Callyptide A, a new cytotoxic peptide from the Red Sea marine sponge Callyspongia species.

In the course of our continuing efforts to allocate bioactive secondary metabolites from Red Sea marine invertebrates, we have investigated the sponge Callyspongia species. The cytotoxic dichloromethane fraction of the methanolic extract of the sponge afforded a new cytotoxic peptide named callyptide A (1). Its structure was determined by extensive 1D and 2D NMR (COSY, HSQC and HMBC) studies and high-resolution mass spectral determination. The configuration of the amino acids was determined by Marfey's analysis. Callyptide A was found to exhibit growth inhibitory activity when tested against d

Afferent sensitization and efferent cytotoxicity in allogeneic tissue responses of the marine sponge Callyspongia diffusa.

Allogeneic branches of the marine sponge Callyspongia diffusa were paired either in parabiosis or as allografts. To examine the specificity of the allogeneic rejection process, naive second sets and third parties were substituted in the midst of primary allogeneic tissue responses. Other experiments were conducted with 0.2-, 0.8-, 3.0-, and 5.0-micron pore size membrane barriers separating various combinations of naive and presensitized allogeneic paired sponges. The results of these experiments demonstrate that both the afferent and efferent phases of allograft rejection in Callyspongia diffu

Characterization of an interleukin 6 cytokine family antagonist protein from a marine sponge, Callyspongia sp.

An inhibitor of IL-6 binding to the human hepatoma line HepG2 and myeloma cell line U266 was identified in a saline extract of the marine sponge, Callyspongia sp. Functional activity, measured through the increase in haptoglobin production by HepG2 cells stimulated with IL-6, could be strongly inhibited by the extract. Similarly, IL-6-induced production of IgM by the B cell line SKW6.4 was substantially reduced. In neither cell line was there evidence of toxicity produced by the extract. Other sponges of the Callyspongia species were found to contain analogous activity. The activity was destro

Callypyrones from marine Callyspongiidae sponge Callyspongia diffusa: antihypertensive bis-γ-pyrone polypropionates attenuate angiotensin-converting enzyme.

Angiotensin I-converting enzyme (ACE) catalyses the biosynthesis of angiotensin II, a potent blood vessel constrictor, from angiotensin I, and ACE inhibitors were recognised as medications for hypertension. Undescribed bis-γ-pyrone polypropionate compounds, callypyrones A and B were purified from the organic extract of Callyspongiidae sponge species Callyspongia diffusa by repeated chromatographic purification. Callypyrone A exhibited significantly greater attenuation potential against ACE (IC50 0.48 mM) than that displayed by callypyrone B (IC50 0.57 mM) and showed comparable activity with st
