Contribution of Proteins to Membrane and Cell Wall Structures in Mycobacterium tuberculosis.

The cell wall of Mycobacterium tuberculosis (Mtb) is an exceptionally complex, multilayered structure that is fundamental to the bacterium's survival, pathogenicity, and resistance to both host immune defenses and antibiotics. This cell envelope consists of several distinct components: a surface capsule, an outer membrane, and a peptidoglycan layer that is covalently attached to arabinogalactan. The arabinogalactan is further esterified with mycolic acids, forming the characteristic mycolyl-arabinogalactan-peptidoglycan (mAGP) complex, which is anchored to the underlying plasma membrane. The a

Computational exploration of viral cell membrane structures for identifying novel therapeutic target.

The membrane proteins of viruses play a critical role, and they shield viruses and takes biochemical mechanisms like sticking to the host cell membrane, merging with them, building new viruses, and breaking free. These steps make sure the virus can infect and multiply. But the membrane proteins of Nipah, Zika, SARS-CoV-2, and Hendra virus can cause special kinds of infections. Nipah and Hendra viruses use their fusion protein to join with the host cell membrane. Their glycoprotein interacts with host receptors. The matrix protein helps to build and support the virus structure. Zika virus relie

Investigating the effects of microwave plasma on bacterial cell structures, viability, and membrane integrity.

Plasma-mediated bacterial inactivation holds great promise but presents several challenges. This study investigates the antibacterial effect of 2.45 GHz non-thermal microwave (MW) plasma on Staphylococcus aureus (S. aureus) and Salmonella abony (S. abony) suspended in phosphate-buffered saline (PBS). A 6-log reduction in both bacterial strains was achieved within 300 s of plasma exposure. The enhanced inactivation is attributed to elevated levels of reactive oxygen species (ROS), particularly ·OH (30.30% in S. aureus, 40.13% in S. abony) and H2O2 (173.27% in S. aureus, and 391.84% in S. abony)

Structure of benzothiadiazine at zwitterionic phospholipid cell membranes.

The use of drugs derived from benzothiadiazine, which is a bicyclic heterocyclic benzene derivative, has become a widespread treatment for diseases such as hypertension (treated with diuretics such as bendroflumethiazide or chlorothiazide), low blood sugar (treated with non-diuretic diazoxide), or the human immunodeficiency virus, among others. In this work, we have investigated the interactions of benzothiadiazine with the basic components of cell membranes and solvents, such as phospholipids, cholesterol, ions, and water. The analysis of the mutual microscopic interactions is of central impo

Structural basis of membrane skeleton organization in red blood cells.

The spectrin-based membrane skeleton is a ubiquitous membrane-associated two-dimensional cytoskeleton underneath the lipid membrane of metazoan cells. Mutations of skeleton proteins impair the mechanical strength and functions of the membrane, leading to several different types of human diseases. Here, we report the cryo-EM structures of the native spectrin-actin junctional complex (from porcine erythrocytes), which is a specialized short F-actin acting as the central organizational unit of the membrane skeleton. While an α-/β-adducin hetero-tetramer binds to the barbed end of F-actin as a fle

Cnm of Streptococcus mutans is important for cell surface structure and membrane permeability.

Streptococcus mutans, a Gram-positive facultative anaerobic bacterium, is a major pathogen of dental caries. The protein Cnm of S. mutans is involved in collagen binding, but its other biological functions are unknown. In this study, a Cnm-deficient isogenic mutant and a complementation strain were generated from a Cnm-positive S. mutans strain to help determine the properties of Cnm. Initially, comparison of the cell surface structure was performed by electron microscopy, which demonstrated that Cnm appears to be localized on the cell surface and associated with a protruding cell surface stru

The structural relationship between mesangial cells and basement membrane of the renal glomerulus.

It has been shown by many studies that mesangial cell contraction exerts considerable influences on glomerular filtration dynamics. However, experimental findings about the geometrical changes within the glomerular tuft going along with mesangial cell contractions are lacking. This study analyzes the geometry of mesangial cells and their relationship to glomerular capillaries, especially to the glomerular basement membrane (GBM). By applying a new staining technique of unosmicated specimens for TEM, the cellular outlines of glomerular cells (mesangial, endothelial and epithelial) and the distr

The physicochemical design, structure and permeability of the JCVI-syn3A minimal cell membrane.

Minimal cells offer a platform to uncover the fundamental physicochemical principles of cellular life. While genomes, proteomes, and metabolic networks of cells have been elucidated, the lipidome, which determines the physicochemical identity of the membrane, remains only partially characterized. Yet, the membrane is a central determinant of cellular viability, regulating solute permeability, mechanical stability, and environmental cues. Here, we investigate membranes that recapitulate the lipid composition of the minimal cell JCVI-syn3A, which is distinguished by an unusually high cholesterol

Specific glycosylation of membrane proteins in epithelial ovarian cancer cell lines: glycan structures reflect gene expression and DNA methylation status.

Epithelial ovarian cancer is the fifth most common cause of cancer in women worldwide bearing the highest mortality rate among all gynecological cancers. Cell membrane glycans mediate various cellular processes such as cell signaling and become altered during carcinogenesis. The extent to which glycosylation changes are influenced by aberrant regulation of gene expression is nearly unknown for ovarian cancer and remains crucial in understanding the development and progression of this disease. To address this effect, we analyzed the membrane glycosylation of non-cancerous ovarian surface epithe

Altered basement membrane structure of the spleen in hairy cell leukaemia. Demonstration of laminin in hairy cells.

Changes in five spleens affected by hairy cell leukaemia were characterized using immunohistochemical staining for the basement membrane (BM) components laminin and type IV collagen. The ring fibres had not been totally destroyed, and were well preserved in places. The spaces filled by abnormal blood or hairy cells were assigned to the categories of altered sinus and two types of pseudosinus. In an altered sinus the staining pattern of the ring fibres had gained attenuations and the sinus was sometimes lined by a continuous BM of uneven thickness. The number of altered sinuses was related to t

Role of gangliosides in behavioral and biochemical actions of alcohol: cell membrane structure and function.

Alcohol exerts its pharmacological effects in adult brain by altering the physicochemical properties of cellular plasma membranes. Although alcohol does induce changes in membrane lipid composition, studies to relate these alterations to the development of behavioral tolerance to alcohol and the withdrawal effects have been unsuccessful. Actions of alcohol on developing brain are even more complex. Some of the reported effects include inhibition of embryogenesis, cell migration, and differentiation, including synaptogenesis. Gangliosides have neuroprotective action against a variety of neural

Alterations in the structure and function of the mammalian cell membrane after exposure to ionizing radiation.

Although the nucleus appears to represent the primary target for the killing of cells by ionizing radiation, the cell membrane may be another important site for radiation injury and modification of the radiation response. In this paper, the effects of radiation on various membrane-associated parameters which are expected to affect cellular viability or function are reviewed. These include the effect of radiation on membrane transport, lectin or hormone receptor sites, cell surface electrokinetics, cell membrane fluidity, cell surface topography, etc. Alternative biochemical mechanisms that may

Cryptosporidium: cellular localization, structural analysis of absorptive cell-parasite membrane-membrane interactions in guinea pigs, and suggestion of protozoan transport by M cells.

In ilea of spontaneously infested guinea pigs, we examined the interface between the plasma membranes of cryptosporidia and absorptive cells using thin section and freeze fracture techniques. Initially, cryptosporidia invaginate microvilli, and the resulting redundant folds of membrane envelop the protozoan, thereby internalizing it in a membrane sac of host cell origin. Subsequently, a pentalaminar membrane fusion site develops at the base of the protozoan between the parasite's outer plasma membrane and the internalized host membrane. The membrane domains isolated by this fusion site are the

Nano-structure of vitronectin/heparin on cell membrane for stimulating single cell in iPSC-derived embryoid body.

Individual cell environment stimulating single cell is a suitable strategy for the generation of sophisticated multicellular aggregates with localized biochemical signaling. However, such strategy for induced pluripotent stem cell (iPSC)-derived embryoid bodies (EBs) is limited because the presence of external stimulation can inhibit spontaneous cellular communication, resulting in misdirection in the maturation and differentiation of EBs. In this study, a facile method of engineering the iPSC membrane to stimulate the inner cell of EBs while maintaining cellular activities is reported. We coa

Cerebral Corpora amylacea are dense membranous labyrinths containing structurally preserved cell organelles.

Corpora amylacea are cell-derived structures that appear physiologically in the aged human brain. While their histological identification is straightforward, their ultrastructural composition and microenvironment at the nanoscale have remained unclear so far, as has their relevance to aging and certain disease states that involve the sequestration of toxic cellular metabolites. Here, we apply correlative serial block-face scanning electron microscopy and transmission electron tomography to gain three-dimensional insight into the ultrastructure and surrounding microenvironment of cerebral Corpo

Outer Hair Cell Lateral Wall Structure Constrains the Mobility of Plasma Membrane Proteins.

Nature's fastest motors are the cochlear outer hair cells (OHCs). These sensory cells use a membrane protein, Slc26a5 (prestin), to generate mechanical force at high frequencies, which is essential for explaining the exquisite hearing sensitivity of mammalian ears. Previous studies suggest that Slc26a5 continuously diffuses within the membrane, but how can a freely moving motor protein effectively convey forces critical for hearing? To provide direct evidence in OHCs for freely moving Slc26a5 molecules, we created a knockin mouse where Slc26a5 is fused with YFP. These mice and four other strai
