Chrysosporium species are saprophytic filamentous fungi commonly found in the soil, dung, and animal fur. Subcutaneous infection caused by this organism is rare in humans. We report a case of subcutaneous fungal infection caused by Chrysosporium keratinophilum in a 38-year-old woman. The patient presented with severe chromoblastomycosis-like lesions on the left side of the jaw and neck for 6 years. She also got tinea corporis on her trunk since she was 10 years old. Chrysosporium keratinophilum was isolated from the tissue on the neck and scales on the trunk, respectively. The patient showed s

A 24-year-old black man presented with a two-month history of swelling of the right ankle. Roentgenograms were initially read as negative. One month later the patient returned with increased pain and swelling, and roentgenograms demonstrated a large lucency involving the distal tibia. Bone scanning revealed extension of the lesion proximally, to 6 cm below the knee. Biopsy of a large tibial abscess demonstrated histocytic giant cells with budding yeasts and septate mycelia. Chrysosporium was isolated from cultures. Amphotericin B was used to treat the patient. Clinically, the infection healed

Equine skin mycoses are a significant concern in Kazakhstan's livestock industry due to the country's historical livestock farming practices, the development of equestrian sports, and food traditions. Skin infections are among the most common fungal infections in horses. Emerging pathogens of equine dermatophytosis include keratinophilic Chrysosporium spp., which can degrade and metabolize keratin found in superficial tissues. This, combined with their thermotolerance, contributes to their pathogenicity. In this study, we investigated the biological properties and pathogenicity of two Chrysosp

During wood degradation, fungi have to deal with toxic and stressful compounds called wood extractives. The identification of the various detoxification strategies developed by fungi, and the molecular targets of these compounds is limited in Basidiomycetes because of the lack of genetic tools. To circumvent this problem, we have developed a direct genetic strategy in the white-rot fungus Phanerochaete chrysosporium. A library of P. chrysosporium UV mutants was generated and screened to isolate mutants resistant to Bagassa guianensis wood extractives (BWE). Wood extractives contribute to B. gu

Chrysosporium is a large genus of saprophytic fungi that is commonly found in the soil. Infection caused by this organism is rare in humans and typically occurs in immunocompromised patients. Primary cutaneous Chrysosporium infection is relatively rare and has been reported in a heart transplant patient. The prognosis is usually favorable, but very poor in the setting of persistent profound immunosuppression. We herein report a case of primary cutaneous Chrysosporium infection following ear piercing in an immunocompetent patient. It is important for clinicians to consider this condition in pat

Chrysosporium species caused an invasive infection in an 18-year-old patient following allogeneic sibling bone marrow transplant for T lineage acute lymphoblastic leukemia. This infection began as a facial swelling and extended into the central nervous system. Fungal disease spread rapidly despite antifungal agents. An autopsy showed fungal involvement of brain, lungs, liver and kidneys.

Perfluorooctanoic acid (PFOA) is becoming a concern due to its persistence, bioaccumulation, and potential harmful effects on humans and the environment. In this study, the fungus Phanerochaete chrysosporium (P. chrysosporium) was used to remove the PFOA in liquid culture system. The results showed that the average activities of laccase (Lac), lignin peroxidase (LiP), and manganese peroxidase (MnP) enzymes secreted by P. chrysosporium were 0.0003 U/mL, 0.013 U/mL, and 0.0059 U/mL, respectively, during the incubation times of 0-75 days. The pH of 3 and incubation time of 45-55 days were the opt

The growing potential of quantum dots (QDs) in biomedical applications has raised considerable concerns regarding their toxicological impact. Consequently, there has been a need to understand the underlying toxicity mechanism of QDs. In this work, we comprehensively investigated the bioaccumulation and toxicity of three CdSe/ZnS QDs (COOH CdSe/ZnS 525, NH2 CdSe/ZnS 525, and NH2 CdSe/ZnS 625) in Phanerochaete chrysosporium (P. chrysosporium) using confocal laser scanning microscopy, reactive oxygen species (ROS) measurements, and cell viability assays. Two cDNA sequences representing putative allelic variants of the Phanerochaete chrysosporium cbhII gene were isolated by hybridization to the Trichoderma reesei cbhII gene. Both of the equivalent genomic sequences were subsequently isolated by the inverse PCR technique. DNA sequencing showed that the cbhII open reading frame of 1,380 bp codes for a putative polypeptide of 460 amino acids which is interrupted by six introns. The domain structure found in T. reesei cbhII is conserved in the equivalent P. chrysosporium protein. The overall similarity between the two gene products is 54%, with the

Aggressive fungal rhinosinusitis with Chrysosporium sp. occurred in a patient with acute lymphocytic leukemia. The infecting organism is an exceedingly rare human pathogen. Usually, human chrysosporial infections are mild and unmarked by symptoms. Most case reports appear in the pathology literature and describe the incidental finding of adiaspores in the pulmonary parenchyma at autopsy. Clinical disease from active growth of chrysosporial mycelia in human tissues has been noted on a porcine aortic valve prosthesis and in a tibial abscess. Hyphal elements were not recovered from any other body

Microorganisms need to resist the hazards posed by heavy metals during the process of metal adsorption. Phanerochaete chrysosporium is a fungus that is efficiently used for heavy-metal biosorption in wastewater treatment. Extraction and analysis of proteins induced by heavy metals can help to understand the regulatory mechanisms of P. chrysosporium in wastewater treatment. In this study, P. chrysosporium was exposed to 50 μM cadmium nitrate. A maximum cadmium adsorption capability of 77.1 mg/g dry biomass was found after 65 h, which was accompanied by a relatively higher protein concentration.

The aim of the present study was to compare the effect of a wide range of culture conditions on production of ligninolytic enzymes by Polyporus sanguineus and Phanerochaete chrysosporium. Lignin peroxidase production by P. sanguineus was comparable with that of P. chrysosporium, although the culture conditions giving the highest yield varied greatly between the two fungi. Highest yield of manganese peroxidase by P. sanguineus obtained in 0.5% malt extract medium and peptone or malt extract supplemented mineral salts broth could not be surpassed by P. chrysosporium in any of the optimization ex

The worldwide occurrence of wheat crown rot, predominantly caused by the pathogen Fusarium pseudograminearum, has a serious impact on wheat production. Numerous microorganisms have been employed as biocontrol agents, exhibiting effectiveness in addressing a wide array of plant pathogens through various pathways. The mycelium of the white-rot fungus Phanerochaete chrysosporium effectively inhibits the growth of F. pseudograminearum by tightly attaching to it and forming specialized penetrating structures. This process leads to the release of intracellular inclusions and the eventual disintegrat

Lignin is a major constituent of lignocellulose and the most abundant aromatic biopolymer on earth. It provides plants with rigidity and protection, but its recalcitrant nature also presents a significant barrier to lignocellulose valorization. The white-rot fungus Phanerochaete chrysosporium is among nature's most efficient lignin degraders, and its ligninolytic capabilities have been subjected to intensive investigations. Genome editing with precision is crucial for elucidating the in vivo mechanisms of its ligninolytic actions, but genetic manipulations of P. chrysosporium are often plagued

The active site amino acid residues of lignin peroxidase are homologous to those of other peroxidases; however, in contrast to other peroxidases, no pH dependence is observed for the reaction of ferric lignin peroxidase with H2O2 to form compound I (Andrawis, A., Johnson, K.A., and Tien, M. (1988) J. Biol. Chem. 263, 1195-1198). Chloride binding is used in the present study to investigate this reaction further. Chloride binds to lignin peroxidase at the same site as cyanide and hydrogen peroxide. This is indicated by the following. 1) Chloride competes with cyanide in binding to lignin peroxid

Phanerochaete chrysosporium is a model white-rot-fungus with exceptional lignolytic activity, but its genetic manipulation has remained a major challenge due to low transformation efficiencies. Here, we report the development of an optimized Agrobacterium-mediated transformation (AMT) protocol optimized for this species. A CRISPR-Cas9 compatible binary vector was modified by replacing the mCherry cassette with a strong sGFP reporter under the constitutive 35S promoter, enabling reliable identification of transformants. Using mycelial disks as starting material and applying a sandwich selection

Quantum dots (QDs) have caused large challenges in clinical tests and biomedical applications due to their potential toxicity from nanosize effects and heavy metal components. In this study, the physiological responses of Phanerochaete chrysosporium (P. chrysosporium) to CdSe/ZnS QDs with either an inorganic sulfide NaHS or an organic sulfide cysteine as antidote have been investigated. Scanning electron microscope analysis showed that the hyphal structure and morphology of P. chrysosporium have obviously changed after exposure to 100 nM of COOH CdSe/ZnS 505, NH2 CdSe/ZnS 505, NH2 CdSe/ZnS 565
