Development of assays to differentiate and quantify three Cercospora species causing Cercospora leaf blight on soybean.

Cercospora leaf blight (CLB) is the most devastating foliar disease of soybean in the US Gulf South, causing significant economic impacts throughout the growing region. The sole causal agent of CLB was thought to be Cercospora kikuchii, but we now recognize that the disease is predominantly caused by Cercospora cf. flagellaris and Cercospora cf. sigesbeckiae in North America. However, a lack of information about the spatial population dynamics of the causal agents and the disease cycle, including the most important sources of inoculum, hinders the development of new and effective management to

Time-Course Comparative Transcriptomics Between Isolates of Cercospora zeae-maydis and Cercospora zeina During Maize Infection.

Gray leaf spot (GLS) is a major foliar disease of maize caused mainly by Cercospora zeae-maydis and Cercospora zeina. Although both species infect maize, the comparative transcriptional programs between isolates of different species during maize colonization remain largely unknown. We conducted time-course pathogen-focused RNA-seq of one Jilin isolate assigned to each species during colonization of maize B73 under a controlled inoculation system. RNA-seq reads were mapped to the public C. zeae-maydis SCOH1-5 and C. zeina CMW25467 v3 reference genomes for transcript quantification and putative

Portable qPCR and ddPCR Diagnostics for Cercospora Leaf Spot: Integrating Field and Laboratory Detection of Cercospora beticola in Beet.

Cercospora beticola, the causal agent of Cercospora leaf spot (CLS), is a major pathogen affecting beet (Beta vulgaris) worldwide. Rapid and accurate detection is critical for effective disease control. This study validated a diagnostic framework combining portable quantitative PCR (qPCR) and laboratory-based digital droplet PCR (ddPCR) assays to detect and quantify C. beticola in leaves, seeds, and soil-associated crop residues. We used two portable qPCR systems (Biomeme Franklin® Three9 and Hyris bCUBE™) for in-field diagnostics, and ddPCR (Bio-Rad Laboratories) for laboratory analysis. Two

Sugarbeet leaf spot disease (Cercospora beticola Sacc.)dagger.

UNLABELLED: SUMMARY Leaf spot disease caused by Cercospora beticola Sacc. is the most destructive foliar pathogen of sugarbeet worldwide. In addition to reducing yield and quality of sugarbeet, the control of leaf spot disease by extensive fungicide application incurs added costs to producers and repeatedly has selected for fungicide-tolerant C. beticola strains. The genetics and biochemistry of virulence have been examined less for C. beticola as compared with the related fungi C. nicotianae, C. kikuchii and C. zeae-maydis, fungi to which the physiology of C. beticola is often compared. Membrane transporters in self resistance of Cercospora nicotianae to the photoactivated toxin cercosporin.

The goal of this work is to characterize membrane transporter genes in Cercospora fungi required for autoresistance to the photoactivated, active-oxygen-generating toxin cercosporin they produce for infection of host plants. Previous studies implicated a role for diverse membrane transporters in cercosporin resistance. In this study, transporters identified in a subtractive cDNA library between a Cercospora nicotianae wild type and a cercosporin-sensitive mutant were characterized, including two ABC transporters (CnATR2, CnATR3), an MFS transporter (CnMFS2), a uracil transporter, and a zinc tr

Development of a Sheath Inoculation Protocol to Screen Rice Varieties for Resistance to Cercospora janseana.

Cercospora janseana (Racib.) O. Const. is a re-emerging fungal pathogen that causes Cercospora net blotch on rice. Previous research on resistance to C. janseana has primarily focused on foliar symptoms. Subsequently, sheath infection remains poorly characterized which hinders disease management efforts. This study developed and validated a reproducible sheath inoculation protocol under controlled conditions. Three inoculation methods (agar disc, spray, and drop) were evaluated with and without mechanical wounding. Lesions only formed with inoculation methods using wounding and the agar disc m

Structural Phylogenetics Identifies Conserved Effector Families and Cell Death-Inducing Proteins from Cercospora zeina.

Cercospora zeina causes gray leaf spot disease on maize in Africa. As a foliar fungal pathogen, C. zeina relies on effectors to facilitate successful infection and colonization of its host. Predicting effector function is therefore essential for understanding the molecular mechanisms underlying this plant-pathogen interaction. Although effector functions are typically predicted using sequence-based analyses, many effectors are subject to strong selection pressure and exhibit high sequence diversity, reducing the accuracy of such predictions. In this study, a structural phylogenetics approach w

Eco-friendly management of cercospora leaf spot disease in sugar beet using ZnO and MgO nanoparticles.

BACKGROUND: Cercospora leaf spot, caused by Cercospora beticola, is the most destructive disease of sugar beet. This study evaluated zinc oxide (ZnO) and magnesium oxide (MgO) nanoparticles (NPs) as sustainable alternatives to chemical fungicides. RESULTS: In vitro experiments, at a concentration of 90 ppm, the fungal inhibition rate of MgO NPs reached 58.33%, higher than the 52.08% recorded for ZnO NPs. SEM observations confirmed that both types of nanoparticles caused severe distortion and deformation of the mycelium of the tested fungi. However, under greenhouse and field conditions, ZnO N

Population Structure and Mating Type Distribution of Cercospora sojina from Soybeans in Indiana, United States.

Frogeye leaf spot on soybeans is traditionally considered as a southern disease in the United States but its impact in North Central USA has been rising in recent years. In this study, we investigated the population structure and mating type distribution in the C. sojina population from Indiana, USA. Based on 27 single nucleotide polymorphism markers, 49 multi-locus genotypes (MLGs) were identified in 234 isolates collected from 29 counties in Indiana in 2020. Bayesian analysis grouped the 49 MLGs into three clusters. This grouping was supported by principal coordinate analysis and, in large p

Modeling the environment-related risk of frogeye leaf spot (Cercospora sojina) in soybean across the United States.

Frogeye leaf spot (FLS), caused by Cercospora sojina, is a common soybean disease across the U.S. Fungicides are a key management tool, particularly when susceptible cultivars are planted; however, widespread QoI resistance has raised concern about overreliance on the remaining effective fungicide classes. Protecting these chemical classes is essential for long-term sustainability, particularly under narrow profit margins. To develop an FLS prediction model that supports more efficient fungicide use, environmental and epidemiological data from multiple site-years were analyzed in 2024 using co

PHL1 of Cercospora zeae-maydis encodes a member of the photolyase/cryptochrome family involved in UV protection and fungal development.

DNA photolyases harvest light energy to repair genomic lesions induced by UV irradiation, whereas cryptochromes, presumptive descendants of 6-4 DNA photolyases, have evolved in plants and animals as blue-light photoreceptors that function exclusively in signal transduction. Orthologs of 6-4 photolyases are predicted to exist in the genomes of some filamentous fungi, but their function is unknown. In this study, we identified two putative photolyase-encoding genes in the maize foliar pathogen Cercospora zeae-maydis: CPD1, an ortholog of cyclobutane pyrimidine dimer (CPD) photolyases described i

Fine Mapping of QTLs/QTNs and Mining of Genes Associated with Race 7 of the Soybean Cercospora sojina by Combining Linkages and GWAS.

Soybean frogeye leaf spot (FLS) disease has been reported globally and is caused by the fungus Cercospora sojina, which affects the growth, seed yield, and quality of soybean. Among the 15 physiological microspecies of C. sojina soybean in China, Race 7 is one of the main pathogenic microspecies. A few genes are involved in resistance to FLS, and they cannot meet the need to design molecular breeding methods for disease resistance. In this study, a soybean recombinant inbred line (RIL3613) population and a germplasm resource (GP) population were planted at two sites, Acheng (AC) and Xiangyang

Regulation of stomatal tropism and infection by light in Cercospora zeae-maydis: evidence for coordinated host/pathogen responses to photoperiod?

Cercospora zeae-maydis causes gray leaf spot of maize, which has become one of the most widespread and destructive diseases of maize in the world. C. zeae-maydis infects leaves through stomata, which is predicated on the ability of the pathogen to perceive stomata and reorient growth accordingly. In this study, the discovery that light was required for C. zeae-maydis to perceive stomata and infect leaves led to the identification of CRP1, a gene encoding a putative blue-light photoreceptor homologous to White Collar-1 (WC-1) of Neurospora crassa. Disrupting CRP1 via homologous recombination re

Genome-Wide Association and Selective Sweep Studies Reveal the Complex Genetic Architecture of DMI Fungicide Resistance in Cercospora beticola.

The rapid and widespread evolution of fungicide resistance remains a challenge for crop disease management. The demethylation inhibitor (DMI) class of fungicides is a widely used chemistry for managing disease, but there has been a gradual decline in efficacy in many crop pathosystems. Reliance on DMI fungicides has increased resistance in populations of the plant pathogenic fungus Cercospora beticola worldwide. To better understand the genetic and evolutionary basis for DMI resistance in C. beticola, a genome-wide association study (GWAS) and selective sweep analysis were conducted for the fi

Genetic diversity, QoI fungicide resistance, and mating type distribution of Cercospora sojina-Implications for the disease dynamics of frogeye leaf spot on soybean.

Frogeye leaf spot (FLS), caused by Cercospora sojina, causes significant damage to soybean in the U.S. One control strategy is the use of quinone outside inhibitor (QoI) fungicides. QoI resistant isolates were first reported in Tennessee (TN) in 2010. To investigate the disease dynamics of C. sojina, we collected 437 C. sojina isolates in 2015 from Jackson and Milan, TN and used 40 historical isolates collected from 2006-2009 from TN and ten additional states for comparison. A subset of 186 isolates, including historical isolates, were genotyped for 49 single nucleotide polymorphism (SNP) mark

Biocontrol of Cercospora leaf spot in sugar beet by a novel Bacillus velezensis KT27 strain: Enhanced antifungal activity and growth promotion in laboratory and field conditions.

Diseases in crops are a major contributor to yield reduction and economic losses. Cercospora leaf spot (CLS), caused by Cercospora beticola, is among the most severe diseases affecting sugar beet and other crops. The increasing resistance of C. beticola to conventional chemical fungicides, along with their excessive application, exacerbates environmental pollution. This study investigates the antagonistic activity of a newly isolated strain, Bacillus velezensis KT27, against Cercospora beticola, Rhizoctonia cerealis, and Fusarium oxysporum under laboratory conditions. Soybean toxin (SBTX) impairs fungal growth by interfering with molecular transport, carbohydrate/amino acid metabolism and drug/stress responses.

Soybean toxin (SBTX) is an antifungal protein from soybeans with broad inhibitory activity against the growth and filamentation of many fungi, including human and plant pathogenic species such as Candida albicans, Candida parapsilosis, Aspergillus niger, Penicillium herquei, Cercospora sojina and Cercospora kikuchii. Understanding the mechanism by which SBTX acts on fungi and yeasts may contribute to the design of novel antifungal drugs and/or the development of transgenic plants resistant to pathogens. To this end, the polymorphic yeast C. albicans was chosen as a model organism and changes i
