Isolation and identification of Clavibacter made easy.

Clavibacter species are the most important gram-positive bacterial plant pathogens in terms of both disease severity and economic importance. As a group of regulated and quarantined plant pathogens, Clavibacter members are slow-growing organisms on general culture media, leading to difficulties in their isolation and identification. In this study, we developed an efficient, straightforward method for the isolation and identification of Clavibacter members from plant materials. First, a semi-selective medium was developed from a modification of conventional YPGA and YDC media, namely ClaYPGA an

Ecogenomic Diversity of Clavibacter nebraskensis in North America.

Goss's wilt and leaf blight of maize is caused by Clavibacter nebraskensis and has reemerged as an important disease in North America. Despite its epidemiological relevance, this species remains poorly characterized in terms of population structure, functional diversity, and ecological differentiation, particularly among strains reported from Mexico. In this study, long-read whole-genome sequencing and phenotypic assays were used to characterize genomic diversity, virulence, and fitness-associated traits in C. nebraskensis. We generated 24 long-read genomes, including 20 contemporary Mexican i

Genetic diversity and population structure of Clavibacter michiganensis subsp. nebraskensis.

Clavibacter michiganensis subsp. nebraskensis (CMN) is a gram-positive bacterium and an incitant of Goss's bacterial wilt and leaf blight or "leaf freckles" in corn. A population structure of a wide temporal and geographic collection of CMN strains (n = 131), originating between 1969 and 2009, was determined using amplified fragment length polymorphism (AFLP) analysis and repetitive DNA sequence-based BOX-PCR. Analysis of the composite data set of AFLP and BOX-PCR fingerprints revealed two groups with a 60% cutoff similarity: a major group A (n = 118 strains) and a minor group B (n = 13 strain

The influence of dihydroquercetin on the cell membrane model of Clavibacter michiganensis subsp. sepedonicus.

Dihydroquercetin (DHQ) exhibited antimicrobial activity associated with its interaction with bacterial cell membranes. However, molecular-level mechanism has not been revealed. Therefore, this study investigated the potential interactions between DHQ and model membrane by constructing a Clavibacter michiganensis subsp. sepedonicus (Cms) membrane model. The optimization of preparation condition was sonicated 45 °C for 20 min determined by dynamic light scattering (DLS). The average particle size of the liposomes showed an average particle size of 162.43 ± 2.41 nm, with zeta potential of -26.77

Colonization and Movement of Green Fluorescent Protein-Labeled Clavibacter nebraskensis in Maize.

Clavibacter nebraskensis causes Goss's bacterial wilt and leaf blight, a major disease of maize. Infected crop residue is the primary inoculum source and infection can occur via wounds or natural openings, such as stomata or hydathodes. The use of resistant hybrids is the primary control method for Goss's wilt. In this study, colonization and movement patterns of C. nebraskensis during infection were examined using green fluorescent protein (GFP)-labeled bacterial strains. We successfully introduced a plasmid to C. nebraskensis via electroporation, which resulted in GFP accumulation. Fluoresce

Unmarked Gene Editing in Clavibacter michiganensis Using CRISPR/Cas9 and 5-Fluorocytosine Counterselection.

Plant-pathogenic bacteria in the genus Clavibacter are important quarantine species that cause considerable economic loss worldwide. The development of effective gene editing techniques and additional selectable markers is essential to expedite gene functional analysis in this important Gram-positive genus. The current study details a highly efficient unmarked CRISPR/Cas9-mediated gene editing system in Clavibacter michiganensis, which couples the expression of cas9 and single-guide RNA with homology-directed repair templates and the negative selectable marker codA::upp within a single plasmid

Effect of leaf surface waxes on leaf colonization by Pantoea agglomerans and Clavibacter michiganensis.

To evaluate the influence of leaf cuticular waxes on bacterial colonization of leaves, bacterial colonization patterns were examined on four glossy maize (Zea mays L.) mutants that were altered in their cuticular wax biosynthesis. Mutant gl3 was indistinguishable from the wild-type maize in its ability to foster colonization by the two bacterial species, Pantoea agglomerans and Clavibacter michiganensis subsp. nebraskensis. In contrast, the other three mutants supported the development of populations that significantly differed in size from those on the wild type. Mutant gl5 gl20 supported sma

Comparative genomics of strains of varying virulence of the gram-positive bacterial pathogen of maize, Clavibacter nebraskensis.

BACKGROUND: Clavibacter nebraskensis (Cn) is a Gram-positive bacterial plant pathogen that infects the vascular tissue of maize, causing Goss's Wilt and Leaf Blight. Detection of highly virulent strains since the mid-2000s suggests that genetic divergence has contributed to the re-emergence of the disease following its effective management and subsequent disappearance in the 1980s. However, few genomic resources exist for this pathogen, and little is known about genomic variation in the species. Our study aimed to characterize genomic diversity across a set of differentially virulent strains i

Invasion of Clavibacter michiganensis into the Xylem and Surrounding Tissues Drives Histological Alterations and Disease Development in Tomato.

Clavibacter michiganensis is a xylem-colonizing and seed-borne phytopathogen that causes bacterial canker disease in tomato plants. In this study, we aimed to determine the colonization patterns of C. michiganensis, as well as the histological changes in tomato plants during infection. To achieve these, GFP-tagged C. michiganensis was inoculated into tomato plants using three methods: stem injection, soil drenching with or without prior root cutting, and cotton-pad inoculation. After inoculation, bacterial colonization was observed using GFP signals, and histological changes were examined usin

Clavibacter michiganensis Reframed: The Story of How the Genomics Era Made a New Face for an Old Enemy.

OBJECTIVE: Bacterial wilt and canker of tomato caused by the gram-positive corynebacterial species Clavibacter michiganensis is an economically important disease threatening the tomato industry in both open-air and greenhouse productions around the world. The disease occurs in many countries, with a particular importance in regions characterised by high temperature and water scarcity. Management of bacterial canker has been a major problem since its original description in 1909. This is due in part to the seedborne nature of the pathogen, allowing the bacterium to be transmitted over long dist

Clavibacter nebraskensis causing Goss's wilt of maize: Five decades of detaining the enemy in the New World.

UNLABELLED: Goss's bacterial wilt and leaf blight of maize (Zea mays) caused by the gram-positive coryneform bacterium Clavibacter nebraskensis is an economically important disease in North America. C. nebraskensis is included within the high-risk list of quarantine pathogens by several plant protection organizations (EPPO code: CORBMI), hence it is under strict quarantine control around the world. The causal agent was reported for the first time on maize in Nebraska (USA) in 1969. After an outbreak during the 1970s, prevalence of the disease decreased in the 1980s to the early 2000s, before t

Detection of Clavibacter michiganensis subsp. michiganensis in viable but nonculturable state from tomato seed using improved qPCR.

Clavibacter michiganensis subsp. michiganensis (Cmm) is a seed-borne pathogen that causes bacterial canker disease of tomato. Cmm is typically detected in tomato seeds using quantitative real-time polymerase chain reaction (qPCR) combined with culture-based isolation. The viable but nonculturable (VBNC) state of Cmm may result in the underestimation or false negative detection of the pathogen. In the present study, propidium monoazide (PMA) and its improved structure PMAxx were used to pretreat Cmm prior to DNA extraction, followed by qPCR. Both PMA and PMAxx could bind to the chromosomal DNA

Identification of beneficial bacterial strains for tomato growth promotion and biocontrol of bacterial canker caused by Clavibacter michiganensis.

Bacterial canker of tomato, caused by Clavibacter michiganensis (Cm), remains difficult to control due to lack of effective management options. In this study, a collection of over 500 bacterial isolates was screened in vitro for antagonistic activity against Cm and plant growth-promoting traits. Based on these results, 32 candidates were evaluated in planta, leading to the identification of three highly effective strains: Pantoea agglomerans SO16PY and two Pseudomonas marginalis sensu lato strains, IRDA16 and SO16PC, which consistently enhanced tomato vegetative growth. Notably, P. agglomerans

PAM-Independent Cas12a Detection of Specific LAMP Products by Targeting Amplicon Loops.

A straightforward approach is suggested to selectively recognize specific products of loop-mediated isothermal amplification (LAMP) with the Cas12a nuclease without a need for a protospacer adjacent motif (PAM) in the sequence of LAMP amplicons (LAMPlicons). This strategy is based on the presence of single-stranded DNA loops in LAMPlicons and the ability of Cas12a to be trans-activated via the binding of guide RNA (gRNA) to single-stranded DNA in the absence of PAM. The approach feasibility is demonstrated on Clavibacter species-multiple bacterial plant pathogens that cause harmful diseases in
