Dedioxygenative Phosphonylation of Carboxylic Acids.

Dedioxygenative transformations of carboxylic acids are faced with the significant challenges in efficiently breaking multiple C-O bonds and achieving broad applicability across diverse carboxylic acids, thereby hampering the development and application of this field. In this work, through the cooperation of multiple catalysts, we realized a dedioxygenative phosphonylation of carboxylic acids that is applicable to both aliphatic and aromatic substrates, showing good scope and functional-group tolerance. This protocol also demonstrates promising performance in the late-stage modification of com

Alpha-hydroxyacids and carboxylic acids.

The carboxylic acids include alpha-hydroxyacids (AHAs), polyhydroxy acids (PHAs), aldobionic acids (ABAs), retinoic acid, vitamin C and azelaic acid. They all have therapeutic actions. AHAs, PHAs and ABAs are organic hydroxyacids, a group of natural and physiological substances which can modulate skin keratinization and increase biosynthesis of dermal components. Because of these effects, AHAs, PHAs and ABAs are therapeutically effective or beneficial for topical treatment of dry skin, rough skin, acne, rosacea, warts, eczema, psoriasis and skin changes associated with ageing, including wrinkl

Cell-free reduction of carboxylic acids with secreted carboxylic acid reductase.

Mycobacterium marinum CAR (MmCAR) is one of the most widely used CARs as the key enzyme for the synthesis of aldehydes, alcohols and further products from the respective carboxylic acids. Herein, we describe the first functionally secreted 131 kDa CAR and its isolated A-domain using Komagataella phaffii and a methanol-free constitutive expression strategy. Precipitated and lyophilized MmCAR (500 µg) was isolated from the culture supernatant and showed no decrease in activity for piperonylic acid (80% conversion), even when stored for up to 3 weeks at 4°C. Lyophilized MmCAR precipitate gave 48%

Ready BODIPY-Tagging of Carboxylic Acids.

Meso-aminoBODIPY and four functionalized analogues are presented as stable and efficient starting materials for the tagging of carboxylic acids via the classical Steglich amidation protocol (DCC, DMAP) at room temperature. Using this methodology, 18 new derivatives were synthesized in moderate-to-high yields (30-86%) from unsubstituted meso-aminoBODIPY, along with four additional analogues obtained from functionalized amino derivatives in yields ranging from 50 to 77%. This approach displayed excellent tolerance toward the functional groups present in the starting carboxylic acids. Photophysic

Photocatalytic hydrofluoroalkylation of alkenes with carboxylic acids.

Incorporation of fluoroalkyl motifs in pharmaceuticals can enhance the therapeutic profiles of the parent molecules. The hydrofluoroalkylation of alkenes has emerged as a promising route to diverse fluoroalkylated compounds; however, current methods require superstoichiometric oxidants, expensive/oxidative fluoroalkylating reagents and precious metals, and often exhibit limited scope, making a universal protocol that addresses these limitations highly desirable. Here we report the hydrofluoroalkylation of alkenes with cheap, abundant and available fluoroalkyl carboxylic acids as the sole reage

Photocaging of carboxylic acids: a modular approach.

Photocaged compounds are important tools for studying and regulating multiple processes, including biological functions. Reported herein is the use of the Passerini multicomponent reaction for modular preparation of photocaged carboxylic acids. The reaction is compatible with several functionalities and proceeds smoothly both in water and dichloromethane. The choice of aldehyde determines the wavelength used for deprotection and enables formation of orthogonally protected products. The isocyanide component can be used for introduction of reactive tags and photosensitizers, as well as for immob

Indolyl carboxylic acids by condensation of indoles with alpha-keto acids.

The novel indole derivatives 2,2-bis(3,3'-indolyl)propionic acid (1); 1,1,1,-tris(3,3',3"-indolyl)ethane (2); and 2,2-bis(3, 3'-indolyl)isocaproic acid (3) were isolated from solvent extracts of indole-supplemented supernatants of Escherichia coli and corynebacteria. The compounds were also obtained by chemical synthesis: compounds 1 and 2 from indole and pyruvic acid and compound 3 from indole and alpha-ketoisocaproic acid, following incubation at 37 degrees C in aqueous medium. Tryptophan and pyruvic acid gave the novel 2-(2-tryptophanyl)lactic acid (4). The condensation reaction between ind

Photochemical phosphorus-enabled scaffold remodeling of carboxylic acids.

The excitation of carbonyl compounds by light to generate radical intermediates has historically been restricted to ketones and aldehydes; carboxylic acids have been overlooked because of high energy requirements and low quantum efficiency. A successful activation strategy would necessitate a bathochromic shift in the absorbance profile, an increase in triplet diradical lifetime, and ease of further functionalization. We present a single-flask transformation of carboxylic acids to acyl phosphonates that can access synthetically useful triplet diradicals under visible light or near-ultraviolet

Deoxygenative Functionalizations of Aldehydes, Ketones and Carboxylic Acids.

The simple and efficient conversion of carbonyl compounds into functionalized alkanes via deoxygenation is highly enabling in chemical synthesis. This Review covers the recent methodology development in carbonyl and carboxyl deoxygenative functionalizations, highlighting some representative and significant contributions in this field. These advances are categorized based on the reactivity patterns of some oxygenated feedstock compounds, including aldehydes, ketones and carboxylic acids. Four types of reactive intermediates arising from aldehydes and ketones during the deoxygenation, namely, bi

Amide synthesis via molecular shuffling of carboxylic acids.

The amide bond is fundamental in chemistry and biology and its classical construction from carboxylic acids and amines is limited by positional connectivity of the carboxylic acid group. Herein, we report an approach to amide synthesis that relies on molecular shuffling of carboxylic acid group. In this reaction, amino benzoic acids engage in oxidative addition to a Pd(0) complex, and CO from decarbonylation inserts into the C-Pd bond formed by alkene insertion before the last reductive elimination, achieving carbonyl shuttle amidation. This amide synthesis is precisely controlled by ligand se

Iron-Catalyzed Decarboxylative Sulfinylation of Alkyl Carboxylic Acids.

Sulfinamides, sulfonamides, and sulfonimidamides are valuable motifs in medicinal chemistry, yet methods to synthesize alkyl variants from simple, readily available feedstocks remain scarce. In this report, we detail the synthesis of these three distinct sulfur functional groups, using readily available and structurally diverse alkyl carboxylic acids as the starting materials. The method harnesses alkyl radical generation from carboxylic acids using commercial iron salts and visible light irradiation, in combination with commercial sulfinylamine reagents, to deliver alkyl sulfinamide products.

Titanium Tetrachloride-Assisted Direct Esterification of Carboxylic Acids.

Ester compounds, widely found in pharmaceutical and natural products, play a crucial role in organic synthesis, prompting the development of numerous methods for their synthesis. An important chemical approach in synthesizing esters from carboxylic acids involves the activation of the carboxyl function, requiring the conversion of the hydroxyl group into a suitable leaving group. This paper presents the findings of our investigations into an efficient method for producing esters from carboxylic acids and alcohols, using the Lewis acid titanium tetrachloride. Titanium tetrachloride has proven h

Transannular C-H functionalization of cycloalkane carboxylic acids.

Cyclic organic molecules are common among natural products and pharmaceuticals1,2. In fact, the overwhelming majority of small-molecule pharmaceuticals contain at least one ring system, as they provide control over molecular shape, often increasing oral bioavailability while providing enhanced control over the activity, specificity and physical properties of drug candidates3-5. Consequently, new methods for the direct site and diastereoselective synthesis of functionalized carbocycles are highly desirable. In principle, molecular editing by C-H activation offers an ideal route to these compoun

Structure-Activity Relationship of Antischistosomal Ozonide Carboxylic Acids.

Semisynthetic artemisinins and other bioactive peroxides are best known for their powerful antimalarial activities, and they also show substantial activity against schistosomes-another hemoglobin-degrading pathogen. Building on this discovery, we now describe the initial structure-activity relationship (SAR) of antischistosomal ozonide carboxylic acids OZ418 (2) and OZ165 (3). Irrespective of lipophilicity, these ozonide weak acids have relatively low aqueous solubilities and high protein binding values. Ozonides with para-substituted carboxymethoxy and N-benzylglycine substituents had high an

Inhibition of the Peroxygenase Lytic Polysaccharide Monooxygenase by Carboxylic Acids and Amino Acids.

Lytic polysaccharide monooxygenases (LPMOs) are widely distributed in fungi, and catalyze the oxidative degradation of polysaccharides such as cellulose. Despite their name, LPMOs possess a dominant peroxygenase activity that is reflected in high turnover numbers but also causes deactivation. We report on the influence of small molecules and ions on the activity and stability of LPMO during catalysis. Turbidimetric and photometric assays were used to identify LPMO inhibitors and measure their inhibitory effect. Selected inhibitors were employed to study LPMO activity and stability during cellu

A Metal-Free Direct Decarboxylative Fluoroacylation of Indole Carboxylic Acids with Fluorinated Acids.

A straightforward preparation of diversified fluorinated indol-3-yl ketones was developed by the direct decarboxylative fluoroacylation of indole carboxylic acids. The reaction could be performed on a gram scale under net conditions. Neither a metal catalyst nor an additive was employed. This methodology featured simple reaction conditions, high efficiency, exclusive selectivity, a broad substrate scope, and easy operation, which allowed it to meet the green chemistry requirement of the modern pharmaceutical industry. Control experiments confirmed that a radical process might be involved in th

Photoinduced Decarboxylative Sulfonylation of Carboxylic Acids with Sodium Sulfinates.

We report photoinduced decarboxylative sulfonylation of carboxylic acids with sodium sulfinates for synthesizing sulfones. The reaction proceeds under mild conditions, directly utilizing carboxylic acids without prefunctionalization, with copper acetate as the oxidant and sodium acetate as the base at room temperature. This method demonstrates excellent substrate compatibility, accommodating simple carboxylic acids, and drug molecules successfully converted into the corresponding sulfones. Furthermore, we developed a photoinduced two-step, one-pot protocol that integrated two carboxylic acids

Trivalent Nickel-Catalyzing Electroconversion of Alcohols to Carboxylic Acids.

The comprehension of activity and selectivity origins of the electrooxidation of organics is a crucial knot for the development of a highly efficient energy conversion system that can produce value-added chemicals on both the anode and cathode. Here, we find that the potential-retaining trivalent nickel in NiOOH (Fermi level, -7.4 eV) is capable of selectively oxidizing various primary alcohols to carboxylic acids through a nucleophilic attack and nonredox electron transfer process. This nonredox trivalent nickel is highly efficient in oxidizing primary alcohols (methanol, ethanol, propanol, b
