A metal-organic framework-derived ruthenium-nitrogen-carbon nanozyme for versatile hydrogen sulfide and cystathionine γ-lyase activity assay.

In this study, a novel approach exploiting the interactions between hydrogen sulfide (H2S) and ruthenium-nitrogen-carbon (Ru-N-C) nanozymes is presented, advancing H2S and cystathionine γ-lyase (CSE) biosensing techniques. Utilizing the intrinsic peroxidase-like activity of Ru-N-C nanozymes and the noticeable inhibition effect caused by H2S, an efficient, simple, and economical assay has been developed. This innovative method allows for the versatile real-time monitoring of H2S from various sources, including specialized donors and native bacterial production. Furthermore, the assay has been a

Phosphonate utilization by the globally important marine diazotroph Trichodesmium.

The factors that control the growth and nitrogen fixation rates of marine diazotrophs such as Trichodesmium have been intensively studied because of the role that these processes have in the global cycling of carbon and nitrogen, and in the sequestration of carbon to the deep sea. Because the phosphate concentrations of many ocean gyres are low, the bioavailability of the larger, chemically heterogeneous pool of dissolved organic phosphorus could markedly influence Trichodesmium physiology. Here we describe the induction, by phosphorus stress, of genes from the Trichodesmium erythraeum IMS101

In Vitro Reconstitution of a Five-Step Pathway for Bacterial Ergothioneine Catabolism.

Ergothioneine is a histidine-derived sulfur metabolite that is biosynthesized by bacteria and fungi. Plants and animals absorb ergothioneine as a micronutrient from their environment or nutrition. Several different mechanisms of microbial ergothioneine production have been described in the past ten years. Much less is known about the genetic and structural basis for ergothioneine catabolism. In this report, we describe the in vitro reconstitution of a five-step pathway that degrades ergothioneine to l-glutamate, trimethylamine, hydrogen sulfide, carbon dioxide, and ammonia. The first two steps

Detoxifying Cyanides Using Cyanase Enzyme Complexes Composed of Carbonic Anhydrase via Irreversible Covalent Bonds.

Cyanase is a possible solution to reduce the environmental impact of cyanide. However, the enzyme's dependence on HCO3- limits its industrial applications. To overcome this problem, carbonic anhydrase is utilized in this study. Three types of Catcher/Tag systems were introduced into the cyanase (psCYN) from Pseudomonas stutzeri and the carbonic anhydrase (hmCA) from Hydrogenovibrio marinus to construct enzyme complexes via irreversible covalent bonds. Initially, a cyanase complex with the aid of scaffolding proteins was designed. The results of cyanase complexes using scaffolding proteins were

Strictosidine synthase, an indispensable enzyme involved in the biosynthesis of terpenoid indole and β-carboline alkaloids.

Terpenoid indole (TIAs) and β-carboline alkaloids (BCAs), such as suppressant reserpine, vasodilatory yohimbine, and antimalarial quinine, are natural compounds derived from strictosidine. These compounds can exert powerful pharmacological effects but be obtained from limited source in nature. the whole biosynthetic pathway of TIAs and BCAs, The Pictet-Spengler reaction catalyzed by strictosidine synthase (STR; EC: 4.3.3.2) is the rate-limiting step. Therefore, it is necessary to investigate their biosynthesis pathways, especially the role of STR, and related findings will support the biosynth

A Key Metabolic Protein in Active Mycobacterium tuberculosis: Insights into Carbon, Nitrogen, and Sulfur Metabolism.

Mycobacterium tuberculosis (Mtb) employs a highly adaptable network of metabolic pathways that are pivotal for its survival and pathogenesis within the host during both exponential growth and persistent infection phases. Central carbon metabolism in Mtb exhibits remarkable flexibility, enabling the bacterium to utilize diverse carbon sources efficiently. In the absence of glucose, Mtb preferentially metabolizes fatty acids as primary carbon substrates. This metabolic shift is supported by the glyoxylate shunt and methyl citrate cycle, which replenish tricarboxylic acid (TCA) cycle intermediate

Ethylene Affects Soil Multifunctionality in Maize Rhizosphere by Driving Sensitive Microbial Communities.

Ethylene modulates plant fitness, but its role in rhizosphere soil multifunctionality remains unclear. This study demonstrates that endogenous ethylene suppresses C- and N-cycling functions while promoting organic P mineralization in the maize rhizosphere, ultimately diminishing the overall soil multifunctionality and attenuating its phenologically driven peak at the tasseling stage. Ethylene reshaped microbial community assembly by enriching opportunistic taxa (e.g., Actinobacteria) while reducing the diversity and relative abundance of sensitive taxa. Community composition (e.g., Bacillus) a

Balancing electron competition in nitrogen oxide reduction via membrane-aerated electroactive biofilms for electron flow regulation and N2O mitigation.

Nitrogen removal in bioelectrochemical systems relies on electrons derived from anodic ammonia oxidation and their utilization for nitrogen oxides reduction at the cathode. However, incomplete anodic oxidation often results in electron competition between NO2- and N2O reduction, even under an applied potential difference. Here, a counter-diffusion electroactive biofilm was constructed to synergistically regulate intracellular carbon metabolism and extracellular electron allocation. Specifically, the electric field activated the microbial glyoxylate shunt (GS) by suppressing isocitrate dehydrog

Biodegradation of the aminopolyphosphonate DTPMP by the cyanobacterium Anabaena variabilis proceeds via a C-P lyase-independent pathway.

Cyanobacteria, the only prokaryotes capable of oxygenic photosynthesis, play a major role in carbon, nitrogen and phosphorus global cycling. Under conditions of increased P availability and nutrient loading, some cyanobacteria are capable of blooming, rapidly multiplying and possibly altering the ecological structure of the ecosystem. Because of their ability of using non-conventional P sources, these microalgae can be used for bioremediation purposes. Under this perspective, the metabolization of the polyphosphonate diethylenetriaminepenta(methylenephosphonic) acid (DTPMP) by the strain CCALA

Engineering stable and reusable biocatalysts through electrostatic immobilization of (S)-norcoclaurine synthase on mesostructured kaolin.

Immobilization of enzymes onto solid supports is a key strategy for enhancing their stability, reusability, and performance under harsh conditions, thereby enabling broader applications in sustainable catalysis. Here, we investigate the immobilization of (S)-Norcoclaurine Synthase ((S)-NCS), a highly promising yet scarcely studied biocatalyst capable of catalyzing the Pictet-Spengler reaction with high enantioselectivity for the synthesis of tetrahydroisoquinolines. We explored its immobilization onto low-cost mesoporous materials, Kaolin and Celite 545, characterized by distinct surface prope

Comparative genomics of ethanolamine utilization.

Ethanolamine can be used as a source of carbon and nitrogen by phylogenetically diverse bacteria. Ethanolamine-ammonia lyase, the enzyme that breaks ethanolamine into acetaldehyde and ammonia, is encoded by the gene tandem eutBC. Despite extensive studies of ethanolamine utilization in Salmonella enterica serovar Typhimurium, much remains to be learned about EutBC structure and catalytic mechanism, about the evolutionary origin of ethanolamine utilization, and about regulatory links between the metabolism of ethanolamine itself and the ethanolamine-ammonia lyase cofactor adenosylcobalamin. The genes and enzymes of phosphonate metabolism by bacteria, and their distribution in the marine environment.

Phosphonates are compounds that contain the chemically stable carbon-phosphorus (C-P) bond. They are widely distributed amongst more primitive life forms including many marine invertebrates and constitute a significant component of the dissolved organic phosphorus reservoir in the oceans. Virtually all biogenic C-P compounds are synthesized by a pathway in which the key step is the intramolecular rearrangement of phosphoenolpyruvate to phosphonopyruvate. However C-P bond cleavage by degradative microorganisms is catalyzed by a number of enzymes - C-P lyases, C-P hydrolases, and others of as-ye

Sulfadiazine degrading bacteria Vibrio alginolyticus isolated from mariculture wastewater: Performance, degradation pathways, and mechanisms based on transcriptomic analysis.

In this study, two strains of novel sulfadiazine (SDZ)-degrading bacteria Vibrio alginolyticus SUL-3 and SUL-5 were isolated from the SDZ contaminated mariculture wastewater. The degradation rate of two strains reached 54.11% ± 0.15% and 63.57% ±0.07% of 1 mg/L SDZ within 4 d, respectively, and could also degrade other 3 sulfonamides (sulfamethoxazole, sulfamonomethoxine, and sulfamethazine). Optimal degradation conditions were identified as: neutral pH (7.0), marine salinity (35‰), and temperature of 30 °C. The addition of exogenous carbon and nitrogen sources was found to enhance bacterial d

An extragenic fix for a broken enzyme.

In this work, we show how exploration of genomic sequence space yields an unexpected evolutionary 'fix' that allows the cell to bypass the phenotypic defects associated with a dysfunctional enzyme. We identified a transposon (Tn) insertion in the gene (argA) encoding N-acetylglutamate synthase (NAGS) that leads to arginine auxotrophy in Pseudomonas aeruginosa. This Tn insertion generates a truncated version of the ArgA protein (ArgA188-432) that retains NAGS activity but, unlike the wild-type protein, is unrestrained by arginine feedback inhibition. However, the Tn::argA mutant readily re-acqu

Enhanced biological nitrogen fixation and nodulation in alfalfa through the synergistic interactions between Sinorhizobium meliloti and Priestia aryabhattai.

Nitrogen fertilizer is crucial for agricultural output. However, prolonged overuse has resulted in nitrate leaching and potential soil acidification. Research on microbial fertilizer has become essential to enhance soil conditions and minimize nitrogen fertilizer usage. In alfalfa cultivation, research on efficient compound microbial agents remains limited, therefore this study concentrates on the investigation of dual microbial combinations. In the screening process, black soil was utilized with alfalfa plants as samples to identify a strain of rhizobacteria, Sinorhizobium meliloti LMGL3-1, e

Optimizing dark fermentation for hydrogen production: lessons from Thermoactinomyces mirandus.

Biological hydrogen (H2) production via dark fermentation (DF) offers a renewable pathway for energy recovery from biomass, but process performance is highly strain dependent and often limited by suboptimal operating conditions. The present study characterizes fermentation physiology and optimizes small-scale H2 production by the recently discovered thermophilic anaerobe Thermoactinomyces mirandus. Batch cultivations at 52 °C in anoxic serum bottles were used to screen carbon sources (glucose, fructose, xylose, arabinose, and lactose), nitrogen sources (yeast extract, casein peptone, casamino

Characterization of toluene and ethylbenzene biodegradation under nitrate-, iron(III)- and manganese(IV)-reducing conditions by compound-specific isotope analysis.

Ethylbenzene and toluene degradation under nitrate-, Mn(IV)-, or Fe(III)-reducing conditions was investigated by compound specific stable isotope analysis (CSIA) using three model cultures (Aromatoleum aromaticum EbN1, Georgfuchsia toluolica G5G6, and a Azoarcus-dominated mixed culture). Systematically lower isotope enrichment factors for carbon and hydrogen were observed for particulate Mn(IV). The increasing diffusion distances of toluene or ethylbenzene to the solid Mn(IV) most likely caused limited bioavailability and hence resulted in the observed masking effect. The data suggests further

Acinetobacter baumannii Catabolizes Ethanolamine in the Absence of a Metabolosome and Converts Cobinamide into Adenosylated Cobamides.

Acinetobacter baumannii is an opportunistic pathogen typically associated with hospital-acquired infections. Our understanding of the metabolism and physiology of A. baumannii is limited. Here, we report that A. baumannii uses ethanolamine (EA) as the sole source of nitrogen and can use this aminoalcohol as a source of carbon and energy if the expression of the eutBC genes encoding ethanolamine ammonia-lyase (EAL) is increased. A strain with an ISAba1 element upstream of the eutBC genes efficiently used EA as a carbon and energy source. The A. baumannii EAL (AbEAL) enzyme supported the growth

A review on remediation of cyanide containing industrial wastes using biological systems with special reference to enzymatic degradation.

Cyanide is a nitrile which is used extensively in many industries like jewelry, mining, electroplating, plastics, dyes, paints, pharmaceuticals, food processing, and coal coking. Cyanides pose a serious health hazard due to their high affinity towards metals and cause malfunction of cellular respiration by inhibition of cytochrome c oxidase. This inhibition ultimately leads to histotoxic hypoxia, increased acidosis, reduced the functioning of the central nervous system and myocardial activity. Different physicochemical processes including oxidation by hydrogen peroxide, alkaline chlorination,
