Silicon central pattern generators for cardiac diseases.

Cardiac rhythm management devices provide therapies for both arrhythmias and resynchronisation but not heart failure, which affects millions of patients worldwide. This paper reviews recent advances in biophysics and mathematical engineering that provide a novel technological platform for addressing heart disease and enabling beat-to-beat adaptation of cardiac pacing in response to physiological feedback. The technology consists of silicon hardware central pattern generators (hCPGs) that may be trained to emulate accurately the dynamical response of biological central pattern generators (bCPGs

An optimality principle for locomotor central pattern generators.

Two types of neural circuits contribute to legged locomotion: central pattern generators (CPGs) that produce rhythmic motor commands (even in the absence of feedback, termed "fictive locomotion"), and reflex circuits driven by sensory feedback. Each circuit alone serves a clear purpose, and the two together are understood to cooperate during normal locomotion. The difficulty is in explaining their relative balance objectively within a control model, as there are infinite combinations that could produce the same nominal motor pattern. Here we propose that optimization in the presence of uncerta

Phylogenetic and individual variation in gastropod central pattern generators.

Gastropod molluscs provide a unique opportunity to explore the neural basis of rhythmic behaviors because of the accessibility of their nervous systems and the number of species that have been examined. Detailed comparisons of the central pattern generators (CPGs) underlying rhythmic feeding and swimming behaviors highlight the presence and effects of variation in neural circuits both across and within species. The feeding motor pattern of the snail, Lymnaea, is stereotyped, whereas the feeding motor pattern in the sea hare, Aplysia, is variable. However, the Aplysia motor pattern is regulariz

Neuromodulation of central pattern generators and its role in the functional recovery of central pattern generator activity.

Neuromodulators play an important role in how the nervous system organizes activity that results in behavior. Disruption of the normal patterns of neuromodulatory release or production is known to be related to the onset of severe pathologies such as Parkinson's disease, Rett syndrome, Alzheimer's disease, and affective disorders. Some of these pathologies involve neuronal structures that are called central pattern generators (CPGs), which are involved in the production of rhythmic activities throughout the nervous system. Here I discuss the interplay between CPGs and neuromodulatory activity,

Modeling synaptic interactions between mammalian breathing and swallowing central pattern generators.

Breathing and swallowing are tightly coupled motor behaviors whose execution depends on the coordinated activity of two brainstem central pattern generators (R-CPG, Sw-CPG). We present a mechanistic neural network model that captures this coordination. The model was built by incrementally expanding the synaptic connectivity of R-CPG and Sw-CPG modules, each composed of neuronal population nodes whose dynamics incorporate firing rate adaptation. The rhythmogenic cores of both CPGs are implemented as half-center oscillators operating independently. In agreement with accompanying experimental rec

Central pattern generators evolved for real-time adaptation to rhythmic stimuli.

For a robot to be both autonomous and collaborative requires the ability to adapt its movement to a variety of external stimuli, whether these come from humans or other robots. Typically, legged robots have oscillation periods explicitly defined as a control parameter, limiting the adaptability of walking gaits. Here we demonstrate a virtual quadruped robot employing a bio-inspired central pattern generator (CPG) that can spontaneously synchronize its movement to a range of rhythmic stimuli. Multi-objective evolutionary algorithms were used to optimize the variation of movement speed and direc

Orofacial Central Pattern Generators: Neuroanatomical and Clinical Insights into Sensory, Motor, Emotional Integration.

Orofacial behaviors such as mastication, swallowing, and phonation are not organized as simple reflex chains but are generated and coordinated by intrinsic brainstem networks known as central pattern generators (CPGs). Although the existence and organization of these circuits are well established in animal models, direct evidence for their microcircuit organization in humans remains limited, and current human interpretations rely largely on lesion-based, neurophysiological, and imaging-based inference. In this review, we synthesize neuroanatomical, neurophysiological, and clinical evidence to

Central Pattern Generators in Spinal Cord Injury: Mechanisms, Modulation, and Therapeutic Strategies for Motor Recovery.

BACKGROUND: Spinal cord injury (SCI) compromises the communication between the brain and spinal circuits involved in locomotion, resulting in severe motor dysfunction. However, currently available therapies have limited effectiveness in restoring motor function after SCI. OBJECTIVE: Recent research has highlighted the importance of the central pattern generator (CPG), a spinal circuitry responsible for generating coordinated patterns of leg motor activity in the absence of brain-derived inputs, in locomotor recovery. Therefore, a highly promising approach for restorative treatment after SCI i

On peripheral control mechanisms acting on the central pattern generators for swimming in the dogfish.

When sinusoidal movements were artificially imposed on the tail region of the curarized spinal dogfish during "fictive locomotion' the coordinated burst pattern recorded in the ventral roots was effectively entrained to follow movement frequencies above as well as below the resting rate. The entrainment was characterized by: (1) a broad range of effective movement frequencies and amplitudes (down to a few degrees); (2) frequency-dependent timing of entrained bursts to the movement; (3) constant burst durations at low and moderate frequencies; (4) incomplete entrainment in response to high or l

Two central pattern generators from the crab, Cancer borealis, respond robustly and differentially to extreme extracellular pH.

The activity of neuronal circuits depends on the properties of the constituent neurons and their underlying synaptic and intrinsic currents. We describe the effects of extreme changes in extracellular pH - from pH 5.5 to 10.4 - on two central pattern generating networks, the stomatogastric and cardiac ganglia of the crab, Cancer borealis. Given that the physiological properties of ion channels are known to be sensitive to pH within the range tested, it is surprising that these rhythms generally remained robust from pH 6.1 to pH 8.8. The pH sensitivity of these rhythms was highly variable betwe

Neuroethological approach to frontolimbic epileptic seizures and parasomnias: The same central pattern generators for the same behaviours.

The aim of this report is not to make a differential diagnosis between epileptic nocturnal seizures and non-epileptic sleep-related movement disorders, or parasomnias. On the contrary, our goal is to emphasize the commonly shared semiological features of some epileptic seizures and parasomnias. Such similar features might be explained by the activation of the same neuronal networks (so-called 'central pattern generators' or CPG). These produce the stereotypical rhythmic motor sequences - in other words, behaviours - that are adaptive and species-specific (such as eating/alimentary, attractive/

Central pattern generators in the brainstem and spinal cord: an overview of basic principles, similarities and differences.

Central pattern generators (CPGs) are generally defined as networks of neurons capable of enabling the production of central commands, specifically controlling stereotyped, rhythmic motor behaviors. Several CPGs localized in brainstem and spinal cord areas have been shown to underlie the expression of complex behaviors such as deglutition, mastication, respiration, defecation, micturition, ejaculation, and locomotion. Their pivotal roles have clearly been demonstrated although their organization and cellular properties remain incompletely characterized. In recent years, insightful findings abo

Contextual modulation of a multifunctional central pattern generator.

The multifunctional buccal central pattern generator in snails, which controls different oral behaviors, has been well characterized. In this work we propose a role for the group of about 40 electrotonically coupled buccal A cluster cells as a context-dependant switch for the buccal central pattern generator, modulating motor patterns that elicit different oral behaviors. We characterize these cells based on location and morphology, and provide evidence for their selective activation under two different stimuli - Listerine perfusion and intestinal nerve stimulation - triggering buccal motor pa

A hormone-activated central pattern generator for courtship.

BACKGROUND: Medicinal leeches (Hirudo spp.) are simultaneous hermaphrodites. Mating occurs after a stereotyped twisting and oral exploration that result in the alignment of the male and/or female gonopores of one leech with the complementary gonopores of a partner. The neural basis of this behavior is presently unknown and currently impossible to study directly because electrophysiological recording techniques disrupt the behavior. RESULTS: Here we report that (Arg(8))-conopressin G and two other members of the oxytocin/vasopressin family of peptide hormones induce in Hirudo verbana a sequenc

On the central pattern generator for the basic breathing rhythmicity.

Recent advances in several laboratories concerning the respiration-related medullary neurons, their locations, projections, interconnections, morphological and physiological properties, and patterns of inhibitory postsynaptic potentials, excitatory postsynaptic potentials, and discharge rate, on the one hand, and the "systems behavior," on the other, have provided the basis for new hypothesis concerning the neural mechanisms underlying the central pattern generator (CPG) for breathing and its different parts. The onset of the "ramp"-like increase in inspiratory activity is due to an abrupt rel

A central pattern generator to control a pyloric-based system.

A central pattern generator (CPG) is built to control a mechanical device (plant) inspired by the pyloric chamber of the lobster. Conductance-based models are used to construct the neurons of the CPG. The plant has an associated function that measures the amount of food flowing through it per unit of time. We search for the best set of solutions that give a high positive flow of food in the maximization function. The plant is symmetric and the model neurons are identical to avoid any bias in the space of solutions. We find that the solution is not unique and that three neurons are sufficient t

Roles for cerebellum and subsumption architecture in central pattern generation.

Within vertebrates, central pattern generators drive rhythmical behaviours, such as locomotion and ventilation. Their pattern generation is also influenced by sensory input and various forms of neuromodulation. These capabilities arose early in vertebrate evolution, preceding the evolution of the cerebellum in jawed vertebrates. This later evolution of the cerebellum is suggestive of subsumption architecture that adds functionality to a pre-existing network. From a central-pattern-generator perspective, what additional functionality might the cerebellum provide? The suggestion is that the adap

Whisker deafferentation and rodent whisking patterns: behavioral evidence for a central pattern generator.

Even in the absence of explicit stimulation, rats emit patterns of rhythmic whisking movements. Because of their stereotyped nature and their persistence after sensory denervation and cortical ablation, whisking movements have been assumed to reflect the output of a central pattern generator (CPG). However, identification of a movement pattern as the product of a CPG requires evidence that its generation, patterning, and coordination are independent of sensory input. To provide such evidence, we used optoelectronic instrumentation to obtain high-resolution records of the movement trajectories

Mouse incising central pattern generator: Characteristics and modulation by pain.

INTRODUCTION: Vertebrate incising and chewing are controlled by a set of neurons comprising the central pattern generator (CPG) for mastication. Mandibular positioning and force generation to perform these tasks is complex and requires coordination of multiple jaw opening and closing muscle compartments located in muscles on both sides of the jaw. The purpose of this study was to determine the characteristics of the CPG by recording mouse incising forces in the home cage environment to evaluate changes in force characteristics with incising frequency and force direction. A second purpose was t
