Molecular mechanism of calcium inhibition in viral channelrhodopsins.

Viral channelrhodopsins (VCR1s) are giant-virus-encoded light-gated channels permeable to monovalent and divalent cations, including Na+ and Ca2+ ions, and inhibited by millimolar Ca2+ concentrations. Here, we combine X-ray crystallography, time-resolved UV-vis spectroscopy, and ATR-FTIR spectroscopy to investigate molecular mechanisms of ion permeation and Ca2+-dependent inhibition in OLPVR1. An atomic resolution structure of OLPVR1 obtained in the presence of 10 mM CaCl2 and 900 mM NaCl reveals a transient intracellular Ca2+ binding site near T87 and T88, close to the retinal cofactor. Stabilized Ion Selectivity Corrects Activation Drift in Kalium Channelrhodopsins.

Effective optogenetic inhibition of neuronal activity requires tools that reliably silence neurons across cell types and conditions. K+-selective channelrhodopsins (KCRs) have emerged as attractive alternatives to chloride-conducting channels for optogenetic inhibition of cellular excitability, but many KCR variants exhibit an ion selectivity shift toward Na+ under prolonged illumination. Through behavioral and electrophysiological analyses in Drosophila, and Caenorhabditis elegans, it is found that both the absolute K+ to Na+ permeability ratio and its stability over time determine the inhibi

Spectrally distinct channelrhodopsins for two-colour optogenetic peripheral nerve stimulation.

Technologies for peripheral nerve stimulation have conventionally relied on the anatomic placement of electrodes adjacent to subsets of sensory fibres or motor fibres that selectively target an end effector. Here, we demonstrate the use of optogenetics to directly target the innervating fibres of an end effector by relying on retrograde transfection of adeno-associated virus serotype 6 to restrict axonal opsin expression to the desired fibre targets. By using an in vivo screen in rats, we identify the first channelrhodopsins as well as a halorhodopsin that respond to red light in the periphera

Optogenetic restoration of high-sensitivity vision using ChRmine- and ChroME-based channelrhodopsins.

Optogenetic gene therapy is a promising mutation-independent treatment that aims to restore visual perception in patients blinded by retinal diseases that cause photoreceptor degeneration. Still, low sensitivity or slow kinetics of currently utilized optogenetic proteins limit the efficacy of such approaches. Here, we evaluated the therapeutic potential of three channelrhodopsin variants: ChRmine, from the algae Rhodomonas lens, ChRmine-T119A, a faster-closing ChRmine variant, and ChroME2s, a second-generation Chronos-based opsin. We expressed these opsins in retinal ganglion cells of rd1 mice,

Characterization of the tail current of Channelrhodopsin-2 variants.

Our study focused on specific ChR2 variants, particularly those with the Step function Opsins (SFO) mutation at the D156-C128 gate. These are widely used in optogenetics due to their heightened sensitivity to light and bi-stable prolonged activation. However, in some ChR2 variants, specifically D156 mutants, a tail current occurs when continuous light exposure is stopped. We specifically examined the D156H-T159S ChR2 variant, which demonstrated a tail current that was somewhat responsive to light and voltage, with a single-channel current of around 9fA, similar to wt-ChR2 as determined by stat

Computational framework and tools for evaluating engineered channelrhodopsin performance in a degenerated retina.

Recent breakthroughs in genetic engineering have paved the way for the development of a range of novel opsins with promising applications in visual restoration through optogenetics. We present a computational model that combines analytical formulations of optogenetic mechanisms with conductance-based representations of cellular and synaptic properties. Through systematic optimization, our model now provides a robust framework for simulating and analyzing the functional dynamics of these newly developed opsins. Moreover, by calibrating our model with the latest preclinical data in optogenetics

Channelrhodopsin fluorescent tag replacement for clinical translation of optogenetic hearing restoration.

Sensory restoration by optogenetic neurostimulation provides a promising future alternative to current electrical stimulation approaches. So far, channelrhodopsins (ChRs) typically contain a C-terminal fluorescent protein (FP) tag for visualization that potentially poses an additional risk for clinical translation. Previous work indicated a reduction of optogenetic stimulation efficacy upon FP removal. Here, we further optimized the fast-gating, red-light-activated ChR f-Chrimson to achieve efficient optogenetic stimulation in the absence of the C-terminal FP. Upon FP removal, we observed a ma

Channelrhodopsin variants for high-rate optogenetic neurostimulation at low light intensities.

Optogenetics allows versatile control of excitable cell networks, which advances basic science research and drives the development of future medical applications. Fast-closing channelrhodopsins (ChRs) are required for high temporal fidelity of neurostimulation, but their short channel open times require sufficient plasma membrane expression and high light intensity, challenging clinical translation. Here, we addressed the need of high-rate neurostimulation by engineering optimized blue-light-sensitive ChR variants. In particular, we report on the ChR2 variant f-ChR2 TC enabling high frequency

Selecting Channelrhodopsin Constructs for Optimal Visual Restoration in Differing Light Conditions.

Channelrhodopsin (ChR)-based optogenetics is one promising approach to restore vision in photoreceptor degenerative diseases such as retinitis pigmentosa (RP) and age-related macular degeneration (AMD). Currently, a large number of ChRs from different alga species as well as engineered variants are available. They vary with their light response properties like peak sensitive wavelength (λmax), current amplitude, and kinetics. Therefore, it is important to choose an appropriate ChR for practical applications, such as vision restoration. Here we describe a standard laboratory protocol for charac

Optogenetic determination of the myocardial requirements for extrasystoles by cell type-specific targeting of ChannelRhodopsin-2.

Extrasystoles lead to several consequences, ranging from uneventful palpitations to lethal ventricular arrhythmias, in the presence of pathologies, such as myocardial ischemia. The role of working versus conducting cardiomyocytes, as well as the tissue requirements (minimal cell number) for the generation of extrasystoles, and the properties leading ectopies to become arrhythmia triggers (topology), in the normal and diseased heart, have not been determined directly in vivo. Here, we used optogenetics in transgenic mice expressing ChannelRhodopsin-2 selectively in either cardiomyocytes or the

When channelrhodopsin goes off-target: biophysical signatures of ectopic expression in neurons.

The Cre-lox system allows targeted expression of optogenetic proteins such as channelrhodopsin-2 (ChR2), but can also produce unintended expression in non-target cells. In a VGLUT1-Cre::ChR2(H134R)-eYFP mouse line, we bred mice that correctly expressed ChR2 in VGLUT1 positive neurons. This was corroborated with whole cell recordings of dopaminergic (DA) neurons of the substantia nigra (SN), which elicited blue light evoked AMPA currents with a delay of a few milliseconds following the onset of light stimulation. However, at some point during breeding we discovered recurring ectopic expression

Intraflagellar transport-20 guides the ciliary membrane trafficking of channelrhodopsin in Chlamydomonas reinhardtii.

BACKGROUND: The cilium is a microtubule-based organelle essential for cellular signaling, whose assembly depends on intraflagellar transport (IFT). IFT20, a unique IFT-B component, localizes to both the Golgi apparatus and cilium/flagellum in mammalian systems and plays a role in ciliary membrane protein trafficking. Here, we analyzed IFT20-mediated ciliary trafficking of channelrhodopsin-1 (ChR1) in Chlamydomonas reinhardtii. METHODS AND RESULTS: Co-immunocytochemistry showed that IFT20 and ChR1 co-localized throughout the flagella of wild-type cells. Using fla8 (kinesin-2) and dhc1b-3 (dyne

Recording Channelrhodopsin-Evoked Field Potentials and Startle Responses from Larval Zebrafish.

Zebrafish are an excellent model organism to study many aspects of vertebrate sensory encoding and behavior. Their escape responses begin with a C-shaped body bend followed by several swimming bouts away from the potentially threatening stimulus. This highly stereotyped motor behavior provides a model for studying startle reflexes and the neural circuitry underlying multisensory encoding and locomotion. Channelrhodopsin (ChR2) can be expressed in the lateral line and ear hair cells of zebrafish and can be excited in vivo to elicit these rapid forms of escape. Here we review our methods for stu

Transgenic approach to express the channelrhodopsin 2 gene in arginine vasopressin neurons of rats.

Optogenetics provides a powerful tool to regulate neuronal activity by light-sensitive ion channels such as channelrhodopsin 2 (ChR2). Arginine vasopressin (AVP; also known as the anti-diuretic hormone) is a multifunctional hormone which is synthesized in the magnocellular neurosecretory cells (MNCs) of the hypothalamus. Here, we have generated a transgenic rat that expresses an AVP-ChR2-enhanced green fluorescent protein (eGFP) fusion gene in the MNCs of the hypothalamus. The eGFP fluorescence that indicates the expression of ChR2-eGFP was observed in the supraoptic nucleus (SON) and in the m

Channelrhodopsin Excitation Contracts Brain Pericytes and Reduces Blood Flow in the Aging Mouse Brain in vivo.

Brains depend on blood flow for the delivery of oxygen and nutrients essential for proper neuronal and synaptic functioning. French physiologist Rouget was the first to describe pericytes in 1873 as regularly arranged longitudinal amoeboid cells on capillaries that have a muscular coat, implying that these are contractile cells that regulate blood flow. Although there have been >30 publications from different groups, including our group, demonstrating that pericytes are contractile cells that can regulate hemodynamic responses in the brain, the role of pericytes in controlling cerebral blood f

Channelrhodopsin ion selectivity determines mechanisms and efficacy of optogenetic defibrillation in human atria and ventricles.

Cardiac re-entrant arrhythmias are a major cause of sudden cardiac death. Optogenetic defibrillation has emerged as a promising anti-arrhythmic strategy based on photoactivation of channelrhodopsins, yet the efficacy and potential adverse consequences of activating different classes of light-gated ion channels have not been systematically evaluated. Previous studies assessed either cation (e.g. ChR2) or anion (e.g. GtACR1) non-selective channels, both of which depolarize resting cardiomyocytes. Recently identified light-gated K+ channels (e.g. WiChR) suppress cardiomyocyte activity while maint

Visual function in mice with photoreceptor degeneration and transgenic expression of channelrhodopsin 2 in ganglion cells.

The progression of rod and cone degeneration in retinally degenerate (rd) mice ultimately results in a complete loss of photoreceptors and blindness. The inner retinal neurons survive and several recent studies using genetically targeted, light activated channels have made these neurons intrinsically light sensitive. We crossbred a transgenic mouse line expressing channelrhodopsin2 (ChR2) under the control of the Thy1 promoter with the Pde6b(rd1) mouse, a model for retinal degeneration (rd1/rd1). Approximately 30-40% of the ganglion cells of the offspring expressed ChR2. Extracellular recordin

Contractions of ex-vivo mouse bladder driven by activation of channelrhodopsin-2 expressed in urothelial cells.

Signaling molecules released from the urothelium by mechanical stretch are known to play a sensory role in bladder contractions via neuronal signaling. It is also theorized that these molecules released from the urothelium could act locally to induce urothelial cell-mediated local bladder contractions. In this study, we specifically stimulated the urothelial cells using optogenetics to investigate how the urothelial released signaling molecules can influence bladder contractions locally. Using an ex-vivo whole bladder preparation, we stimulated the urothelial cells by activating channelrhodops

Bidirectional optogenetic modulation of peripheral sensory nerve activity: Induction vs. suppression through channelrhodopsin and halorhodopsin.

In this study, we investigated the potential of optogenetics for modulating activity of peripheral sensory nerves, particularly tactile and proprioceptive afferents. Using adeno-associated virus serotype 9 vector, we selectively transduced channelrhodopsin (ChR2(H134R)) and halorhodopsin (eNpHR3.0) into large-diameter sciatic nerve afferents of rats. Diverging from conventional dorsal root ganglion (DRG) approaches, we applied optical stimulation at the distal portion of the nerve. Results show successful induction and suppression of activity in large-diameter afferents via optical stimulation

Characterization of Optically and Electrically Evoked Dopamine Release in Striatal Slices from Digenic Knock-in Mice with DAT-Driven Expression of Channelrhodopsin.

Fast-scan cyclic voltammetry (FCV) is an established method to monitor increases in extracellular dopamine (DA) concentration ([DA]o) in the striatum, which is densely innervated by DA axons. Ex vivo brain slice preparations provide an opportunity to identify endogenous modulators of DA release. For these experiments, local electrical stimulation is often used to elicit release of DA, as well as other transmitters, in the striatal microcircuitry; changes in evoked increases in [DA]o after application of a pharmacological agent (e.g., a receptor antagonist) indicate a regulatory role for the tr
