Genetic Divergence and Antibody Expression Influence the N-Glycomes of CHO-K1 and CHO-S Cells.

While Chinese hamster ovary (CHO) cells continue to be the workhorse of recombinant therapeutic protein production, decades of genetic divergence in the industrially relevant CHO-K1 and CHO-S cell lines, are likely to have resulted in differing glycosylation capabilities. Glycosylation can influence the efficacy, serum half-life, and safety of biologics, and as a critical quality attribute of glycoprotein biopharmaceuticals, it is essential to better understand how major CHO cell manufacturing platforms diverge. We used matrix-assisted laser desorption ionization-time of flight mass spectromet

G1 Synchronization of CHO Cells by Isoleucine Deprivation.

INTRODUCTIONThis protocol provides a method for the synchronization of a monolayer culture of CHO cells in G(1) using isoleucine deprivation. Since CHO cells can also be adapted to grow in suspension culture, this procedure can be used to obtain larger quantities of cells. When isoleucine is replaced, the cells resume growth and begin to enter S phase ~4 hours later. This method arrests almost 100% of the CHO cells in G(1), and upon reversal, leads to rapid recovery of cell growth and very high cell viability. This method has been used with other hamster cell lines, some transformed mouse cell

Antiapoptosis Engineering for Improved Protein Production from CHO Cells.

Improving the time integral of viable cell concentration by overcoming cell death, namely apoptosis, is one of the most widely used strategies for the efficient production of therapeutic proteins. By establishing stable cell lines that overexpress antiapoptotic genes or downregulate proapoptotic genes, the final product yields can be enhanced as cells become more resistant to environmental stresses. From the selection of high-expressing clones to verification of antiapoptotic activity, the method to construct a stable antiapoptotic cell line is discussed in this chapter.

[Expression of ATR-Fc fusion protein in CHO cells].

ATR-Fc is a fusion protein consisting of extracellular domain of human anthrax toxin receptor (ATR) and a fragment (hinge, CH2, and CH3 domains) of the Fc of human IgG1. The aim of ATR-Fc expression is to get an antibody-like molecule binding to protective antigen (PA), a component of anthrax toxins, this fusion protein may compete with cell surface receptor for PA binding, and block the transport of lethal factor (LF) and edema factor (EF) into cells, thereby act as an antitoxin to prevent and treat anthrax infection. A DNA fragment encoding N-terminal amino acids 1-227 of ATR and human IgG1

Comprehensive genome-scale CRISPR knockout screening of CHO cells.

Chinese hamster ovary (CHO) cells play a pivotal role in the production of recombinant therapeutics. In the present study, we conducted a genome-scale pooled CRISPR knockout (KO) screening using a virus-free, recombinase-mediated cassette exchange-based platform in CHO-K1 host and CHO-K1 derived recombinant cells. Genome-wide guide RNA (gRNA) amplicon sequencing data were generated from cell libraries, as well as short- and long-term KO libraries, and validated through phenotypic assessment and gRNA read count distribution. Additionally, we obtained gRNA amplicon sequencing data from the highl

Glycosylation control strategies for recombinant therapeutic proteins in CHO cells.

Chinese hamster ovary (CHO) cells are widely employed as host cells for the production of recombinant therapeutic proteins (RTPs). Glycosylation, a crucial post-translational modification, significantly affects the function and quality of RTPs expressed in CHO cells. This modification not only influences protein folding, stability, and immunogenicity, but also dictates their biological activity and half-life. This review summarizes the biological role of glycosylation within CHO cells, analyzes both intrinsic and extrinsic factors affecting recombinant protein glycosylation, and explores recen

Toward a 4D genome annotation of CHO cells for biomanufacturing.

Biotechnology and biopharma rely on detailed genome annotations for cell-line engineering, yet most production hosts are nonmodel organisms with limited resources linking sequence to physiology across space and time. A complete four-dimensional genome annotation for Chinese hamster ovary (CHO) cells that links sequence, networks, spatial constraints, process state, and passaging history does not yet exist; current efforts instead provide partial layers that must be connected into an actionable framework. In this opinion article, we illustrate this framework for CHO cells, the dominant platform

Global structure and determinants of transcriptomic adaptation in CHO cells.

Transcriptomic adaptation plays a central role in the phenotypic plasticity of Chinese hamster ovary (CHO) cells. While various gene expression studies have provided insights into condition-specific responses, a comprehensive, systems-level understanding of how CHO cell transcriptomes dynamically adapt across diverse experimental contexts remains limited. Here, we present a large-scale investigation of more than 600 RNA-seq samples across diverse cell lines, culture conditions and experimental designs to systematically dissect stable and adaptive components of the transcriptome. We consolidate

CHO cells for virus-like particle and subunit vaccine manufacturing.

Chinese Hamster Ovary (CHO) cells, employed primarily for manufacturing monoclonal antibodies and other recombinant protein (r-protein) therapeutics, are emerging as a promising host for vaccine antigen production. This is exemplified by the recently approved CHO cell-derived subunit vaccines (SUV) against respiratory syncytial virus (RSV) and varicella-zoster virus (VZV), as well as the enveloped virus-like particle (eVLP) vaccine against hepatitis B virus (HBV). Here, we summarize the design, production, and immunogenicity features of these vaccine and review the most recent progress of othe

Stress-induced increase of monoclonal antibody production in CHO cells.

Monoclonal antibodies (mAbs) are of great interest to the biopharmaceutical industry due to their widely used application as human therapeutic and diagnostic agents. As such, mAb require to exhibit human-like glycolization patterns. Therefore, recombinant Chinese hamster ovary (CHO) cells are the favored production organisms; many relevant biopharmaceuticals are already produced by this cell type. To optimize the mAb yield in CHO DG44 cells a corelation between stress-induced cell size expansion and increased specific productivity was investigated. CO2 and macronutrient supply of the cells dur

Large-scale isolation of UV-sensitive clones of CHO cells.

We have isolated 54 ultraviolet light (UV) sensitive clones of Chinese hamster ovary (CHO) cells, including two from a parent cell line which is hypersensitive to ethyl methanesulfonate (EMS) and is also sensitive to X rays. A replica plating technique was used for the isolation of two of the clones, and a semiautomated technique was used for the isolation of the other 52 clones. We have observed UV sensitization of up to 5-fold in the mutants relative to the parent in terms of the slopes of the survival curves. Seven of the clones were examined for DNA repair competence using a repair replica

Advancing recombinant protein production in CHO cells through metabolic engineering.

Chinese hamster ovary (CHO) cells serve as the predominant platform for producing recombinant therapeutic proteins in biopharmaceutical manufacturing, the production capacity of which relies heavily on efficient protein synthesis, folding, and secretion pathways. However, during high-density and prolonged cultivation, these cells frequently encounter bottlenecks-including excessive lactate and ammonia accumulation, redox imbalance, and endoplasmic reticulum (ER) stress-which ultimately constrain both the yield and quality of target protein. To overcome these limitations, metabolic engineering

Identification of Loci with High Transgene Expression in CHO Cells.

Chinese Hamster Ovary (CHO) cells are commonly used for producing therapeutic proteins in the biopharmaceutical industry. Targeted integration (TI) of therapeutic protein-encoding transgenes into predetermined high and stably expressing genomic loci can simplify the cell line development processes for biologics production. Establishing a successful TI system requires identifying genomic loci that allow a high expression of the integrated transgenes. In this work, we demonstrated that the Thousands of Reporters Integrated in Parallel (TRIP) technology can identify such transcriptional hotspots

Combating viral contaminants in CHO cells by engineering innate immunity.

Viral contamination in biopharmaceutical manufacturing can lead to shortages in the supply of critical therapeutics. To facilitate the protection of bioprocesses, we explored the basis for the susceptibility of CHO cells to RNA virus infection. Upon infection with certain ssRNA and dsRNA viruses, CHO cells fail to generate a significant interferon (IFN) response. Nonetheless, the downstream machinery for generating IFN responses and its antiviral activity is intact in these cells: treatment of cells with exogenously-added type I IFN or poly I:C prior to infection limited the cytopathic effect

Stimulation of adenylate cyclase by amylin in CHO-K1 cells.

The CHO-K1 cell line responds to the peptide amylin by a rapid elevation of cAMP. The related peptide calcitonin gene-related peptide (CGRP) is 100 times less potent at stimulating adenylate cyclase than is amylin. The actions of amylin at this receptor are concentration dependent and not antagonized by the CGRP antagonist CGRP-(8-37). Although these cells have receptors for calcitonin, amylin is unable to take part in any high affinity interaction with these receptors, as assessed by radioligand binding. The CHO-K1 cell line has receptors for amylin that are distinct from those for calcitonin

Endocytosis and recycling of the fibronectin receptor in CHO cells.

An anti-fibronectin receptor monoclonal antibody preferentially labels the leading edges of freshly plated CHO fibroblasts, suggesting that this receptor circulates through the endocytic cycle. Using a new labelling reagent, I show that this receptor is indeed endocytosed at 37 degrees C and then returned to the cell surface. These findings imply that fibronectin receptors are recirculated to the leading edge of a motile cell by the endocytic cycle, and establish that the processes of endocytosis/exocytosis and cell locomotion are intimately linked.

High frequency of mutation to tubercidin resistance in CHO cells.

The acquisition of high-level resistance to tubercidin (an adenosine analog) in CHO cells occurs in a single step at high frequency (10(-3) to 10(-4)) without mutagenesis. Analysis of a large number of independent mutants by a fluctuation test (Luria and Delbruk, 1943) indicates that they arise independently of the selection medium and all fall into the same complementation group. All mutants tested lack detectable adenosine kinase activity. An analysis of hybrids between mutant and wild-type cells indicates that resistance to tubercidin is a recessive marker which segregates as would be expec

A replica plating method for CHO cells using nylon cloth.

A new technique of replica plating for CHO cells has been developed in which the pattern of colonies growing in a tissue culture plate is reproduced on a nylon cloth. The efficiency of replica plating onto the nylon cloth is affected by the colony size and the time of cloth-to-plate contact. Maximum replica plating efficiency (80-90%) occurs using 5- to 6-day-old colonies and a contact time of 4 h. The method easily allows 10(4) colonies to be examined for mutant phenotypes. The effectiveness of the technique is demonstrated by isolation of a clone deficient in activity of glucose-6-phosphate

Co-expression enhances cloning efficiency and protein production in CHO cells.

Chinese hamster ovary (CHO) cell lines are widely used in the biopharmaceutical industry, particularly for the production of monoclonal antibodies and recombinant protein drugs. Recombinant cell lines are typically derived from single-cell clones to ensure product consistency and stability. However, the difficult-to-express recombinant proteins may impair single-cell proliferation, thereby significantly reducing cloning efficiency. In this study, co-expression of insulin-like growth factor-1 (IGF-1) was used to improve the single-cell cloning efficiency. Co-expression of IGF-1 significantly im

Identification of ICR170-induced XPD mutations in UV-sensitive CHO cells.

We highlight selected contributions of Dr. Richard Setlow that contributed to our earlier understanding of excision repair processes and set the stage for dissecting nucleotide excision repair (NER) in mammalian cells through molecular genetics. More than 20 years ago, large-scale screens for UV-sensitive mutants of hamster CHO cells isolated approximately 200 mutants, many of which were assigned to the XPD/ERCC2 complementation group, but the nature of the mutations was not determined. The XPD protein performs not only an essential viability function as a structural component of transcription
