Chitin synthase inhibitors as antifungal agents.

Increased risk of fungal diseases in immunocompromised patients, emerging fungal pathogens, limited repertoire of antifungal drugs and resistance development against the drugs demands for development of new and effective antifungal agents. With greater knowledge of fungal metabolism efforts are being made to inhibit specific enzymes involved in different biochemical pathways for the development of antifungal drugs. Chitin synthase is one such promising target as it is absent in plants and mammals. Nikkomycin Z, a chitin synthase inhibitor is under clinical development. Chitin synthesis in fung

[Chitin Synthase 2 (CHS2) gene of Malassezia species].

Malassezia species have been recognized as members of the microbiological flora of human and animal skin; they are also considered to play an important role in the pathogenesis of folliculitis, atopic dermatitis and otitis externa. Therefore, the molecular characteristics were investigated to clarify the epidemiology and the pathogenesis of diseases associated with Malassezia species in human and animals. Molecular investigation was made of 105 clinical isolates of M. pachydermatis from dogs and cats by random amplification of polymorphic DNA (RAPD) and chitin synthase 2 (CHS2) gene sequence a

Chitin synthase in the filarial parasite, Brugia malayi.

Fragments of putative chitin synthase (chs) genes from two filarial species (Brugia malayi and Dirofilaria immitis) were amplified by PCR using degenerate primers. The full genomic and cDNA sequences were obtained for the B. malayi chs gene (Bm-chs-1); the predicted amino acid sequence is highly similar, over a large region, to two CHS sequences of the nematode Caenorhabditis elegans and also to two insect CHS sequences. Bm-chs-1 is abundantly transcribed in B. malayi adult females, independent of their fertilization status, but is also expressed in males and microfilariae. Oocytes and early e

Fungal Chitin Synthases: Structure, Function, and Regulation.

Chitin is an essential polysaccharide of the fungal cell wall, critical for structural integrity, cell division and, in pathogenic fungi, virulence. As chitin is absent in both plant and mammalian systems, chitin synthases are considered attractive targets for the specific control of fungal pathogens. Yet despite decades of research, structural information on chitin synthases was lacking and inhibitors have failed to gain approval in the clinic. Current inhibitors are also ineffective against major agricultural pathogens such as Aspergillus and Fusarium species, largely due to the presence of

Chitin synthase, a fungal glycosyltransferase that is a valuable antifungal target.

During the last 30 past years, more life-threatening fungal infections have appeared due to the increasing frequency of patients with weakened immune systems. Inhibition of fungal enzymes involved in the biosynthesis of sterols is considered to be a safe and effective option for antifungal therapy. However, the intensive use of sterol-biosynthesis inhibitors for years has resulted in resistance development. Consequently, the search for alternative therapeutics must be intensified. In this context, the biosynthesis of chitin, an essential component of the fungal cell wall that is absent in mamm

Evaluation of the susceptibility of Coccidioides immitis to lufenuron, a chitin synthase inhibitor.

The activity of the chitin synthase inhibitor lufenuron was evaluated in vitro using the spherule-endospore (SE) phase of Coccidioides immitis. The lufenuron was also used to treat mice infected with C. immitis by the respiratory route. In vitro, lufenuron had no effect upon fungal cell growth. Two formulations of lufenuron were evaluated in vivo. Neither the oral nor the injectable lufenuron extended the survival of mice infected with C. immitis when compared with placebo-treated mice.

A putative role for calmodulin in the activation of Neurospora crassa chitin synthase.

The possible role of calmodulin in cell wall formation and chitin synthesis was studied in Neurospora crassa by examining the effects of anti-calmodulin agents on protoplast regeneration and possible associations between chitin synthase and calmodulin related proteins in microsomal isolates. Protoplast regeneration was inhibited by trifluoperazine (> 20 microM), an anticalmodulin agent. Chitin synthase activity in microsomes was associated with that of calmodulin-dependent protein kinase and inhibited by trifluoperazine (100 microM). In vitro activity of chitin synthase was enhanced upon inclu

RNAi Targeting Chitin synthase Genes Specifically Affects Larval Performance in Tuta absoluta.

An RNA interference (RNAi)-based management strategy has a great potential for control of the tomato leafminer Tuta absoluta, a microlepidoptera pest damaging a broad range of Solanaceae hosts. Nevertheless, whether RNAi efficiency of the target genes is affected by dsRNA reachability and mRNA levels remains to be established in T. absoluta. Two chitin synthase genes, TaChSA and TaChSB, were highly expressed in ectodermally-derived cells and midgut respectively, facilitating to test dsRNA reachability using oral delivery. According to the 2- ΔCT value, TaChSA average level was much higher than

Discovery and activity evaluation of fungal chitin synthase inhibitors via virtual screening.

Chitin synthase is a fungal-specific and highly selective antifungal target, and its inhibitors represent a promising direction for antifungal drug development. In this study, a structure-based high-throughput virtual screening was performed against a compound library comprising 1.6 million molecules, using the high-resolution cryo-electron microscopy structure of Candida albicans chitin synthase 2 (CaChs2) as the molecular target. Based on the virtual screening results, a machine learning-based classification model was constructed to identify key structural features associated with high CaChs

Morphology engineering of Penicillium chrysogenum by RNA silencing of chitin synthase gene.

Chitin synthases, that catalyze the formation of chitin the major component of cell walls in most filamentous fungi, play crucial roles in the growth and morphogenesis. To investigate the roles of chitin synthase in Penicillium chrysogenum, we developed an RNAi system to silence the class III chitin synthase gene chs4. After transformation, mutants had a slow growth rate and shorter but highly branched hyphae. All transformants either were unable to form conidia or could form only a few. Changes in chs4 expression could lead to a completely different morphology and eventually cause distinct pe

Identification of Sporothrix schenckii based on sequences of the chitin synthase 1 gene.

Sporothrix schenckii is pathogenic to human and animals. To detect S. schenckii in the tissue, we designed specific oligonucleotide primers based on the chitin synthase gene. Amplification products were selectively obtained from S. schenckii DNA. Polymerase chain reaction analysis with the primer pair S2-R2 was able to detect 10 pg genomic DNA of S. schenckii with ethidium bromide staining. This detection system will be useful as a microbiological tool for the diagnosis of human and animal sporotrichosis.

Recent progress in the chemotherapy of human fungal diseases. Emphasis on 1,3-β-glucan synthase and chitin synthase inhibitors.

Systemic fungal infections of humans and economically important animals are increasingly common throughout the world. These infections are severe and often hard to treat with existing safe, oral medications. Thus there has been increasing research on alternatives resulting in study of natural and synthetic inhibitors of 1,3-β-Glucan synthase (GS) and chitin synthase (CS)-enzymes important in the biosynthesis of fungal cell walls that are not utilized in human biochemistry. Some such agents have recently been introduced into parenteral clinical use. There is hope that safe agents of this type w

AOP Report: Inhibition of Chitin Synthase 1 Leading to Increased Mortality in Arthropods.

Arthropods (including insects, crustaceans, and arachnids) rely on the synthesis of chitin to complete their life cycles (Merzendorfer 2011). The highly conserved chitin synthetic process and the absence of this process in vertebrates make it an exploitable target for pest management and veterinary medicines (Merzendorfer 2013; Junquera et al. 2019). Susceptible, nontarget organisms, such as insects and aquatic invertebrates, exposed to chitin synthesis inhibitors may suffer population declines, which may have a negative impact on ecosystems and associated services. Hence, it is important to p

Chitin synthase A: a novel epidermal development regulation gene in the larvae of Bombyx mori.

Chitin synthase is the key regulatory enzyme for chitin synthesis and excretion in insects, as well as a specific target of insecticides. The chitin synthase A gene (BmChsA) cloned from Bombyx mori, the model species of lepidopteran, is an epidermis-specific expressed gene during the molting stage. Knockdown BmChsA gene in 3rd instar larvae increased the number of non-molting and abnormal molting larvae. Exposure to nikkomycin Z, a chitin synthase inhibitor downregulated the expression of BmChsA and decreased the amount of epidermis chitin during the molting process. The thickness of the new e

Fusarium verticillioides chitin synthases CHS5 and CHS7 are required for normal growth and pathogenicity.

Fusarium verticillioides is both an endophyte and a pathogen of maize and is a health threat in many areas of the world because it can contaminate maize with fumonisins, a toxic secondary metabolite. We identified eight putative chitin synthase (CHS) genes in F. verticillioides genomic sequence, and phylogenetic evidence shows that they group into seven established CHS gene classes. We targeted two CHSs (CHS5 and CHS7) for deletion analysis and found that both are required for normal hyphal growth and maximal disease of maize seedlings and ears. CHS5 and CHS7 encode a putative class V and clas

Identification of the conserved coding sequences of three chitin synthase genes in Fonsecaea pedrosoi.

Primers having designs based on highly conserved stretches in the deduced amino acid sequences of chitin synthase (CHS) genes were used in PCR reactions to amplify 600 bp and 366 bp products from the genomic DNA of three major causal agents of chromoblastomycosis. Cloning and sequencing of the PCR products of one of these fungi, Fonsecaea pedrosoi, identified three CHS sequences designated as FpCHS1, FpCHS2 and FpCHS3. FpCHS1 and FpCHS2 were homologous to regions of CHS1 and CHS2 of Saccharomyces cerevisiae, and their derived amino acid sequences fell into chitin synthase classes I and II, res

Design, synthesis, and biological evaluation of quinoline derivatives as dual-target inhibitors of chitin synthase and glucosamine-6-phosphate synthase.

Two series of compounds were designed and synthesized based on a glycine methyl ester-quinoline core scaffold combined with a fragment containing nitrogen using either an isoxazole or a piperidine carboxamide fragment as a linker as dual inhibitors targeting both chitin synthase (CHS) and glucosamine-6-phosphate synthase (GlmS). The inhibition percentage (IP) values of these compounds against CHS ranged from 29.9% to 84.5%, while their inhibition percentages against GlmS spanned 38.1% to 86.9% at the concentration of 300 μg mL-1. Inhibition of Phytophthora infestans chitin synthase via cyclic peptide targeting for sustainable disease control.

Phytophthora spp. are devastating plant pathogens causing significant economic losses in agriculture, but unlike true fungi, oomycetes have cell walls composed predominantly of cellulose and glucans, with little to no detectable chitin. Despite this, one to two putative chitin synthase (Chs) genes are present in Phytophthora spp. suggesting a functional role in its biology. In this study, we characterized the single chitin synthase of P. infestans (PiChs) to better understand its function and explore its potential as a target for novel fungicidal treatments. Unlike typical chitin synthases tha
