Inhibition of mitochondrial bioenergetics by carbaryl is only evident for higher concentrations -- Relevance for carbaryl toxicity mechanisms.

Although pesticides have been useful in agriculture pest control, there is a considerable risk for human health and damage to ecosystems. Carbaryl is a carbamate often taken as a safe insecticide, although data on metabolic activities is still scarce, viz. mitochondrial toxicity. Therefore, it is the goal of this work to assay the compound on isolated mitochondria, a biochemical model already used with other pesticides. Mitochondria isolated from the livers of Wistar rats were assayed for bioenergetic parameters, namely mitochondrial respiration, membrane potential, membrane integrity and enzy

Pralidoxime in carbaryl poisoning: an animal model.

INTRODUCTION: Poisoning from organophosphates and carbamates is a significant cause of morbidity and mortality worldwide. Concerns have been expressed over the safety and efficacy of the use of oximes such as pralidoxime (2-PAM) in patients with carbamate poisoning in general, and more so with carbaryl poisoning specifically. The goal of the present study was to evaluate the role of 2-PAM in a mouse model of lethal carbaryl poisoning. METHODS: Female ICR Swiss Albino mice weighing 25-30 g were acclimated to the laboratory and housed in standard conditions. One hundred and ten mice received an

The effect of ozonation on the degradation of carbaryl in aqueous solution.

The aim of this study is to investigate the synergetic effect of ozonation on ultraviolet irradiation on the degradation of carbaryl (CBL) (1-naphthyl-N-methylcarbamate) in aqueous solutions (40 mg L-1). The degradation of CBL and the intermediates formed during ozonation were determined by gas chromatography-mass spectrometer. The change in formaldehyde, dissolved oxygen, pH, and total acidity were determined as a function of ozonation time. It was found that the dissolved oxygen and total acidity were increased with the increasing ozonation time but the pH was decreased with the increasing o

[Analysis of carbaryl and other pesticides by capillary electrophoresis].

Five pesticides, i.e. carbaryl, simazine, atrazine, arnetryne and prometryn were separated by capillary electrophoresis technique. The effects of experimental parameters including pH, the concentration of surfactant and organic modifier on the separation were investigated. From the experiments, we found the optimum electrophoretic conditions were as follows: 20 mmol/L borate-50 mmol/L sodium dodecyl sulfate-10% methanol (volume fraction) as the running buffer (pH 9.2), applied voltage of 25 kV, detection wavelength of 225 nm. Under the optimum conditions, baseline-separation of each component

Effects of carbaryl on some dopaminergic behaviors in rats.

1. The effects of acute oral administration of carbaryl (10-80 mg/kg), a carbamate insecticide, on some experimental models for detecting dopaminergic activity were examined in rats. Also, serum biochemical variables following carbaryl treatments were determined. 2. Carbaryl (20 and 40 mg/kg) significantly increased the number of apomorphine-induced yawns and at dose of 80 mg/kg it prolonged the duration time of haloperidol-induced catalepsy. Pretreatment with carbaryl failed to affect apomorphine-induced stereotypes. 3. Carbaryl significantly reduced blood cholinesterase activity and elevated

A teratology study of carbaryl and malathion mixtures in rat.

Recently, much controversy has developed in relation to the use of pesticides alone and in mixtures. Mixtures are used as a more efficient method of pest control. Carbaryl and malathion are one of many combinations used today. A limited amount of work has been done with these two pesticides in relation to their teratogenic potential. This study examined the teratological parameters of carbaryl and malathion in combination compared to the effect of the individual insecticides. Formulation grade carbaryl (0, 1, 10, and 100 mg/kg), formulation grade malathion (1 and 50 mg/kg), and a formulation g

Subacute neurotoxicity following long-term exposure to carbaryl.

Carbaryl, a widely used insecticide, is reputed to have a wide safety margin. It can induce acute cholinesterase poisoning, which is rapidly reversible on discontinuation of exposure. Long-term sequelae from long-term exposure have not previously been described in humans. This report describes the experience of a 75-year-old man who had long-term excessive exposure to carbaryl and in whom a debilitating syndrome, including headaches, memory loss, proximal muscle weakness, muscle fasciculation, muscle cramps, and anorexia with marked weight loss, developed. At the time of diagnosis, serum pseud

Tumorigenic potential of carbaryl in the heterozygous p53 knockout mouse model.

The heterozygous p53 knockout mouse model was used to assess whether vascular tumors noted in a 2-year carcinogenicity study in CD-1 mice with carbaryl were induced through a genotoxic mechanism. This knockout mouse model was selected for carbaryl because of the high sensitivity of this model to genotoxic events and its low spontaneous incidence of tumors until 9-12 months of age. Carbaryl was administered continuously via the diet to groups of 20 male heterozygous p53 knockout mice at concentrations of 0, 10, 30, 100, 300, 1000 and 4000 ppm for 180 days. Carbaryl exposure and incident cancer in the Agricultural Health Study.

Carbaryl is a carbamate insecticide with a broad spectrum of uses in agricultural, commercial and household settings. It has previously been linked with non-Hodgkin lymphoma (NHL) but studies of cancer risk in humans are limited. We examined occupational carbaryl use and risk of all cancers in the Agricultural Health Study, a prospective study of a cohort of pesticide applicators in North Carolina and Iowa. This analysis included 21,416 subjects (1,291 cases) enrolled from 1993-1997 and followed for cancer incidence through 2003. Pesticide exposure and other data were collected using self-admi

Diurnal variation in thermoregulatory response to chlorpyrifos and carbaryl in the rat.

Time of day of exposure is rarely considered in the study of insecticide toxicology. It would be expected that the circadian temperature rhythm (CTR) as well as the circadian rhythms of other physiological processes would affect the efficacy of anticholinesterase (antiChE) insecticides. The ability of antiChE insecticides to alter core temperature (T(c)) could be affected by time of exposure in relation to the CTR. To this end, we assessed time of exposure on the efficacy of the antiChE insecticides chlorpyrifos (CHP) and carbaryl (CAR) to alter T(c) in the rat. T(c) and motor activity (MA) we

Improved photocatalytic decomposition of carbaryl pesticide in wastewater using ZnO nanorods.

This study explores the enhanced photocatalytic performance of ZnO nanorods (ZnO-R) for degrading the carbaryl pesticide (CB) in wastewater. For comparison, commercial ZnO (ZnO-C) was used to evaluate the differences in the photocatalytic decomposition of CB between ZnO-R and ZnO-C. The results regarding the material properties demonstrated that ZnO-R enhances CB removal performance due to its unique rod shape, which extends light absorption and improves electron-hole separation. The removal rates of the carbaryl pesticide from the aqueous solution using ZnO-R and ZnO-C were 98.2% and 87.3%, r

Carbaryl use and incident cancer in the Agricultural Health Study: an updated analysis.

Carbaryl is a common carbamate insecticide in the United States (USA). Previous epidemiologic investigations, including within the Agricultural Health Study (AHS), have suggested potential associations between carbaryl use and cancer risk. The AHS is a prospective cohort study of licensed pesticide applicators in North Carolina (NC) and Iowa (IA), USA. Information on lifetime pesticide use was reported at enrollment (1993-1997) and follow-up (1999-2005). We evaluated cancer risks associated with ever- and intensity-weighted lifetime days (IWLD) of carbaryl use. Among 52,625 applicators, 8713 i

Developmental neurotoxic effects of two pesticides: Behavior and biomolecular studies on chlorpyrifos and carbaryl.

In recent times, an increased occurrence of neurodevelopmental disorders, such as neurodevelopmental delays and cognitive abnormalities has been recognized. Exposure to pesticides has been suspected to be a possible cause of these disorders, as these compounds target the nervous system of pests. Due to the similarities of brain development and composition, these pesticides may also be neurotoxic to humans. We studied two different pesticides, chlorpyrifos and carbaryl, which specifically inhibit acetylcholinesterase (AChE) in the nervous system. The aim of the study was to investigate if the p

The nature of Cl- secretion, induced by carbaryl, across the isolated skin of Rana esculenta.

1. The pesticide carbaryl induces Cl- secretion through the isolated frog skin. 2. This effect is due to the activation of both processes responsible for this phenomenon: (a) Na+/K+/2Cl- cotransport on the serosal membrane; (b) Cl- selective channels on the external membrane. 3. Cl- outflux is inhibited by bumetanide (10(-5) M) on the serosal side and by diphenylamine-2-carboxylic acid (DPC) (10(-3) M) on the external side. 4. The DPC action is not mimicked by Naproxen, a specific inhibitor of cyclooxygenase. 5. A comparison with isoprenaline, demonstrates that the carbaryl action is, paradoxi

Effects of atrazine, metolachlor, carbaryl and chlorothalonil on benthic microbes and their nutrient dynamics.

Atrazine, metolachlor, carbaryl, and chlorothalonil are detected in streams throughout the U.S. at concentrations that may have adverse effects on benthic microbes. Sediment samples were exposed to these pesticides to quantify responses of ammonium, nitrate, and phosphate uptake by the benthic microbial community. Control uptake rates of sediments had net remineralization of nitrate (-1.58 NO3 µg gdm⁻¹ h⁻¹), and net assimilation of phosphate (1.34 PO4 µg gdm⁻¹ h⁻¹) and ammonium (0.03 NH4 µg gdm⁻¹ h⁻¹). Metolachlor decreased ammonium and phosphate uptake. Chlorothalonil decreased nitrate remine

High-performance liquid chromatographic determination of carbaryl and 1-naphthol in biological fluids.

A simple and sensitive method for the simultaneous analysis of carbaryl and 1-naphthol in whole blood by reversed-phase high-performance liquid chromatography and fluorescence detection is described. Spiked blood (heparinized) containing an internal standard was hemolyzed and extracted with ethyl acetate. After centrifugation the extractant was removed and taken to dryness. Reconstitution and subsequent high-performance liquid chromatography-fluorescence analysis yielded linear standard curves for carbaryl and 1-naphthol. Linear response vs. concentration profiles were obtained for carbaryl an
