Naturally Occurring Cardenolides Affecting Schistosoma mansoni.

Schistosomiasis is a neglected tropical disease of considerable public health burden. We recently discovered a micromolar activity of several cardenolides against newly transformed schistosomula (NTS) of the parasitic flatworm Schistosoma mansoni in a small compound screen including different substance classes of both natural products as well as synthetic molecules. In further experiments, a focused library of naturally occurring and synthetic steroids was explored against NTS and adult S. mansoni, revealing seven cardenolides with comparable activities as known anthelminthics such as praziqua

Cardenolides: Insights from chemical structure and pharmacological utility.

Cardiac glycosides (CGs) are a class of naturally occurring steroid-like compounds, and members of this class have been in clinical use for more than 1500 years. They have been used in folk medicine as arrow poisons, abortifacients, heart tonics, emetics, and diuretics as well as in other applications. The major use of CGs today is based on their ability to inhibit the membrane-bound Na+/K+-ATPase enzyme, and they are regarded as an effective treatment for congestive heart failure (CHF), cardiac arrhythmia and atrial fibrillation. Furthermore, increasing evidence has indicated the potential cy

Are endogenous cardenolides controlled by atrial natriuretic peptide.

Endogenous cardenolides are digoxin-like substances and ouabain-like substances that have been implicated in the pathogenesis of hypertension and mood disorders in clinical and pre-clinical studies. Regulatory signals for endogenous cardenolides are still unknown. These endogenous compounds are believed to be produced by the adrenal gland in the periphery and the hypothalamus in the central nervous system, and constitute part of an hormonal axis that may regulate the catalytic activity of the α subunit of Na(+)/K(+)-ATPase. A review of literature suggests that there is great overlap in physiol

Cytotoxic cardenolides from the latex of Calotropis procera.

Three new cardenolides (3, 9 and 10), along with eight known ones, were isolated from the latex of Calotropis procera. The structural determination was accomplished by the 1D- and 2D-NMR spectra as well as HRESIMS analysis. The growth inhibitory activity of the latex and its sub-fractions as well as isolated compounds was evaluated against human A549 and Hela cell lines. The results exhibited that latex had strong growth inhibitory activity with IC50s of (3.37 μM, A-549) and (6.45 μM, Hela). Among the four extracts (hexane, chloroform, ethyl acetate and aqueous), chloroform extract displayed t

19-Nor- and 18,20-epoxy-cardenolides from the leaves of Calotropis gigantea.

Two new cardenolides (1 and 2) along with 12 known compounds were isolated from the dichloromethane extract of the leaves of Calotropis gigantea. The structural elucidation was accomplished by spectroscopic methods. Some of the isolates were evaluated for cytotoxic activity against KB, BC, and NCI-H187 cancer cell lines, and all cardenolides tested were found to possess strong inhibitory effects. The presence of a deoxysugar at C-3, a formyl group at C-10, and an alpha,beta-unsaturated gamma-lactone was crucial for cytotoxic activity.

Cardenolide toxin diversity impacts monarch butterfly growth and sequestration.

In coevolutionary interactions, host plants accrue novel chemical defenses that specialist herbivores counter by detoxification and sometimes sequestration. We recently found unusual nitrogen- and sulfur-containing (N,S-) cardenolides in some milkweeds-highly toxic compounds that monarch butterflies (Danaus plexippus) detoxify during sequestration. We hypothesized that the N,S-cardenolides in Asclepias curassavica (uscharin and voruscharin) would reduce caterpillar performance and sequestration more than other abundant related cardenolides (15-hydroxy-calotropin, frugoside, calactin). Cardenol

Anti-Neuroinflammatory Effects of Cardenolides in IL-1β-Activated SK-N-SH Cells.

Neuroinflammation is a key feature of neurological disorders such as Alzheimer's or Parkinson's disease, depression or amyotrophic lateral sclerosis. This study investigates the anti-neuroinflammatory effects of three cardenolide derivatives, digitoxigenin, digitoxigenone, and coroglaucigenin (all at concentrations of 1, 5, 10, and 25 µM), in IL-1β-stimulated human neuroblastoma SK-N-SH cells. All compounds significantly reduced PGE2 production without affecting cell viability by downregulation of cyclooxygenase (COX)-2 and microsomal prostaglandin E synthases (mPGES)-1 expression and synthesi

Elucidation of the mechanism of anti-herpes action of two novel semisynthetic cardenolide derivatives.

Human herpesviruses are among the most prevalent pathogens worldwide and have become an important public health issue. Recurrent infections and the emergence of resistant viral strains reinforce the need of searching new drugs to treat herpes virus infections. Cardiac glycosides are used clinically to treat cardiovascular disturbances, such as congestive heart failure and atrial arrhythmias. In recent years, they have sparked new interest in their potential anti-herpes action. It has been previously reported by our research group that two new semisynthetic cardenolides, namely C10 (3β-[(N-(2-h

Semisynthetic Cardenolides Acting as Antiviral Inhibitors of Influenza A Virus Replication by Preventing Polymerase Complex Formation.

Influenza virus infections represent a major public health issue by causing annual epidemics and occasional pandemics that affect thousands of people worldwide. Vaccination is the main prophylaxis to prevent these epidemics/pandemics, although the effectiveness of licensed vaccines is rather limited due to the constant mutations of influenza virus antigenic characteristics. The available anti-influenza drugs are still restricted and there is an increasing viral resistance to these compounds, thus highlighting the need for research and development of new antiviral drugs. In this work, two semis

Genome Assembly and Annotation of the Medicinal Plant Calotropis gigantea, a Producer of Anticancer and Antimalarial Cardenolides.

Calotropis gigantea produces specialized secondary metabolites known as cardenolides, which have anticancer and antimalarial properties. Although transcriptomic studies have been conducted in other cardenolide-producing species, no nuclear genome assembly for an Asterid cardenolide-producing species has been reported to date. A high-quality de novo assembly was generated for C. gigantea, representing 157,284,427 bp with an N50 scaffold size of 805,959 bp, for which quality assessments indicated a near complete representation of the genic space. Transcriptome data in the form of RNA-sequencing

Development and application of targeted UHPLC-MS/MS methods for the analysis of 31 groups of cardenolide glycosides.

Cardenolides are steroidal glycosides characterized by structural complexity and both medicinal and ecological relevance, necessitating precise and reliable analytical methods for their detection and quantitation. Although cardenolide MS/MS fragmentation has been extensively studied, no method has previously enabled simultaneous detection and quantitation of all glycosides derived from a given genin. In this study, we developed group-specific MS/MS methods for comprehensive screening of cardenolide glycosides containing 31 distinct genin backbones, i.e. aglycones. Application of these 31 genin

A simple chemical method for synthesizing malonyl hemiesters of 21-hydroxypregnanes, potential intermediates in cardenolide biosynthesis.

A simple and versatile method for the chemical synthesis of 21-hydroxypregnane 21-O-malonyl hemiesters which may be important intermediates of cardenolide biosynthesis is described. Starting from commercial beta-methyldigitoxin, acid hydrolysis followed by 3beta-O-acetylation and ozonolysis with reductive cleavage of the ozonides afforded 3beta-acetoxy-5beta-pregnane-14beta,21-diol-20-one which was finally converted into the target compound by treatment with malonyl chloride. The malonylation protocol was optimized using deoxycorticosterone (DOC) as the pregnane educt.

Monarch Performance and Cardenolide Sequestration Decline on Naturally Derived Hybrid Milkweeds (Asclepias syriaca x speciosa).

Plant hybridization often alters herbivore-relevant traits, yet the effects of hybrid feeding on specialist herbivores, especially those that co-opt host-plant defenses, are largely unknown. We examined how naturally derived hybrids between Asclepias syriaca and A. speciosa (A. syriaca x speciosa) affect monarch butterfly (Danaus plexippus) performance and cardenolide sequestration. Across three independent greenhouse experiments, monarch caterpillars reared on genetically heterogeneous A. syriaca x speciosa hybrid milkweeds grew more slowly than caterpillars reared on either parental species.

Diagnostic Ion-Guided Isolation and Characterization of Trace Periplocin-Derived Cardenolide Metabolites from Rat Urine.

Cardiac glycosides from Periplocae Cortex are bioactive constituents, but isolating and structurally confirming their trace in vivo metabolites remains challenging because authentic standards are often unavailable. We developed a steroid-core fragment-guided strategy based on the diagnostic ion at m/z 355.2270 that integrates metabolite screening, targeted purification, and structural characterization. Periplocin-related analytes were detected in rat urine and plasma after oral periplocin administration using HPLC-QQQ-MS/MS and UPLC-Q-TOF-MS/MS. Guided by the diagnostic ion, 13 L of rat urine

Strophanthus sarmentosus Extracts and the Strophanthus Cardenolide Ouabain Inhibit Snake Venom Proteases from Echis ocellatus.

Strophanthus sarmentosus is recognised for various ethnomedicinal applications, including treatment after snakebites. However, only limited scientific evidence exists on its antivenomous capabilities. This study investigates the efficacy of methanol and ethylacetate extracts from S. sarmentosus leaves and roots against Echis ocellatus venom. A non-toxic range for the extracts was determined in rats, and assays were performed to test their anti-hemorrhagic and anti-hemolytic activity as well as their influence on venom-induced blood clotting. In all of these experiments, the extracts demonstrat
