Chromosome banding studies in Philadelphia chromosome positive myeloid leukaemia.

A cytogenetic study of Ph1 positive myeloid leukaemia in both chronic and acute phases had been made by a chromosome banding technique. The translocation (t(9;22)(q34;q11), designated t(Ph1) was present in the myeloid cells of 43 of 44 patients; the exceptional case had normal number 9 chromosomes and a different translocation (t(19;22)(q13;q11)). A translocation additional to that involving the Ph1 was found as a stable abnormality present in all myeloid cells in 4 patients, chromosome 17 being involved in 2. The association of isochromosome number 17 with blast crisis was confirmed. New data

Two cases of ring chromosome 13. Chromosome banding patterns and mosaic configuration.

Two ring chromosomes 13 were studied by means of various cytogenetic techniques in two psychomotorically retarded male infants. Common features of our patients include microcephalia, hypertelorism, wide and prominent nose bridge, and cryptorchism. Various configurations of the aberrant chromosomes could be identified in cultured skin fibroblasts and peripheral lymphocytes from both patients. Chromosome heteromorphisms and analysis of silver stained nucleolus organizer regions (Ag-NOR) substantiated the parental origin of the ring chromosomes. The more severely affected patient showed a break p

Giemsa banding of chromosome 1gh+ and linkage analysis.

A four-generation transmission of 1qh+ chromosome was ascertained by routine chromosome analysis of a mildly dysmorphic and retarded 61/2-year-old female. Concordance between synophrys and the 1qh+ marker was the only consistent phenotypic relationship. The variant chromosome did not appear uncoiled, and Giemsa centromeric staining (C-bands) revealed an increased width of the heterochromatin commensurate with the increased length of the long arm. Giemsa banding of the entire chromosome (G-bands) revealed two heterochromatin bands, identical in appearance, in the centromeric region with the rem

High resolution chromosome banding in the Rubinstein-Taybi syndrome.

We studied eight cases of the Rubinstein-Taybi syndrome to determine if a detectable chromosome change is associated with this syndrome. High resolution G-banded analysis of prometaphase chromosomes was carried out on venous blood with two to four karyotypes analyzed for each patient. Initial study of two patients suggested an abnormality in chromosome 15q, but blinded analysis and review of the karyotypes proved to be normal. Therefore, all eight patients had normal G-banded prometaphase chromosome studies. Because of technical difficulties and observer bias, we urge caution in using high res

Chromosome changes in 17 human neoplasms studied with banding.

Chromosome abnormalities from 17 malignancies that were studied using direct preparations and G-, Q-, and C-banding methods are presented and the findings assessed. The material used was from 6 primary cervical carcinomas, 5 primary and metastatic ovarian carcinomas and 6 other metastatic carcinomas. In general, chromosome 1 was found to be involved more frequently than the others in structural aberrations. These included short arm deletions--the most frequent marker over all the cases--long arm deletions, long arm duplications and various translocations including more complex rearrangements i

Chromosome banding analysis of gynecomastias and breast carcinomas in men.

Male breast cancer is 100 times less frequent than its female counterpart and accounts for less than 1% of all cancers in men. Although men with breast cancer also often have gynecomastia, it is still unknown whether gynecomastia per se predisposes the male breast to malignant disease. We describe the cytogenetic analysis of three gynecomastias and four breast cancers in men. No chromosome abnormalities were detected in two cases of gynecomastia, with no other concomitant breast disease. The third gynecomastia sample, taken from a site where a breast carcinoma had previously been removed, had

Proteins in chromosome banding. II. Effect of R- and C-banding treatments on the proteins of isolated nuclei.

SDS polyacrylamide gel electrophoresis was used to study the proteins extracted from, and those remaining in isolated, fixed, air-dried nuclei subjected to a variety of R- and C-banding techniques. The R-banding procedures, involving exposure to hot Earle's BSS or NaH2PO4, had the least effect of any of the banding techniques on the extraction of proteins from isolated nuclei. Only small amounts of 5 nonhistone proteins were detected in the Earle's BSS extract, and no proteins were found in the NaH2PO4 solution after treatment. The residual proteins remaining in the nuclei after either treatme

The X1X2Y sex chromosome system in the fish Hoplias malabaricus. I. G-, C- and chromosome replication banding.

Hoplias malabaricus, a widely distributed neotropical fish (Central America to Argentina), may represent a group of distinct species showing diversified cytotypes with respect to chromosome number, morphology and sex systems. One of these karyotypic forms is characterized by an X1X1X2X2/X1X2Y sex chromosome system, with 2n = 40 and 39 chromosomes in females and males respectively. Analyses with G-, C- and chromosome replication banding permitted a better characterization of the sex chromosomes in this cytotype. The Y chromosome, unique in males, resulted from a translocation event between two

Chromosome-band-specific painting: chromosome in situ suppression hybridization using PCR products from a microdissected chromosome band as a probe pool.

We describe a chromosome-band-specific painting method that involves (1) microdissection of the chromosome, chromosomal region or band, (2) amplification of a variety of chromosome/region/band-specific DNA fragments with the polymerase chain reaction (PCR), and (3) chromosome in situ suppression hybridization (CISS) with the direct use of the PCR products as a probe pool. With this method, it was possible 1) to paint an entire X or Y chromosome, a distal one-fourth of 2q, and only a band at 8q24.1, 2) to identify the origin of a minute marker chromosome in a mentally retarded patient, 3) to de

Study of two cases of ring 13 chromosome using high-resolution banding.

The chromosomes of two patients with ring 13 (r13) were studied using high-resolution RBG banding of prometaphase cells. The rings of the two patients differ slightly in breakpoints. Cell with multiple single, double-sized rings, quadruple-sized rings, rod- and ring-shaped fragments, and fragments showing varied states of condensation were seen, as were cells monosomic for chromosome 13. The evolution of these cell lines as a result of sister chromatid exchange, nondisjunction, ring breakage, and premature chromosome condensation is discussed. Clinical features of these patients reflect the he

Chromosome analysis on 930 consecutive newborn children using quinacrine fluorescent banding technique.

Chromosome analysing using quinacrine fluorescence was performed on 930 consecutive newborn infants. The total incidence of major chromosome aberrations including numerical changes of the sex chromosomes, and structural changes of autosomes, was 0.54%. Incidence of XYY (0.4%) and XXY (0.2%) were relatively higher as compared to other studies. About 0.75% of the newborn infants were found to have a variable bright fluorescent band located on the proximal area of the short arm (p11) rather than on the proximal long arm (q11) of chromosome No. 3. Attempts were also made to record the variable flu

Impact of chromosomal aberrations detected by chromosome banding analysis in symptomatic Waldenström's macroglobulinemia.

The clinicopathologic features and prognostic impact of MYD88 L265P (MYDL265P) and CXCR4 mutations (CXCR4Mut) have been well reported, although little is known regarding the impact of chromosomal aberrations (CA) detected by chromosome banding analysis (CBA) in symptomatic Waldenström's macroglobulinemia (sWM). Thus, we investigated the clinicopathologic features and prognostic impact in sWM with CAs identified by CBA. We retrospectively analyzed the clinicopathologic results and genomic alterations by droplet digital PCR, fluorescence in situ hybridization (FISH), and CBA using the G-banding

BR-ChromNet: Banding resolution localization of chromosome structural abnormality with conditional random field.

Detecting chromosome structural abnormalities in medical genetics is essential for diagnosing genetic disorders and understanding their implications for an individual's health. However, existing computational methods are formulated as a binary-class classification problem trained only on representations of positive/negative chromosome pairs. This paper introduces an innovative framework for detecting chromosome abnormalities with banding resolution, capable of precisely identifying and masking the specific abnormal regions. We highlight a pixel-level abnormal mapping strategy guided by banding

High-resolution chromosome banding and fragile site studies in von Hippel-Lindau syndrome.

von Hippel-Lindau syndrome is an autosomal dominant disorder that predisposes to the development of benign and malignant tumors. The gene for von Hippel-Lindau syndrome has not yet been localized and the cytogenetics of this cancer-prone genetic disease have not been fully explored. Therefore, we did high-resolution chromosome banding of lymphocytes from patients from 14 kindreds with von Hippel-Lindau syndrome. There were 18 patients (eight male, and ten female). None of the male patients showed a detectable chromosome abnormality. However, three of the ten female patients had 45,X/46,XX/47,X

Q-banding of chromosomes in human spontaneous abortions.

Chromosome studies of 242 spontaneous abortions were carried out by Q-banding technique. The abortuses were selected for study because they were phenotypically abnormal, had not progressed beyond 12 weeks development or were from women with repeated abortions. Chromosome anomalies were found in 126 (52%) of the abortuses. Of these, 71 (56%) were trisomies. Trisomies were found for all the autosomes except Nos. 1, 3, 5, 11, 17 and 18. Triploidy was the second commonest anomaly in this series, making up 26 (21%) of the total anomalies. About 70% of these had an XXY sex chromosome complement. Onl

Chromosomal banding patterns in acute nonlymphocytic leukemia.

Bone marrow chromosomes obtained from 50 of 55 consecutive adult patients with acute nonlymphocytic leukemia were analyzed with quinacrine fluorescence. Twenty-five patients showed a normal karyotype and 25 an abnormal karyotype on the initial samples available for analysis. Among the 25 patients with abnormalities, the marrow cells contained 48 chromosomes in one case, 47 in two, 46 in ten, 45 in nine, 43 in two, and 42 chromosomes in one case. Seven of the ten patients with 46 chromosomes had abnormalities, primarily balanced translocations, that were not detected with the standard Giemsa st

CABAND: Classification of aberrations in multicolor banded chromosomes.

A system is presented to describe aberrations in chromosomes painted with the mBAND methodology. The CABAND (classification of aberrations in multicolor banded chromosomes) system is based on the fact that only banded parts of aberrations and not DAPI stained parts can be described accurately.
