Chlordecone-induced hepatic dysfunction.

Chlordecone (Kepone) is a decachloroketone analog of the dodecachlorohydrocarbon mirex and is used as a stomach poison insecticide. Despite the structural similarity to mirex, chlordecone is unlike mirex in general organ-specific toxic properties. Chlordecone is primarily accumulated in the liver, where it causes a variety of morphological and biochemical alterations. Although less effective than mirex as a hepatotoxin, it causes liver enlargement, focal necrosis, mitochondrial changes, fatty infiltration of hepatocytes, and proliferation of endoplasmic reticulum. Chlordecone accumulation and

Potentiation of CCl4 lethality by chlordecone.

Previous studies have dealt with the propensity of chlordecone (CD) to potentiate the hepatotoxicity of CCl4. The effect of this interaction upon CCl4 lethality has never been reported. The objective of the present study was to evaluate the effect of CD on CCl4 lethality and several parameters associated with cytochrome P-450 activity. Male Sprague-Dawley rats (150-200 g) were fed powdered diets containing 0 to 10 ppm CD or 225 ppm phenobarbital (PB) for 15 days. On day 15, rats from each group received an i.p. injection of CCl4 in corn oil and the 48-h LD50 determined. Additional animals were

Effects of prenatal chlordecone on sexually differentiated behavior in adult rats.

In rodents, exposure to estrogens during early development masculinizes the structure and function of the brain. The effects of early exposure to estrogens or estrogenic compounds can be evaluated by neurobehavioral testing after puberty. In this study, the effect of developmental exposure to the chlorinated pesticide, chlordecone (CD) on sexually differentiated behaviors in adults was investigated because CD binds to estrogen receptors and causes estrogenic effects in the reproductive tract of humans and rodents at relatively high doses. Pregnant Sprague-Dawley rats were exposed to 5 mg/kg CD

Induction of cytochrome P-450 isozymes by mirex and chlordecone.

The effect of the insecticides, mirex and chordecone (Kepone), on the cytochrome P-450 monooxygenase system in C57BL/6N mouse liver microsomes was studied. Mice were treated intraperitoneally with low (6 mg/kg) and high (30 mg/kg) doses of mirex and chlordecone in corn oil for 2 days. For comparison, mice were also treated with either phenobarbital (PB) or 3-methylcholanthrene (3-MC). All treatments significantly increased the hepatic microsomal P-450 content over that of controls. Benzphetamine N-demethylase, ethoxyresorufin O-deethylase, benzo[a]pyrene hydroxylase, and acetanilide hydroxylas

Chlordecone, a persistent pollutant: analytical challenges and sustainable remediation solutions.

Chlordecone, a persistent organochlorine insecticide, was extensively applied in the French West Indies between 1972 and 1993. Decades after its ban, it continues to contaminate soils and waters, cycling through food webs and resulting in chronic exposure of both ecosystems and human populations. Its environmental persistence, strong affinity for organic matter, and adverse effects on human health have made it a pressing public health concern. The region now exhibits the highest incidence of prostate cancer globally, reflecting complex interactions between environmental exposure and genetic su

Tolerance of aged Fischer 344 rats against chlordecone-amplified carbon tetrachloride toxicity.

We have investigated the effects of chlordecone 1(CD)+CCl4 combination in adult (3 months), middle aged (14 months), and old aged (24 months) male Fischer 344 (F344) rats. After a non-toxic dietary regimen of CD (10 ppm) or normal powdered diet for 15 days, rats received a single non-toxic dose of CCl4 (100 microl/kg, i.p., 1:4 in corn oil) or corn oil (500 microl/kg, i.p.) alone on day 16. Liver injury was assessed by plasma ALT, AST, and histopathology during a time course of 0-96 h. Liver tissue repair was measured by [3H-CH3]-thymidine (3H-T) incorporation into hepatic nuclear DNA and prol

Chlordecone-induced hepatotoxicity and fibrosis are mediated by the proteasomal degradation of septins.

Chlordecone (CLD) is a pesticide persisting in soils and contaminating food webs. CLD is sequestered in the liver and poorly metabolized into chlordecol (CLDOH). In vitro liver cell models were used to investigate the fate and mechanistic effects of CLD and CLDOH using multiomics. A 3D-cell model was used to investigate whether CLD and CLDOH can affect susceptibility to the metabolic dysfunction-associated steatotic liver disease (MASLD). Hepatocytes were more sensitive to CLD than CLDOH. CLDOH was intensively metabolized into a glucuronide conjugate, whereas CLD was sequestered. CLD but not C

Ultra-trace quantification method for chlordecone in human fluids and tissues.

Chlordecone is an organochlorine pesticide (OCP) considered as a Persistent Organic Pollutant (POP) as it persists in the environment, bio-accumulates through the food web, causes adverse effects to human health and the environment and transports across international boundaries far from its sources. The atypical physico-chemical properties of chlordecone make its inclusion in classical analytical approaches non applicable. The aim of our work was to include chlordecone in a multi organochlorine residue method preventing any degradation during the analytical process and thus allowing quantifica

Mechanism of the lethal interaction of chlordecone and CCl4 at non-toxic doses.

There is significant interest in the possibility of unusual toxicity due to interaction of toxic chemicals upon environmental or occupational exposures even though such exposures may involve levels ordinarily considered harmless individually. While many laboratory and experimental models exist for such interactions, progress in this area of toxicology has suffered for want of a model where the interactants are individually non-toxic. We developed such a model where prior exposure to non-toxic levels of the pesticide Kepone (chlordecone) results in a 67-fold amplification of CCl4 lethality in e

Chlordecone exposure and its hidden burden on human health: mechanisms, epidemiology, and biomonitoring.

Chlordecone is a highly persistent organochlorine pesticide that has caused long-term environmental contamination, particularly in the French West Indies, where it was widely used on banana plantations between the 1970s and the early 1990s. Its chemical stability, high lipophilicity, and strong affinity for organic matter have led to prolonged retention in soil, water, and food chains, resulting in chronic exposure of local populations. Experimental studies indicate that chlordecone acts as a neurotoxin, hepatotoxin, and endocrine disruptor, and is involved in biological processes consistent w

Articulations of poisoning: historicity, relationality, and contextuality of Chlordecone pollution in the French Antilles.

Chlordecone, a highly persistent organochlorine insecticide, was widely applied to banana plantations in Martinique and Guadeloupe from 1972 until being banned in 1993. Decades later, its residues still permeate soil, waterways, food systems, and human bodies in the two islands. Classified as both a carcinogenic and an endocrine disruptor, the chemical compound has generated widespread protests, epidemiological debates, and political critique. Drawing on six months of non-consecutive ethnographic fieldwork conducted in Martinique between 2017 and 2022, this article examines how people interpre

Ovarian dysfunction following prenatal exposure to an insecticide, chlordecone, associates with altered epigenetic features.

Chlordecone (CD) is an insecticide that was used in the French West Indies for several years to control the banana root borer pest. Given its nonsignificant degradation, it persists in the environment. CD is a carcinogenic compound with reproductive and developmental toxicity and is a recognized endocrine-disrupting chemical. In this study, we examined the effects of CD on female reproductive system of mice with the focus on epigenetic features in ovary. Our data show that gestational exposure to low dose of CD affects meiotic double-strand breaks repair in female embryos. In adult mice derive

Estradiol-chlordecone (Kepone) interactions: additive effect of combinations for uterotropic and embryo implantation functions.

Chlordecone (Kepone) is a polychlorinated hydrocarbon that has a low affinity for the estrogen receptor. Although the compound has been shown to be an estrogen antagonist for some responses within the central nervous system, and in hypophyseal gonadotrophic cells, such an action for other responses has not been reported. In the present study ovariectomized immature rats were used to determine the uterotropic effects of chlordecone in the presence or absence of estradiol. Administration of 30 mg/kg chlordecone per day for 3 days increased the response to 0.01, 0.1 and 1 microgram/kg per day of

Chlordecone potentiates hepatic fibrosis in chronic liver injury induced by carbon tetrachloride in mice.

Chronic liver damage due to viral or chemical agents leads to a repair process resulting in hepatic fibrosis. Fibrosis may lead to cirrhosis, which may progress to liver cancer or a loss of liver function, with an associated risk of liver failure and death. Chlordecone is a chlorinated pesticide used in the 1990s. It is not itself hepatotoxic, but its metabolism in the liver triggers hepatomegaly and potentiates hepatotoxic agents. Chlordecone is now banned, but it persists in soil and water, resulting in an ongoing public health problem in the Caribbean area. We assessed the probable impact o
