Plant transcriptome data mining identified twenty-two putative novel taxa in the family Closteroviridae.

The family Closteroviridae comprises filamentous, RNA genome-containing viruses that infect plants. In the present study, public domain SRA libraries derived from plants were mined for novel closteroviral sequences, resulting in the identification of twenty-two putative novel closterovirids across twenty-two plant genera. The identified viruses were represented by nineteen coding-complete and three partial genomes. Based on genome organization, pairwise sequence identity and phylogenetic analysis, the viruses were classified in the following genera: Ampelovirus (6), Bluvavirus (1), Closterovir

A Novel Genetic Variant of Grapevine leafroll-associated virus-3 (GLRaV-3) from Idaho Grapevines.

Grapevine leafroll-associated virus-3 (GLRaV-3) is a major constraint on profitable grapevine cultivation. The virus is transmitted efficiently by mealybugs and soft scale insects, or through vegetative propagation by cuttings, and is present worldwide, wherever grapevines are grown. GLRaV-3 exists as a complex of genetic variants currently classified in several phylogenetic groups that can differ from each other by as much as 30% in nucleotide sequence of the whole genome. In the course of the GLRaV-3 testing of wine grapes in southern Idaho, plants of two grapevine cultivars were found to ha

Characterization of a new closterovirus infecting Ficus carica.

Figs (Ficus carica L.) are economically important worldwide, valued for both fruit production and ornamental purposes. Due to the widespread use of vegetative propagation, diverse viruses have accumulated in fig germplasm. To date, more than 14 viruses have been described associated to fig trees, including eight members of the family Closteroviridae, although their role in the etiology of fig mosaic disease (FMD) is unclear. Characteristic symptoms of FMD include mosaic and chlorotic spots on the leaves, along with deformation of fruits and leaves. Fig mosaic virus (FMV) is the causal agent of

Occurrence of Major Grapevine Viruses in Oklahoma and Genetic Diversity Assessment of Field Isolates of Grapevine Leafroll-Associated Virus 3 and Grapevine Red Blotch Virus.

Oklahoma grapevine production interests are on the rise, but very little is known about the occurrence of major grapevine-infecting viruses in the state. To address this knowledge gap, a total of 85 grapevine samples were collected from eight vineyards across six counties in Oklahoma during the spring and fall of 2024. Total RNA extracts from each sample were assayed for 14 major viruses by RT-PCR, including grapevine leafroll-associated virus 1 (GLRaV-1), GLRaV-2, GLRaV-3, GLRaV-4, grapevine fanleaf virus (GFLV), grapevine red blotch virus (GRBV), tobacco ringspot virus, tomato ringspot virus

Analysis of genetic variability in grapevine leafroll-associated virus 13 and the characteristics of the p23.

Grapevine leafroll-associated virus 13 (GLRaV-13) is a newly discovered virus that infects grapevines; however, prior research on its sequence features and gene functions has been minimal. In this study, GLRaV-13 was detected in 34 of 234 grapevine samples, and the complete genome sequences of three Chinese isolates of GLRaV-13 were successfully obtained through high-throughput sequencing (HTS), overlapping RT-PCR, and RACE. Additionally, the interactions among GLRaV-13 encoded proteins were revealed by yeast two-hybrid (Y2H) and bimolecular fluorescence complementation (BiFC) assays. There ar

Real-time detection of cucurbit chlorotic yellows virus by RT-LAMP.

Cucurbit chlorotic yellows virus (CCYV) of the genus Crinivirus and family Closteroviridae, is an emerging infectious agent transmitted by whiteflies that mainly infects cucumber, melon and watermelon plants. Here, we developed a procedure based on reverse transcription (RT) followed by loop-mediated isothermal amplification (LAMP) to detect CCYV in real-time by using a fluorescent dye and incubating at 65 °C. This method detected CCYV in RNA extracts at about 10 min and in nylon membrane-filtered crude extracts between 55 and 95 min. The detection was sensitive, ten times higher than RT follo

Field evaluation of Cucurbit yellow stunting disorder virus transmission by Bemisia tabaci.

Cucurbit yellow stunting disorder virus (CYSDV) is a whitefly-transmitted Crinivirus (Closteroviridae) that impacts melon production in many parts of the world including the USA. It has been responsible for melon crop loss in the southwestern U.S. since 2006 when it was first identified. Control strategies have revolved mainly around chemical control, but research to identify suitable products and approaches to implementing them have lagged. The current study investigated the performance of four systemic insecticides in the field while concurrently tracking CYSDV disease progression after cont

Antiviral activity of tiazofurin and mycophenolic acid against Grapevine leafroll-associated virus 3 in Vitis vinifera explants.

The ability to control plant viral diseases with chemicals has great potential value for agriculture, but few chemicals are available to date due to the difficulty in obtaining effective drugs. IMP dehydrogenase is an enzyme which catalyzes the conversion of inosine 5'-monophosphate to xanthosine 5'-monophosphate in the de novo purine nucleotide synthetic pathway, and is considered a sensitive target for antiviral drugs. Two IMPDH inhibitors, tiazofurin (TR) and mycophenolic acid (MPA), were tested for their inhibitory effect on Grapevine leafroll-associated virus 3 (GLRaV-3) in in vitro grape

Nucleotide sequence, genome organization and phylogenetic analysis of Strawberry pallidosis associated virus, a new member of the genus Crinivirus.

The complete nucleotide sequence of Strawberry pallidosis associated virus (SPaV), a newly identified member of the genus Crinivirus, family Closteroviridae has been determined. RNA 1 is 8067 nucleotides long and encodes at least three open reading frames (ORFs). The first ORF (ORF 1a) specifies a multifunctional protein that has papain-like proteinase, methyltransferase and RNA helicase domains. The RNA-dependent-RNA polymerase is encoded in ORF 1b and is probably expressed by a +1 ribosomal frameshift. The 3' ORF of RNA 1 encodes a small protein with two potential transmembrane helices. RNA

Silencing Suppressor Protein p26 of Areca Palm Velarivirus 1 (APV1) Interacts With SGS3 and Promotes Its Degradation Via the Ubiquitination Pathway.

RNA silencing is a conserved defence mechanism against viral infections in eukaryotes, whereas many plant viruses evolved viral suppressors of RNA silencing (VSRs) to counteract the antiviral silencing in hosts. Areca palm velarivirus 1 (APV1), a member of genus Velarivirus, family Closteroviridae, is a causal agent of yellow leaf disease affecting Areca catechu in Hainan, China. In this work, we identified the APV1-encoded p26 protein as a multifunctional VSR that suppresses local silencing induced by single-stranded RNA (ssRNA), systemic silencing induced by ssRNA and double-stranded RNA (ds

Physalis angulata: A New Natural Host of Tomato chlorosis virus in Brazil.

Tomato chlorosis virus (ToCV) and Tomato infectious chlorosis virus (TICV) are the two Solanaceae-infecting Crinivirus species (family Closteroviridae) of worldwide importance. In Brazil, only ToCV has been detected under natural conditions infecting tomato (Solanum lycopersicum), sweet pepper (Capsicum annuum), and potato (S. tuberosum), causing foliar chlorosis (1, 3). However, there are no formal reports of alternative weed hosts of ToCV. During crop surveys in Capão Bonito, São Paulo State, Brazil (May 2011), a high incidence (above 20%) of plants of the weed, cut leaf ground cherry (Physa
