Recently reported research from this laboratory has demonstrated the autotrophic growth of certain hydrogen-uptake-positive strains of Rhizobium japonicum and defined minimal conditions for such growth. Ribulose 1,5-bisphosphate carboxylase has been detected in autotrophically growing cells, but at low specific activity. Moreover, growth rates were low, and growth ceased at low cell densities. We report here improved autotrophic growth rates of R. japonicum SR through the use of a modified mineral salts/vitamins medium and a programmed increase in oxygen tension as autotrophic growth proceeds.

Rhizobium japonicum strain SR grows chemoautotrophically on a mineral salts medium when incubated in an H2- and CO2-containing atmosphere. Mutant strains unable to grow or that grow very poorly chemoautotrophically with H2 have been isolated from strain SR. The mutant isolation procedure involved mutagenesis with ethyl methane sulfonate, penicillin selection under chemoautotrophic growth conditions, and plating of the survivors onto medium containing carbon. The resulting colonies were replica plated onto medium that did not contain carbon, and the plates were incubated in an H2- and CO2-conta

This study investigated the production of microbial protein (MP) using a novel microbial consortium composed of microalgae and hydrogen-oxidizing bacteria (HOB). This photo-chemoautotrophic consortium was aimed at capturing and valorising carbon dioxide (CO2) while overcoming typical limitations of each microbial group. Batch tests were run under varying gas mixtures of oxygen (O2), CO2 and hydrogen (H2) to assess process performance in terms of biomass growth, nitrogen assimilation, gas consumption, final biomass and microbial community composition. The consortium was compared to the single m

The effect of nitrate, nitrite, nitric oxide (NO), and nitrous oxide on a mixed, mesophilic (35 degrees C) methanogenic culture was investigated. Short-term inhibition assays were conducted at a concentration range of 10-350 mg N/L nitrate, 17-500 mg N/L nitrite, 0.02-0.8 mg N/L aqueous NO, and 19-191 mg N/L aqueous nitrous oxide. Simultaneous methane production and N-oxide reduction was observed in 10 and 30 mg N/L nitrate and 0.02 mg N/L aqueous NO-amended cultures. However, addition of N-oxide resulted in immediate cessation of methanogenesis in all other cultures. Methanogenesis completely

The "wooden-steps" hypothesis [Distel DL, et al. (2000) Nature 403:725-726] proposed that large chemosynthetic mussels found at deep-sea hydrothermal vents descend from much smaller species associated with sunken wood and other organic deposits, and that the endosymbionts of these progenitors made use of hydrogen sulfide from biogenic sources (e.g., decaying wood) rather than from vent fluids. Here, we show that wood has served not only as a stepping stone between habitats but also as a bridge between heterotrophic and chemoautotrophic symbiosis for the giant mud-boring bivalve Kuphus polythal

Deep continental subsurface hosts diverse microbial ecosystems that are primarily driven by chemolithoautotrophy. In this study, we investigated deep continental igneous rock-hosted bacterial populations enriched under microoxic (ME) and anoxic (AE) conditions. Metataxonomic, metagenomics and metabolomics approaches, along with physiological analyses, were performed to elucidate community composition, CO2 utilization and possible bioconversion potential of subsurface rock enrichment cultures under chemolithoautotrophic conditions. Following prolonged incubation, ME enrichments resulted in high

Chemolithoautotrophic bacteria represent a diverse group of microorganisms capable of utilising carbon dioxide (CO2) as their sole carbon source, deriving the necessary energy for growth and metabolism from redox reactions involving inorganic compounds such as hydrogen, sulfur, or iron. These organisms have attracted substantial scientific and industrial interest due to their dual potential to function as biological CO2 sinks, mitigating greenhouse gas accumulation, and as microbial platforms for the sustainable biosynthesis of value-added compounds. Their natural CO2 fixation pathways, includ

Flue gas mitigation technologies aim to reduce the environmental impact of flue gas emissions, particularly from industrial processes and power plants. One approach to mitigate flue gas emissions involves bio-mitigation, which utilizes microorganisms to convert harmful gases into less harmful or inert substances. The review thus explores the bio-mitigation efficiency of chemolithotrophic interactions with flue gas and their potential application in bio-reactors. Chemolithotrophs are microorganisms that can derive energy from inorganic compounds, such as carbon dioxide (CO2), nitrogen oxides (N

BACKGROUND: Symbiotic relationships between microbes and their hosts are widespread and diverse, often providing protection or nutrients, and may be either obligate or facultative. However, the genetic mechanisms allowing organisms to maintain host-symbiont associations at the molecular level are still mostly unknown, and in the case of bacterial-animal associations, most genetic studies have focused on adaptations and mechanisms of the bacterial partner. The gutless tubeworms (Siboglinidae, Annelida) are obligate hosts of chemoautotrophic endosymbionts (except for Osedax which houses heterotr

Permanently anoxic regions in the ocean are widespread and exhibit unique microbial metabolic activity exerting substantial influence on global elemental cycles and climate. Reconstructing microbial metabolic activity rates in these regions has been challenging, due to the technical difficulty of direct rate measurements. In Cariaco Basin, which is the largest permanently anoxic marine basin and an important model system for geobiology, long-term monitoring has yielded time series for the concentrations of biologically important compounds; however, the underlying metabolite fluxes remain poorl

Mixotrophy offers a promising strategy for biosynthesis by simultaneously utilizing organic carbon and CO2; however, mixotrophic microorganisms are rarely isolated outside of photoautotrophic microalgae. In this study, the chemoautotroph Cupriavidus necator H16 was found to preferentially consume fructose when coexisting with CO2 and H2, switching to utilization of only CO2 and H2 after fructose depletion. Transcriptomic analysis revealed significant differences in genes involved in energy metabolism, electron generation, and the respiratory chain. The molecular mechanism underlying the inabil

In almost all respiratory organisms, organic substrates are degraded via catabolic processes to the central metabolite acetyl-CoA which is then oxidized to CO2 for energy metabolism or used as a building block for anabolism. Most microorganisms have either the closed tricarboxylic acid cycle or the complete Wood-Ljungdahl-pathway for acetyl-CoA oxidation, but the sulfate-reducing, naphthalene-degrading culture N47 possesses both completely. Combining 13C- labeled substrates and mass-specific GC-MS analysis of amino acids and fatty acids with enzyme activity assays suggests that N47 has a chemo

Autotrophic organisms fix dissolved inorganic carbon (DIC: CO2, HCO3-, CO3-2) into biomass, introducing organic carbon into the global carbon cycle. Many lineages of autotrophs developed CO2 concentrating mechanisms (CCMs) to grapple with the catalytic constraints of the carboxylase of the Calvin-Benson-Bassham cycle, ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO). Carboxysomes and DIC transporters act together to form CCMs, which facilitate DIC fixation by autotrophic bacteria when DIC is scarce. Autotroph Thiomicrospira pelophila has carboxysomes, with an unusual carbonic anhydras

Current challenges in the anaerobic bioremediation of benzene are the lack of capable cultures and limited knowledge on the biodegradation pathway. Under methanogenic conditions, benzene may be mineralized by syntrophic interactions between microorganisms, which are poorly understood. The present study developed an optimized formula for anoxic medium to successfully promote the growth of the putative benzene degrader Deltaproteobacterium Hasda-A and enhance the benzene degradation activity of methanogenic enrichment cultures. Within 70 d of incubation, the benzene degradation activity and re

The reduction of elemental sulfur is an important energy-conserving pathway in prokaryotes inhabiting geothermal environments, where sulfur respiration contributes to sulfur biogeochemical cycling. Despite this, the pathways through which elemental sulfur is reduced to hydrogen sulfide remain unclear in most microorganisms. We integrated growth experiments using Thermovibrio ammonificans, a deep-sea vent thermophile that conserves energy from the oxidation of hydrogen and reduction of both nitrate and elemental sulfur, with comparative transcriptomic and proteomic approaches, coupled with scan

Chemolithotrophic ammonia-oxidizing bacteria and Thaumarchaeota are central players in the global nitrogen cycle. Obligate ammonia chemolithotrophy has been characterized for bacteria; however, large gaps remain in the Thaumarchaeotal pathway. Using batch growth experiments and instantaneous microrespirometry measurements of resting biomass, we show that the terrestrial Thaumarchaeon Nitrososphaera viennensis EN76(T) exhibits tight control over production and consumption of nitric oxide (NO) during ammonia catabolism, unlike the ammonia-oxidizing bacterium Nitrosospira multiformis ATCC 25196(T

Autotrophic CO₂ fixation is a fundamental metabolic process that enables microorganisms to inhabit carbon-limited environments. Multiple pathways mediate this process, with variants distributed across diverse taxa and some genes shared among pathways, making their identification from genomic data challenging. Here, we present a curated resource comprising pathway-specific KEGG Orthology marker genes and a lightweight, rule-based tool AutoFixMark for predicting the presence of seven known CO₂ fixation pathways in microbial genomes. To support marker gene identification and benchmarking, we comp

This study evaluated the effect of three sulfate salt-based culture media on the reprecipitation of sulfur under the action of two types of bacterial inoculum, a pure strain of Acidithiobacillus ferrooxidans (ATCC 23270) and a consortium of this strain and Acidithiobacillus thiooxidans (ATCC 15494), in a biodesulfurization process for coal (particle size < 0.25 mm) from the 'La Guacamaya' mine (Puerto Libertador, Córdoba, Colombia). All of the experiments were periodically monitored, with measurements taken of pH, cell concentration, iron concentration, and pyrite oxidation. Additionally, mine

As the non-photosynthetic microbial community (NPMC) isolated from seawaters utilized inorganic carbon sources for carbon fixation, the concentrations and ratios of Na2CO3, NaHCO3, and CO2 were optimized by response surface methodology design. With H2 as the electron donor, the optimal carbon sources were 270 mg/L Na2CO3, 580 mg/L NaHCO3, and 120 mg/L CO2. The carbon fixation efficiency in response to total organic carbon (TOC) was up to 30.59 mg/L with optimal carbon sources, which was about 50% higher than that obtained with CO2 as the sole carbon source. The mixture of inorganic carbon sour

The hydrothermal vent tubeworm Riftia pachyptila is a dominant member of many hydrothermal vent communities along the East Pacific rise and is one of the fastest growing metazoans known. Riftia flourish in diffuse hydrothermal fluid flows, an environment with high spatial and temporal heterogeneity in physical and chemical conditions. To date, physiological and biochemical studies of Riftia have focused on Riftia's adaptations to its chemoautotrophic bacterial symbionts. However the relation between in situ physico-chemical heterogeneity and Riftia host and symbiont metabolism, in particular s
