Novel Properties for Endoglucanase Acquired by Cell-Surface Display Technique.

In order to improve the stability of endoglucanase under thermal and acidic conditions, the endoglucanase gene was fused to the N-terminus of the Saccharomyces cerevisiae pir gene, encoding the cell wall protein PIR. The fusion gene was transformed into Pichia pastoris GS115 for expression. A resulting strain with high expression and high activity was identified by examining resistance to Geneticin 418, Congo red staining, and quantitative analysis of enzyme activity. SDS-PAGE analysis revealed that the endoglucanase was successfully displayed on the yeast cell surface. The displayed endogluca

Phage display-derived human antibodies in clinical development and therapy.

Over the last 3 decades, monoclonal antibodies have become the most important class of therapeutic biologicals on the market. Development of therapeutic antibodies was accelerated by recombinant DNA technologies, which allowed the humanization of murine monoclonal antibodies to make them more similar to those of the human body and suitable for a broad range of chronic diseases like cancer and autoimmune diseases. In the early 1990s in vitro antibody selection technologies were developed that enabled the discovery of "fully" human antibodies with potentially superior clinical efficacy and lowes

Characterization of dental allergy associated polymer surfaces by phage display technique.

Many materials are used during a dental treatment, which can cause dental allergy in susceptible patients. Behind the immune response there are multistage biochemical processes, in which protein interactions can be investigated by directed evolutionary, phage display technique. Aim of this study is to determine the frequent amino acid sequences on polystyrene surface as dental base thermoplastic polymers by phage display technique. Positive control was streptavidin (Str) covered polymer, the negative control was bovine serum albumin blocked polystyrene. Based on our results it was found that t

CEN-Display: Construction and optimization of a surface display system in Saccharomyces cerevisiae CEN.PK2-1C.

Over the past decades, yeast surface display (YSD) technology has emerged as a powerful biotechnological tool with broad applications in biomedicine, industrial catalysis, and environmental science. However, its efficiency, stability and applicability are often limited by proteolytic degradation during secretion in commonly used strains and steric hindrance associated with anchoring architectures. In this study, we developed a high-performance platform, "CEN-Display," using a systematic engineering strategy on the Saccharomyces cerevisiae CEN.PK2-1C strain to expand the YSD toolbox for new cha

INTRODUCTION/OBJECTIVE: Antibodies have broad applications in various fields, such as biology and medicine. The screening and preparation of highly specific and sensitive antibodies are essential research areas. Several techniques for the preparation of mouse-derived antibodies have been developed, but limited studies on rabbit-derived antibodies with a broader antibody profile and easier humanization are reported. An improved yeast surface display technique was used for rapid screening of rabbit-derived Fab antibodies. METHODS: After RNA extraction from peripheral rabbit blood, a cDNA librar

Peptidic inhibitors of insulin-degrading enzyme with potential for dermatological applications discovered via phage display.

Insulin-degrading enzyme (IDE) is an atypical zinc-metalloendopeptidase that hydrolyzes insulin and other intermediate-sized peptide hormones, many of which are implicated in skin health and wound healing. Pharmacological inhibitors of IDE administered internally have been shown to slow the breakdown of insulin and thereby potentiate insulin action. Given the importance of insulin and other IDE substrates for a variety of dermatological processes, pharmacological inhibitors of IDE suitable for topical applications would be expected to hold significant therapeutic and cosmetic potential. Phage Display-Derived Monoclonal Antibodies Against Internalins A and B Allow Specific Detection of Listeria monocytogenes.

Listeria monocytogenes is the causative agent of listeriosis, a highly lethal disease initiated after the ingestion of Listeria-contaminated food. This species comprises different serovars, from which 4b, 1/2a, and 1/2b cause most of the infections. Among the different proteins involved in pathogenesis, the internalins A (InlA) and B (InlB) are the best characterized, since they play a major role in the enterocyte entry of Listeria cells during early infection. Due to their covalent attachment to the cell wall and location on the bacterial surface, along with their exclusive presence in the pa

Whole-cell surface-display system in attenuated Salmonella Enteritidis for slide microagglutination-based antibody detection.

BACKGROUND: Microagglutination is a sensitive, rapid, and cost-effective serological assay widely used for disease diagnosis and surveillance in animals and humans. However, it traditionally relies on culturing live pathogenic bacteria as antigens, posing biosafety risks and limiting its application primarily to bacterial pathogens. Bacterial surface display technology offers a promising alternative by enabling the presentation of heterologous proteins or peptides on the bacterial surface, avoiding the need to culture the target pathogen, simplifying antigen preparation, and enabling potential

Pitfalls to avoid when using phage display for snake toxins.

Antivenoms against bites and stings from snakes, spiders, and scorpions are associated with immunological side effects and high cost of production, since these therapies are still derived from the serum of hyper-immunized production animals. Biotechnological innovations within envenoming therapies are thus warranted, and phage display technology may be a promising avenue for bringing antivenoms into the modern era of biologics. Although phage display technology represents a robust and high-throughput approach for the discovery of antibody-based antitoxins, several pitfalls may present themselv

[Microenvironmental regulation strategies and applications of yeast surface display systems].

Yeast surface display (YSD) is an important technological platform integrating protein engineering, synthetic biology, and industrial biocatalysis. Its catalytic efficiency and stability highly depend on the dynamic synergy of the three-tier microenvironments: the cell wall, intracellular environment, and extracellular compartments. This review systematically elaborates on the core components of the YSD microenvironment, including the cell wall, anchor proteins, intracellular secretion pathways, and extracellular physicochemical properties, and clarifies their critical roles in the folding, ac

Application of phage display for T-cell receptor discovery.

The immune system is tasked to keep our body unharmed and healthy. In the immune system, B- and T-lymphocytes are the two main components working together to stop and eliminate invading threats like virus particles, bacteria, fungi and parasite from attacking our healthy cells. The function of antibodies is relatively more direct in target recognition as compared to T-cell receptors (TCR) which recognizes antigenic peptides being presented on the major histocompatibility complex (MHC). Although phage display has been widely applied for antibody presentation, this is the opposite in the case of

Bridging discovery and therapy in precision medicine-Phage display as a catalyst for nanobody innovation: A review.

Monoclonal antibodies, particularly immunoglobulin G (IgG)-based formats, have shown significant efficacy in targeted therapies; however, they face limitations such as poor tissue penetration, complex manufacturing, and antigen escape. These limitations have driven the development of alternative therapeutic strategies, such as bispecific antibodies, antibody-drug conjugates, and chimeric antigen receptor T cells, each with their own technical and translational challenges. Nanobodies (Nbs), derived from camelid heavy-chain-only antibodies, are the smallest functional antibody fragments and are

Brain shuttle peptides derived from phage display.

Vascular beds within different tissues, including the brain, exhibit diverse molecular heterogeneity. Screening of peptide libraries using in vivo phage display exploits this heterogeneity, enabling the discovery of peptide ligands which selectively home to specific tissues. A prime example is identification of the iron-mimicking peptide CRTIGPSVC which undergoes receptor-mediated transport across the blood-brain barrier (BBB) by binding to the transferrin/transferrin receptor (TfR) complex. Various other brain-homing and BBB-crossing peptides have been discovered by in vivo phage display as w

A tethered yeast surface display system facilitates enriching epitope-specific binding interactions.

Engineering proteins for therapeutic applications is a field that has seen substantial growth in the past two decades, but challenges remain. A major deficiency in current strategies is the lack of a means to efficiently screen for binding to key epitopes to yield a desired function. To meet this challenge, we designed a tethered yeast surface display construct that leverages high local concentrations by tethering a candidate protein binder to a target protein of interest (POI) via a flexible peptide linker. These high local concentrations enable screening of epitope-specific binders based on

NGS and the design of an optimized phage display workflow for peptide discovery.

Phage display is a powerful technology that has demonstrated great potential in identifying peptides with high binding affinity and specificity toward a broad spectrum of biological targets. The undesired enrichment of nonspecific binders remains a major challenge in phage display selection. The integration of next-generation sequencing (NGS) into phage display has expanded the horizons of ligand discovery by significantly enhancing our ability to interrogate the massive sequence space of combinatorial phage display libraries and providing quantitative information about their composition and e

cDNA phage display for the discovery of theranostic autoantibodies in rheumatoid arthritis.

Rheumatoid arthritis (RA) is the world's most common autoimmune disease mainly characterized by a chronic inflammation of multiple synovial joints. Rheumatologists now have a whole range of treatment options including glucocorticoids (GCs), classical synthetic and biological disease-modifying antirheumatic drugs (cs- and bDMARDS), resulting in a tremendous improvement in treatment outcomes for RA patients over the last two decades. Despite this progress, the choice of treatment regimen to achieve stable remission at the individual patient level still largely depends on trial and error. In this

Discovery of Landscape Phage Probes Against Cellular Communication Network Factor 1 (CCN1/Cyr61).

Detection of cancer biomarkers at the earliest stages of disease progression is commonly assumed to extend the overall quality of life for cancer patients as the result of earlier clinical management of the disease. Therefore, there is an urgent need for the development of standardized, sensitive, robust, and commonly available screening and diagnostic tools for detecting the earliest signals of neoplastic pathology progression. Recently, a new paradigm of cancer control, known as multi-cancer detection (MCD), evolved, which measures the composition of cancer-related molecular analytes in the

Identification of chalcopyrite-binding peptides for flotation applications using phage display and deep sequencing.

Phage display is a powerful technology for identifying new ligands, including mineral-selective peptides. However, it is prone to several biases that may lead to false positives or false negatives, particularly due to non-specific binding, amplification bias, and the limited number of clones typically sequenced. By combining phage display with deep sequencing, we detected hundreds of thousands of chalcopyrite-binding peptides across different rounds of selection. Adsorption assays confirmed the strong affinity of the LNSSLRL peptide when produced de novo by chemical synthesis, and demonstrated

Phage Display Biopanning Strategy for Discovery of Nanoparticle-Binding Peptides.

The phage display technology allows to select peptides, exposed on the surface of bacteriophage virions, which can specifically bind to virtually any materials, including proteins, lipids, carbohydrates, and other organic macromolecules, as well as metals or metal oxides. In this chapter, we describe and discuss procedures for effective selection of peptides characterized with affinity to metallic or metal oxide nanoparticles. The use of a peptide library displayed on virions of the M13 bacteriophage, the most commonly used phage display system, is presented. Specific notes indicate what are c

Intelligent antigen design combined with yeast display enhances targeted immune responses.

Inducing strong and targeted humoral immune responses, a central goal of next-generation vaccine development, involves a complex series of interconnected steps. However, a comprehensive study that addresses and integrates all steps is still missing. Here, we present a study that addresses these steps to elicit precise neutralizing antibody (nAb) responses targeting focused epitope motifs. We present a new protein design model called EpiTopoFold for constructing a topology that precisely accommodates and stabilizes the epitope motif. After ten thousand in silico simulations, we designed a de no
