Study of rabies virus by Differential Scanning Calorimetry.

Differential Scanning Calorimetry (DSC) has been used in the past to study the thermal unfolding of many different viruses. Here we present the first DSC analysis of rabies virus. We show that non-inactivated, purified rabies virus unfolds cooperatively in two events centered at approximately 62 and 73 °C. Beta-propiolactone (BPL) treatment does not alter significantly viral unfolding behavior, indicating that viral inactivation does not alter protein structure significantly. The first unfolding event was absent in bromelain treated samples, causing an elimination of the G-protein ectodomain,

Differential scanning calorimetry and scanning thermal microscopy analysis of pharmaceutical materials.

Micro-thermal analysis (microTA) by scanning thermal microscopy is being used increasingly for the analysis of pharmaceutical dosage forms. However, there is currently little evidence to show that microTA data can compare directly with that from the established approach of differential scanning calorimetry (DSC). This work compares DSC and microTA data from an active vitamin B6 analogue, pyridoxal hydrochloride, and two commonly used pharmaceutical excipients, Mannitol and Avicel which are used in its formulation. It is found that microTA provides precise and accurate micro-thermal analytical

Intrinsic affinity of protein - ligand binding by differential scanning calorimetry.

Differential scanning calorimetry (DSC) determines the enthalpy change upon protein unfolding and the melting temperature of the protein. Performing DSC of a protein in the presence of increasing concentrations of specifically-binding ligand yields a series of curves that can be fit to obtain the protein-ligand dissociation constant as done in the fluorescence-based thermal shift assay (FTSA, ThermoFluor, DSF). The enthalpy of unfolding, as directly determined by DSC, helps improving the precision of the fit. If the ligand binding is linked to protonation reactions, the intrinsic binding const

Thermal transition studies of a mature xanthelasma by differential scanning calorimetry.

A large xanthelasma which had been present for at least 5 years was removed surgically from a normolipaemic female age 54 years, and examined in the fresh state by differential scanning calorimetry. Thermal transitions recorded over the range 30-40 degrees C suggest that the lipid present, predominantly esterified cholesterol, is not bound to protein or other tissue components, and that the chronicity of mature xanthelasmata as with ectopic lipid deposits at other sites is enhanced by chemical modification of lipid and effects on component phase behaviour, which are significant at local skin t

Interaction of F-actin with phosphate analogues studied by differential scanning calorimetry.

The thermal unfolding of F-actin and the changes induced in it by the binding of phosphate analogues were studied by differential scanning calorimetry. It is shown that the conformation of actin is drastically altered by interaction with beryllium fluoride or aluminium fluoride, while the effects of vanadate and phosphate are negligible. The effect of beryllium fluoride on the F-actin structure, as reflected in a significant increase of the actin thermal stability, is much more pronounced in the presence of Mg2+ than in the case of F-actin polymerized by KCl or LiCl in the absence of Mg2+. It

Characterization of different strains of poliovirus and influenza virus by differential scanning calorimetry.

Vaccines against poliomyelitis and influenza contain inactivated forms of poliovirus and influenza virus. These antigens are generated on an industrial scale from the purified active viruses that have been analysed in this study by DSC (differential scanning calorimetry). Multiple unfolding transitions are seen for influenza virus A/New Caledonia/20/99 (H1N1), A/Panama/2007/99 (H3N2) and B/Shangdong/7/97. These data, combined with previously reported data on other influenza viruses, indicates that each influenza virus strain has a characteristic unfolding behaviour. Only minor changes were see

Compatibility study between ibuproxam and pharmaceutical excipients using differential scanning calorimetry, hot-stage microscopy and scanning electron microscopy.

Differential scanning calorimetry (DSC) was used as a screening technique for assessing the compatibility of ibuproxam with some currently employed pharmaceutical excipients. The influence of processing effects (simple blending, cogrinding or kneading) on drug stability was also evaluated. On the basis of DSC results, ibuproxam was found to be compatible with corn starch, avicel and sodium carboxymethylcellulose. Some drug-excipient interaction was observed with polyethyleneglycol 4000, palmitic acid, stearic acid, Ca and Mg stearate. Actual solid-phase interactions of the drug with polyvinylp

Benzodiazepines co-crystals screening using FTIR and Raman spectroscopy supported by differential scanning calorimetry.

Co-crystals, which are defined as "solids that are crystalline materials composed of two or more molecules in the same crystal lattice" have recently been the focus of increased interest in the pharmaceutical industry since co-crystallization can improve unfavorable physicochemical properties of active pharmaceutical ingredients. Thus, the quest for new co-crystal screening methods has become an issue of importance. The aim of this work was, therefore, to show to what extent expanded methodology based on FTIR and Raman spectroscopy supported by the DSC method can be used as a reliable tool to

[Examination of warm and cold ischemic injury of small intestine by differential scanning calorimetry].

INTRODUCTION/AIM: Small bowel is extremely sensitive to ischemia/reperfusion injury. Therefore, we compared the effect of warm and cold ischemia on the intestinal structural changes by differential scanning calorimetry (DSC) method. MATERIALS AND METHODS: Warm and cold ischemia groups were established on Wistar rats with 1, 3 and 6 h ischemic times (n = 30). Intestinal biopsies were collected after laparotomy and at the end of the ischemia periods. DSC measurement was performed on the mucosa, muscular layer and total intestinal wall. RESULTS: Our DSC data confirmed that longer warm ischemia

Solvent-Based Separation of PA6 and PET Using Solubility Data from Differential Scanning Calorimetry.

To minimize plastic pollution and prevent further extraction of fossil feedstock, the circular economy for polymers offers a promising solution. As an integral part of the circular economy to close material loops, solvent-based recycling enables the separation of mixed plastic waste into pure component streams without altering the polymer structure but requires reliable solubility data of polymers in solvents. We present a standardized differential scanning calorimetry (DSC) protocol to determine decrystallization and precipitation temperatures of polymers in various solvents and apply it to t

pH-Dependent Stability and Cooperativity of Protein Unfolding from Differential Scanning Calorimetry Using FitFoldData.

Protein folding is highly sensitive to environmental conditions such as pH, which can influence internal hydrogen bonding and interactions with the solvent. In this study, we use our FitFoldData online tool to analyze published differential scanning calorimetry (DSC) data on the unfolding of four proteins measured across different pH values. For each dataset, we examine the fitted thermodynamic parameters, including the intrapeptide and peptide-solvent hydrogen-bonding energies (h and hps) and the Zimm-Bragg cooperativity parameter (σ). In the analyzed datasets, under both acidic and alkaline

Differential Scanning Calorimetry (DSC) in Solid-State Characterization of Solid Dispersions: A Practical Guide.

Differential Scanning Calorimetry (DSC) is one of the most widely employed and rapid thermal analysis techniques for early-stage solid-state characterization, particularly in the development of solid dispersions. As an indirect method, DSC detects transitions in solid-state properties through heat flow measurements, providing valuable insights into crystallinity, polymorphism, and miscibility. However, the accuracy and interpretation of DSC thermograms are highly sensitive to various experimental parameters, including heating and cooling rates, sample preparation, moisture content, and the the

Torn human rotator cuff tendons have reduced collagen thermal properties on differential scanning calorimetry.

The cause of the high failure rates often observed following rotator cuff tendon repairs, particularly massive tears, is not fully understood. Collagen structural changes have been shown to alter tendon thermal and mechanical properties. This study aimed to form a quantitative rather than qualitative assessment, of whether differences in collagen structure and integrity existed between small biopsies of normal, small, and massive rotator cuff tears using differential scanning calorimetry. Thermal properties were measured for 28 human biopsies taken intra-operatively from normal, small, and mas

Direct analysis for cholesterol in gallstones by use of differential scanning calorimetry transition phase.

A cholesterol transition phase at 37.5 degrees C has been shown by use of a Differential Scanning Calorimeter. The transition, reversible and reproducible, is useful for analytical purpose. We propose an analytical method for cholesterol in gallstones based on this transition. The method requires no intermediate reactions and cholesterol can be assayed directly in the powdered stone, with no other pretreatment or reagents. The analysis is highly specific, requires about 10 min, and is quite precise (CV about 1.5%) and accurate (about 2%).

Characterization of amorphous solids with weak glass transitions using high ramp rate differential scanning calorimetry.

Measurement of the glass transition temperature (T(g)) of proteins and other high molecular weight polymers in the amorphous state is often difficult, since the transition is extremely weak, that is, the DeltaC(p) at the glass transition temperature is small. For example, little is known about the solid-state properties of hydroxyethyl starch (HES), which is beginning to become more commonly evaluated as a bulking agent in pharmaceutical products. For weak thermal events, such as the change in heat capacity at the T(g) of a pure protein or large synthetic polymer, increased heating rate should

Preliminary use of differential scanning calorimetry of cerebrospinal fluid for the diagnosis of glioblastoma multiforme.

Thermal stability signatures of complex molecule interaction in biological fluids can be measured using a new approach called differential scanning calorimetry (DSC). The thermal stability of plasma proteome has been described previously as a method of producing a disease-specific "signature," termed thermogram, in several neoplastic and autoimmune diseases. We describe the preliminary use of DSC performed on cerebrospinal fluid (CSF) as a diagnostic tool for the identification of patients with glioblastoma multiforme (GBM). Samples of CSF from nine patients with confirmed GBM were evaluated u

A Simultaneous Differential Scanning Calorimetry-X-ray Diffraction Study of Olanzapine Crystallization from Amorphous Solid Dispersions.

Amorphous solid dispersions (ASDs) of class II and IV biopharmaceutics classification system drugs in water-miscible polymers are a well-recognized means of enhancing dissolution, while such dispersions in hydrophobic polymers form the basis of micro- and nanoparticulate technologies. However, drug recrystallization presents significant problems for product development, and the mechanisms and pathways involved are poorly understood. Here, we outline the use of combined differential scanning calorimetry (DSC)-synchrotron X-ray diffraction to monitor the sequential appearance of polymorphs of ol
