Figs (Ficus carica L.) are economically important worldwide, valued for both fruit production and ornamental purposes. Due to the widespread use of vegetative propagation, diverse viruses have accumulated in fig germplasm. To date, more than 14 viruses have been described associated to fig trees, including eight members of the family Closteroviridae, although their role in the etiology of fig mosaic disease (FMD) is unclear. Characteristic symptoms of FMD include mosaic and chlorotic spots on the leaves, along with deformation of fruits and leaves. Fig mosaic virus (FMV) is the causal agent of

RNA silencing, a conserved gene-regulatory mechanism mediated by small interfering RNAs (siRNAs), is a major component of plant antiviral immunity. The plant RNA-binding protein SUPPRESSOR OF GENE SILENCING 3 (SGS3) forms condensates that drive siRNA body assembly and promote siRNA biogenesis however, the regulators of SGS3 condensate formation and their roles in antiviral responses remain largely unknown. Here, we show that the cold-regulated 15-kDa dehydrin protein (COR15) homologs from Citrus aurantifolia and Nicotiana benthamiana interact with the citrus tristeza virus (CTV)-encoded p20 pr

Viroporins are viral proteins that form ion channels in the membranes of the host and, thus, alter the host ion homeostasis to create more favorable environments for the virus. Since the discovery of the ion channel activity of the M2 protein encoded by influenza virus A (species: Alphainfluenzavirus influenzae), many additional viral proteins have also been characterized as viroporins. However, most viroporins known thus far belong to animal viruses, while the discovery of plant virus viroporins has significantly lagged. In this work, we present evidence that the p33 protein, a membrane-assoc

Citrus Tristeza Virus (CTV) is the most economically significant viral pathogen of citrus worldwide, transmitted in a semipersistent manner by several aphid species, most efficiently by Aphis [Toxoptera] citricidus. Transmission efficiency varies substantially among CTV genotypes, but the viral determinants underlying these differences remain poorly understood. In this study, we investigated the roles of three viral genes, p33, p18, and p13, in aphid transmission using a genetically engineered CTV-T36 mutant (CTV-T36Δp33Δp18Δp13) as a backbone. This mutant is derived from the poorly transmissi

Four stable hybridoma cell lines secreting monoclonal antibodies to grapevine closterovirus A (GVA) were obtained by fusing spleen cells of immunized BALB/c mice with mouse myeloma cell line Sp2/0-Ag 14. In ELISA all MAbs reacted with virus in leaf extracts from Nicotiana benthamiana, glass-house-, field-, or in vitro-grown grapevines, or with cortical shavings from mature grape canes. In IEM tests, only one of the MAbs (PA3.F5) decorated virus particles on the entire surface. This MAb was likely induced by a surface antigenic determinant, whereas the other three MAbs (PA3.D 11, PA3.C 6, and P

Citrus can host a number of important vector- and graft-transmissible pathogens which cause severe diseases. Citrus disease management and clean stock programs require pathogen detection systems which must be economical and sensitive to maintain a healthy citrus industry. Rapid diagnostic tests for simultaneous detection of major graft-transmissible disease agents enable reduction of cost and time. The genetic and biological features of viruses and viroids can vary according to the strains/variants, with severe and mild strains described within the same species. The use of diagnostic tests tha

This study reports a simple template-based reverse transcription-polymerase amplification assay (ST-RT-RPA) for detection of citrus tristeza virus (CTV) from crude plant extract lysed in NaOH:EDTA (1:1) without the need of tedious RNA isolation. The developed assay showed versatility in its usage as amplification can be performed at wide temperature range (14°C to 42°C) and incubation time (4 to 32 min), although the best conditions were 38°C for 30 min. The developed ST-RT-RPA assay could detect the CTV up to 10-8 dilution of crude plant extract of NaOH:EDTA and up to 0.01 fg µl-1 of RNA of C

The family Closteroviridae comprises filamentous, RNA genome-containing viruses that infect plants. In the present study, public domain SRA libraries derived from plants were mined for novel closteroviral sequences, resulting in the identification of twenty-two putative novel closterovirids across twenty-two plant genera. The identified viruses were represented by nineteen coding-complete and three partial genomes. Based on genome organization, pairwise sequence identity and phylogenetic analysis, the viruses were classified in the following genera: Ampelovirus (6), Bluvavirus (1), Closterovir

Apscaviroid nanocitri (citrus dwarfing viroid, CDVd) is a graft-transmissible pathogen that frequently coexists with other citrus viroids and affects tree vigor. This study evaluated the occurrence of citrus viroids in small growers and commercial orchards in Chile and characterized CDVd strains obtained from urban Citrus spp. trees, with unknown propagation history following the discontinuation of the national citrus certification program. A total of 301 samples from five citrus-producing regions were analyzed, revealing that CDVd is widely distributed and commonly associated with Hostuviroid

Due to the low titer or uneven distribution of Citrus tristeza virus (CTV) in field samples, detection of CTV by using conventional detection techniques may be difficult. Therefore, in the present work, the cadmium-telluride quantum dots (QDs) was conjugated with a specific antibody against coat protein (CP) of CTV, and the CP were immobilized on the surface of gold nanoparticles (AuNPs) to develop a specific and sensitive fluorescence resonance energy transfer (FRET)-based nanobiosensor for detecting CTV. The maximum FRET efficiency for the developed nano-biosensor was observed at 60% in AuNP

This article details combined portable sample preparation and colorimetric detection using loop-mediated isothermal amplification (RT-LAMP) of Citrus tristeza virus (CTV) from citrus leaves. The sample preparation employs an OmniLyse micro-homogenizer and cellulose paper disks to extract and isolate total nucleic acids in <15 min. Primer and dimethyl sulfoxide (DMSO) concentrations were optimized to minimize RT-LAMP assay reaction times and maximize the delay in the appearance of false positives due to non-specific amplification, respectively. The CTV primers were assessed against a panel of 2

Trifoliate orange (Poncirus trifoliata), a deciduous close relative of evergreen Citrus, has important traits for citrus production, including tolerance/resistance to citrus greening disease (Huanglongbing, HLB) and other major diseases, and cold tolerance. It has been one of the most important rootstocks, and one of the most valuable sources of resistance and tolerance genes for citrus. Here we present a high-quality, chromosome-scale genome assembly of P. trifoliata. The 264.9-Mb assembly contains nine chromosomal pseudomolecules with 25 538 protein-coding genes, covering 97.2% of the estima

During infection, Citrus tristeza virus (CTV) produces a non-coding subgenomic RNA referred to as low-molecular-weight tristeza 1 (LMT1), which for a long time has been considered as a by-product of the complex CTV replication machinery. In this study, we investigated the role of LMT1 in the virus infection cycle using a CTV variant that does not produce LMT1 (CTV-LMT1d). We showed that lack of LMT1 did not halt virus ability to replicate or form proper virions. However, the mutant virus demonstrated significantly reduced invasiveness and systemic spread in Nicotiana benthamiana as well as an
