A minimal cellulosome-like system in Cellulosilyticum lentocellum.

Cellulosomes are efficient enzymatic nanomachines which have arisen for the degradation of cellulosic biomass. They are found abundantly in soil-dwelling microbes and bacteria which thrive in the stomachs of ruminant mammals. Two protein domains, cohesins and dockerins, characterise cellulosomes. These domains interact with each other to form, in many cases, enormous complexes with as many as 160 individual proteins. However, genome annotation of Cellulosilyticum lentocellum DSM 5427 revealed a single cohesin domain (encoded by Clole_2599) and a single dockerin domain (Clole_2598). Therefore,

Enhanced biodegradation of sticky deposits through cutinase-lipase synergy in designer cellulosomes.

Sticky deposits hinder the recycling of waste paper by causing equipment clogging and poor paper quality. Enzymatic degradation is constrained by poor enzyme stability and limited degradation capacity. Herein, Humicola insolens cutinase and Archeoglobus fulgidus carboxylesterase, were selected to construct a cellulosome-based complex (namely HACOM) to enhance the binding and degradation of sticky deposits and the stability of enzymes. Computational analysis and protein adsorption experiments revealed that the carbohydrate-binding module could interact with the substrate poly (vinyl acetate) (P

Mechanistic partitioning and functional synergy of bacterial cellulosome and fungal cellulases in cellulose saccharification.

Synergistic saccharification involving bacterial cellulosome and fungal cellulases holds significant promise for industrial applications; however, this integrated approach remains largely under-investigated. This study dissects the synergy from structural characterization, adsorption analysis, crystallinity evolution, and hydrolysis performance evaluation. The cellulosome exhibited superior enhancement of substrate accessibility after enzymatic hydrolysis of sugarcane bagasse compared with fungal cellulases. The distinct changes in crystallinity observed during the hydrolysis of microcrystalli

Creating a large designer cellulosome in yeast to boost ethanol production.

Cellulosic biomass represents a promising feedstock for biofuel and biochemical production. However, its recalcitrant structure strongly hinders enzymatic degradation. Cellulosomes are large multienzyme complexes, highly efficient at degrading cellulose. A cellulase in a cellulosome has a dockerin domain that binds to a cohesin module on the CipA (cellulosome integrating protein A). In a native cellulosome all cohesins are identical, so that the cellulase types and their positions in a CipA cannot be controlled. Here, we constructed the largest designer CipA known to date. Using innovative tec

Multidomain structure and cellulosomal localization of the Clostridium thermocellum cellobiohydrolase CbhA.

The nucleotide sequence of the Clostridium thermocellum F7 cbhA gene, coding for the cellobiohydrolase CbhA, has been determined. An open reading frame encoding a protein of 1,230 amino acids was identified. Removal of a putative signal peptide yields a mature protein of 1,203 amino acids with a molecular weight of 135,139. Sequence analysis of CbhA reveals a multidomain structure of unusual complexity consisting of an N-terminal cellulose binding domain (CBD) homologous to CBD family IV, an immunoglobulin-like beta-barrel domain, a catalytic domain homologous to cellulase family E1, a duplica

Single-cell analysis of yeast surface display for designer cellulosome applications using a fluorescent protein complex.

Designer cellulosomes (DCs) are precisely engineered multi-enzyme complexes aimed at lignocellulose saccharification. Achieving spontaneous designer cellulosome display on yeast cell surfaces has been a long-term objective to enhance consolidated bioprocesses with concurrent ethanol production. A "self-assembly" approach involves simultaneous scaffoldin display and docking enzyme secretion. However, challenges arise from the size and complexity of designer cellulosomes, coupled with the yeast cells' limited capacity for heterologous protein expression. A comprehensive examination of Saccharomy

AlphaFold-driven structural proteomics reveals extensive cellulosome machinery in human ruminococcal symbionts.

Cellulosomes are large, surface-displayed enzyme complexes that enable anaerobic bacteria to degrade recalcitrant plant polysaccharides, yet cellulosome-expressing bacteria are thought to be rare in the human gut. Here, we show that extensive sequence divergence obscures the detection of many ruminococcal cellulosomes by conventional sequence homology-based methods. Using proteome-scale AlphaFold2 structural predictions, we uncovered a substantially expanded set of putative cellulosome-producing Ruminococcus species, including six previously unrecognized human symbionts. Structure-based cluste

Cellulosome engineering as biological macromolecular assembly for lignocellulose deconstruction: Structure, interactions, and functional design.

Lignocellulosic biomass is a heterogeneous solid matrix whose biological deconstruction is limited by cellulose accessibility, lignin exposure, pore-scale transport, and enzyme stability at solid-liquid interfaces. Although bacterial cellulosomes provide a natural strategy for organizing multiple enzymes through scaffoldin-mediated cohesin-dockerin interactions, their engineering value depends on more than enzyme colocalization. This review frames cellulosome engineering as a process-aware macromolecular assembly problem, in which catalytic balance, substrate targeting, assembly size, inter-do

Improving cellulosic ethanol production by an engineered yeast consortium displaying a pentafunctional mini-cellulosome.

As a traditional ethanol-producing microorganism, Saccharomyces cerevisiae is an ideal host for consolidated bioprocessing. However, when overloaded cellulase genes are expressed in yeast, the metabolic burden on cells may greatly affect cell growth and cellulosic ethanol production. In this study, we developed a yeast consortium system that secretes and assembles five types of cellulases on the yeast cell surface to improve cellulosic ethanol production. This system involves one display strain, which provides the scaffoldin on the surface and several secretion strains that secrete each cellul

Reconstruction of a resource balance analysis model of Clostridium thermocellum examines the metabolic cost of glycolytic and cellulosome enzymes.

Clostridium thermocellum is an increasingly well-studied organism with considerable advantages for consolidated bioprocessing towards ethanol production. Here, a genome-scale resource balance analysis (RBA) model of C. thermocellum, ctRBA, is reconstructed based on a recently published stoichiometric model (iCTH669), global proteomics, and 13C MFA datasets to analyze proteome allocation and the burden imposed on metabolism with regard to ethanol yield and titer. Glycolytic and fermentation enzyme concentrations were accurately quantified by the model, with glyceraldehyde-3-phosphate dehydrogen

The cellulase/hemicellulase system of the anaerobic fungus Orpinomyces PC-2 and aspects of its applied use.

Anaerobic fungi, first described in 1975 by Orpin, live in close contact with bacteria and other microorganisms in the rumen and caecum of herbivorous animals, where they digest ingested plant food. Seventeen distinct anaerobic fungi belonging to five different genera have been described. They have been found in at least 50 different herbivorous animals. Anaerobic fungi do not possess mitochondria, but instead have hydrogenosomes, which form hydrogen and carbon dioxide from pyruvate and malate during fermentation of carbohydrates. In addition, they are very oxygen- and temperature-sensitive, a

Genome Wide Analysis Reveals the Extrinsic Cellulolytic and Biohydrogen Generating Abilities of Neocallimastigomycota Fungi.

Ruminating animals, especially cattle lack the carbohydrate active enzyme encoding genes which are required for the degradation of the glycosidic linkages of plant cell wall carbohydrates (such as cellulose, hemicellulose, lignin and pectin). Thus, ruminating animals are completely dependent on the microorganisms (anaerobic bacteria and fungi, methanogenic archaea and protozoa) residing in their rumen (hindgut). In this study, we have retrieved and analyzed the complete genome wide annotations of the Neocallimastigomycota division fungi such as Anaeromyces robustus, Neocallismatix californiae,

Metabolic engineering of Ruminiclostridium papyrosolvens for 2,3-butanediol production from lignocellulose.

Ruminiclostridium papyrosolvens is an anaerobic, mesophilic, and cellulolytic bacterium capable of secreting cellulosomes for efficient lignocellulose degradation, making it a promising candidate for consolidated bioprocessing (CBP). However, its native cellulolytic activity is repressed by cellobiose, and its natural fermentation products are limited to low-value compounds. In this study, we firstly discovered that the cellulosomal cip-cel gene cluster is subject to cellobiose-mediated carbon catabolite repression (CCR). By replacing its native promoter with the endogenous cellobiose/cellodex

The curdlan-type exopolysaccharide produced by Cellulomonas flavigena KU forms part of an extracellular glycocalyx involved in cellulose degradation.

The genus Cellulomonas is comprised of a group of Gram-positive, soil bacteria capable of utilizing cellulose as their sole source of carbon and energy. Cellulomonas flavigena KU was originally isolated from leaf litter and subsequently shown to produce large quantities of a curdlan-type (beta-1,3-glucan) exopolysaccharide (EPS) when provided with an excess of glucose or other soluble carbon-source. We report here that curdlan EPS is also produced by Cellulomonas flavigena KU when growing on microcrystalline cellulose in mineral salts-yeast extract media. Microscopic examination of such cultur

Enhancement of novel cello-xylosome saccharification performance via microbial surface display on lignocellulosic feedstocks.

Cellulosomes are regarded as the most intricate naturally occurring cellulolytic systems. This study reports the design and surface assembly of a novel artificial cello-xylosome on Escherichia coli BL21:ΔCsgA cells. This synthetic complex incorporates two highly active enzymes previously sourced from sheep rumen microbiota, namely the glucanase IDSGLUC5-28 and the xylanase IDSXYN10-1. The structural integrity of the designer cello-xylosome was confirmed using a combined approach involving western blot analysis, zymography, and immunofluorescence microscopy. Alkaliphilic microbial cellulases: sources, structural insights, and bioengineering approaches for industrial applications.

As the biofuel/biorefinery/green economy advances quickly, the need for durable cellulases to operate under varying conditions in the industrial sector has increased. A specific type of cellulase that has emerged from this demand is alkaliphilic cellulase (ALKC). Due to their stability and high catalytic activity at elevated pH ranges (e.g., pH 8-11), ALKCs are being developed as biocatalysts for alkaline pretreatment and other harsh, high-pH industrial processes. The purpose of this review is to provide an updated overview of ALKCs focusing on their microbial origins, structural characteristic

Engineering a high-performance isopropyl alcohol biosensor using a bio-nanocomposite multienzyme platform for real-time bioprocess monitoring.

The spatial organization of multi-enzyme cascades offers a powerful strategy to overcome diffusion limitations and cofactor regeneration bottlenecks in industrial biocatalysis. In this study, a cellulosome-inspired complex was engineered to facilitate the efficient, self-sufficient oxidation of isopropanol (IPA). By exploiting high-affinity cohesin-dockerin interactions, a dockerin-fused secondary alcohol dehydrogenase (S-ADH_doc) was co-immobilized with a dockerin-fused alkyl hydroperoxide reductase (AhpF_doc), serving as a robust NADH regeneration partner. Synergic action of an inserted carbohydrate-binding module in a glycoside hydrolase family 5 endoglucanase.

Most known cellulase-associated carbohydrate-binding modules (CBMs) are attached to the N- or C-terminus of the enzyme or are expressed separately and assembled into multi-enzyme complexes (for example to form cellulosomes), rather than being an insertion into the catalytic domain. Here, by solving the crystal structure, it is shown that MtGlu5 from Meiothermus taiwanensis WR-220, a GH5-family endo-β-1,4-glucanase (EC 3.2.1.4), has a bipartite architecture consisting of a Cel5A-like catalytic domain with a (β/α)8 TIM-barrel fold and an inserted CBM29-like noncatalytic domain with a β-jelly-rol

The gastrointestinal microbiome in dairy cattle is constrained by the deterministic driver of the region and the modified effect of diet.

BACKGROUND: Dairy cattle (Bos taurus), especially Holstein cows, which are the highest-producing dairy animals and are widely bred to provide milk products to humans, rely critically on their associated gastrointestinal tract (GIT) microbiota to digest plant feed. However, the region-specific taxonomic composition and function of the GIT microbiome in dairy cattle and the mechanistic basis for the diet-induced effects remain to be elucidated.  RESULTS: We collected 120 digesta samples from 10 GIT regions of 12 Holstein cows fed forage- and grain-based diets and characterized their GIT microbio

Combining sequence-based approaches with anaerobic microbiology and modelling to understand gut microbial communities.

Gut micro-organisms possess biochemical capabilities that far exceed those of their mammalian hosts, particularly in the ability to gain energy from the breakdown of diet-derived plant material (fibre). This article reviews investigations into gut microbial communities conducted by Harry Flint and his research group. First, extracellular cellulosome and amylosome enzyme complexes were found to mediate the breakdown of plant cell walls and resistant starch by specialised Firmicutes bacteria, both in the human colon and in the rumen. In contrast, Bacteroidetes (Bacteroides, Prevotella spp.) rely
