Cartilage organoids for cartilage development and cartilage-associated disease modeling.

Cartilage organoids have emerged as powerful modelling technology for recapitulation of joint embryonic events, and cartilage regeneration, as well as pathophysiology of cartilage-associated diseases. Recent breakthroughs have uncovered "mini-joint" models comprising of multicellular components and extracellular matrices of joint cartilage for development of novel disease-modifying strategies for personalized therapeutics of cartilage-associated diseases. Here, we hypothesized that LGR5-expressing embryonic joint chondroprogenitor cells are ideal stem cells for the generation of cartilage orga

Engineered cartilage utilizing fetal cartilage-derived progenitor cells for cartilage repair.

The aim of this study was to develop a fetal cartilage-derived progenitor cell (FCPC) based cartilage gel through self-assembly for cartilage repair surgery, with clinically useful properties including adhesiveness, plasticity, and continued chondrogenic remodeling after transplantation. Characterization of the gels according to in vitro self-assembly period resulted in increased chondrogenic features over time. Adhesion strength of the cartilage gels were significantly higher compared to alginate gel, with the 2-wk group showing a near 20-fold higher strength (1.8 ± 0.15 kPa vs. 0.09 ± 0.01 k

Performance parity in cartilage repair: SPMK-g-PEEK versus cartilage-cartilage interfaces.

Effective fluid exudation and rehydration are essential for the low-friction function of healthy articular cartilage, facilitating interstitial fluid pressurisation, solute transport, and aqueous lubrication. However, current metallic biomaterials used in focal cartilage repair or hemiarthroplasty compromise this fluid-pressure dependent load support, leading to the erosion of the interfacing cartilage. This study investigates bioinspired hydrophilic 3-sulfopropyl methacrylate potassium salt (SPMK) polymer grafted onto a PEEK substrate (SPMK-g-PEEK) as a potential solution. SPMK-g-PEEK aims to

Cartilage Tissue and Therapeutic Strategies for Cartilage Repair.

High incidence of articular cartilage defects is still a major challenge in orthopedic and trauma surgery worldwide. It also has great socioeconomic effects as it is the major cause of disability in industrialized countries. This highlights the essential need for new treatments. Knowledge about the factors that have been implicated in the pathogenesis of cartilage diseases, including changes in the composition and structure of cartilaginous extracellular matrix (ECM), molecular factors and environmental signaling pathways could help the development of innovative therapeutic strategies. It is c

Uncalcified cartilage resorption in human fetal cartilage canals.

In the human fetus, epiphyses appear as a solid avascular cartilaginous mass until the eleventh week of development. Around the third fetal month of development, vascular canals coming from the perichondrium are recognized in the mineralized epiphyseal cartilage. Whether cartilage canals develop by passive inclusion or active chondrolysis is still a matter of controversy. We studied the relationships between the intracanalar cells and the surrounding matrix on human fetal epiphyses embedded in glycol methacrylate. At the blind end of canals both stellate fibroblast-like cells and vacuolated ma

Proteoglycan aggregation in injured articular cartilage. A comparison of healing lacerated cartilage with osteoarthritic cartilage.

Canine knee cartilage which had been damaged by laceration with a scalpel down to subchondral bone (Model A) and knee cartilage from dogs in whom osteoarthritis had been induced by transection of the anterior cruciate ligament by arthrotomy (Model B) were compared 7 weeks after the surgical procedures. Cartilage from the operated knees in Model A was hypocellular and showed extensive fibrosis, while samples from Model B showed moderate fibrillation and osteophyte formation. (In both cases water content was increased and uronic acid content decreased in cartilage from the operated knees.) Despi

Cartilage diseases.

Hyaline cartilages, fibrocartilages and elastic cartilages play multiple roles in the human body including bearing loads in articular joints and intervertebral discs, providing joint lubrication, forming the external ears and nose, supporting the trachea, and forming the long bones during development and growth. The structure and organization of cartilage's extracellular matrix (ECM) are the primary determinants of normal function. Most diseases involving cartilage lead to dramatic changes in the ECM which can govern disease progression (e.g., in osteoarthritis), cause the main symptoms of the

Cartilage tympanoplasty.

This chapter describes two techniques for cartilage reconstruction of the tympanic membrane: the perichondrium/cartilage island flap, which uses tragal cartilage, and the palisade technique, which uses cartilage from the tragus or cymba. The perichondrium/cartilage island flap is preferred for management of the atelectatic ear and the high-risk perforation. The palisade technique is preferred in cases of cholesteatoma and when ossicular reconstruction is needed in the malleus-present situation. Descriptions of the modifications that should be taken in response to specific surgical indications

Cartilage proteoglycans.

The structure of the protein core of the high molecular weight aggregating proteoglycan from pig laryngeal cartilage has been investigated. Mild trypsin digestion of proteoglycan aggregates released a large (Mr approximately equal to 150K) protein-rich fragment that contained the hyaluronate-binding region (Mr 66K). Rotary-shadowing electron microscopy of this preparation showed it to contain 'double globe' structures, similar to those seen with intact proteoglycans. Interaction studies and immunochemical evidence showed that one of the globular domains was the binding region. The second globu

Analysis of cartilage creep-recovery of human osteoarthritic cartilage.

BACKGROUND: Articular cartilage of weight-bearing joints restores its thickness after loading through viscoelastic recovery. Creep-recovery is a key parameter for assessing cartilage biomechanical properties and is relevant for osteoarthritis (OA) diagnosis and cartilage replacement development. This study aimed to evaluate creep-recovery in human osteoarthritic cartilage. METHODS: Full-thickness osteochondral explants (n = 34) were obtained from tibial plateaus of 14 patients undergoing total knee replacement. Samples were graded histologically as normal (n = 6), mild (n = 18), moderate (n =

Cartilage Conundrum: Investigating Outcomes in Knee Cartilage Restoration Techniques.

BACKGROUND: Management of chondral injury can be difficult due to the avascularity of articular cartilage and the limited ability to proliferate and promote repair. Untreated lesions can lead to early development of arthritis and the need for joint replacement. For intermediate-sized lesions, osteochondral autograft transfer and autologous cell-based cartilage treatment have been accepted as the most effective treatment options. PURPOSE: To investigate the rates of complications within 2 years of initial surgery in patients who undergo primary autologous cell-based cartilage treatment and ost

Characteristics of cartilage engineered from human pediatric auricular cartilage.

In the repair of cartilage defects, autologous tissue offers the advantage of lasting biocompatibility. The ability of bovine chondrocytes isolated from hyaline cartilage to generate tissue-engineered cartilage in a predetermined shape, such as a human ear, has been demonstrated; however, the potential of chondrocytes isolated from human elastic cartilage remains unknown. In this study, the authors examined the multiplication characteristics of human auricular chondrocytes and the ability of these cells to generate new elastic cartilage as a function of the length of time they are maintained i

Metal deposition at the bone-cartilage interface in articular cartilage.

There is a growing interest being shown in the changes occurring in elemental distribution at the bone-cartilage interface, the changes either being a result of mechanical damage or disease. In particular, such investigations have tended to concern the elemental alterations associated with the osteoarthritic wear and tear damage occurring to the cartilage and subchondral bone of synovial joints or that associated with disease processes such as rheumatic arthritis. Present studies examine sections of femoral head obtained from total hip replacement surgery, use being made of micro-proton-induce

Exploring cartilage development and disease models: applications of cartilage organoids.

Cartilage is an avascular and aneural connective tissue vital for joint function, yet its limited intrinsic regenerative capacity poses significant challenges for treating injuries and degenerative diseases such as osteoarthritis (OA). Conventional models-including animal studies, cell lines, and primary chondrocyte cultures-exhibit considerable limitations in accurately replicating the native cartilage microenvironment, species-specific relevance, and differentiation potential. Recently, cartilage organoids have emerged as transformative tools that closely emulate the architecture, cellular h

Osteophyte Cartilage as a Potential Source for Minced Cartilage Implantation: A Novel Approach for Articular Cartilage Repair in Osteoarthritis.

Osteoarthritis (OA) is a common joint disorder characterized by cartilage degeneration, often leading to pain and functional impairment. Minced cartilage implantation (MCI) has emerged as a promising one-step alternative for large cartilage defects. However, the source of chondrocytes for MCI remains a challenge, particularly in advanced OA, as normal cartilage is scarce. We performed in vitro studies to evaluate the feasibility of MCI using osteophyte cartilage, which is present in patients with advanced OA. Osteophyte and articular cartilage samples were obtained from 22 patients who underwe

Proteoglycan 4 reduces friction more than other synovial fluid components for both cartilage-cartilage and cartilage-metal articulation.

OBJECTIVE: The clinical success of focal metallic resurfacing implants depends largely on the friction between implant and opposing cartilage. Therefore, the present study determines the lubricating ability of the synovial fluid components hyaluronic acid (HA), proteoglycan 4 (PRG4) and a surface-active phospholipid (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine, POPC), on the articulation between cartilage and a Cobalt Chromium Molybdenum (CoCrMo) implant surface, compared with two cartilage surfaces.
