Natural thermometers: chloroplasts as heat sensors.

All organisms need to sense their surroundings and respond. This is especially true for plants, which are rooted to the ground and must contend with daily fluctuations of both above- and belowground environments. Of these, temperature is perhaps the most variable, with shifts throughout the day and microenvironments that affect one part of the plant differently from another (e.g., a leaf in the canopy vs. one in the shade). Heat stress is of particular concern and negatively affects basic cellular processes including protein folding and function, membrane fluidity, and cytoskeletal organizatio

ARSENATE INDUCED CHLOROSIS 1/ TRANSLOCON AT THE OUTER ENVOLOPE MEMBRANE OF CHLOROPLASTS 132 Protects Chloroplasts from Arsenic Toxicity.

Arsenic (As) is highly toxic to plants and detoxified primarily through complexation with phytochelatins (PCs) and other thiol compounds. To understand the mechanisms of As toxicity and detoxification beyond PCs, we isolated an arsenate-sensitive mutant of Arabidopsis (Arabidopsis thaliana), arsenate induced chlorosis1 (aic1), in the background of the PC synthase-defective mutant cadmium-sensitive1-3 (cad1-3). Under arsenate stress, aic1 cad1-3 showed larger decreases in chlorophyll content and the number and size of chloroplasts than cad1-3 and a severely distorted chloroplast structure. The

Removal of Mn from spinach chloroplasts by sodium cyanide and the binding of Mn2+ to Mn-depleted chloroplasts.

Manganese and copper were released from spinach chloroplasts by NaCN-treatment, though iron was not affected. The Hill reaction activity was also inhibited by this treatment, but was partially recovered by the addition of either Mn2+ or Cu2+, but not of Fe3+. The interaction of Mn2+ with manganese-depleted chloroplasts by NaCN-treatment was studied using 54Mn2+. A Scatchard plot shows the high and low affinity binding sites of Mn2+ on NaCN-treated chloroplast membrane; high affinity binding being specific for NaCN-treated chloroplast with a binding constant, KH, of 1.9 X 10(5) M-1, and a maxim

Preparation and characterization of phospholipid-depleted chloroplasts.

Spinach class II chloroplasts were treated with snake venom phospholipase A2 in the presence of bovine serum albumin, and separated by sucrose-density centrifugation. The treatment yielded phospholipid-depleted chloroplasts which had lost 82.6% of the original phospholipids. About 20% of the phospholipids of chloroplasts were resistant to enzyme attack. These results suggest that phospholipids exist in two states in chloroplast membranes. In spite of considerable phospholipid depletion, the chloroplast preparations retained a large portion of their photoactivities, i.e. light-induced electron

Studies on the manganese of the chloroplast.

Manganese deficiency of green plants is known to affect preferentially the activity of the oxygen evolving system in the photosynthetic apparatus. Our studies showed that the time needed to reactivate photosynthesis in Mn-deficient algae varies with each culture, and is often very short when Mn is added not before illumination but during the light period. The recent finding by Cheniae and Martin that the reactivation requires light, is confirmed. The plain incorporation of (54)Mn into deficient algae as distinguished from reactivation was barely affected by light, yet was inhibited by uncouple

Cold-sensitive chloroplast development: advances from chloroplast ribosome-deficient mutants.

Cold-sensitive chloroplast ribosome-deficient mutants reveal that low temperature disrupts chloroplast ribosome biogenesis, chloroplast translation, and nascent polypeptide protection, thereby impairing chloroplast biogenesis and delaying chloroplast development in young leaves. In nature, cold stress commonly delays leaf greening in juvenile leaves of susceptible plant species. Accumulating evidence suggests that cold stress impairs chloroplast development, primarily by suppressing translational processes within chloroplasts, which play a pivotal role in their biogenesis. Although wild-type m

Chloroplastic Aspartyl-tRNA Synthetase Is Required for Chloroplast Development, Photosynthesis and Photorespiratory Metabolism.

Photorespiration is a complex metabolic process linked to primary plant metabolism and influenced by environmental factors, yet its regulation remains poorly understood. In this study, we identified the asprs3-1 mutant, which displays a photorespiratory phenotype with leaf chlorosis, stunted growth, and diminished photosynthesis under ambient CO2, but normal growth under elevated CO2 conditions. Map-based cloning and genetic complementation identified AspRS3 as the mutant gene, encoding an aspartyl-tRNA synthetase. AspRS3 is localised in both chloroplasts and mitochondria, with the chloroplast

The chloroplast import receptor is an integral membrane protein of chloroplast envelope contact sites.

A chloroplast import receptor from pea, previously identified by antiidiotypic antibodies was purified and its primary structure deduced from its cDNA sequence. The protein is a 36-kD integral membrane protein (p36) with eight potential transmembrane segments. Fab prepared from monospecific anti-p36 IgG inhibits the import of the ribulose-1,5-bisphosphate carboxylase small subunit precursor (pS) by interfering with pS binding at the chloroplast surface. Anti-p36 IgGs are able to immunoprecipitate a Triton X-100 soluble p36-pS complex, suggesting a direct interaction between p36 and pS. This im

Photoinduction of cyclosis-mediated interactions between distant chloroplasts.

Communications between chloroplasts and other organelles based on the exchange of metabolites, including redox active substances, are recognized as a part of intracellular regulation, chlororespiration, and defense against oxidative stress. Similar communications may operate between spatially distant chloroplasts in large cells where photosynthetic and respiratory activities are distributed unevenly under fluctuating patterned illumination. Microfluorometry of chlorophyll fluorescence in vivo in internodal cells of the alga Chara corallina revealed that a 30-s pulse of localized light induces

Optimal disk packing of chloroplasts in plant cells.

Photosynthesis is essential for ecosystem survival, but while plants require light, excessive exposure can damage cells. Chloroplasts, photosynthetic organelles, respond via self-organized motion within cells to optimize light absorption. These disk-shaped organelles must balance two competing needs: dense packing to enhance absorption under dim light and rapid spatial rearrangement to avoid damage from excess light. Using microscopy, we show that plant cell shape and chloroplast size achieve both goals: dense monolayer packing for optimal absorption in low light and sidewall packing for light

Engineered chloroplasts as vaccine factories to combat bioterrorism.

Bacillus anthracis is ideal for making biological weapons, but the licensed anthrax vaccine is unsuitable for widespread public administration. Recombinant subunit-vaccine candidates offer potential alternatives, and plant-based production systems facilitate the inexpensive bulking of target antigens. A recent report demonstrates expression of anthrax protective antigen in tobacco chloroplasts--this material is immunogenic and protective when injected into mice. Provided an economic purification scheme can be developed, this technology holds promise for an improved vaccine.

Engineering Rubisco condensation in chloroplasts to manipulate plant photosynthesis.

Although Rubisco is the most abundant enzyme globally, it is inefficient for carbon fixation because of its low turnover rate and limited ability to distinguish CO2 and O2, especially under high O2 conditions. To address these limitations, phytoplankton, including cyanobacteria and algae, have evolved CO2-concentrating mechanisms (CCM) that involve compartmentalizing Rubisco within specific structures, such as carboxysomes in cyanobacteria or pyrenoids in algae. Engineering plant chloroplasts to establish similar structures for compartmentalizing Rubisco has attracted increasing interest for i

Expression of tetanus toxin Fragment C in tobacco chloroplasts.

Fragment C (TetC) is a non-toxic 47 kDa polypeptide fragment of tetanus toxin that can be used as a subunit vaccine against tetanus. Expression of TetC in Escherichia coli and yeast was dependent on the availability of synthetic genes that were required to improve translation efficiency and stabilize the mRNA. To explore the feasibility of producing TetC in tobacco leaves, we attempted expression of both the bacterial high-AT (72.3% AT) and the synthetic higher-GC genes (52.5% AT) in tobacco chloroplasts. We report here that the bacterial high-AT mRNA is stable in tobacco chloroplasts. Signifi

The Ultrastructure of Chloroplasts in Mineral-deficient Maize Leaves.

The ultrastructure of mesophyll chloroplasts in full-nutrient and mineral-deficient maize (Zea mays) leaves was examined by electron microscopy after glutaraldehyde-osmium tetroxide fixation. Nitrogen, calcium, magnesium, phosphorus, potassium, and sulfur deficiencies were induced by growing the plants in nutrient culture. Distinctive chloroplast types were observed with each deficiency. Chloroplasts from nitrogen-deficient plants were reduced in size and had prominent osmiophilic globules and large grana stacks. Magnesium deficiency was characterized by the accumulation of osmiophilic globule

Chloroplast genome sequence of triploid Toxicodendron vernicifluum and comparative analyses with other lacquer chloroplast genomes.

BACKGROUND: Toxicodendron vernicifluum, belonging to the family Anacardiaceae, is an important commercial arbor species, which can provide us with the raw lacquer, an excellent adhesive and painting material used to make lacquer ware. Compared with diploid, triploid lacquer tree has a higher yield of raw lacquer and stronger resistance to stress. Triploid T. vernicifluum was a newly discovered natural triploid lacquer tree. However, the taxonomy of triploid T. vernicifluum has remained uncertain. Here, we sequenced and analyzed the complete chloroplast (cp) genome of triploid T. vernicifluum a

Arabidopsis Chloroplast protein for Growth and Fertility1 (CGF1) and CGF2 are essential for chloroplast development and female gametogenesis.

BACKGROUND: Chloroplasts are essential organelles of plant cells for not only being the energy factory but also making plant cells adaptable to different environmental stimuli. The nuclear genome encodes most of the chloroplast proteins, among which a large percentage of membrane proteins have yet to be functionally characterized. RESULTS: We report here functional characterization of two nuclear-encoded chloroplast proteins, Chloroplast protein for Growth and Fertility (CGF1) and CGF2. CGF1 and CGF2 are expressed in diverse tissues and developmental stages. Proteins they encode are associate

Chloroplast SecA functions as a membrane-associated component of the Sec-like protein translocase of pea chloroplasts.

Protein cross-linking studies with a thylakoid membrane translocation intermediate were used to demonstrate that chloroplast SecA functions as a membrane-associated component of the Sec-like ATP-dependent protein translocase of pea chloroplasts. In assays with isolated thylakoids, it was observed that translocation of the 33-kDa protein of the oxygen-evolving complex of photosystem II (OE33) decreased when the ATP concentration was low, and that the protein accumulated as a bound precursor. The bound precursor was able to be translocated into the lumen when the ATP concentration was raised, in

Sustainable production of myoglobin meat protein in plant chloroplasts.

Alternative routes for producing animal proteins are crucial for reducing the reliance on traditional livestock farming, which contributes significantly to greenhouse gas emissions, deforestation, and water consumption. Myoglobin (Mb) is an important oxygen-binding hemoprotein found in vertebrate muscle which enhances the nutritional and sensorial properties of meat. Due to its unique functionality, Mb has been heterologously expressed in a variety of organisms, although only transient expression in Nicotiana benthamiana has been reported for higher plants. In this study, we used chloroplast t
