Understanding Lignin Oxidation by a Two-Domain Multicopper Oxidase from Cellvibrio japonicus.

Bacterial two-domain multicopper oxidases (2dMCOs) represent a structurally distinct class of trimeric multicopper oxidases. Differing considerably from well-characterized monomeric three-domain laccases, type B 2dMCOs have a T1 active site positioned in a tunnel in the trimer center. The biochemical properties and roles in lignin conversion of 2dMCOs remain poorly understood. Here, we present a comprehensive biochemical characterization of a type B 2dMCO from Cellvibrio japonicus (CjMCO) and discuss links between its structural organization and activity. The T1 copper of CjMCO had a redox pot

Flavonoid-associated microbial defense and Cellvibrio sp. C74 pretreatment collectively enhance straw degradation by Stropharia rugosoannulata.

Stropharia rugosoannulata efficiently utilizes straw in natural habitats, yet how it simultaneously defends against competing environmental microorganisms and degrades lignocellulose remains unclear. Here, transcriptomic, metabolomic, and microbiome analyses were integrated with flavonoid antibacterial assays and bacterial pretreatment experiments to characterize fungus-bacterium interactions during straw degradation. The flavonoid biosynthesis-related genes chi, fns, and fls were significantly upregulated during degradation, and the total flavonoid content reached 101.06 μg/g dry weight at th

Insights into the molecular determinants of substrate specificity in glycoside hydrolase family 5 revealed by the crystal structure and kinetics of Cellvibrio mixtus mannosidase 5A.

The enzymatic hydrolysis of the glycosidic bond is central to numerous biological processes. Glycoside hydrolases, which catalyze these reactions, are grouped into families based on primary sequence similarities. One of the largest glycoside hydrolase families is glycoside hydrolase family 5 (GH5), which contains primarily endo-acting enzymes that hydrolyze beta-mannans and beta-glucans. Here we report the cloning, characterization, and three-dimensional structure of the Cellvibrio mixtus GH5 beta-mannosidase (CmMan5A). This enzyme releases mannose from the nonreducing end of mannooligosacchar

Unifying themes and distinct features of carbon and nitrogen assimilation by polysaccharide-degrading bacteria: a summary of four model systems.

Our current understanding of enzymatic polysaccharide degradation has come from a huge number of in vitro studies with purified enzymes. While this vast body of work has been invaluable in identifying and characterizing novel mechanisms of action and engineering desirable traits into these enzymes, a comprehensive picture of how these enzymes work as part of a native in vivo system is less clear. Recently, several model bacteria have emerged with genetic systems that allow for a more nuanced study of carbohydrate active enzymes (CAZymes) and how their activity affects bacterial carbon metaboli

Integrated Adaptive Responses of Rhizosphere Microbial Communities and Functional Genes to Cadmium Stress in Ryegrass (Lolium perenne L.).

Heavy metal pollution impacts on soil microorganisms are of growing concern. This study employed high-throughput sequencing and qPCR to investigate rhizosphere microbial communities and functional genes responses in ryegrass (Lolium perenne L.) under cadmium (Cd) stress. Results showed dose-dependent shifts in microbial diversity, metabolic pathways and functional gene abundance. The relative abundances of bacterial classes Alphaproteobacteria and Bacteroidia and genera Cellvibrio and Algoriphagus increased with Cd levels, suggesting roles in Cd adaptation. The classes Actinobacteria, Sacchari

Differential Microbial Signature Associated With Benign Prostatic Hyperplasia and Prostate Cancer.

Apart from other risk factors, chronic inflammation is also associated with the onset of Prostate Cancer (PCa), wherein pathogen infection and tissue microbiome dysbiosis are known to play a major role in both inflammatory response and cancer development. However, except for a few studies, the link between microbes and PCa remained poorly understood. To explore the potential microbiome signature associated with PCa in Indian patients, we investigated differential compositions of commensal bacteria among patients with benign prostatic hyperplasia (BPH) and PCa using 16S rRNA amplicon sequencing

Custom fabrication of biomass containment devices using 3-D printing enables bacterial growth analyses with complex insoluble substrates.

Physiological studies of recalcitrant polysaccharide degradation are challenging for several reasons, one of which is the difficulty in obtaining a reproducibly accurate real-time measurement of bacterial growth using insoluble substrates. Current methods suffer from several problems including (i) high background noise due to the insoluble material interspersed with cells, (ii) high consumable and reagent cost and (iii) significant time delay between sampling and data acquisition. A customizable substrate and cell separation device would provide an option to study bacterial growth using optica

Toxicological Evaluation of Zingiber purpureum Extract and Its Effects on Gut Microbiota in Zebrafish.

Zingiber purpureum Roscoe (Phlai) is renowned in Southeast Asia for its anti-inflammatory, antioxidant, and antibacterial activities, yet its safety and effects on gut health in vivo are not well characterized. This study investigated the biological effects of ethanolic-soluble Phlai extract (ESPE) in zebrafish (Danio rerio), focusing on chemical marker standardization in phenylbutanoids and cucurminoids, developmental toxicity, gut microbiota investigation, and tissue histopathology. Zebrafish embryos were exposed to ESPE to assess toxicity while adult females received an ESPE-enriched diet c

Effects of vermicompost leachate on nitrogen, phosphorus, and microbiome in a food waste bioponic system.

Food waste is rich in nutrients, such as nitrogen and phosphorus, and can be integrated with bioponics, a closed-loop agricultural system that combines hydroponics with biological nutrient recovery. Vermicompost leachate (VCL) supplementation has been shown to improve the co-composting of organic waste (i.e., compost quality) and the biodegradation of organic compounds. Thus, VCL has high potential for enhancing nutrient availability in bioponics from food waste. However, the understanding of nitrogen and phosphorus availability in food waste-based bioponics is limited, both with and without V

Soil microbiome perturbation impedes growth of Bouteloua curtipendula and increases relative abundance of soil microbial pathogens.

Bouteloua curtipendula (sideoats grama) is a valuable prairie grass for livestock forage, supporting food webs of herbivorous insects, reducing soil erosion, and limiting weed infiltration in urban grasslands. Efficient establishment of B. curtipendula in prairie restorations and urban plantings could drastically improve long-term functionality of the space. Soil microbial communities have been linked to plant germination, growth, and drought tolerance in many plant species, however little is known about the factors contributing to B. curtipendula germination and early growth. In this study, w

[Effects of Soil Amendments on the Bacterial Diversity and Abundances of Pathogens and Antibiotic Resistance Genes in Rhizosphere Soil Under Drip Irrigation with Reclaimed Water].

Due to reclaimed water, irrigation can cause human health and environmental risks. Soil amendments are applied to reveal the abundance of pathogens and antibiotic resistance genes in rhizosphere soil irrigated by reclaimed water and to better understand the effects of environmental factors on the rhizosphere soil bacterial composition, which has guiding significance for the reasonable use of soil amendments. In this study, the effects of biochar, bioorganic fertilizer, humic acid, loosening soil essence, and corn vinasse on bacterial community diversity and certain gene abundances in rhizosphe
