[Ceruletide--a sensible adjuvant measure in ERCP?].

The gastro-intestinal hormone Ceruletide is used for diagnostic and therapeutic purposes. Apart from other effects, Ceruletid brings about a relaxation of the Sphincter of Oddi. In the present double-blind study, it was examined on 70 patients whether the effect of this drug can facilitate the cannulation of the Papilla Vateri. Using a dosage of 50 ng/kg body weight Ceruletid in i.m. application the examination time required by the most experienced examiner up to the cannulation of the Papilla Vateri was shortened by half with respect to the collective control group treated with placebo. Effect of ceruletide on discriminated avoidance behavior in rats.

To clarify the central action of peripherally administered ceruletide, we examined the effects of ceruletide and four different classes of neuroleptics, i.e. haloperidol, chlorpromazine, oxypertine and sulpiride, on the discriminated avoidance response (DAR) in rats. Ceruletide and sulpiride did not suppress the DAR over a broad range of doses, whereas other three neuroleptics caused a dose-related decrease in both avoidance and response rates. In addition, the combined administration of ceruletide (100 micrograms/kg s.c.) and neuroleptics at critical doses that suppress the DAR caused a signi

Ceruletide acts in the abdomen, not in the brain, to produce satiety.

Ceruletide (caerulein), a decapeptide extracted from the skin of the frog, Hyla caerulea, is very similar in structure to the C-terminal octapeptide of cholecystokinin (CCK-8). Although ceruletide and CCK-8 act through similar or identical receptors to produce the same visceral effects, previous studies in the rat suggested that peripherally administered ceruletide acted directly on the ventromedial hypothalamic (VMH) area to decrease food intake, but peripherally administered CCK-8 acted at a vagally innervated abdominal site to decrease food intake. Since it is unprecedented for these two pe

[Effects of ceruletide in the treatment of postoperative paralytic ileus].

After a short analysis of the pathogenetic causes which can lead to paralytic ileus and the drugs at present available for treatment of this serious morbid form, the paper reports results obtained with Ceruletide, a decapeptide which has proved active on intestinal peristalsis in experimental and clinical trials. The results of this study, carried out on 20 patients subjected to surgical operation on the abdomen, treated with Ceruletide by intramuscular route 24-48 hours after the operation, showed that this drug is capable of leading to a quick recovery of intestinal motor activity without ca

[Cholecystography with ceruletide. Results and evaluation of its routine use].

Cholecystography has been up to day used as unique or first instance examination in patients suspected to have chronic gallbladder diseases. On the other hand, cholecystosonography is proposed with increasing frequence as first examination for simple and rapid detection of cholelithiasis. Results of high standard cholecystography are therefore welcomed in order to assess it present role in respect to diagnostic ultrasounds. On this basis, the authors reports the results of systematic administration of ceruletide in 300 clinically unselected patients with a good opacification at cholecystograph

[Improvement of oral cholecystography and cholangiography with ceruletid (author's transl)].

Following oral cholecystography in 100 patients, the conventional "fatty meal" was replaced by an intramuscular injection of Ceruletid in a dose of 0.4 microgram/kg body weight. The synthetic decapetide Ceruletid is a substance with a hormone-like effect on the gastro-intestinal tract. It contracts smooth muscle in the gut and stimulates secretion in the stomach and the exocrine pancreas. Compared with other substances producing contraction which are given orally, Ceruletid acts more quickly and more powerfully in producing contraction of the gall bladder muscle. In 86% of positive cholecystog

Sonography of the common bile duct and the gallbladder during ceruletid infusion.

In the present study, we investigated a new sonographic test to confirm or exclude partial common bile duct (CBD) obstruction, hereinafter called "dynamic cholangio-cholecysto sonography (DCCS)." Healthy controls (6) and patients with low- to intermediate probability for partial CBD obstruction (17) were investigated. DCCS started with three baseline masurements of gallbladder volume and CBD diameter, which were then repeated every 2 to 3 min for 45 min during a 30-min infusion of ceruletid. According to CBD diameter change during gallbladder contraction, DCCS was considered positive (> 1 mm),

After the failure of other medicamentous treatment attempts for paralytic ileus, Ceruletid (Takus) was used in 20 surgical patients. Ceruletid was applied as an infusion of 40 microgram in 250 ml or 500 ml physiological saline solution over a period of 2-3 hours. During this time the occurrence of bowel sounds, flatulence and defecation was observed. The most important clinical and serological parameters were measured before and after treatment. Treatment was successful in 17 cases, in 3 cases no success was achieved. Severe side effects did not occur. The use of Ceruletid in the described for

Ceruletide increases threshold and tolerance to experimentally induced pain in healthy man.

UNLABELLED: Previous studies suggested that ceruletide might be endowed with analgesic and sedative properties. To investigate the effects of ceruletide on experimentally induced pain and on central nervous functions, two studies, each involving 24 healthy subjects, were carried out in random double-blind fashion. Every subject participated in three experiments one week apart. In study 1, 120 and 60 ng/kg/hr ceruletide IV increased threshold and tolerance to electrically and threshold to thermally induced cutaneous pain significantly more than saline (p less than 0.001), the higher dose being

Cholecystokinin octapeptide (CCK-8), ceruletide and analogues of ceruletide: effects on tremors induced by oxotremorine, harmine and ibogaine. A comparison with prolyl-leucylglycine amide (MIF), anti-Parkinsonian drugs and clonazepam.

Cholecystokinin octapeptide (CCK-8), ceruletide (caerulein, CER) and 10 analogues of ceruletide, were studied in mice for antagonism of the tremors induced by harmine (5 mg/kg, s.c.), ibogaine (20 mg/kg, s.c.) and oxotremorine (0.2 mg/kg, s.c.). The following reference drugs were tested for comparison: prolyl-leucylglycine amide (MIF), atropine, haloperidol, biperiden, ethopropazine, trihexyphenidyl, methixene and clonazepam. All treatments were subcutaneous, the antagonists being given 10 min (in some trials 30 min) before the tremorogen. Tremorolytic potency (ED50) was calculated from dose-r

Effect of ceruletide on rest pain in patients with arterial insufficiency of the lower extremity.

The effect of ceruletide (CRL), a synthetic decapeptide analogue of cholecystokinin, on rest pain and arterial blood flow was evaluated in 8 patients with advanced, occlusive atherosclerosis of the lower extremities. CRL 1, 2, or 4 ng kg-1 or placebo were infused intravenously in random order, and in a double-blind fashion. Pain relief, assessed by a scoring system, was significantly better (p less than 0.01) following the 2 and 4 ng kg-1 doses of CRL (2.71 and 2.66, respectively) than following placebo (0.75). Arterial blood flow was not affected by either CRL in any dose or by placebo. Pretr

[Behavioral effects of ceruletide in Wriggle Mouse Sagami--in comparison with in Rolling Mouse Nagoya].

The behavioral effects of ceruletide in Wriggle Mouse Sagami (WMS) and Rolling Mouse Nagoya (RMN) were observed in an open-field study. Ceruletide decreased the movements of both types of mice, improving the ataxic gait in WMS; however, no such improvement was observed in RMN. There may be different mechanisms underlying the ataxic manifestations in WMS and RMN.

Palatal myoclonus following Behçet's disease ameliorated by ceruletide, a potent analogue of CCK octapeptide.

Systemic administration of ceruletide, a potent analogue of CCK octapeptide (0.8 microgram/kg, i.m.) markedly improved palatal myoclonus. CCK analogues may be useful agents for the treatment of palatal myoclonus. This report suggests that dopaminergic system may also contribute to the underlying biochemical mechanism of palatal myoclonus.

Effect of cholecystokinin octapeptide and ceruletide on release of acetylcholine from cerebral cortex of the rat in vivo.

The effect of cholecystokinin octapeptide (CCK-8) and its analogue, ceruletide on release of acetylcholine (ACh) from the cerebral cortex was investigated in urethane-anaesthetized and in unanaesthetized rats. Cholecystokinin octapeptide and ceruletide markedly stimulated output of ACh at doses of 1.5 and 5.0 micrograms/kg (i.p.), respectively. This effect was prevented by proglumide (160 mg/kg i.p.), a specific cholecystokinin receptor antagonist. At doses of 10 micrograms/kg (i.p.) and more, both CCK-8 and ceruletide decreased output of ACh from the cerebral cortex. The decrease was prevente

Antistereotype effects of ceruletide and some neuroleptics differentiated by interactions with clonazepam, muscimol, scopolamine and clonidine.

Compulsive gnawing was produced in mice by administration of either methylphenidate or (after sensitizing pretreatment with the neuroleptic, tetflutixol) apomorphine. Drugs which antagonise stereotypy, such as ceruletide (CER, a sulphated decapeptide related to cholecystokinin octapeptide), haloperidol, zuclopenthixol and fluphenazine were applied in equipotent doses (reducing stereotypy by 80%). Clonazepam, muscimol, clonidine and scopolamine (but not methylscopolamine) antagonized to a different extent the antistereotype effect of ceruletide and the neuroleptics.
