Enteric Virome and Bacterial Microbiota in Children With Ulcerative Colitis and Crohn Disease.

OBJECTIVES: We examined the fecal virome and bacterial community composition of children with Crohn disease (CD), ulcerative colitis (UC), and healthy controls to test the hypothesis that unique patterns of viral organisms and/or presence of bacterial pathogens may be identified that could contribute to the pathogenesis of pediatric inflammatory bowel disease (IBD). METHODS: Fecal samples from 24 children (mean 12.2 years) with CD (n = 7) or UC (n = 5) and similar aged controls (n = 12) were processed to determine individual viromes. Viral sequences were identified through translated protein

Abolishment of morphology-based taxa and change to binomial species names: 2022 taxonomy update of the ICTV bacterial viruses subcommittee.

This article summarises the activities of the Bacterial Viruses Subcommittee of the International Committee on Taxonomy of Viruses for the period of March 2021-March 2022. We provide an overview of the new taxa proposed in 2021, approved by the Executive Committee, and ratified by vote in 2022. Significant changes to the taxonomy of bacterial viruses were introduced: the paraphyletic morphological families Podoviridae, Siphoviridae, and Myoviridae as well as the order Caudovirales were abolished, and a binomial system of nomenclature for species was established. In addition, one order, 22 fami

Interaction between gut virome and microbiota on inflammatory bowel disease.

Inflammatory bowel disease (IBD), encompassing Crohn's disease and ulcerative colitis, is a chronic condition marked by recurring gastrointestinal inflammation. While immune, genetic, and environmental factors are well-studied, the gut virome has received less attention. This editorial highlights the work which investigates the gut virome's role in IBD and its interactions with the bacterial microbiome and host immune system. The gut virome consists of bacteriophages, eukaryotic viruses, and endogenous retroviruses. Among these, Caudovirales bacteriophages are predominant and influence bacteri

Staphylococcus species infected by a bacteriophage with a tail that is both curved and contractile.

Using a selective plating strategy for staphylococci, we surveyed the local community wastewater and purified 16 independent isolates representing the following seven species of Staphylococcus: S. cohnii, S. equorum, S. lentus, S. nepalensis, S. sciuri, S. shinii, and S. xylosus. Staphylococcus aureus was not detected. The wastewater also served as a source to identify a bacteriophage (phage), referred to here as JS1, that could infect all these species of Staphylococcus, as well as a range of clinical S. aureus strains, including methicillin-resistant isolates. The class Caudoviricetes are ta

Genetic analysis of the cold-sensitive growth phenotype of Burkholderia pseudomallei/thailandensis bacteriophage AMP1.

Bacteriophages related to phage Bp_AMP1 are the most widely spread group of phages infecting Burkholderia pseudomallei-the causative agent of melioidosis. These viruses are also infective against the nonpathogenic host Burkholderia thailandensis, allowing experimental work with them without any special safety precautions. The indirect data as well as the results of the mathematical modelling suggest that the AMP1-like viruses may act as natural biocontrol agents influencing the population levels of B. pseudomallei in soil and water habitats in endemic regions. The cold sensitivity of the lytic

Disease-specific alterations in the enteric virome in inflammatory bowel disease.

Decreases in the diversity of enteric bacterial populations are observed in patients with Crohn's disease (CD) and ulcerative colitis (UC). Less is known about the virome in these diseases. We show that the enteric virome is abnormal in CD and UC patients. In-depth analysis of preparations enriched for free virions in the intestine revealed that CD and UC were associated with a significant expansion of Caudovirales bacteriophages. The viromes of CD and UC patients were disease and cohort specific. Importantly, it did not appear that expansion and diversification of the enteric virome was secon

Isolation and characterization of a bacteriophage targeting Proteus mirabilis for use in culture media.

Proteus mirabilis is an important opportunistic bacterium that affects both animal and human health. One of the key characteristics of this bacterium is its distinctive zonal spreading on solid media. This movement, known as swarming, complicates the isolation and identification of pathogenic agents present alongside P. mirabilis in clinical samples. The aim of this study was to isolate a lytic bacteriophage targeting P. mirabilis to prevent its growth and evaluate the use of these phages in cultures of clinical samples contaminated with Proteus. A phage designated as P5663 was isolated using

Isolation and Characterization of New Bacteriophages against Staphylococcal Clinical Isolates from Diabetic Foot Ulcers.

Staphylococcus sp. is the most common bacterial genus in infections related to diabetic foot ulcers (DFUs). The emergence of multidrug-resistant bacteria places a serious burden on public health systems. Phage therapy is an alternative treatment to antibiotics, overcoming the issue of antibiotic resistance. In this study, six phages (SAVM01 to SAVM06) were isolated from effluents and were used against a panel of staphylococcal clinical samples isolated from DFUs. A genomic analysis revealed that the phages belonged to the Herelleviridae family, with sequences similar to those of the Kayvirus g

Identification of the biological properties of three novel lytic bacteriophages capable of infecting Bacillus anthracis: Novel members of the class Caudoviricetes.

Bacillus anthracis spores can remain viable for many years in regions where anthrax cases are reported or where deceased animals are buried. Anthrax occurrences may be more common in areas where livestock farming is a primary source of livelihood. In this study, 43 Bacillus anthracis isolates were used, which were isolated from samples collected in different provinces of Türkiye. Three lytic bacteriophages(BaTR8, BaTR13, and BaTR16) were isolated from environmental soil samples. These three lytic bacteriophages' biological properties (host range, burst size, and phage morphology) were defined,

Unraveling a new Klebsiella phage KP47 against Klebsiella spp.

BACKGROUND: The increase in cases of infectious diseases related to multidrug-resistant bacteria and the spread of multidrug-resistant genes have driven the search for therapeutic alternatives that can circumvent this global phenomenon. One of these alternatives is the use of bacteriophages, which are viruses capable of infecting and killing specific bacteria. Klebsiella pneumoniae species complex (KpSC) is an important opportunistic group associated with multidrug resistance. This group includes Klebsiella quasipneumoniae subsp. similipneumoniae, a species recognized as a clinically relevant
