Gastric adenocarcinoma with chief cell differentiation.

A case of adenocarcinoma with chief cell differentiation, a novel entity in the stomach, is presented. An 82-year-old woman who had undergone distal gastrectomy, was scheduled for upper gastrointestinal endoscopy to clarify mechanical ileus. A protruding tumor 16 x 14 x 9 mm in size was found in the cardia of the remnant stomach. Histological examination indicated a well-differentiated tubular adenocarcinoma composed of basophilic columnar or cuboidal cells with occasional coarse eosinophilic granules. Immunohistochemical analysis revealed strong expression of pepsinogens I and II and Runt-rel

Gastric chief cell mass in duodenal ulcer.

Chief cell mass and serum Pepsinogen I were calculated in 25 duodenal ulcer patients and in 40 healthy controls. Comparisons were also made with parietal cell mass and the stimulated acid secretion. Chief cell mass was expressed bay a Zymogenous Index (ZI) obtained by multiplying the number of chief cells per mm2 for the thickness of the glandular layer. The analysis of the data shows a significant decrease of ZI in duodenal ulcer patients in comparison with controls. The grouping of the subjects according to the age (over and under fifty years), confirms that ZI decreases significantly in duo

Secretion of intrinsic factor from cultured rat gastric chief cells.

Intrinsic factor (IF) is a vitamin B12 binding protein that is secreted from the gastric mucosa. We tested secretagogues which stimulate IF secretion in rat gastric perfusion and found that carbachol and cholecystokinin octapeptide (CCK-8) stimulated secretion, but histamine and tetragastrin did not. To confirm these results, we examined IF secretion from isolated rat chief cells. For this purpose, we established an enzyme immunoassay (EIA) using an avidin-biotin peroxidase complex to measure small amounts of IF. To prepare an anti-rat IF, IF was isolated from the stomach, and was injected int

An easy method to highlight chief cells in gastric biopsies.

BACKGROUND: Hematoxylin and eosin (H&E) is not an optimal stain to discriminate chief cells from parietal cells in gastric biopsies MATERIALS AND METHODS: Fifteen sets of biopsies from the gastric corpus were consecutively stained with H&E and toluidine blue stains; chief cells stained deep blue with toluidine blue, thus contrasting with lightly-stained parietal cells. In well-oriented sections, the continuity of the chief cell zone was studied at × 4 magnification and its thickness in one field at × 10 magnification. RESULTS: Toluidine blue stained fundic sections that exhibited normal muco

[Chief cell mass in gastric ulcer: cyto-secretory correlations].

The aim of this experience has been to evaluate the chief cell mass and serum pepsinogen I in gastric ulcer patients. Comparisons were also made with parietal cell mass and acid secretion. Chief cell mass and serum pepsinogen I are not only influenced by the localization of ulcer but, also, by the histological condition of fundic mucosa. In fact, the behaviour of serum pepsinogen I and chief cell mass in type I gastric ulcer is the same observed in case of fundic chronic gastritis without gastric ulcer. In case of gastric ulcer type I with superficial fundic gastritis it emerges normozymogenis

M3 muscarinic receptors mediate pepsinogen secretion via polyphosphoinositide hydrolysis in guinea pig gastric chief cells.

The muscarinic receptor system involved in pepsinogen secretion from isolated guinea pig gastric chief cells was investigated by evaluating the effect of muscarinic receptor antagonists on carbamylcholine (CCh)-stimulated chief cell responses. CCh stimulated the hydrolysis of polyphosphoinositide in chief cells at the same concentrations as it stimulated pepsinogen secretion. Each of five different muscarinic receptor antagonists, atropine, pirenzepine, 4-diphenylacetoxy-N-methylpiperidine methiodide (4-DAMP), AF-DX116 and scopolamine, inhibited both pepsinogen secretion and inositol phosphate

[Epithelial formula and relationship between the chief and parietal cells of fundal gastric glands in gastric and duodenal ulcer].

The fundic glands of 143 stomachs resected in connection with peptic ulcer, including those localized in the stomach (70 observations) and in the duodenum (73 observations) were studied. The number of epithelial cells in the longitudinal section of the fundic gland in ulcer of the stomach was most often in the range of 100 to 149 (in 37 cases of 70), in duodenal ulcer--150 to 199 (in 36 observations of 73). In patients with duodenal ulcer, as distinct from those with gastric ulcer, the epithelial formula of the fundic gland was characterized by a lower content of non-differentiated and mucous

IP6K1 interacts with the syndecan SDC4 to support secretory granule biogenesis in gastric chief cells.

Inositol hexakisphosphate kinases (IP6Ks) catalyze the synthesis of the inositol pyrophosphate 5-InsP7, and regulate diverse physiological processes. Ip6k1-/- mice display reduced body weight despite normal food intake, elevated fecal protein, and reduced skeletal muscle mass compared with Ip6k1+/+ mice, suggesting that IP6K1 may support protein digestion. IP6K1 is expressed throughout the mouse gastrointestinal tract, and is especially enriched in the cytoplasm of chief cells in the stomach, which are responsible for the storage and secretion of digestive enzymes. Proliferation and Differentiation of Gastric Mucous Neck and Chief Cells During Homeostasis and Injury-induced Metaplasia.

BACKGROUND & AIMS: Adult zymogen-producing (zymogenic) chief cells (ZCs) in the mammalian gastric gland base are believed to arise from descending mucous neck cells, which arise from stem cells. Gastric injury, such as from Helicobacter pylori infection in patients with chronic atrophic gastritis, can cause metaplasia, characterized by gastric cell expression of markers of wound-healing; these cells are called spasmolytic polypeptide-expressing metaplasia (SPEM) cells. We investigated differentiation and proliferation patterns of neck cells, ZCs, and SPEM cells in mice. METHODS: C57BL/6 mice

Multimerin1 and not Galectin-8 tempers WNT signaling to promote gastric chief cell differentiation.

Galectins are a family of proteins that bind galactose-containing glycans. One member, galectin-8, preferentially binds galactose that contains a terminal sulfate. Aberrant expression and secretion of sulfated glycosylation epitopes, such as 3'-Sulfo-LeA/C, is a feature of high-risk human foregut metaplasias. In addition, recent work has demonstrated that 3'-Sulfo-LeC is a marker of mature murine zymogenic chief cells of the stomach and that 3'-Sulfo-LeC epitope is secreted via cathartocytosis during the cellular transition to a metaplastic state. Based on those findings, we used Lgals8-/- mic

Cystine/Glutamate Antiporter (xCT) Is Required for Chief Cell Plasticity After Gastric Injury.

BACKGROUND & AIMS: Many differentiated epithelial cell types are able to reprogram in response to tissue damage. Although reprogramming represents an important physiological response to injury, the regulation of cellular plasticity is not well understood. Damage to the gastric epithelium initiates reprogramming of zymogenic chief cells into a metaplastic cell lineage known as spasmolytic polypeptide-expressing metaplasia (SPEM). The present study seeks to identify the role of xCT, a cystine/glutamate antiporter, in chief cell reprogramming after gastric injury. We hypothesize that xCT-dependen

Gastric adenocarcinoma with chief cell differentiation: a proposal for reclassification as oxyntic gland polyp/adenoma.

Gastric adenocarcinoma with chief cell differentiation (GA-CCD) has been reported as a new, rare variant of gastric adenocarcinoma. Only 12 cases in Japanese patients have been described to date, but they demonstrate distinct clinicopathologic features. To further characterize these lesions, we have collected 10 additional cases. Patients ranged in age from 44 to 79 years (mean, 64.2 y) with a relatively equal sex distribution (6 women and 4 men). Stratified by race, 4 patients were Hispanic, 2 were White, 2 were African American, 1 was Asian (Chinese), and the race was unknown for 1 patient.

Gastric chief cell mass in chronic gastritis. Count and relationships to parietal cell mass and functional indices.

The chief cell mass, expressed as the Zymogenous Index (number of cells per mm2 multiplied by the thickness of the glandular layer) was calculated in 42 subjects with chronic gastritis, and in 40 subjects with normal gastric mucosa, and was compared with the serum concentration of Pepsinogen I (PG I), with the parietal cell mass (expressed as Parietal Index: number of cells per mm2 multiplied by the thickness of the glandular layer), and with the acid output. The results showed that there are no significant variations in the chief cells in comparison with healthy controls in the case of superf

Gastric adenocarcinoma of the fundic gland (chief cell-predominant type): A review of endoscopic and clinicopathological features.

Gastric adenocarcinoma of the fundic gland (chief cell-predominant type, GA-FG-CCP) is a rare variant of well-differentiated adenocarcinoma, and has been proposed to be a novel disease entity. GA-FG-CCP originates from the gastric mucosa of the fundic gland region without chronic gastritis or intestinal metaplasia. The majority of GA-FG-CCPs exhibit either a submucosal tumor-like superficial elevated shape or a flat shape on macroscopic examination. Narrow-band imaging with endoscopic magnification may reveal a regular or an irregular microvascular pattern, depending on the degree of tumor exp

Dynamic role of gastric stem cells and chief cells in precancerous lesions of gastric cancer: global knowledge mapping and emerging trends based on bibliometric analysis from 2004 to 2024.

BACKGROUND: Gastric stem cells (GSCs) and chief cells are vital for maintaining gastric epithelial homeostasis. However, under pathological conditions, these cells undergo significant functional changes, contributing to the progression of precancerous lesions of gastric cancer (PLGC). Dysregulation of key signaling pathways such as WNT, NF-κB, and YAP leads to aberrant cellular behaviors, which are implicated in the early stages of gastric carcinogenesis. This study aimed to elucidate the roles of GSCs and chief cells in maintaining gastric epithelial integrity, their contributions to the deve

Gastric adenocarcinoma of fundic gland type (chief cell predominant type) treated with endoscopic aspiration mucosectomy.

Upper endoscopy screening in an asymptomatic 56-year-old man showed a small, yellowish elevated lesion with a central depression on the posterior wall in the gastric cardia. Biopsy specimens from this lesion were suspicious of carcinoid tumor. We suspected this lesion to be a sporadic gastric carcinoid tumor with a diameter of 5 mm, limited to the mucosal layer. We then performed an endoscopic aspiration mucosectomy with a cap-fitted endoscope. Microscopically, the lesion obtained from the resected specimen was minimally invasive to the submucosa and showed highly differentiated columnar cells

Effect of hydrochloric acid and prostaglandins on pepsinogen synthesis and secretion in canine gastric chief cell monolayer cultures.

The actions of hydrochloric acid, natural PGE1, PGE2 and a synthetic commercial PGE1 preparation, Enisoprost (Searle) on pepsinogen synthesis and secretion were studied in canine chief cell monolayer cultures. Hydrochloric acid, applied directly to the apical surface of chief cells using culture plate inserts (Millipore) had no effect on secretion, nor did it affect the action of any secretagogue in the basolateral medium. All prostaglandins tested showed significant stimulation of pepsinogen secretion. Basal secretion of pepsinogen after 90 min was 9.4 (1.3)% to total initial monolayer conten

[Total peptic activity in gastric juice and peptic cell mass (chief fundic cells and mucopeptic fundic-antral cells): cellular-secretory correlations in normal subjects].

The aim of this experience has been to evaluate cytosecretory correlations between total peptic activity in gastric juice and gastric peptic cell mass (chief cells and fundic-antral mucopeptic cells) in normal subjects. Gastric peptic cell mass and total peptic activity do not change depending on sex. Concerning age, in spite of a significant decrease of gastric peptic cell mass, peptic activity does not significantly decrease. This apparent disagreement between decreasing anatomic data and functional data, remaining unchanged, may be explained by analyzing the behaviour of fundic and antral p
