Range expansion and first observation of Tridacna noae (Cardiidae: Tridacninae) in American Sāmoa.

Giant clams are ecologically important, benefitting species of all trophic levels. But numerous populations have declined drastically in numbers due to past intensive exploitation that led to their listing in both CITES Appendix II and IUCN Red List of Threatened Species. However, giant clams are notoriously difficult to identify, and recent molecular work has revealed that morphological misidentification of giant clams have confounded current population assessments and extinction risk. The most recent study of the status of giant clams in the Samoan Archipelago was published in a journal over

How costly are metacercarial infections in a bivalve host? Effects of two trematode species on biochemical performance of cockles.

Bivalve stocks have been decreasing in the last decades largely due to emergent diseases and consequent mass mortality episodes. Cerastoderma edule (the edible cockle) is one of the most exploited bivalves in Europe and is among the most common hosts for trematodes, the most prevalent macroparasites in coastal waters but yet poorly studied. Therefore, in the present study, this bivalve species was used as host model to determine if trematode infection exerts a negative effect on bivalve energy metabolism and balance and if the tissues targeted by different trematodes influence the metabolic co

First Report of Haplosporidium edule Infection in the Olive-Green Cockle (Cerastoderma glaucum) from the Northern Adriatic Sea: Expanding Host Range and Geographic Distribution.

Haplosporidium edule is a haplosporidian parasite originally described in the common edible cockle (Cerastoderma edule) along the European Atlantic coast. In this study, we report the first detection of H. edule in the olive-green cockle (Cerastoderma glaucum) from the northern Adriatic Sea, representing both a novel host record and a new geographic occurrence. During a cross-sectional study conducted in May 2019, 90 C. glaucum specimens were collected from three lagoon sites in northeastern Italy. Histological examination of soft tissues revealed haplosporidian developmental stages, including

Somatic evolution of marine transmissible leukemias in the common cockle, Cerastoderma edule.

Transmissible cancers are malignant cell lineages that spread clonally between individuals. Several such cancers, termed bivalve transmissible neoplasia (BTN), induce leukemia-like disease in marine bivalves. This is the case of BTN lineages affecting the common cockle, Cerastoderma edule, which inhabits the Atlantic coasts of Europe and northwest Africa. To investigate the evolution of cockle BTN, we collected 6,854 cockles, diagnosed 390 BTN tumors, generated a reference genome and assessed genomic variation across 61 tumors. Our analyses confirmed the existence of two BTN lineages with hemo

Trematode genetic patterns at host individual and population scales provide insights about infection mechanisms.

Multiple parasites can infect a single host, creating a dynamic environment where each parasite must compete over host resources. Such interactions can cause greater harm to the host than single infections and can also have negative consequences for the parasites themselves. In their first intermediate hosts, trematodes multiply asexually and can eventually reach up to 20% of the host's biomass. In most species, it is unclear whether this biomass results from a single infection or co-infection by 2 or more infective stages (miracidia), the latter being more likely a priori in areas where preva

Trematode infection modulates cockles biochemical response to climate change.

Resulting mainly from atmospheric carbon dioxide (CO2) build-up, seawater temperature rise is among the most important climate change related factors affecting costal marine ecosystems. Global warming will have implications on the water cycle, increasing the risk of heavy rainfalls and consequent freshwater input into the oceans but also increasing the frequency of extreme drought periods with consequent salinity increase. For Europe, by the end of the century, projections describe an increase of CO2 concentration up to 1120 ppm (corresponding to 0.5 pH unit decrease), an increase in the water

Molecular detection reveals infection phenology of a host - Trematode system (Cerastoderma edule - Bucephalus minimus) in southwestern France.

Accurate detection of parasites in their hosts is essential for pathogen control and ecosystem studies. The trematode Bucephalus minimus (Digenea: Bucephalidae) infects the edible cockle Cerastoderma edule, a commercially fished and ecologically important bivalve that serves as its first intermediate host, in which it can be difficult to detect. We investigated detection accuracy and infection phenology of this host-parasite system in Arcachon Bay, France. Cockles were collected monthly over one year to compare detection methods: stereomicroscopy of tissues versus molecular detection by quanti

Effect of light on the trematode Himasthla elongata: from cercarial behaviour to infection success.

The cockle Cerastoderma edule, a socioeconomically important bivalve of the northeast Atlantic, is host to several trematodes, including Himasthla elongata. In the life cycle of this trematode, cercariae (free-living stages) emerge from the first intermediate host, a snail, to infect cockles as second intermediate hosts. During their lifespan (less than 2 d), cercariae must ensure successful host-to-host transmission via the surrounding water and therefore are exposed to and impacted by different environmental conditions, including abiotic factors. Given that the light:dark cycle is one of the

Physiological responses to ocean acidification and warming synergistically reduce condition of the common cockle Cerastoderma edule.

The combined effect of ocean acidification and warming on the common cockle Cerastoderma edule was investigated in a fully crossed laboratory experiment. Survival of the examined adult organisms remained high and was not affected by elevated temperature (+3 °C) or lowered pH (-0.3 units). However, the morphometric condition index of the cockles incubated under high pCO2 conditions (i.e. combined warming and acidification) was significantly reduced after six weeks of incubation. Respiration rates increased significantly under low pH, with highest rates measured under combined warm and low pH co

A validated DNA metabarcoding approach for routine authentication of processed bivalves.

Species mislabeling in commercial bivalves represents a growing concern for food authenticity. Given the diversity of traded bivalves, broad taxonomic coverage is essential for untargeted screening. To address this challenge, we developed and validated a DNA metabarcoding approach employing two PCR assays, a singleplex and a duplex, to amplify mitochondrial 16S rDNA fragments (160-203 bp) across seven families: Ostreidae, Pectinidae, Mytilidae, Pharidae, Veneridae, Glycymerididae, and Cardiidae. Taxa were identified at the species or genus level in 38 reference samples and four model food samp

Decoding the gene expression response of Cerastoderma edule to chronic trematode infection: A comparison among host tissues.

Parasites can induce gene expression changes in their hosts, either benefiting the parasite or the host. In particular, trematodes are not only one of the most ubiquitous groups of aquatic parasites, they also have huge impacts on individual hosts with significant ecological and economic repercussions. The trematode Bucephalus minimus infects Cerastoderma edule (the edible cockle), a socioeconomically and ecologically important bivalve, as its first intermediate host. This parasite is one of the most harmful parasites infecting cockles, affecting their ability to reproduce, grow, and survive,

Are metallothioneins equally good biomarkers of metal and oxidative stress?

Several researchers investigated the induction of metallothioneins (MTs) in the presence of metals, namely Cadmium (Cd). Fewer studies observed the induction of MTs due to oxidizing agents, and literature comparing the sensitivity of MTs to different stressors is even more scarce or even nonexistent. The role of MTs in metal and oxidative stress and thus their use as a stress biomarker, remains to be clearly elucidated. To better understand the role of MTs as a biomarker in Cerastoderma edule, a bivalve widely used as bioindicator, a laboratory assay was conducted aiming to assess the sensitiv

Interactive effects of contamination and trematode infection in cockles biochemical performance.

Anthropogenic activities, especially those involving substances that pollute the environment can interfere with bivalve populations, as well as parasitism, a fundamental ecological interaction often neglected. In marine environments, organisms are concomitantly exposed to pollutants and parasites, a combination with synergistic, antagonistic or additive effects representing a potential threat to aquatic communities sustainability. In the present study, Cerastoderma edule (the edible cockle)-Himasthla elongata (trematode) was used as host-parasite model. Cockles are worldwide recognized as good

Investigations into matrix components affecting the performance of the official bioassay reference method for quantitation of paralytic shellfish poisoning toxins in oysters.

Significant differences previously observed in the determination of paralytic shellfish poisoning toxins (PSTs) in oysters using official method AOAC 2005.06 and 959.08 were investigated in detail with regard to possible matrix effects. Method AOAC 2005.06 gave results 2-3 times higher than the mouse bioassay method, 959.08, differences thought to be due to underestimation of PSTs by the mouse bioassay. In order to prove the cause of these large differences, work was conducted here to examine the presence and effects of matrix components on the performance of each of the two assays. A range of

Expression regulation of Fad and Mboat gene families to cold stress in the noble scallop Chlamys nobilis: A comparative analysis across bivalves.

Cold waves exacerbated by global warming and the resultant sharp drops in coastal water temperature pose serious threats to the sustainability of bivalve aquaculture. The fatty acid desaturase (Fad) and membrane-bound O-acyltransferase (Mboat) gene families play essential roles in maintaining membrane lipid homeostasis and facilitating cold adaptation. Although Fad genes have been investigated in several bivalves, the role of Mboat genes in cold stress and how these two families coordinately regulate lipid remodeling under cold stress remain largely unclear. This study performed a comprehensiv
