Although porcine circovirus type 2 (PCV2)-associated diseases have been evaluated for known immune evasion strategies, the pathogenicity of these viruses remained concealed for decades. Surprisingly, the same viruses that cause panzootics in livestock are widespread in young, unaffected animals. Recently, evidence has emerged that circovirus-like viruses are also linked to complex diseases in humans, including children. We detected PCV2 genome-carrying cells in fetal pig thymi. To elucidate virus pathogenicity, we developed a new pig infection model by in vivo transfection of recombinant PCV2

Circoviruses are a group of small circular, single-stranded DNA viruses that belong to the family Circoviridae. They are known to infect a wide variety of animals. Rhizomys sinensis is a species of rodent that is the reservoir of many zoonotic pathogens. Our previous study identified many sequencing reads mapped to the genome of viruses in Circoviridae in R. sinensis. However, little is known about the circulation and genetic characterization of circoviruses in R. sinensis. This study identified three different circoviruses in samples from 195 R. sinensis. First, the bamboo rat circovirus is w

Circoviruses belong to the family Circoviridae, which is classified into two genera, Circovirus and Cyclovirus. Some circoviruses have been identified in various organisms and/or their fecal samples and might be associated with diseases in their hosts. However, few circoviruses are reported in human. Recently, two novel circoviruses HCirV-1 and HCirV-2 were identified in humans through next-generation sequencing and were defined as the species of Circovirus human in the family Circoviridae. Both viruses are suspected to be associated with liver diseases, particularly among immunosuppressed peo

Neonatal porcine diarrhoea of uncertain aetiology has been reported from a number of European countries. The aim of the present study was to use viral metagenomics to examine a potential viral involvement in this diarrhoea and to describe the intestinal virome with focus on eukaryotic viruses. Samples from the distal jejunum of 50 diarrhoeic and 19 healthy piglets from 10 affected herds were analysed. The viral fraction of the samples was isolated and nucleic acids (RNA and DNA fractions) were subjected to sequence independent amplification. Samples from diarrhoeic piglets from the same herds

Canine circovirus (CanineCV) is a relatively new viral species, belonging to the family Circoviridae, whose pathogenic role is still uncertain. Since its first description in one domestic dog in 2011 from the USA, several reports have been documenting its distribution worldwide. Recently, CanineCV was also detected in wild animals such as wolves, foxes and badgers. In order to investigate the presence and the genetic characteristics of CanineCV in foxes of Arctic and Sub-Arctic regions, the presence of CanineCV DNA in internal organs (liver and spleen) of 51 arctic foxes (Vulpes lagopus) from

The common carp (Cyprinus carpio) is an invasive species in Argentinian Patagonia. This study aimed to investigate the presence of Circoviridae-related viruses associated with its population in the Negro River. Total DNA samples extracted from the organ pools of 50 individuals were evaluated using a broad-spectrum PCR, which led in a single case to the detection of a circo-like virus. Based on the partial rep sequence obtained, phylogenetic analysis showed a relation between the virus found and those that have been identified in other cyprinid fishes, such as gudgeon (Gobio gobio) and fathead

Occupational exposure to pathogens can pose health risks. This study investigates the viral exposure of workers in a wastewater treatment plant (WWTP) and a swine farm by analyzing aerosol and surfaces samples. Viral contamination was evaluated using quantitative polymerase chain reaction (qPCR) assays, and target enrichment sequencing (TES) was performed to identify the vertebrate viruses to which workers might be exposed. Additionally, Quantitative Microbial Risk Assessment (QMRA) was conducted to estimate the occupational risk associated with viral exposure for WWTP workers, choosing Human

Infections with goose circovirus (GoCV) are associated with growth retardation and developmental problems in farmed geese. An indirect immunofluorescence assay for detecting virus-specific serum antibody was developed for diagnostic and epidemiological purposes. In the absence of a method for growing GoCV in cell culture, the assay was based on the reaction of antibodies with the GoCV capsid protein produced within baby hamster kidney cells using the eukaryotic Semliki forest virus expression vector. Using an optimized test that involved screening sera at 1:50 dilution and the use of a fluores

Waterfowl circovirus (WFCV) is an important member of the genus Circovirus within the family Circoviridae, encompassing duck circovirus (DuCV) and goose circovirus (GoCV). As a pathogen that poses a serious threat to the global waterfowl industry, WFCV induces significant immunosuppression in its hosts, leading to growth retardation, feather abnormalities, and increased susceptibility to other pathogens. Consequently, it results in markedly elevated mortality rates among affected birds and causes substantial economic losses. In recent years, with the advancement of molecular biology techniques

Raccoon dogs as an ancient species of Canidae are the host of many viruses, including rabies virus, canine distemper virus, severe acute respiratory syndrome coronavirus, and so on. With the development of raccoon dog breeding in recent years, some viruses which infected poultry or pigs were also detected from raccoon dogs. At present, the fecal virome of raccoon dogs has been rarely studied. Using an unbiased viral metagenomic approach, we investigated the fecal virome in raccoon dogs collected from one farm of Jilin Province, China. Many DNA or RNA viruses identified in those fecal samples w

Heparin is one of the main pharmaceutical products manufactured from raw animal material. In order to describe the viral burden associated with this raw material, we performed high-throughput sequencing (HTS) on mucus samples destined for heparin manufacturing, which were collected from European pigs. We identified Circoviridae and Parvoviridae members as the most prevalent contaminating viruses, together with viruses from the Picornaviridae, Astroviridae, Reoviridae, Caliciviridae, Adenoviridae, Birnaviridae, and Anelloviridae families. Putative new viral species were also identified. The loa

Metagenomic analysis of fecal samples from diarrheic pigs in China identified 10 complete circular single-stranded DNA viral genomes previously designated as putative porcine circovirus 5 (PCV5)-like viruses. Genome characterization revealed a typical cressdnavirus organization with bidirectionally oriented Rep and Cap genes. Phylogenetic analyses based on complete genomes and encoded proteins showed that these viruses do not cluster with members of Circoviridae but instead form a distinct lineage within the family Pecoviridae, closely related to the genus Macochavirus. Sequence identity, gene

Human immunodeficiency virus-1 (HIV-1) infection disrupts the homeostatic equilibrium between the host and commensal microbes. However, the dynamic changes of plasma commensal viruses and their role in HIV/simian immunodeficiency virus (SIV) pathogenesis are rarely reported. Here, we investigated the longitudinal changes of plasma virome, inflammation levels, and disease markers using an SIV-infected Macaca leonina model. Large expansions of plasma Anelloviridae, Parvoviridae, Circoviridae and other commensal viruses, and elevated levels of inflammation and D-dimer were observed since the chro

Porcine circovirus (PCV) comprises four types, PCV1, PCV2, PCV3, and PCV4, which belong to the Circovirus genus of the family Circoviridae. PCV1 is nonpathogenic, whereas PCV2, PCV3, and PCV4 can infect pigs and cause disease. However, due to a lack of experimental evidence, whether vaccines based on PCV capsid (Cap) can induce cross-reactivity against PCVs remains controversial. In this study, recombinant truncated capsids (rCaps) of PCV2, PCV3, and PCV4 were highly and efficiently expressed and purified, followed by the development and evaluation of antibodies against PCVs. The results showe

The International Committee on Taxonomy of Viruses (ICTV) holds a ratification vote annually after review of newly proposed taxa by ICTV Study Groups and members of the virology community. In March 2025, the vote outcome of the 11 proposals within the mandate of the Animal DNA Viruses and Retroviruses Subcommittee was made public. Here, we provide a summary of the newly accepted proposals. These include reorganization of taxa in the realm Varidnaviria, classification of the 'polinton-like' viruses into a new family (Phypoliviridae) within a new order Archintovirales; establishment of a new phy

Viromics studies are allowing us to understand not only the enormous diversity of the virosphere, but also the potential threat posed by the emerging viruses. Regarding the latter, the main concern lies in monitoring the presence of RNA viruses, but the zoonotic potential of some DNA viruses, on which we have focused in the present study, should also be highlighted. For this purpose, we analyzed 160 fecal samples from 14 species of small terrestrial mammals, 9 of them belonging to the order Rodentia. This allowed us to identify a total of 25 complete or near-complete genomes belonging to the f

Circoviruses are small viruses in the Circoviridae family that associate with a wide spectrum of disease from asymptomatic to lethal in different animal. In the current study, a novel swine-origin circovirus with unique genomic architecture was determined from both oropharyngeal swabs and fecal samples collected from febrile pigs in Jiangsu Province, China. The complete genome sequence of this novel porcine circovirus strain (named as PCV-CH-ZJ01) was determined to be 2544 nt in length, containing four major open reading frames: Rep1, 498-833 nt, 112 aa; Rep2, 1055-1546 nt, 164 aa; Cap1, 1756-

Wild animals are considered reservoirs for emerging and reemerging viruses, such as the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Previous studies have reported that bats and ticks harbored variable important pathogenic viruses, some of which could cause potential diseases in humans and livestock, while viruses carried by reptiles were rarely reported. Our study first conducted snakes' virome analysis to establish effective surveillance of potential transboundary emerging diseases. Consequently, Adenoviridae, Circoviridae, Retroviridae, and Parvoviridae were identifie

Cycloviruses, small ssDNA viruses belonging to the Circoviridae family, have been suggested as possible causes of enteric, respiratory and neurological disorders in human patients. One of these species, cyclovirus-Vietnam (CyCV-VN), initially isolated from cerebrospinal fluid samples of patients with unexplained neurological disorders, has since been reported in serum samples from chronically patients infected with HBV, HCV or HIV, in Italy. On the other hand, CyCV-VN was not detected in serum samples from healthy individuals. Here, we report on a high prevalence of 43.4% (40/92) of CyCV-VN in

This study is focused on the identification of the faecal virome of healthy chickens raised in high-density, export-driven poultry farms in Brazil. Following high-throughput sequencing, a total of 7743 de novo-assembled contigs were constructed and compared with known nucleotide/amino acid sequences from the GenBank database. Analyses with blastx revealed that 279 contigs (4 %) were related to sequences of eukaryotic viruses. Viral genome sequences (total or partial) indicative of members of recognized viral families, including Adenoviridae, Caliciviridae, Circoviridae, Parvoviridae, Picobirna
