Disease modification and symptom relief in osteoarthritis using a mutated GCP-2/CXCL6 chemokine.

We showed that the chemokine receptor C-X-C Motif Chemokine Receptor 2 (CXCR2) is essential for cartilage homeostasis. Here, we reveal that the CXCR2 ligand granulocyte chemotactic protein 2 (GCP-2) was expressed, during embryonic development, within the prospective permanent articular cartilage, but not in the epiphyseal cartilage destined to be replaced by bone. GCP-2 expression was retained in adult articular cartilage. GCP-2 loss-of-function inhibited extracellular matrix production. GCP-2 treatment promoted chondrogenesis in vitro and in human cartilage organoids implanted in nude mice in

Constitutive expression of the antibacterial CXC chemokine GCP-2/CXCL6 by epithelial cells of the male reproductive tract.

The reproductive tract is continuously challenged by potential pathogens present in the environment. Therefore, robust host defense mechanisms are essential both for the health of the individual and for fertilization. Antibiotic innate immunity peptides possess broad antimicrobial activity. Recently, we found that the CXC chemokine, granulocyte chemotactic protein (GCP)-2/CXCL6, possesses antibacterial activity. In the present study, we investigated, therefore, the presence of GCP-2/CXCL6 in the human male reproductive system. GCP-2/CXCL6 was detected at 19nM (mean; range: 5-47nM; n=14) in sem

Adipose Mesenchymal Stem Cells Overexpressing Chemokine (CXCL6) Showed Strong Angiogenesis and Anti-Inflammatory Ability in the Treatment of Pulmonary Hypertension.

Pulmonary arterial hypertension (PAH) is a progressive disease characterized by vascular remodeling and right ventricular failure, with limited effective treatments. This study aimed to evaluate therapeutic effects of adipose-derived mesenchymal stem (ADMS) cells engineered to overexpress chemokine CXCL6 in monocrotaline-induced PAH rats. ADMS cells were transduced with a CXCL6-expressing lentivirus and characterized by flow cytometry, migration assays, and tube formation tests. Following establishment of PAH model, rats were administered intravenous injections of CXCL6-ADMS or control cells.

Ethanol promoting the upregulation of C-X-C Motif Chemokine Ligand 1(CXCL1) and C-X-C Motif Chemokine Ligand 6(CXCL6) in models of early alcoholic liver disease.

Alcoholic liver disease (ALD) denotes a series of liver diseases caused by ethanol. Recently, immune-related genes (IRGs) play increasingly crucial role in diseases. However, it's unclear the role of IRGs in ALD. Bioinformatic analysis was used to discern the core immune-related differential genes (IRDGs) in the present study. Subsequently, Cell Counting Kit-8 say, oil red O staining, and triglyceride detection were employed to explore optimal experimental conditions of establishing hepatocellular models of early ALD. Ultimately, real-time reverse transcription-PCR and immunohistochemistry/imm

CXCL6: A potential therapeutic target for inflammation and cancer.

Chemokines were originally defined as cytokines that affect the movement of immune cells. In recent years, due to the increasing importance of immune cells in the tumor microenvironment (TME), the role of chemokines has changed from a single "chemotactic agent" to a key factor that can regulate TME and affect the tumor phenotype. CXCL6, also known as granulocyte chemoattractant protein-2 (GCP-2), can recruit neutrophils to complete non-specific immunity in the process of inflammation. Cancer-related genes and interleukin family can promote the abnormal secretion of CXCL6, which promotes tumor

CXCL6 Reshapes Lipid Metabolism and Induces Neutrophil Extracellular Trap Formation in Cholangiocarcinoma Progression and Immunotherapy Resistance.

The chemokine CXCL6 is identified as a pivotal regulator of biological processes across multiple malignancies. However, its function in cholangiocarcinoma (CCA) is underexplored. Tumor profiling for CXCL6 is performed using a public database. Both in vitro and in vivo experiments are utilized to evaluate the oncogenic effects of CXCL6 on CCA. Additionally, RNA-Seq is employed to detect transcriptomic changes related to CXCL6 expression in CCA cells and neutrophils. Molecular docking, fluorescence colocalization, and Co-IP are used to elucidate a direct interaction between JAKs and CXCR1/2. Add

ELR positive CXCL chemokines are highly expressed in an animal model of ulcerative colitis.

Background: The presence of neutrophil-rich inflammation in colon tissues of patients with ulcerative colitis (UC) is one of the most important histological characteristics of this disease. However, the expression of CXCL chemokines governing the infiltration of neutrophils in UC has not been well elucidated. Materials and methods: In this experimental study, the UC model was induced in Wistar rats by administration of 2 mL 4% acetic acid into the large colon through the rectum. Animals were anesthetized after 48 hrs; their colon tissue samples were isolated for macroscopic and histopathologic

Pirfenidone exerts anti-inflammatory effects by inhibiting CXCL6 to alleviate idiopathic pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) poses a health challenge with the destruction of lung architecture. It is featured by limited effective therapeutic options and a complex pathological mechanism. The molecular mechanism underlying the antifibrotic agent pirfenidone (PFD) remains to be fully determined. This study investigated the anti-inflammatory effects of pirfenidone in the treatment of IPF and explored its regulation of the C-X-C motif chemokine ligand 6 (CXCL6), a potent neutrophil chemoattractant. Using the murine model, pirfenidone has been identified to limit lung inflammation and ne

Anti-inflammatory effects of the GAG-binding CXCL9(74-103) peptide in dinitrofluorobenzene-induced contact hypersensitivity in mice.

BACKGROUND: To recruit leucocytes to an inflammatory site, chemokine binding to glycosaminoglycans (GAGs) is critical. Therefore, strategies to interfere with this interaction, aiming at the production of anti-inflammatory agents, were developed. These include production of modified chemokines without affinity for G protein-coupled receptors but with enhanced affinity for GAGs. Such modified chemokines compete with functional chemokines for GAG binding, prevent chemokine immobilization and presentation, and inhibit leucocyte migration. In addition to modified chemokines, a GAG-binding peptide

Scleroderma dermal microvascular endothelial cells exhibit defective response to pro-angiogenic chemokines.

OBJECTIVES: Angiogenesis plays a critical role in SSc (scleroderma). The aim of this study was to examine the expression of growth-regulated protein-γ (Gro-γ/CXCL3), granulocyte chemotactic protein 2 (GCP-2/CXCL6) and their receptor CXCR2 in endothelial cells (ECs) isolated from SSc skin and determine whether these cells mount an angiogenic response towards pro-angiogenic chemokines. The downstream signalling pathways as well as the pro-angiogenic transcription factor inhibitor of DNA-binding protein 1 (Id-1) were also examined. METHODS: Skin biopsies were obtained from patients with dcSSc. E

Modulation of the bovine trophoblastic innate immune response by Brucella abortus.

Brucellosis is still a widespread zoonotic disease. Very little is known about the interaction between Brucella abortus and trophoblastic cells, which is essential for better understanding the pathogenesis of the Brucella-induced placentitis and abortion, a key event for transmission of the disease. The goal of this study was to evaluate the profile of gene expression by bovine trophoblastic cells during infection with B. abortus. Explants of chorioallantoic membranes were inoculated with B. abortus strain 2308. Microarray analysis was performed at 4 h after infection, and expression of cytoki

Nickel exposure aggravates aortic dissection by exacerbating neutrophil recruitment and NETosis to compromise endothelial barrier.

Nickel exposure elevates aortic dissection (AD) risk, yet its pathogenic mechanisms remain unclear. Here, we demonstrate that nickel accelerates AD progression, particularly in hypertensive individuals. Bioinformatics analysis of GEO datasets identified chemokine-mediated endothelial-neutrophil crosstalk as a key pathway. Using a neutrophil-endothelial co-culture model, nickel activated the CXCL6/CXCL8-CXCR2 axis, triggering endothelial secretion of CXCL6/8 and upregulating neutrophil CXCR2 expression. This dual effect promoted neutrophil recruitment, adhesion, and migration, validated in nick

High-affinity ELR+ CXC chemokines display balanced G protein-, GRK- and β-arrestin-associated transducer engagement at the viral G protein-coupled receptor ORF74.

Open reading frame 74 (ORF74) is the single G protein-coupled receptor (GPCR) encoded in the genome of Kaposi's sarcoma-associated herpesvirus (KSHV). Due to its high constitutive activity and broad chemokine binding profile, ORF74 contributes to KSHV-associated diseases through autocrine and paracrine mechanisms that promote cell proliferation, inflammation and angiogenesis. Previous studies showed that ELR+ CXC chemokines (i.e., CXCL1-3 and CXCL5-8), which contain a conserved Glu-Leu-Arg motif, bind ORF74 with high affinity. Among these chemokines, CXCL1-3 and CXCL6-8 act as full or partial
