcus granulosus sensu lato (s.l.), the cestode parasite responsible for cystic echinococcosis (CE). This is nota

Intermittent fasting alters the antigen-specific CD5+ B-1 cell natural antibody repertoire in male and female mice.

Natural antibodies (NAbs), primarily produced by CD5+ B-1 cells, provide critical early protection against infections such as Streptococcus pneumoniae. The structure of these natural antibodies, germline-like with minimal N-additions, is essential for their protective capacity. The protective capacity and germline status of NAbs are compromised in aged male mice but maintained in aged female mice. CD5+ B-1 cells are maintained through self-renewal, necessary for maintaining germline-like natural antibodies, and this process depends on autophagy. Intermittent fasting enhances autophagy and stem

Characterizing Kupffer Cell Production of CD5 Antigen-Like and Its Function on Regulating Migration of Natural Killer T Cells.

CD5 antigen-like (CD5L) is a multifunctional glycoprotein characterized for its role in the lipid metabolism, particularly within macrophages. In the liver, CD5L strongly correlates with liver injury. This study explored the role of CD5L on liver lipid accumulation and inflammatory response. CD5L promoted lipid uptake in hepatocytes and stellate cells. In multiple models of liver injury, expression of Cd5l was associated with that of Clec4f, a marker for liver macrophages, consistent with its role as a macrophage survival factor. Transwell assay was used to demonstrate a novel function of CD5L

IL-23p19 and CD5 antigen-like form a possible novel heterodimeric cytokine and contribute to experimental autoimmune encephalomyelitis development.

Among various cytokines, interleukin (IL)-12 family cytokines have very unique characteristics in that they are composed of two distinct subunits and these subunits are shared with each other. IL-23, one of the IL-12 family cytokines, consists of p19 and p40 subunits, is mainly produced by antigen-presenting cells, and plays a critical role in the expansion and maintenance of pathogenic helper CD4+ T (Th)17 cells. Since we initially found that p19 is secreted in the culture supernatant of activated CD4+ T cells, we have further investigated the role of p19. p19 was revealed to associate with C

Role of B-cell antigen receptor-associated molecules and lipid rafts in CD5-induced apoptosis of B CLL cells.

A total of 40 patients with B-CLL were investigated for CD5-triggered apoptosis and categorized as 20 resistant (group I) and 20 sensitive patients (group II). The densities of surface IgM (sIgM) and CD5 were lower in group I than group II, as were the percentages of CD79b+, CD38+, and Zap70-expressing B cells. CD5 signaling was mediated through the BCR in group II B cells, as established by coimmunoprecipitation of CD5 and CD79a and tyrosine phosphorylation of CD79a. Following colocalization of CD5 and sIgM in membrane lipid rafts (LRs), Syk became associated with these molecules, whereas SHP

Adoptive transfer of NK cells engineered with a CD5-based chimeric antigen receptor (SRCD5CAR) to treat invasive fungal infections.

The treatment of invasive fungal infections (IFIs) poses a major health challenge due to their severity, the scarcity and toxicity of current antifungal drugs, and the emergence of drug-resistant strains, thus forcing the search for novel strategies. Approaches involving adoptive transfer of immune cells harboring chimeric antigen receptors (CARs) are gaining momentum in several clinical settings. Based on the fungal β-glucan-binding properties of the lymphocyte scavenger receptor CD5, we assayed NK and T cells engineered with a CD5-based second-generation CAR (SRCD5CAR) for the treatment of I

Single-Cell Dissection Reveals Immune Dysregulation After CD5 or CD7-Directed Chimeric Antigen Receptor T-Cell Therapy.

CD5- and CD7-directed chimeric antigen receptor T-cell (5CAR and 7CAR) therapies for T-cell malignancies carry the risk of life-threatening infection. Although depletion of target-positive lymphocytes is expected, the contribution of residual cell dysfunction to infection risk remains unclear. This work uses single-cell sequencing to investigate immune dysregulation after 5CAR or 7CAR therapy in patients with T-cell acute lymphoblastic leukemia. 5CAR induces marked T-cell exhaustion linked to CD5 loss and B lymphocyte-induced maturation protein 1 upregulation.
