Chromosome positioning and male infertility: it comes with the territory.

The production of functional spermatozoa through spermatogenesis requires a spatially and temporally highly regulated gene expression pattern, which in case of alterations, leads to male infertility. Changes of gene expression by chromosome anomalies, gene variants, and epigenetic alterations have been described as the main genetic causes of male infertility. Recent molecular and cytogenetic approaches have revealed that higher order chromosome positioning is essential for basic genome functions, including gene expression. This review addresses this issue by exposing well-founded evidences whi

Chromosome (re)positioning in spermatozoa of fathers and sons - carriers of reciprocal chromosome translocation (RCT).

BACKGROUND: Non-random chromosome positioning has been observed in the nuclei of several different tissue types, including human spermatozoa. The nuclear arrangement of chromosomes can be altered in men with decreased semen parameters or increased DNA fragmentation and in males with chromosomal numerical or structural aberrations. An aim of this study was to determine whether and how the positioning of nine chromosome centromeres was (re)arranged in the spermatozoa of fathers and sons - carriers of the same reciprocal chromosome translocation (RCT). METHODS: Fluorescence in situ hybridization

Changes in chromosome positioning may contribute to the development of diseases related to X-chromosome aneuploidy.

The radial positions of the centromeric regions of chromosomes 1 and X were determined in normal male fibroblasts (XY) and in fibroblasts from a patient with a rare case of XXXXY polysomy. The centromeric regions and presumably the whole territories of active X chromosomes were demonstrated to occupy similar, although not identical, positions in XY and XXXXY cells. The centromeres of inactive X chromosomes (Barr bodies) were located closer to the nuclear periphery as compared with the centromeres of active X chromosomes. In addition, it was established that the nuclear radial position of gene-

Interphase chromosome positioning in in vitro porcine cells and ex vivo porcine tissues.

BACKGROUND: In interphase nuclei of a wide range of species chromosomes are organised into their own specific locations termed territories. These chromosome territories are non-randomly positioned in nuclei which is believed to be related to a spatial aspect of regulatory control over gene expression. In this study we have adopted the pig as a model in which to study interphase chromosome positioning and follows on from other studies from our group of using pig cells and tissues to study interphase genome re-positioning during differentiation. The pig is an important model organism both econom

Impact of sperm fractionation on chromosome positioning, chromatin integrity, DNA methylation, and hydroxymethylation level.

BACKGROUND: Sperm chromosomes are nonrandomly organized in the cell nucleus, which plays an important role in the regulation of early embryo development, which is determined by the specific localization of sperm chromosomal regions carrying genes with expression crucial at the first contact with ooplasm during fertilization. Thus, the aim of this study is to determine whether the application of selective methods providing high-quality spermatozoa with good motility and/or morphology can increase the frequency of gametes with a specific positioning of chromosomes. For the first time, we used a

Three-Dimensional Confocal Analysis of Chromosome Positioning Coupled with Immunofluorescence in Mouse Sperm Nuclei.

Male infertility is associated with several causes affecting the paternal nucleus such as DNA lesions (breaks, deletions, mutations, ...) or numerical chromosome anomalies. More recently, male infertility has also been associated with changes in the sperm epigenome, including modification in the topology of chromatin (Olszewska et al., Chromosome Research 16:875-890, 2008; Alladin et al., Syst Biol Reprod Med 59: 146-152, 2013) ref with number 1, 2. Indeed, the positioning of chromosomes in the sperm nucleus is nonrandom and defines chromosome territories (Champroux et al., Genes (Basel) 9:501

Chromosome banding studies in Philadelphia chromosome positive myeloid leukaemia.

A cytogenetic study of Ph1 positive myeloid leukaemia in both chronic and acute phases had been made by a chromosome banding technique. The translocation (t(9;22)(q34;q11), designated t(Ph1) was present in the myeloid cells of 43 of 44 patients; the exceptional case had normal number 9 chromosomes and a different translocation (t(19;22)(q13;q11)). A translocation additional to that involving the Ph1 was found as a stable abnormality present in all myeloid cells in 4 patients, chromosome 17 being involved in 2. The association of isochromosome number 17 with blast crisis was confirmed. New data

Asciminib for Philadelphia chromosome-positive leukemias.

Twenty-five years after the introduction of imatinib, we have entered a new era of therapy for chronic myeloid leukemia (CML). Despite the development of second- and third-generation tyrosine kinase inhibitors (TKI), their impact has been incremental in improving outcomes for CML patients. While frontline use of second-generation TKI has improved molecular response rates and reduced progression to blast phase, there has been no improvement in overall survival compared to that achieved with imatinib, likely due to the higher toxicity and consequent higher non-CML-related mortality. Imatinib rem

The 3D positioning of ANT2 and ANT3 genes within female X chromosome territories correlates with gene activity.

The three-dimensional positioning of the X-chromosomal adenine nucleotide translocase genes, ANT2 and ANT3, were compared in the active and inactive X chromosome territories (Xa and Xi) of female human amniotic fluid cell nuclei. ANT2 is located in Xq24-q25 and is transcriptionally active on Xa, but inactive on Xi. ANT3 is located in the pseudoautosomal region Xp22.3 and escapes X-inactivation. Three-color fluorescence in situ hybridization, confocal laser scanning microscopy, and three-dimensional image analysis revealed that transcriptionally active ANT2 and ANT3 genes were positioned more p

Philadelphia chromosome-positive thrombocythemia and megakaryoblast leukemia.

A case of well-documented and -illustrated megakaryoblastic transformation is described in a patient with thrombocythemia passing through a stage of myelofibrosis without features of chronic granulocytic leukemia. Immunocytologic studies with the use of conventional and monoclonal antibodies against platelet membrane glycoproteins and electron microscopic investigations, demonstrating bull's-eye granules and platelet peroxidase positivity, proved the megakaryocytic differentiation of the blast cells. From the onset of the disease as well as during the megakaryoblastic transformation, the Phila

Ponatinib in refractory Philadelphia chromosome-positive leukemias.

BackgroundResistance to tyrosine kinase inhibitors in patients with chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph-positive ALL) is frequently caused by mutations in the BCR-ABL kinase domain. Ponatinib (AP24534) is a potent oral tyrosine kinase inhibitor that blocks native and mutated BCR-ABL, including the gatekeeper mutant T315I, which is uniformly resistant to tyrosine kinase inhibitors.MethodsIn this phase 1 dose-escalation study, we enrolled 81 patients with resistant hematologic cancers, including 60 with CML and 5 with Ph-positive A

Non random position of metaphasic chromosomes: a study of radiation induced and constitutional chromosome rearrangements.

The rearranged chromosomes derived from reciprocal translocations or dicentric-acentric formations, observed 48 h after their induction by irradiation at Go phase, have a clear tendency to be closer together than their normal homologues. This tendency disappears in longer cultures, and does not exist when many different constitutional reciprocal translocations are considered together. It indicates that the chromosomes having exchanged segments remain adjacent at the following metaphase, and thus, that metaphase plates reflect, at least partially the interphase arrangement of chromosomes.

Evolving Paradigms in the Treatment of Philadelphia Chromosome-Positive ALL.

The treatment of adults with Philadelphia chromosome-positive ALL (Ph+ ALL) has evolved significantly over the past 25 years. These changes have been driven largely by the development of potent BCR::ABL1 tyrosine kinase inhibitors (TKIs) such as ponatinib, more accurate risk stratification and monitoring of measurable residual disease (MRD) and, most recently, the frontline use of blinatumomab-based, chemotherapy-free regimens. Although the historical standard of care for newly diagnosed Ph+ ALL was intensive chemotherapy followed by allogeneic hematopoietic stem-cell transplantation (allo-HSC

Characterization of Asciminib-Resistant Philadelphia Chromosome-Positive Cells.

BACKGROUND: Asciminib is approved for treating patients with chronic-phase chronic myeloid leukemia who were previously treated with two or more tyrosine kinase inhibitors or those with T315I mutation. However, the mechanisms underlying asciminib resistance remain unclear. METHODS: In this study, we established a new asciminib-resistant cell line. We examined BCR::ABL1 gene mutation analysis and the effects of conventional chronic myelogenous leukemia inhibitors. RESULTS: Direct sequencing revealed Y139D and T315I mutations in asciminib-resistant cells. Ponatinib and omacetaxine were effecti

Philadelphia chromosome-positive lymphoblastic lymphoma-Is it rare or underdiagnosed?

Lymphoblastic lymphomas (LBLs) are neoplasms of precursor B and T cells; they are considered in the same spectrum as precursor B and T cell acute lymphoblastic leukemia (ALL). The World Health Organization classification classifies both LBL and ALL as one disease entity. While chromosome abnormalities are well defined with all of their therapeutic and prognostic implications in ALL, these are not well studied in LBL. Here, we describe a case of Philadelphia chromosome-positive LBL and review the available literature regarding this entity.

Treatment of Philadelphia chromosome-positive acute lymphoblastic leukemia.

Philadelphia chromosome positive (Ph(+)) acute lymphoblastic leukemia (ALL) includes at least one-quarter of all adults with ALL. Until recently, conventional chemotherapy programs that have been effective in other precursor B-cell ALL cases have been unable to cure patients with this diagnosis. Allogeneic stem cell transplantation early in first remission has been the recommended therapy. The availability of imatinib mesylate and other tyrosine kinase inhibitors and small molecules that affect the BCR/ABL signaling pathways may be changing the treatment paradigm and the prognosis for these pa
