Oligomeric cholates: amphiphilic foldamers with nanometer-sized hydrophilic cavities.

The hydroxyl at the C-3 of cholic acid was converted to an amino group, and the resulting amino-functionalized cholic acid was used as a monomer to prepare amide-linked oligomeric cholates. These cholate oligomers fold into helical structures with nanometer-sized hydrophilic internal cavities in solvent mixtures consisting of mostly nonpolar solvents such as carbon tetrachloride or ethyl acetate/hexane and 2-5% of a polar solvent such as methanol or DMSO. The conformations of the foldamers were studied by UV, fluorescence, fluorescence quenching, and fluorescence resonance energy transfer. The

Cholate Shunt, Oral Cholate Challenge and Endoscopic Lesions of Portal Hypertension: The SHUNT-V Study.

BACKGROUND: The accuracy of current criteria for ruling out large oesophageal varices (LEV) and other endoscopic lesions of portal hypertension (PH) may be compromised by obesity and MASLD/MASH. AIMS: In the US multicentre SHUNT-V study, we evaluated the disease severity index (DSI) for detecting LEV and other lesions of PH at endoscopy. METHODS: Subjects were adults with compensated cirrhosis scheduled for endoscopy to screen for varices. DSI was calculated from clearances of labelled cholates after oral and intravenous administration. DSI ≤ 18.3 was evaluated as a cut-off for ruling out LE

Sodium cholate ameliorates nonalcoholic steatohepatitis by activation of FXR signaling.

Non-alcoholic steatohepatitis (NASH) has become a major cause of liver transplantation and liver-associated death. The gut-liver axis is a potential therapy for NASH. Sodium cholate (SC) is a choleretic drug whose main component is bile acids and has anti-inflammatory, antifibrotic, and hepatoprotective effects. This study aimed to investigate whether SC exerts anti-NASH effects by the gut-liver axis. Mice were fed with an high-fat and high-cholesterol (HFHC) diet for 20 weeks to induce NASH. Mice were daily intragastric administrated with SC since the 11th week after initiation of HFHC feedin

Dietary cholate is required for antiatherogenic effects of ethanol in mouse models.

BACKGROUND: Human consumption of moderate amounts of ethanol is associated with reduced cardiovascular events. Studies examining the effect of ethanol on atherosclerosis in mouse models have yielded conflicting results that may be due to differences in dietary fat and cholate content. To determine if dietary cholate influences ethanol's effect on atherosclerosis, we fed apolipoprotein E-/- and low-density lipoprotein receptor (LDLR)-/- mice different liquid diets with or without ethanol. METHODS: Apolipoprotein E-/- mice were fed a low-fat or high saturated fat, cholate-containing diet with o

Mild hepatic fibrosis in cholesterol and sodium cholate diet-fed rats.

To date, the majority of research on hypercholesterolemia has focused on the effects of a high cholesterol diet on atherosclerosis and coronary heart disease. The toxic effects of cholesterol on the liver and the relationship between the intake of a high cholesterol diet and hepatic fibrosis, however, have not been investigated clearly or histopathologically. Male Wistar rats were fed a diet supplemented with 1.0% cholesterol and 0.3% sodium cholate for 12 weeks. Rats were sacrificed and analyzed via blood biochemistry, traditional microscopy and immunohistochemistry. Following the feeding of

The ratio between the fast and slow forms of bovine cytochrome c oxidase is changed by cholate or nucleotides bound to the cholate-binding site close to the cytochrome a3/CuB binuclear centre.

We determined the fraction of 'slow' and 'fast' conformations of bovine cytochrome c oxidase, following the kinetics of cyanide binding to the oxidized enzyme. We investigated whether treatment of heart mitochondrial particles with different commercially available types of cholate (standard and ultrapure) can affect the fraction of cytochrome c oxidase in the two states. Compared to standard cholate, the use of ultra-pure cholate for solubilization of heart mitochondrial particles significantly increased the fraction of the fast enzyme. Complete homogeneity (approximately 100% fast) was observ

Alterations in Liver Perfusion in Adults With Fontan Circulation as Assessed by Dual Cholate Clearance.

BACKGROUND: Fontan circulation (FC) in complex congenital heart disease is characterized by altered hemodynamics and associated with Fontan-associated liver disease. Patients with FC may exhibit abnormalities in cholate clearance due to abnormal perfusion. We aimed to compare cholate clearance in adults with FC to healthy controls and explore associations between cholate clearance and clinical features. METHODS AND RESULTS: This is a prospective cohort study of patients with FC ≥18 years of age between 2019 and 2022. Systemic and portal hepatic clearance of cholate was assessed using a dual c

[The bile cholate-cholesterol coefficient for assessing the causes of a protracted convalescence in patients with viral hepatitis].

Changes of the content of bile acids, cholate-cholesterol coefficient, bile acid fraction ratios in the bile were assessed in patients with viral hepatitis and chronic cholecystitis. A scheme is proposed for evaluation of bile bladder function considering four states: functional hypocholia, cholecystitis, threat of cholelithiasis. It is shown that in 23-28% protracted convalescence in viral hepatitis and manifested in prolonged blood serum ALAT activity is caused by the presence of subclinical form of cholecystitis.

Hepatic cholate clearance as assessed by HepQuant-SHUNT is associated with clinical outcomes in individuals with Fontan circulation.

BACKGROUND: Fontan-associated liver disease [FALD] is universal in individuals with Fontan circulation [FC]. The dual cholate clearance test is a noninvasive, flow-dependent measure of liver function. We aim to explore the association between cholate clearance and clinical outcomes in this population. METHODS: Prospective study of adults with FC ≥ 18 years from University of Pennsylvania [Penn] and Northwestern University [NU]. Hepatic clearance of oral d4-cholate and intravenous 13C-cholate were measured from peripheral venous blood and calculated as portal hepatic filtration rate [HFR], sys

Genetic Evidence for a Putative B Cell-Cholate Immunometabolic Axis in Pericarditis: A Mendelian Randomization Study.

Background: Pericarditis is an inflammatory heart disease with unclear causal mechanisms. Immune dysregulation is implicated, but observational studies cannot reliably infer causality. Whether plasma metabolites mediate immune-pericarditis pathways remains unknown. Methods: A two-sample Mendelian randomization (MR) study was performed using publicly available summary-level genome-wide association study data. Genetic instruments were selected via significance screening, linkage disequilibrium clumping, and strength evaluation (F-statistic > 10). A relaxed instrument selection threshold of p < 1

Uranyl-ion Directed Hydrogelation of Cholate for Enhanced Emission Resolution and Energy Transfer-driven Sensing.

In this work, we report the first example of an actinide (UO2 2+) induced hydrogel formation. We discovered that the gel phase led to a remarkable improvement in the resolution of the emission from the uranyl cation. This phenomenon was previously seen only in sophisticated uranyl complexes in organic solvents or in the solid-state. The organized and confined gel microenvironment facilitated efficient energy transfer from UO2 2+→ Eu3+ in the hydrogel matrix. Additionally, this "turn-on" energy transfer strategy provided a simple method for uranyl sensing using commercially available ingredient

Cost-effectiveness of an oral cholate challenge test for the management of patients at risk for large esophageal varices.

AIMS: The dual oral cholate challenge test (DuO) quantifies liver function and portal-systemic shunting. Herein we report the economic impact of the use of the DuO Disease Severity Index (DSI) in the clinical management of patients with chronic liver disease suspected of having large esophageal varices. METHODS: A Markov health state transition model of 100,000 patients with chronic liver disease suspected of having varices was populated with previously reported epidemiological, utility, and price data to assess the cost-effectiveness of employing the DuO test against the standard of care. Diabetes downregulates peptide transporter 1 in the rat jejunum: possible involvement of cholate-induced FXR activation.

Peptide transporter 1 (PepT1), highly expressed on the apical membrane of enterocytes, is involved in energy balance and mediates intestinal absorption of peptidomimetic drugs. In this study, we investigated whether and how diabetes affected the function and expression of intestinal PepT1. Diabetes was induced in rats by combination of high-fat diet and low dose streptozocin injection. Pharmacokinetics study demonstrated that diabetes significantly decreased plasma exposures of cephalexin and acyclovir following oral administration of cephalexin and valacyclovir, respectively. A novel protein-preserving passive tissue clearing approach using sodium cholate and urea for whole-organ imaging.

The recent advancements in tissue-clearing techniques have opened new possibilities for non-invasive three-dimensional (3D) volumetric imaging of a wide range of biological specimens. Passive tissue-clearing methods use diffusion-based processes to infiltrate clearing reagents into samples without mechanical forces or energy input, aiming to minimize the sample disruption while preserving the tissue architecture and molecular information. Nevertheless, these methods often rely on sodium dodecyl sulfate (SDS) as a delipidating detergent, which has a risk of causing tissue damage and protein dis
