A new optimized regularized Stokeslet model reveals the effects of multicellular protozoan colony configuration on hydrodynamic performance.

Many microbial eukaryotes have unicellular life stages, but can also form multicellular colonies. We explored hydrodynamic consequences of colony morphology, which affects swimming and flux of prey-carrying water to cells in a colony, using the choanoflagellate, Choanoeca flexa, which forms cup-like colonies that can turn inside-out so flagella line the cup's interior or cover its outside surface. Detailed hydrodynamic models incorporating cell morphologies are not feasible for colonies with many cells. Therefore, we designed a reduced model of each cell using regularized-force-dipoles with pa

The architecture of cell differentiation in choanoflagellates and sponge choanocytes.

Although collar cells are conserved across animals and their closest relatives, the choanoflagellates, little is known about their ancestry, their subcellular architecture, or how they differentiate. The choanoflagellate Salpingoeca rosetta expresses genes necessary for animal development and can alternate between unicellular and multicellular states, making it a powerful model for investigating the origin of animal multicellularity and mechanisms underlying cell differentiation. To compare the subcellular architecture of solitary collar cells in S. rosetta with that of multicellular 'rosette'

Diversity and spatial segregation of TRP channels in the closest relatives of animals.

Sensory systems, built around specialized cell types, are central to how animals perceive and respond to their environments. However, the evolutionary origin of these systems remains unclear. Here, we show that choanoflagellates, the sister group of animals, possess remarkably diverse repertoires of transient receptor potential (TRP) channels, an ancient superfamily of sensory ion channels. Comparative analyses indicate that most major animal TRP channel families predate the origin of animals and reveal extensive radiations of the TRPM and TRPW families within choanoflagellates, with the latte

Evolutionarily diverse caveolins share a common structural framework built around amphipathic disks.

Caveolins are a unique family of membrane remodeling proteins present broadly across animals (Metazoa), and in vertebrates form flask-shaped invaginations known as caveolae. While human caveolin-1 assembles into an amphipathic disk composed of 11 spirally packed protomers, the structural basis underlying caveolin function across animals remains elusive. Here, we predicted structures for 73 caveolins spanning animal diversity, as well as a newly identified choanoflagellate caveolin from Salpingoeca rosetta. This analysis revealed seven conserved structural elements and a propensity to assemble

Characterization of an NO-Activated Homodimeric Soluble Guanylate Cyclase from a Choanoflagellate.

Soluble guanylate cyclases are gas-sensing enzymes in eukaryotes that catalyze the formation of cyclic GMP from GTP. While commonly studied sGCs from insects and vertebrates are heterodimers, there are additional classes of homodimeric gas-sensing sGCs in eukaryotes that are not as well characterized. Reported here is the characterization of Cf sGC1 isolated from the organism Choanoeca flexa, a single-celled eukaryote. Cf sGC1 is a homodimeric sGC that exhibits a three-state activation profile in response to NO similar to that observed with heterodimeric NO-responsive sGCs. Cf sGC1 was isolate

Nitric oxide signaling controls collective contractions in a colonial choanoflagellate.

Although signaling by the gaseous molecule nitric oxide (NO) regulates key physiological processes in animals, including contractility,1-3 immunity,4,5 development,6-9 and locomotion,10,11 the early evolution of animal NO signaling remains unclear. To reconstruct the role of NO in the animal stem lineage, we set out to study NO signaling in choanoflagellates, the closest living relatives of animals.12 In animals, NO produced by the nitric oxide synthase (NOS) canonically signals through cGMP by activating soluble guanylate cyclases (sGCs).13,14 We surveyed the distribution of the NO signaling

Riding the Storm: Microeukaryote Community Responses to Hurricane Disturbance.

Sandy beaches are dynamic ecosystems where terrestrial and marine organisms coexist, resulting in diverse communities where microeukaryotes (protists, microalgae, and meiofauna) are key drivers. Understanding their diversity patterns and responses to stressors is essential for elucidating ecological processes, yet information on this subject remains limited. Herein, microeukaryotic diversity in a sandy beach in Acapulco, Guerrero, was analyzed via ITS1 metabarcoding assessing community structure changes following hurricane Otis. Overall, the number of ASVs decreased substantially from 260 befo

Characterization of the tandem CWCH2 sequence motif: a hallmark of inter-zinc finger interactions.

BACKGROUND: The C2H2 zinc finger (ZF) domain is widely conserved among eukaryotic proteins. In Zic/Gli/Zap1 C2H2 ZF proteins, the two N-terminal ZFs form a single structural unit by sharing a hydrophobic core. This structural unit defines a new motif comprised of two tryptophan side chains at the center of the hydrophobic core. Because each tryptophan residue is located between the two cysteine residues of the C2H2 motif, we have named this structure the tandem CWCH2 (tCWCH2) motif. RESULTS: Here, we characterized 587 tCWCH2-containing genes using data derived from public databases. We catego

Evolutionary Proteomics Uncovers Ancient Associations of Cilia with Signaling Pathways.

Cilia are organelles specialized for movement and signaling. To infer when during evolution signaling pathways became associated with cilia, we characterized the proteomes of cilia from sea urchins, sea anemones, and choanoflagellates. We identified 437 high-confidence ciliary candidate proteins conserved in mammals and discovered that Hedgehog and G-protein-coupled receptor pathways were linked to cilia before the origin of bilateria and transient receptor potential (TRP) channels before the origin of animals. We demonstrated that candidates not previously implicated in ciliary biology locali
