Modeling Osteoporotic Spongy Bone Using Cellular Structures.

The mechanical properties of spongy bone are a function of the tissue's microstructure including the strength and microarchitecture of the trabeculae. The thickness and number of trabeculae (morphological data) are the most important parameters that can indicate the porosity of tissue. Osteoporosis is known as a decrease in the thickness or loss of trabeculae that causes a reduction in the density and strength of bone. This research aims to model osteoporosis using cellular (lattice) structures and also presents mechanical relationships to predict tissue behavior during osteoporosis using morp

Actions of ultraviolet light on cellular structures.

Solar radiation is the primary source of human exposure to ultraviolet (UV) radiation. Overexposure without suitable protection (i.e., sunscreen and clothing) has been implicated in mutagenesis and the onset of skin cancer. These effects are believed to be initiated by UV-mediated cellular damage, with proteins and DNA as primary targets due to a combination of their UV absorption characteristics and their abundance in cells. UV radiation can mediate damage via two different mechanisms: (a) direct absorption of the incident light by the cellular components, resulting in excited state formation

The Riveting Cellular Structures of Apicomplexan Parasites.

Parasitic protozoa of the phylum Apicomplexa cause a range of human and animal diseases. Their complex life cycles - often heteroxenous with sexual and asexual phases in different hosts - rely on elaborate cytoskeletal structures to enable morphogenesis and motility, organize cell division, and withstand diverse environmental forces. This review primarily focuses on studies using Toxoplasma gondii and Plasmodium spp. as the best studied apicomplexans; however, many cytoskeletal adaptations are broadly conserved and predate the emergence of the parasitic phylum. After decades cataloguing the co

Alteration of skeletal muscle cellular structures by potassium depletion.

After rats had been fed a low-potassium diet for 4 to 8 weeks, skeletal muscle showed ultrastructural changes involving membranous organelles. Mitochondria were often swollen, condensed, or disintegrated. The transverse tubules were disoriented, focally dilated, and tortuous. The sarcoplasmic reticulum showed various degrees of dilatation. Vacuoles of different sizes occurred frequently. Whirls of membranes were closely associated with any of the membranous organelles, especially near an orifice of a transverse tubule. Supplementation of potassium reversed these changes. These findings are ver

[Reparative and neoplastic spheroid cellular structures and their mathematical model].

Spheroid cell structures in the cell cultures have been described and are used for studying cell-cell and cell- matrix interactions. At the same time, spheroid cell structure participation in the repair and development of cancer in vivo remains unexplored. The aim of this study was to investigate the cellular composition of spherical structures and their functional significance in the repair of squamous epithelium in human papilloma virus-associated cervical pathology--chronic cervicitis and cervical intraepithelial neoplasia 1-3 degree, and also construct a mathematical model to explain the d

Saturated fatty acids: simple molecular structures with complex cellular functions.

PURPOSE OF REVIEW: This review summarizes recent findings on the biological functions of saturated fatty acids. Some of these findings suggest that saturated fatty acids may have important and specific regulatory roles in the cells. Until now these roles have largely been outweighed by the negative impact of dietary saturated fatty acids on atherosclerosis biomarkers. Elucidated biochemical mechanisms like protein acylation (N-myristoylation, S-palmitoylation) and putative physiological roles are described. RECENT FINDINGS: The review will focus on the following topics: new aspects on the met

Advances in Small Molecules in Cellular Reprogramming: Effects, Structures, and Mechanisms.

The method of cellular reprogramming using small molecules involves the manipulation of somatic cells to generate desired cell types under chemically limited conditions, thus avoiding the ethical controversy of embryonic stem cells and the potential hazards of gene manipulation. The combinations of small molecules and their effects on mouse and human somatic cells are similar. Several small molecules, including CHIR99021, 616452, A83-01, SB431542, forskolin, tranylcypromine and valproic acid [VPA], have been frequently used in reprogramming of mouse and human somatic cells. This indicated that

Effects of ionizing radiation on cellular structures, induced instability and carcinogenesis.

Ionizing radiation is perhaps the most extensively studied human carcinogen. There have been a number of epidemiological studies on human populations exposed to radiation for medical or occupational reasons, as a result of protracted environmental exposures due to radiation accidents, or after atomic bombings. As a result of these studies exposure to ionizing radiation has been unambiguously linked to cancer causation. While cancer induction is the primary concern and the most important somatic effect of exposure to ionizing radiation, potential health risks do not only involve neoplastic dise

Tau physiology and pathology: impacts on cellular structures and neurodegenerative diseases.

This review explores the crucial roles of the tau protein in neuronal integrity and its dysregulation in neurodegenerative diseases (NDs), particularly tauopathies. Key features include abnormal tau phosphorylation, leading to insoluble aggregates and neuronal dysfunction. Various therapeutic strategies, such as reducing tau phosphorylation, inhibiting aggregation, and enhancing clearance through autophagy and immunotherapies, are discussed. Promising candidates such as anle138b and methylene blue display efficacy in preclinical models. The interplay between tau and Aβ pathology is also highli

Annulate lamellae - an underexplored cellular structure.

Annulate lamellae (AL) are endoplasmic reticulum (ER) subdomains harbouring a subset of nucleoporins (Nups), the proteins that assemble into the nuclear pore complexes (NPCs) on the nuclear envelope (NE). AL have been observed in a variety of cell types, including oocytes, spermatids, embryonic cells, somatic cells and tumour cells, as well as in multiple cell lines. Some studies propose that AL derive from the NE, whereas studies in Drosophila egg chambers indicate that AL can assemble through differential condensation of soluble Nups, thus implying that modes of AL assembly can vary dependin

Cellular structure and biology of Dupuytren's disease.

Numerous studies support the idea that the myofibroblast is a key cell responsible for the tissue contraction in Dupuytren's disease. In vitro models have been developed to study the underlying cellular basis of myofibroblast differentiation and contraction. Studies suggest that the growth factor TGF-beta 1 combined with mechanical stress can promote the differentiation of fibroblasts into myofibroblasts. Agonists, such as LPA and thrombin, can promote the contraction of myofibroblasts through specific intracellular signaling pathways that regulate levels of phosphorylated myosin light chain.

A cellular basis for mapping behavioural structure.

To flexibly adapt to new situations, our brains must understand the regularities in the world, as well as those in our own patterns of behaviour. A wealth of findings is beginning to reveal the algorithms that we use to map the outside world1-6. However, the biological algorithms that map the complex structured behaviours that we compose to reach our goals remain unknown. Here we reveal a neuronal implementation of an algorithm for mapping abstract behavioural structure and transferring it to new scenarios. We trained mice on many tasks that shared a common structure (organizing a sequence of

Structural and cellular biology of rhabdovirus entry.

Rhabdoviruses are enveloped viruses with a negative-sense single strand RNA genome and are widespread among a great variety of organisms. In their membrane, they have a single glycoprotein (G) that mediates both virus attachment to cellular receptors and fusion between viral and endosomal membranes allowing viral genome release in the cytoplasm. We present structural and cellular aspects of Rhabdovirus entry into their host cell with a focus on vesicular stomatitis virus (VSV) and rabies virus (RABV) for which the early events of the viral cycle have been extensively studied. Recent data have

Tunneling Nanotubes are Novel Cellular Structures That Communicate Signals Between Trabecular Meshwork Cells.

PURPOSE: The actin cytoskeleton of trabecular meshwork (TM) cells plays a role in regulating aqueous humor outflow. Many studies have investigated stress fibers, but F-actin also assembles into other supramolecular structures including filopodia. Recently, specialized filopodia called tunneling nanotubes (TNTs) have been described, which communicate molecular signals and organelles directly between cells. Here, we investigate TNT formation by TM cells. METHODS: Human TM cells were labeled separately with the fluorescent dyes, DiO and DiD, or with mitochondrial dye. Fixed or live TM cells were

A comparison of anatomic and cellular transcriptome structures across 40 human brain diseases.

Genes associated with risk for brain disease exhibit characteristic expression patterns that reflect both anatomical and cell type relationships. Brain-wide transcriptomic patterns of disease risk genes provide a molecular-based signature, based on differential co-expression, that is often unique to that disease. Brain diseases can be compared and aggregated based on the similarity of their signatures which often associates diseases from diverse phenotypic classes. Analysis of 40 common human brain diseases identifies 5 major transcriptional patterns, representing tumor-related, neurodegenerat

Expansion Microscopy reveals changes in cellular and extracellular structures between healthy and perturbed tendons.

Tendons transmit mechanical forces between muscles and bones through their highly aligned, collagen-rich extracellular matrix. When damaged, resident cells help restore this matrix. However, in tendinopathies, this repair response fails, leading to loss of proper tendon function. How altered mechanical states reshape tendon cell and matrix architecture remains poorly understood because existing methods do not readily capture tendon structure across relevant length scales. Here, we combine Fluorescent Labeling of Abundant Reactive Entities (FLARE) with Expansion Microscopy (ExM) to visualize ce
