Improving cell encapsulation through size control.

Capsules based on the polyelectrolyte complexation between the polyanions sodium alginate and sodium cellulose sulphate with the polycation poly(methylene-co-guanidine) hydrochloride in the presence of calcium chloride have previously shown important advantages for cell encapsulation. However, in vivo long-term applications require capsule features that are well suited for the functionality of encapsulated cells. These should be targeted to the site of implantation with an appropriate size, a relative stability, and suitable diffusion properties. This study shows the effect of capsule size red

Orthogonally crosslinked alginate conjugate thermogels with potential for cell encapsulation.

Hydrogels with more than one mode of crosslinking have gained interest due to improved control over hydrogel properties such as mechanical strength using multiple stimuli. In this work, sodium alginate was covalently conjugated onto thermoresponsive polyurethanes to prepare hybrid polymers (EPC-Alg) that are responsive to both temperature and Ca2+, forming orthogonally crosslinked hydrogels which are non-toxic to cells. Notably, the crosslinks are fully reversible, allowing for gel strength to be modulated via selective removal of either stimulus, or complete deconstruction of the hydrogel net

Thread- reinforced encapsulation devices (THRED) for therapeutic stem cell delivery.

INTRODUCTION: Mesenchymal stem cells (MSCs) have been studied as potential therapeutic agents for various diseases. However, their widespread application has been limited, in part, due to the concern for tumorigenic transformation and lack of longevity in host environments secondary to immune-mediated destruction. Therefore, we set out to study an MSC encapsulation and delivery method with potential application in the treatment of intestinal ischemic diseases. METHOD: Human vertebra bone adherent mesenchymal stem cells (vBA-MSCs) were encapsulated in an alginate-derived Thread Reinforced Enca

[Cell encapsulation for the treatment of central nervous system disorders].

INTRODUCTION: One of the main matters of drug administration into the central nervous system (CNS) is to cross the blood/brain barrier. For this reason, in some cases the drug does not reach the therapeutic level or is necessary to use some special systems to get a right administration. AIM: To present a technological system which allow continuous and direct administration of the drug into the CNS. It consists of enclosing secreting-cells of the desired product within a semipermeable polymeric matrix. Furthermore, this device could be implanted easily into the CNS which improves the access of

Stem Cell-Derived Beta-Cell Therapies: Encapsulation Advances and Immunological Hurdles in Diabetes Treatment.

Diabetes mellitus encompasses a heterogeneous group of metabolic disorders defined by abnormalities in insulin secretion, function, or both. Exogenous insulin therapy has long been the principal treatment strategy for patients with type 1 diabetes and for those in advanced stages of type 2 diabetes. Stem cell therapy has gained significant attention in recent years as a potential curative approach for several life-threatening disorders. In this review, we focus on the use of induced pluripotent stem cells as an alternative source for beta-cell generation, offering a solution to organ scarcity

Emerging Strategies for Beta Cell Encapsulation for Type 1 Diabetes Therapy.

Diabetes is a prevalent chronic disease affecting millions of people globally. To address this health challenge, advanced beta cell therapy using biomaterials-based macroscale, microscale, and nanoscale encapsulation devices must tackle various obstacles. First, overcoming foreign body responses is a major focus of research. Strategies such as immunomodulatory materials and physical immunoshielding are investigated to reduce the immune response and improve the longevity of the encapsulated cells. Furthermore, oxygenating strategies, such as the use of oxygen-releasing biomaterials, are develop

Poly(vinyl alcohol) synthetic polymer foams as scaffolds for cell encapsulation.

Poly(vinyl alcohol) (PVA) foams were used as scaffolds in hollow fiber membrane-based cell encapsulation devices. The surrounding permselective membrane serves as an immunoisolation barrier while allowing metabolites and other small molecules to be freely transported. The internal matrix defines the microenvironment for the encapsulated cells. PC12 cell-containing devices represent one possible strategy for safe transplantation of dopamine-secreting cells for the treatment of dopamine-deficient diseases such as Parkinson's disease. Artificial cells with emphasis on cell encapsulation of genetically engineered cells.

Artificial cells are prepared in the laboratory for medical and biotechnological applications. Encapsulated cells are being studied for the treatment of diabetes, liver failure, and other conditions. More recently, there have been extensive studies into the use of encapsulated genetically engineered cells for gene therapy. We recently found that daily orally administered artificial cells, each containing a genetically engineered microorganism, can lower the elevated urea level in uremic rats to normal levels. This may solve the final obstacle of the lack of an effective oral urea removal syste

Neovascularized implantable cell homing encapsulation platform with tunable local immunosuppressant delivery for allogeneic cell transplantation.

Cell encapsulation is an attractive transplantation strategy to treat endocrine disorders. Transplanted cells offer a dynamic and stimulus-responsive system that secretes therapeutics based on patient need. Despite significant advancements, a challenge in allogeneic cell encapsulation is maintaining sufficient oxygen and nutrient exchange, while providing protection from the host immune system. To this end, we developed a subcutaneously implantable dual-reservoir encapsulation system integrating in situ prevascularization and local immunosuppressant delivery, termed NICHE. NICHE structure is 3

Reversible light-responsive protein hydrogel for on-demand cell encapsulation and release.

The design of biomaterials that can reconfigure on-demand in response to external stimuli is an emerging area in materials research. However, achieving reversible assembly of protein-based biomaterials by light input remains a major challenge. Here, we present the engineering of a new protein material that is capable of switching between liquid and solid state reversibly, controlled by lights of different wavelengths. The materials are created by incorporating a light-responsive mutant Dronpa protein domain into the backbone of Elastin-Like Proteins (termed DELPs). We show that the DELP materi

Encapsulated mesenchymal stromal cells for in vivo transplantation.

Immunomodulatory human mesenchymal stromal cells (hMSC) have been incorporated into therapeutic protocols to treat secondary inflammatory responses post-spinal cord injury (SCI) in animal models. However, limitations with direct hMSC implantation approaches may prevent effective translation for therapeutic development of hMSC infusion into post-SCI treatment protocols. To circumvent these limitations, we investigated the efficacy of alginate microencapsulation in developing an implantable vehicle for hMSC delivery. Viability and secretory function were maintained within the encapsulated hMSC p

Intrasplenic transplantation of encapsulated cells: a novel approach to cell therapy.

Cell therapy is likely to succeed clinically if cells survive at the transplantation site and are protected against immune rejection. We hypothesized that this could be achieved with intrasplenic transplantation of encapsulated cells because the cells would have instant access to oxygen and nutrients while being separated from the host immune system. In order to provide proof of the concept, primary rat hepatocytes and human hepatoblastoma-derived HepG2 cells were used as model cells. Rat hepatocytes were encapsulated in 100-kDa hollow fibers and cultured for up to 28 days. Rat spleens were im

Personalized Alginate Encapsulation: The Role of Autologous Blood Additives in Parathyroid Cell Transplantation.

The only therapeutic intervention for hypoparathyroidism is parathyroid transplantation, but graft rejection is a concern. This study sought to mitigate this problem by utilizing the patient's blood, serum, or plasma in the transplant. To accomplish this objective, blood additives derived from Sprague-Dawley rats are incorporated within alginate, and human parathyroid cells are encapsulated. The prepared microbeads are monitored for mechanical properties, followed by xenotransplantation into rats for the evaluation of cell function and immunological response. Biodegradation data showed that th

Geotrichum candidum arthrospore cell wall particles as a novel carrier for curcumin encapsulation.

We report for the first time that curcumin is successfully encapsulated into a new natural pre-formed carrier, which was derived from arthrospore cell wall particles (APs) of probiotic Geotrichum candidum LG-8 and mainly composed of beta-1,4-glucan. Vacuum infusion process was used for efficient encapsulation of curcumin. The results showed that the encapsulation efficiency and yield of APs were 36.5 ± 0.9 % and 730.6 ± 26.5 μg/g (wet basis), respectively. Compared with the other probiotic carriers such as Saccharomyces cerevisiae, it could more effectively maintain the antioxidant property an

Injectable Acylhydrazone-Linked RAFT Polymer Hydrogels for Sustained Protein Release and Cell Encapsulation.

A new class of temperature responsive polymer, termed PADO, is synthesized by reversible addition-fragmentation chain-transfer polymerization. Synthesized from copolymerization of diacetone acrylamide (DAAM), di(ethylene glycol) ethyl ether acrylate, and oligo(ethylene glycol) methyl ether acrylate, PADO polymer phase separates at temperature above its lower critical solution temperature (36-42 °C) due to enhanced hydrophobic interactions between the short ethylene glycol side chains. Solution of PADO polymers exhibit injectable shear-thinning properties and reach sol-gel transition rapidly (<

Polymeric Approaches to Reduce Tissue Responses Against Devices Applied for Islet-Cell Encapsulation.

Immunoisolation of pancreatic islets is a technology in which islets are encapsulated in semipermeable but immunoprotective polymeric membranes. The technology allows for successful transplantation of insulin-producing cells in the absence of immunosuppression. Different approaches of immunoisolation are currently under development. These approaches involve intravascular devices that are connected to the bloodstream and extravascular devices that can be distinguished in micro- and macrocapsules and are usually implanted in the peritoneal cavity or under the skin. The technology has been subjec

Thiol-Methylsulfone Crosslinked Hydrogels for Cell Encapsulation: Molecular Scale Modulation of Physiochemical Properties.

Hydrogels mimicking the mechanical and biochemical features of the cellular microenvironment allow cell encapsulation and facilitate in vitro 3D culture. In addition to biocompatibility and reactivity in physiological conditions, a key criterion for crosslinking chemistry is appropriate gelation kinetics to allow mixing and homogeneous distribution of cells with the hydrogel precursors. We have previously presented aryl methylsulfone/thiol (MS/SH) reaction as a thiol-reactive cross-linking system for cell encapsulation in star polyethylene glycol (PEG4) hydrogels with a gelation kinetics in mi

Active invasion and/or encapsulation? A reappraisal of host-cell parasitism by Cryptosporidium.

Host-cell invasion by Cryptosporidium is a complex process that requires many different factors derived from both the parasite and the host cell. However, the exact natures of the processes have yet to be resolved. Here, research on different components of the invasion process is put in context, and the sequence of events and pathways associated with the establishment of Cryptosporidium in its unique niche is clarified. In addition, initial parasite-host contact, host-cell invasion and host-cell responses are described. The roles of parasite and host-cell-derived components in the invasion pro
