Citrinin and endosulfan induced teratogenic effects in Wistar rats.

Dietary exposures to food pollutants such as mycotoxin(s) or pesticide(s) are most significant due to their adverse effects on the production and reproduction in animals and the human population. The present investigation was conducted to evaluate the teratogenic potential of citrinin (CIT) and endosulfan either alone or in combination in pregnant rats during gestational days 6-20. Endosulfan (1 mg kg(-1) body weight, by oral intubation) and CIT (10 mg kg(-1) feed, through diet) when administered either alone or in combination in pregnant rats caused significant teratogenic effects in the deve

Interaction of potassium dichromate with the nephrotoxins, mercuric chloride and citrinin.

Potassium dichromate enhanced the effects of mercuric chloride or citrinin on renal slice transport of the organic ions p-aminohippurate (PAH) and tetraethylammonium (TEA) under certain experimental conditions. At the doses employed and times studied no effects, or only minimal effects, were observed on renal slice transport when the toxins were tested individually. Potassium dichromate (10 mg/kg) administered subcutaneously (s.c.) in combination with mercuric chloride (4 mg/kg, s.c.) or citrinin 35 or 55 mg/kg administered intraperitoneally (i.p.), resulted in a marked depression of organic i

PRESENCE OF CITRININ IN GRAINS AND ITS POSSIBLE HEALTH EFFECTS.

BACKGROUND: Citrinin is a mycotoxin produced by several species of the genera Aspergillus, Penicillium and Monascus and it occurs mainly in stored grain. Citrinin is generally formed after harvest and occurs mainly in stored grains, it also occurs in other plant products. Often, the co-occurrence with other mycotoxins is observed, especially ochratoxin A, which is usually associated with endemic nephropathy. At the European Union level, systematic monitoring of Citrinin in grains began with the aim of determining its highest permissible amount in food. Thus, far the systematic monitoring of th

Toxic effects of citrinin on the male reproductive system in mice.

This study investigated the effects of citrinin (CTN) on male mouse reproductive organs. Adult male mice were exposed to intraperitoneal injection of CTN at 0-6.25 mg/kg body weight daily for 7 days, and then mated with sexually mature untreated female mice. Reproductive organ relative weights, semen quality, serum testosterone concentrations and fertility of treated mice were assessed. CTN significantly increased relative weights of the testes, epididymis, seminal vesicle and preputial gland, increased the number of abnormal spermatozoa and decreased the number of live spermatozoa. A signific

Citrinin exposure disrupts organelle distribution and functions in mouse oocytes.

Citrinin (CTN) is a secondary fungal metabolite produced by several species of Aspergillins and Penicillins, and it is widely found in vegetable-derived foods such as cereals and fermented rice-based food supplements. Previous studies indicated that CTN had immunotoxicity, hepatotoxicity, nephrotoxicity, and reproductive toxicity, which caused severe effects on human and animal health. However, the potential toxicity of CTN on the organelles of mouse oocytes is still unclear. In this study, we showed that the exposure to 30 μM CTN significantly reduced the developmental capacity of mouse oocyt

Citrinin inhibits the function of Leydig cells in male rats in prepuberty.

Citrinin, a mycotoxin existing in fruits, has nephrotoxicity, hepatotoxicity and embryotoxicity. The effects of citrinin on Leydig cell development in prepuberty remains unclear. Male Sprague-Dawley rats were gavaged with 0, 1, 2.5, and 5 mg/kg citrinin from postnatal days 21-28. Citrinin at 5 mg/kg significantly decreased serum testosterone levels, while increasing serum LH and FSH levels. Citrinin at 1-5 mg/kg markedly downregulated Hsd17b3 and HSD17B3 expression, while upregulating Srd5a1 (SRD5A1) and Akr1c14 (AKR1C14) expression at 2.5 and/or 5 mg/kg. Citrinin at 5 mg/kg also significantly

Studies on the sequential development and pathogenesis of citrinin mycotoxicosis in turkeys and ducklings.

The toxic effects of citrinin in turkeys and ducklings was studied in four trials. Citrinin dissolved in dimethyl sulfoxide-70% ethanol solution (3:1, volume/volume) was administered by gavage to male turkey poults and male white Pekin ducklings. When seven-day-old ducklings were given doses of citrinin between 30 to 110 mg/kg body weight, most of the treated ducklings which died (49/80) did so within four to 12 hours. Blood samples were collected sequentially at 3, 6, 12, and 24 hours after administration from seven-day-old ducklings given the single lethal dose (LD50). The alterations includ

Combination effect of citrinin and other chemicals on rat kidney tumorigenesis.

Histological studies were made on the nephrotoxic effect of citrinin on the kidneys of rats, with or without previous treatment with the nephrotoxic chemicals, N-(3,5-dichlorophenyl)succinimide (NDPS) and N-nitrosodimethylamine (DMN). Oral administration of 0.02% or 0.05% citrinin alone caused signs of kidney injury but did not induce kidney tumors. On treatment with DMN alone, 8 of 14 rats (57.1%) developed kidney tumors; two (14.3%) were renal cell tumors, eight (57.1%) embryonal cell tumors, and one (7.1%) hemangioendothelioma. On the other hand, kidney tumors developed in 18 of 19 rats (94

Panax ginseng extract reduces citrinin production and attenuates nephrotoxicity in Monascus purpureus.

AIMS: Monascus purpureus is a widely used filamentous fungus in food and medicinal fermentation, yet the co-production of citrinin, a nephrotoxic mycotoxin, poses significant safety concerns. In this study, we established a co-growth system of M. purpureus with Panax ginseng to investigate the regulatory effects of P. ginseng on citrinin biosynthesis and related toxicity. METHODS AND RESULTS: HPLC analysis revealed that P. ginseng supplementation significantly reduced citrinin production by approximately 68% without impairing fungal biomass accumulation. RT-qPCR results showed that the expres

Citrinin Induces Hepatic Inflammatory Injury through the PERK-CHOP-NLRP3 Axis-Mediated Pyroptosis.

Citrinin (CTN), a widespread food and feed contaminant, poses a significant health risk, yet its hepatic toxicity remains unclear. Here, we investigated the role of endoplasmic reticulum (ER) stress-mediated pyroptosis in CTN-induced liver injury using mice and HL-7702 cells. CTN exposure disrupted the hepatic cord structure, induced hepatocyte swelling with karyolysis, and promoted inflammatory infiltration. Liver injury markers and pro-inflammatory cytokines IL-1β and IL-18 were significantly elevated in both models, confirming inflammatory liver injury. Mechanistically, CTN activated pyropt

Citrinin Exposure Induced Testicular Damage and Spermatogenesis Disorder by Triggering Endoplasmic Reticulum Stress.

Damage to the reproductive system is the key factor leading to male infertility. Citrinin (CTN) is produced by Penicillium and Aspergillus in nature, and is definitely found in food and animal feed. Studies have revealed that CTN can cause damage to male reproductive organs and reduce fertility, but the mechanism of toxicity has not been revealed. In the present study, male Kunming mice were given different doses of CTN (0, 1.25, 5 or 20 mg/kg BW) by intragastric administration. The results demonstrated that CTN exposure caused disorder of androgen, a decline in sperm quality, and histopatholo

Safety evaluation of sub-chronic oral exposure to ochratoxin A and citrinin in Wistar rats.

Ochratoxin A (OTA) and citrinin (CTN) are naturally occurring mycotoxins produced by filamentous fungi that frequently contaminate food and pose risks to human and animal health. This study evaluated the systemic toxicity of subchronic oral exposure to OTA and CTN in Wistar rats following Organization for Economic Co-operation and Development (OECD) Test Guideline 408. Fifty rats (25 males and 25 females) were randomly assigned to five groups: vehicle control, OTA (25 or 100 μg/kg), and CTN (25 or 100 μg/kg). Animals were orally dosed for 90 days, with clinical signs monitored daily and body w

Combined Inhibitory Effects of Citrinin, Ochratoxin-A, and T-2 Toxin on Aquaporin-2.

Aquaporins form a large family of transmembrane protein channel that facilitates selective and fast water transport across the cell membrane. The inhibition of aquaporin channels leads to many water-related diseases such as nephrogenic diabetes insipidus, edema, cardiac arrest, and stroke. Herein, we report the molecular mechanism of mycotoxins (citrinin, ochratoxin-A, and T-2 mycotoxin) inhibition of aquaporin-2 (AQP2) and arginine vasopressin receptor 2.
