Rat thymocytes were separated into five density layers by isopycnic centrifugation on a discontinuous-density Ficoll gradient and each fraction was tested for metabolic functions at different times during a 96-h culture period in various conditions. Element assimilation rates or the DNA replication rate of microbial taxa can be measured in environmental samples through quantitative DNA-stable isotope probing (DNA-qSIP). Here, we introduce a set of DNA standards that may be used to quantify the density of DNA extracted from environmental samples after isopycnic centrifugation. We describe a rapid, efficient method for purifying Loa loa microfilariae from blood that does not affect their longterm motility. It is based on the separation of microfilariae on a performed discontinuous Percoll gradient, followed by filtration on cellulose ester membranes. The parasite population, after isolation, is close to 100% pure, 100% viable and remains motile for at least 20 days. Recovery from original blood samples is typically around 90%. A large-scale procedure for purification and concentration of Japanese encephalitis virus vaccine by a continuous-flow isopycnic banding technique in a sucrose density gradient solution, using a K-III zonal centrifuge rotor, is presented. The quality of zonal-purified vaccine was compared with commercial and Japanese National Institutes of Health reference vaccines for antigenicity, immunodiffusion, and allergic encephalitogenicity tests to show its high purity.

In previous experiments on growth and aging in the yellow-fever mosquito, Aedes aegypti, a low mol. wt. (500000) DNA species was found in the supernatant fraction after ultracentrifugation of homogenates of rapidly-growing larvae. This DNA species, "sDNA", constituted 30-40% of total DNA in 2-4-day-old larvae, but was less than 5% in older larvae, pupae and adults. Jejunal biopsies from six patients having the small bowel enteropathy associated with common variable immunodeficiency have been subjected to analytical subcellular fractionation and enzymic and regulatory peptide microassay to define the organelle pathology of this syndrome. Purification of epizootic hemorrhagic disease virus (EHDV) from sonicated cell culture supernatant was effected using polyethylene glycol (PEG) precipitation and two CsCl density gradient centrifugations. During purification procedures the pH was maintained at values close to pH 8.0 as pH stability for EHDV was maximal between pH 7.2 and pH 9.0. We reported previously that painful crises in patients with sickle cell anemia are accompanied by striking decreases in the percent of densest red cells (fraction 4) when studied with isopyknic Percoll-Stractan gradients. We report that an alternative to density gradients is the red cell distribution width (RDW), an estimate of red cell size variation measured with a Coulter counter. Representative preparations of partially purified Creutzfeldt-Jakob disease (CJD), including disaggregated density gradient fractions, were treated with a variety of nucleases. Replicative bypass repair of UV damage to DNA was studied in a wide variety of human, mouse and hamster cells in culture. Coxiella burneti phase I, purified from a formalin-inactivated yolk-sac vaccine, was separated into two bands of morphologically distinct cell types when subjected to sucrose gradient centrifugation. In this study, we present a detailed workflow for the isolation, quantitation, and evaluation of mucin proteins.
