[The human genome--chromosome 20].

There is an increasing interest on one of the smallest human chromosomes as it is shown by the First International Symposium on the Human Chromosome 20 and by the genetic map prepared by EUROGEN. The conserved part of the long arm of human chromosome 20 is synthenic with the distal part of the mouse chromosome 2 allowing for some analogies between them. Human chromosome 20 contains several important genes for the human pathology. Mutations of one of them, the vasopressin-neurophysin II gene, are responsible for hereditary neurohypophyseal diabetes insipidus. Severe combined immunodeficiency du

Deletions of chromosome 20q and the pathogenesis of myeloproliferative disorders.

Myeloproliferative disorders (MPD) and myelodysplastic syndromes (MDS) represent an overlapping spectrum of clonal preleukaemic conditions in which mutations have resulted in dysregulation of multipotent haemopoietic progenitors. An interstitial deletion of the long arm of chromosome 20 is a recurring abnormality associated with both of these classes of disorders. The association of 20q deletions with 'stem cell' disorders suggests that the deletion may mark the site of one or more genes, loss or inactivation of which perturbs the regulation of haemopoietic progenitors. The identification and

Management of ring chromosome 20 syndrome: Narrative review and consensus recommendations.

Ring chromosome 20 (ring 20) is a rare genetic condition usually presenting as developmental and epileptic encephalopathy. The disease is caused by fusion of the long and short arms of chromosome 20. Patients are symptomatic even if there is no loss of genetic material. Epilepsy in ring 20 is usually drug-resistant, with seizures often significantly debilitating and severely impacting quality of life. We assembled a taskforce of clinician-scientists with expertise in ring 20, as well as caregivers of individuals with ring 20. We then reviewed published literature on ring 20 with a focus on man

New association between ring chromosome 20 syndrome and hypomelanosis of Ito.

Hypomelanosis of Ito is an uncommon neuroectodermal disease associated with a wide range of cytogenetic abnormalities. Ring chromosome 20 is a rare chromosomal disorder characterized by severe, refractory epilepsy, cognitive delay, and unspecific dysmorphic traits. An association between the hypomelanosis of Ito and ring chromosome 20 syndrome was never reported peviously. We describe a young girl who has ring chromosome 20 and who also has clinical symptoms of hypomelanosis of Ito. After her diagnosis of epilepsy, she was submitted to neurologic and genetic testing, a skin biopsy, and repeate

The map of chromosome 20.

The number of gene assignments to human chromosome 20 has increased slowly until recently. Only seven genes and one fragile site were confirmed assignments to chromosome 20 at the Ninth Human Gene Mapping Workshop in September 1987 (HGM9). One fragile site, 13 additional genes, and 10 DNA sequences that identify restriction fragment length polymorphisms (RFLPs), however, were provisionally added to the map at HGM9. Five mutated genes on chromosome 20 have a relation to disease: a mutation in the adenosine deaminase gene results in a deficiency of the enzyme and severe combined immune deficienc

Trisomy 2, trisomy 20, and del(17p) as sole chromosomal abnormalities in three cases of hepatoblastoma.

Short-term cultures of three hepatoblastomas were analyzed cytogenetically. Trisomy 2, trisomy 20, and a deletion of 17p were found as the sole abnormalities, yielding the karyotypes 47,XY, + 2; 47,XX, + 20; and 46,XX,del(17)(p12)/46,XX. This is the first reported case of deletion of 17p as the sole chromosomal abnormality in a hepatoblastoma and the first reported case of trisomy 20 without double minute chromosomes as a sole chromosomal abnormality in hepatoblastoma.

A FEMALE PATIENT WITH DUPLICATION OF 7p13-pter ASSOCIATED WITH DEL 20p13pter RESULTING FROM MALSEGREGATED PATERNAL 7;20 BALANCED TRANSLOCATION.

Duplication of the short arm of chromosome 7 is a genomic disorder presenting with distinctive facies including hypertelorism, large anterior fontanel, and intellectual disability. A 2½-year-old Egyptian girl was referred because of cleft palate and dysmorphic features. She showed clinical manifestations of duplication of 7p, along with atypical features of corpus callosum hypogenesis and skeletal anomalies. Chromosome analyses revealed unbalanced translocations involving the short arms of chromosomes 7 and 20 due to malsegregation of a paternal balanced translocation 7;20. Fluorescence in sit

Significance of extra copies of chromosome 20 and the long arm of chromosome 2 in hepatoblastoma.

Cytogenetic analysis of both primary and metastatic hepatoblastoma revealed the following abnormal chromosomal complements respectively: 46,XX,der(2)t(2;2)(p25;q21),der(22)t(1;22)(q22;p13) [6]/47,XX,der(2)t(2;2)(p25;q21),+20,der(22)t(1;22)q22;p13)[4]/47,XX,der (2)t(2;2)(p25;q21), +20[1], and 48,XX,der(2)t(2;2)(p25;q21),+12,+17, -18,+20,der(22)t(1;22)(q22;p13)[9]/50,XX,der(2)t(2;2)(p25;q21), +8,+12,+17,+20[4]. Two abnormalities, the chromosome 2 derivative and trisomy 20, are recurrent in hepatoblastoma, but the derivative involving chromosomes 1 and 22 is a novel abnormality.

Trisomy 1q, 2, and 20 in a case of hepatoblastoma: possible significance of 2q35-q37 and 1q12-q21 rearrangements.

Combined cytogenetic, chromosome painting, and spectral karyotyping (SKY) analyses in a case of hepatoblastoma revealed a karyotype of 49,XY,+Y,+der(2)t(2;3)(q35;q25),der(3)t(1;3)(q12; q25),+20. Trisomy 1q, 2, and 20 identified in the present case are consistent with the previously reported cytogenetic alterations in hepatoblastoma. The breakpoints at 1q12 and 2q35 identified in this case have also been reported previously as nonrandom changes. The frequent occurrence of these rearrangements in hepatoblastoma suggests that they may be of pathogenic significance.

Congenital acute monoblastic leukemia with double translocation (8;16) (p11;p13) and (16;20) (q13;p13).

A new case of congenital acute monoblastic leukemia (AML5) with a rare (8;16) translocation is described: this translocation (or a variant) was previously reported only in 4 infants and 10 older children or adults, practically always in association with AML5 or acute myelomonocytic leukemia. The authors discuss the possible causative implication of band 8p11 with some cytological particularities, i.e. erythrophagocytosis by some blasts and strong peroxidase and esterase activities, observable here and in most of the cases with the t(8;16). The t(8;16) may be added to the other chromosomal abno

Meiotic segregation analysis in spermatozoa of pericentric inversion carriers using fluorescence in-situ hybridization.

BACKGROUND: Pericentric inversions are structural chromosomal abnormalities resulting from two breaks, one on either side of the centromere, within the same chromosome, followed by 180 degrees rotation and reunion of the inverted segment. They can perturb spermatogenesis and lead to the production of unbalanced gametes through the formation of an inversion loop. METHODS: We report here the analysis of the meiotic segregation in spermatozoa from six pericentric inversion carriers by multicolour fluorescence in-situ hybridization (FISH) and review the literature. RESULTS: The frequencies of th

Identification of novel regions of deletion in familial Wilms' tumor by comparative genomic hybridization.

Wilms' tumor, an embryonic renal neoplasm diagnosed primarily in young children, can occur in either a noninheritable (sporadic) or a familial form, with the latter presenting earlier and more often at bilateral sites. Although familial Wilms' tumor is thought to develop through inherited and acquired mutational inactivation of the two alleles of predisposing tumor suppressor genes, only a small percentage of cases can be accounted for by mutations affecting the WT1 gene or linkage to the Beckwith-Weidemann syndrome of the BWS region on the short arm of chromosome 11. To find chromosomal regio

Cytogenetic findings in untreated patients with essential thrombocythemia.

Essential thrombocythemia (ET) is a chronic myeloid disorder that is characterized by persistent thrombocytosis, thrombohemorrhagic symptoms and a low risk of transformation to leukemia. Chromosomal abnormalities in ET are very rare and most of the patients studied were either in leukemic transformation or they had received treatment with cytotoxic agents. The number of cases studied at the time of diagnosis is very limited. In the present study, 67 cases with ET, at the time of diagnosis, were cytogenetically studied by a G-banding technique. Among them, only four presented chromosomal abnorm
