Separation of tetracyclines by high-performance capillary electrophoresis and capillary electrochromatography.

Separations of various tetracycline mixtures by high-performance capillary electrophoresis (HPCE) and a new form of electrochromatography (CEC) are compared. The new CEC method involves etching the inner wall of the capillary surface with an appropriate reagent (ammonium dihydrogen fluoride) in order to produce a significant increase in surface area. The etched surface is then modified by a silation/hydrosilation reaction sequence to first produce a hydride intermediate which is then further reacted to attach a C18 moiety. The bare and hydride capillaries are tested under HPCE conditions while

A new open-tubular approach to capillary electrochromatography.

While most capillary electrochromatography is based on the use of packing materials similar to those for HPLC, this method creates a new surface on the inner wall of the fused-silica tubing that is then modified with various organic moieties. In this format there is no need for frits, and bubble formation is not a serious problem. With 50-microns-i.d. capillaries, molecules are separated by a combination of differences in electrophoretic mobility and solute/bonded phase interactions. A number of examples are presented, which include tetracyclines, peptides, and proteins. Peaks for all compound

Chiral Separations by Using Capillary Electrochromatography: An Overview.

Capillary electrochromatography is a miniaturized technique offering some advantages over conventional ones in separation science. It combines the best features of capillary zone electrophoresis (high efficiency of separation) and liquid chromatography (high selectivity). Because of the low flow rates, minute volumes of mobile phases can be applied, and therefore, it can be considered a green analytical technique. Capillary electrochromatography has been successfully used to separate many classes of compounds, including peptides, proteins, drugs, antibiotics, pollutants, enantiomers, etc. In t

Capillary electrochromatography of hydrophobic amines on continuous beds.

The capillary electrochromatographic separation performance of hydrophobic amines and a related quaternary ammonium compound on continuous beds based on polymers of acrylamide has been studied. The chromatographic bed is polymerized in situ and the character of the polymers with regard to hydrophobicity and charge has been systematically changed by regulating its content of isopropyl and sulfonate ligands, respectively. The best performance was obtained for columns with a molar ratio of 1:80 for the sulfonate and isopropyl groups, and resulted in efficiencies up to 200000 plates per meter. The

Applications of capillary electrochromatography analysis for formulated pesticide products.

The feasibility of using capillary electrochromatography (CEC) as a high-efficiency reversed-phase separation technique has been demonstrated for the analysis of some pesticide formulation products. Some operating parameters of CEC analysis (organic modifier content, pH of the buffer, and sample diluent) were studied using commercially available capillaries packed with Hypersil (Phenomenex, Torrance, CA, U.S.A.) octadecylsilic (ODS) particles. It was found that the resolution decreases in linear fashion with the increase in percent acetonitrile in the sample diluent for neutral components if a

Open-tubular capillary electrochromatography using a capillary coated with octadecylamine-capped gold nanoparticles.

Octadecylamine-capped gold nanoparticles (ODA-Au-NPs) were prepared and characterized by using UV-Vis adsorption spectrum, transmission electron chromatography (TEM), SEM, and FT-IR. A simple but robust hydrophobic coating was easily developed by flushing a capillary with a solution of ODA-Au-NPs, because the positive charges were carried by the nanoparticles which strongly adsorb to the negatively charged inner surface of a fused-silica capillary via electrostatic and hydrophobic interactions. The chromatographic characteristics of the coated capillary was investigated by varying the experime

S-citalopram imprinted monolithic columns for capillary electrochromatography enantioseparations.

In this study, the molecular imprinting method was used to separate enantiomeric forms of chiral antidepressant drug, R,S-citalopram (R,S-CIT) in aqueous solution by CEC system combining the advantages of capillary electrophoresis (CE) and high-performance liquid chromatography (HPLC). For that, an amino acid-based molecularly imprinted monolithic capillary column was designed and used as a stationary phase for selective separation of S-citalopram (S-CIT) for the first time. S-CIT was selectively separated from the aqueous solution containing the other enantiomeric form of R-CIT, which is the

Electrochromatography in chemically modified etched fused-silica capillaries.

A new method of electrochromatography is described in which a 50-microns capillary is etched with ammonium hydrogen difluoride, followed by modification of the new surface via a silation reaction with triethoxysilane to produce a hydride intermediate, and then subsequently subjected to hydrosilation using 1-octadecene in the presence of a platinum complex catalyst. The C18 bonded phase is then compared with bare capillaries, etched bare capillaries and the hydride etched capillary to determine if any solute-bonded phase interactions are present. With bradykinin as a test solute, peak efficienc

A drying step in the protocol to pack capillary columns by centripetal forces for capillary electrochromatography.

Capillary columns have been packed for capillary electrochromatography (CEC) using centripetal forces. The packed columns were maintained under wet conditions or they were dried with nitrogen gas prior to forming the retaining frits. Upon fabrication of the retaining frits, the dried columns were resolvated with the mobile phase. The performance of the columns was evaluated to determine the effect of the drying step during the packing procedure. The columns submitted to the drying step showed improved separation efficiencies and stronger retention characteristics than those kept under wet cond

Enantiomer separation by capillary electrochromatography on a cyclodextrin-modified monolith.

A chiral monolithic stationary phase was prepared by packing a capillary with bare porous silica and sintering the silica bed at high temperature. The resulting silica monolith was polymer-coated with Chirasil-Dex, a permethylated beta-cyclodextrin covalently linked via an octamethylene spacer to dimethylpolysiloxane. Subsequently, Chirasil-Dex was thermally immobilized on the silica support and a chiral monolith of very high stability (30 kV, more than 400 bar pressure) was obtained. Fused-silica capillary columns were packed with ethylene chlorotrifluoroethylene (ECTFE) particles for use in capillary electrochromatography (CEC). Electroosmotic flow (EOF) was generated in these columns using acetonitrile-water mixtures as the mobile phase. Electroosmotic mobilities of 1.6 x 10(-4) cm2 V(-1) s(-1) (linear velocities of 1 mm s(-1)) were observed using a mobile phase without an electrolyte present. Dynamics of capillary electrochromatography experimental study on the electrosmotic flow and conductance in open and packed capillaries.

For capillary electrochromatography (CEC) to become an analytical separation technique of high speed and resolution the factors determining the conductivity of the column as well as the generation and control of electrosmotic flow (EOF) in porous media have to be understood. In the present study the conductance of capillaries packed with a variety of stationary phases was evaluated with respect to the conductance of the open capillary and the data were interpreted in the light of the Tobias equation. However, the consistently observed reduction of the EOF when a capillary having a charged inne

Novel Separation Media with Metal Oxide Nanostructures for Capillary Electrochromatography.

Zinc oxide nanowires (ZnO nanowire, ZnO NWs) are nanostructures that have drawn attention as separation media for efficient biomolecules because of high biological compatibility and low cost. Development of the capillary column (ZnO column) using a ZnO NW to an inner wall has been reported, although there are only a few studies about molecular recognition of a ZnO NW regardless of numerous studies reporting ZnO NWs. In our previous studies, we conducted fundamental research to elucidate molecular recognition of ZnO NW and develop a novel liquid phase separation field. Consequently, we achieved

Improved quantification limits in chiral capillary electrochromatography by peak compression effects.

The peak compression effect has been applied to improve quantification limits in chiral capillary electrochromatography (CEC). A stationary phase based on the chiral selector vancomycin (Chirobiotic V) was used for separations of the enantiomers of mianserin. By adding solvents with a low dielectric constant, e.g. 2-propanol or tetrahydrofuran, to the sample solution, peak compression could be induced. The plate numbers for the minor enantiomer increased from approximately 100,000 to 1.4-1.6 million plates/m, when the composition of the mobile phase was adjusted so that the analyte eluted with
