The giant (2-3 × 10-2 m long) internodal cells of the aquatic plant, Chara, exhibit a rapid (>100 × 10-6 m s-1) cyclic cytoplasmic streaming which stops in response to mechanical stimuli. Since the streaming - and the stopping of streaming upon stimulation - is easily visible with a stereomicroscope, these single cells are ideal tools to investigate mechanosensing in plant cells, as well as the potential for these cells to be anesthetized. We found that dropping a steel ball (0.88 × 10-3 kg, 6 × 10-3 m in diameter) through a 4.6 cm long tube (delivering ca. 4 × 10-4 J) reliably induced mechani

Pectin, a normal constituent of cell walls, caused growth rates to accelerate to the rates in living cells when supplied externally to isolated cell walls of Chara corallina. Because this activity was not reported previously, the activity was investigated. Turgor pressure (P) was maintained in isolated walls or living cells using a pressure probe in culture medium. Pectin from various sources was supplied to the medium. Ca and Mg were the dominant inorganic elements in the wall. EGTA or pectin in the culture medium extracted moderate amounts of wall Ca and essentially all the wall Mg, and wall

The hydraulic resistance (the reciprocal of the hydraulic conductivity Lp) Lp-1 was measured in cells of Chara corallina by the method of transcellular osmosis. Treatment of cells with 100 mM KCl decreased Lp-1 significantly. Subsequent treatment of the cells with 70 mM CaCl2 recovered the decreased Lp-1 to the original value. To know whether K+ or Ca2+/Mg2+ acts on the cell wall and/or the membrane, the hydraulic resistances of the cell wall (Lpw-1) and that of the membrane (Lpm-1) were determined in one and the same cell. For this, a pair of cells (twin cells) were made from an internodal ce

BACKGROUND: Visceral leishmaniasis (VL) is a parasitic disease that affects public health. It is described by weight reduction, irregular fever bouts, anemia, and amplification of the spleen and liver.
MATERIALS AND METHODS: Three concentrations (15.6, 31.2, and 62.5 μg/mL) were used to find the potency of an aqueous extract of Chara vulgaris algae in the treatment of VL. A cytotoxicity assay was performed to show the cytotoxic effect of this extract on human cells. High-performance liquid chromatography (HPLC) test was done to determine the active compounds in the extract. Histopathological

Electrical response upon wounding was analyzed in Chara corallina. A specimen comprising two adjoining internodal cells was prepared. One cell (victim cell) was killed by cutting and any changes in the membrane potential of the neighboring cell (the receptor cell) were measured. Upon cutting the victim cell, the receptor cell generated four kinds of depolarizations: (1) rapid component, (2) slow and long-lasting component, (3) action potential and (4) small spike. Rapid and slow components were observed in most cells. On the other hand, the action potential and small spike were not always ubiq

Oxidative stress and chronic inflammation are major contributors to skin photoaging, highlighting the need for natural antioxidants with multifunctional bioactivities. This study developed an ultrasound-assisted extraction (UAE) strategy to recover an astaxanthin-rich extract from the underutilized freshwater macroalga Chara corallina and evaluated its antioxidant, anti-inflammatory, and anti-photoaging properties. Different solvent systems were evaluated for extraction efficiency, and astaxanthin-equivalent recovery was quantified by high-performance liquid chromatography with photodiode arra

Oxygenic photosynthesis in streptophytic algae, such as Charophyceae, is often impeded by low CO2 levels in aquatic habitats. Consequently, many algal groups evolved a CO2-concentrating mechanism (CCM). However, its presence in Charophyceae remains controversial. To explore this, we analyzed the acclimation of photosynthesis, carbon isotope composition, and gene expression in Chara braunii under varying inorganic carbon (Ci) conditions. The photosynthetic activity changed complementarily under low- or high-Ci levels. Notably, the Ci compensation point of photosynthesis was significantly lower

Inositol phosphates (InsPs) and inositol pyrophosphates (PP-InsPs) are conserved signalling molecules, but their evolutionary origin and diversification in the green lineage remain poorly understood. Here we investigated the InsP network in the streptophyte alga Chara braunii, a key lineage close to the origin of land plants. Using capillary electrophoresis-electrospray ionization mass spectrometry, we detected a broad spectrum of InsP and PP-InsP species from InsP3 to InsP8, including multiple positional isomers. Developmental profiling across dormant oospores, young thalli and mature thalli

Endogenous biological rhythms are central to temporal organization in land plants, yet their evolutionary origins in streptophyte algae remain unresolved. We investigated whether rhythmic morpho-physiological processes in Chara braunii are consistent with endogenous regulation and how continuous light intensity modulates these responses. In C. braunii, time-lapse imaging, pigment quantification, chlorophyll fluorescence measurements, and metabolomic profiling were conducted under continuous low (dim) light and continuous higher (regular) light conditions. Circadian oscillations in branchlet an

Pectin is a normal constituent of cell walls of green plants. When supplied externally to live cells or walls isolated from the large-celled green alga Chara corallina, pectin removes calcium from load-bearing cross-links in the wall, loosening the structure and allowing it to deform more rapidly under the action of turgor pressure. New Ca(2+) enters the vacated positions in the wall and the externally supplied pectin binds to the wall, depositing new wall material that strengthens the wall. A calcium pectate cycle has been proposed for these sub-reactions. In the present work, the cycle was t

In the cells of Chara corallina, permeant monohydric alcohols including methanol, ethanol and 1-propanol increased the hydraulic resistance of the membrane (Lpm-1). We found that the relative value of the hydraulic resistance (rLpm-1) was linearly dependent on the concentration (Cs) of the alcohol. The relationship is expressed in the equation: rLpm-1 = ρmCs + 1, where ρm is the hydraulic resistance modifier coefficient of the membrane. Ye et al. (2004) showed that membrane-permeant glycol ethers also increased Lp-1. We used their data to estimate Lpm-1 and rLpm-1. The values of rLpm-1 fit the

Biological and environmental effects of lanthanide series of elements have received much attention recently due to their wide applications. In this study, effects of La(3+) treatments on calcium and magnesium concentrations as well as cytoplasmic streaming of internodal cells of Chara corallina were investigated. At all treatment concentrations (10, 100, and 1,000 μM), La(3+) significantly decreased calcium concentrations in the cell-wall fractions after 5-h treatments. Calcium concentrations in the cell contents and magnesium concentrations in the cell-wall fractions were reduced by 100 and 1

BACKGROUND: Chara vulgaris (C. vulgaris), a green macroalga containing antioxidant and antibacterial compounds, shows potential in treating infections and inflammation. Enterococcus faecalis is an opportunistic Gram-positive pathogen responsible for skin and soft tissue infections, especially in compromised patients. Due to rising antibiotic resistance, new treatments are needed. Selenium nanoparticles (SeNPs) have enhanced antimicrobial and antioxidant properties. This study explores C. vulgaris-derived selenium nanoparticles (CV-SeNPs) as a therapeutic agent against E. faecalis infection, ai

It is generally agreed that solute transport across the Chara plasma membrane is energized by a proton electrochemical gradient maintained by an H(+)-extruding ATPase. Nonetheless, as deduced from steady-state current-voltage (I-V) measurements, the kinetic and thermodynamic constraints on H(+)-ATPase function remain in dispute. Uncertainties necessarily surround long-term effects of the relatively nonspecific antagonists used in the past; but a second, and potentially more serious problem has sprung from the custom of subtracting, across the voltage spectrum, currents recorded following pump

BACKGROUND: Bioactive compounds with a wide range of chemical compositions and biological functions are found in the Chlorophyceae family. The present work investigated the anticancer effect of ethanolic extract from Chara vulgaris (C. vulgaris) and its selenium nanoformulation (CvSeNPs) against solid Ehrlich carcinoma (SEC).
METHODS: Gas chromatography-mass spectroscopy was used to analyze C. vulgaris, and many analytical methods were used to characterize the biosynthesized CvSeNPs, including zeta potential, particle size, polydispersity index (PDI), SEM, TEM, and FTIR. In addition, mice wit

The impact of unstirred layers (USLs) during cell pressure probe experiments with Chara corallina internodes has been quantified. The results show that the hydraulic conductivity (Lp) measured in hydrostatic relaxations was not significantly affected by USLs even in the presence of high water flow intensities ('sweep-away effect'). During pressure clamp, there was a reversible reduction in Lp by 20%, which was explained by the constriction of water to aquaporins (AQPs) in the C. corallina membrane and a rapid diffusional equilibration of solutes in arrays where water protruded across AQPs. In

CYP73 and CYP98 are two cytochrome P450 monooxygenase families present in bryophytes and tracheophytes, and they play key roles in the formation of phenolic compounds containing a caffeic acid moiety and more complex phenolic compounds. CYP73 encodes the second enzyme in the core phenylpropanoid pathway and is the first enzyme family in this pathway so far exclusively found in land plants and not in any water-living sister clades, like the streptophyte algae. That makes CYP73 a prime candidate for investigations into necessary adaptations during terrestrialization. This study aims to support o
