Genetic modification of Clostridium kluyveri for heterologous n-butanol and n-hexanol production.

UNLABELLED: The mesophilic microbe Clostridium kluyveri serves as the most commonly used model microbe to elucidate the physiology and biochemistry of ethanol-based chain elongation via reverse β-oxidation. In this pathway, ethanol and acetate are converted into short- and medium-chain carboxylates. However, to date, no genetic system has been published in a peer-reviewed publication. Here, we report the development of versatile genetic tools for C. kluyveri, utilizing the pMTL Clostridia shuttle vector system and thiamphenicol as a selective marker. We identified the native restriction-modifi

Cu2ZnSnS4-Sporomusa ovata photobiohybrids coupled with Clostridium kluyveri fermentation for CO2 conversion to C4-C6 fatty acids.

The conversion of greenhouse CO2 into long-chain chemicals remains a major challenge in artificial photosynthesis. Here, we present a novel semi-biological platform that integrates microbial photocatalysis with fermentation to produce fatty acids from CO2. The process begins with light-driven CO2 reduction to acetate and ethanol, enabled by a photobiohybrid composed of the earth-abundant, non-toxic semiconductor Cu2ZnSnS4 (CZTS) and the CO2-fixing electrotroph Sporomusa ovata (S. ovata). In this CZTS-S. ovata hybrid, the CZTS nanoparticles act as light absorbers, generating reducing equivalent

Synergistic acetate-butyrate co-utilization enhances caproate production by Clostridium kluyveri CICC 24509 under high substrate loads.

Microbial chain elongation presents a compelling circular economy strategy for converting low-value waste streams into valuable chemicals like caproate. However, realizing its full potential is hampered by challenges, including limited microbial tolerance to industrially relevant high substrate concentrations and suboptimal electron donor-acceptor utilization. This study addresses these limitations by evaluating the performance of a previously uncharacterized chain-elongating bacterium, Clostridium kluyveri CICC 24509, under high-substrate conditions using ethanol as the electron donor. We sys

Medium-Chain Fatty Acids (MCFA) Production Through Anaerobic Fermentation Using Clostridium kluyveri: Effect of Ethanol and Acetate.

Medium-chain fatty acids (MCFA) are saturated monocarboxylic acids and can be used as antimicrobials, corrosion inhibitors, precursors in biodiesel, and bioplastic production. In the present study, MCFA production was evaluated with acetate and ethanol using the bacteria Clostridium kluyveri. Effects of substrate, electron donor, and methane inhibitor on MCFA production were evaluated. Bacteria successfully converted the ethanol and acetate to butyrate (C4), caproate (C6), and caprylate (C8) by chain elongation process. The highest concentrations of butyrate (4.6 g/l), caproate (3.2 g/l), and

A novel mechanistic insight into CO2 effects on chain elongation in Clostridium kluyveri based on transcriptomic and ¹³C-targeted metabolomic analyses.

Chain elongation is an important technology for processing organic wastewater (such as brewing wastewater), and Clostridium kluyveri is one of the most crucial microorganisms involved. This study aims to investigate the regulatory mechanism of inorganic carbon (IC) on the growth and metabolic pattern of Clostridium kluyveri. Results revealed that the production of caproate by Clostridium kluyveri from ethanol and acetate is a growth-coupled fermentation type and regulated by IC. With increased IC concentration, both the efficiency of caproate synthesis and the growth rate of Clostridium kluyve

Self-sufficient fermentation paradigm for cassava stillage valorization into C6 carboxylic acids: regulatory mechanisms and novel microbe identification.

Cassava stillage (CS), a carbohydrate-rich byproduct of bioethanol production, holds significant untapped potential as a renewable resource. Upcycling this problematic wastewater offers great promise for addressing both environmental challenges and the demand for sustainable biochemicals. Here, this study proposed a self-sufficient biotechnological paradigm that directly valorizes CS into medium-chain carboxylic acids (MCCAs, e.g., caproic acid) by integrating lactic acid/butyric acid-type fermentation with microbial chain elongation (CE) by two phase fermentation regulatory. Prokaryotic community diversity, assembly processes, and potential metabolic pathways of a hexanoic acid-producing microbiota constructed from Chinese baijiu fermentation pit mud.

Hexanoic acid (HA), a vital platform chemical, can be produced by various mixed cultures using ethanol or ethanol-containing waste. However, systematic research remains lacking on its ecological succession, community assembly, and core synthetic pathways. In this study, an ethanol-based HA-producing synthetic community (SynCom) was constructed from the pit mud using a top-down strategy, and dominated by Clostridium sensu stricto 12 (primarily Clostridium kluyveri, 86%), Clostridium sensu stricto 18 and Pseudoclavibacter. A highly significant positive correlation (P < 0.01) was observed between

Engineering Clostridium Tyrobutyricum for High Butanol Production through Induction Expression of Exogenous NADPH-Dependent HBD.

Clostridium tyrobutyricum Δcat1::adhE2 is a promising cell factory for butanol production because of its robustness, high butanol tolerance, and minimal butyrate production. However, excessive acetate and ethanol production remains a major bottleneck limiting its butanol yield. Coexpressing an exogenous hbd(Ck) encoding the NADPH-dependent 3-hydroxybutyryl-CoA dehydrogenase (HBD) from Clostridium kluyveri with adhE2 could increase the C4 carbon flux, resulting in increased butanol and decreased acetate and ethanol production. However, constitutively overexpressing hbd(Ck) in Δcat1::adhE2 shows

Development of novel microbial synthetic consortia for the production of antimicrobial fermentates containing caproate.

Short- and medium chain carboxylic acids (SCCA/MCCA) are natural antimicrobials produced by fermentation and chain elongation, but currently only a few SCCA/MCCA are used in food-related applications. With the aim to diversify the SCCA/MCCA profile of fermentates for biopreservation, we designed bioprocesses employing bacterial or multi-kingdom consortia to produce caproate-containing fermentates using a targeted cross-feeding strategy. We combined Limosilactobacillus reuteri, Clostridium kluyveri, and Saccharomyces cerevisiae, and quantified substrates utilization and metabolites. The antimic

Metabolic Engineering of Clostridium tyrobutyricum for High-Yield n-Butanol Production by Increasing Intracellular Reducing Equivalent with NADPH-Dependent 3-Hydroxybutyryl-CoA Dehydrogenase.

Clostridium tyrobutyricum was engineered to overexpress adhE2 encoding the aldehyde/alcohol dehydrogenase and an exogenous NADPH-dependent 3-hydroxybutyryl-CoA dehydrogenase from Clostridium kluyveri (Ckhbd) for n-butanol production. In general, large amounts of butyrate, acetate, and ethanol are also produced from glucose when butanol biosynthesis is hindered by limited intracellular NADH pools. In silico flux balance analysis showed that coupling NADP+/NADPH turnover with butanol production increased the reducing equivalent supply and butanol selectivity over ethanol and acids, thus increasi

Enhancing microbial electrosynthesis of fatty acids from carbon dioxide by combining bioaugmentation of an acetogenic enrichment and Fe-Sn oxide cathode coating.

This study evaluated the impact of bioaugmenting a mixed acetogenic enrichment with Clostridium kluyveri on CO2 reduction to fatty acids, specifically butyrate and caproate, in a microbial electrosynthesis (MES) cell equipped with an Fe-Sn oxide - coated cathode. Without bioaugmentation, at a volumetric CO2 inflow of 1.67 LCO2 (Lc d)-1 (per cathode chamber volume) approximately 86% of the CO2 was converted to the target products, leading to maximum production rates of butyrate and caproate of 875 mg (Lc d)-1 and 94 mg (Lc d)-1, respectively. Constructing simplified microbial consortia that couple lactic acid and ethanol utilization to highly produce caproic acid from liquor-making wastewater.

Converting biodegradable carbon in wastewater into medium-chain fatty acids (MCFAs) through stable microbiota is highly attractive. In this study, we utilized a top-down approach for constructing MCFA-producing microbial consortia. Specifically, an enrichment and plating-screening strategy employing lactic acid and ethanol as selective carbon sources was applied to isolate simplified caproic acid-producing microbial consortia from liquor-making pit mud. The representative microbial consortium SimpCom3 demonstrated high level of caproic acid production (14.62 ± 0.48 g/L) in a semi-synthetic med

Implementation of a Clostridium luticellarii genome-scale model for upgrading syngas fermentations.

Syngas fermentation is a powerful platform for converting waste streams into sustainable carboxylic acid precursors for value-added biochemicals. Steel mills produce significant syngas, yet industrial microbial syngas valorization remains unrealized. The most promising syngas-converting biocatalysts consist of Clostridia species, such as Clostridium kluyveri, Clostridium autoethanogenum, and Clostridium ljungdahlii. Clostridium luticellarii, a recently discovered species, shares close phylogenetic ties with these organisms. Preliminary metabolic studies suggest its potential for syngas acetoge

A metabolic relay for caproic acid production in Huangshui: Cooperative interaction between Clostridium tyrobutyricum and Caproiciproducens.

Microbial chain elongation for medium-chain fatty acid production has gained significant attention. This study elucidated how initial pH and carbon sources regulate microbial caproic acid production from organic waste. By successively subculturing pit mud with Huangshui, lactic acid, or ethanol at different pH values (4.0-6.0), we obtained two enriched consortia: NatCom H (from Huangshui) and NatCom E (from ethanol), both of which produce 10 g/L caproic acid at pH 6.0. Key functional divergence was observed: NatCom H maintained production from pH 5.0 to 6.0, whereas NatCom E was functional onl

[Formation of so-called byproducts (propanols, butanols et al.) from ethanol by microorganisms].

Microbiological literature implies and furnishes evidence that aliphatic alcohols and the corresponding carboxylates as well as acetone can be produced from ethanol during microbial metabolic processes. Propionate/propanol-1 followed by butyrate can be obtained by means of step-by-step reductive carboxylation of acetyl-CoA. Both butyrate/butanol-1 and caproate/hexanol-1 are typical fermentation products of Clostridium kluyvery. In cases where butyrate decomposition is disrupted up to 50% of butyrate is isomerised to isobutyrate. In addition to ethanol, butyrate and butanol-1, isopropanol and a

Enhanced electron transfer mediated by surface functional groups of targetedly modified sludge-based biochar for sustainable microbial chain elongation.

While unmodified sludge-based biochar (SBBC) exhibits limited electron transfer capacity in chain elongation (CE), its surface functional groups are recognized as critical factor influencing direct interspecies electron transfer (DIET). This study employed three modification methods, i.e., H2O2, HNO3, and anthraquinone sulfonate (AQS), to explore the effects of modified SBBC on CE process. Results indicated that the AQS-modified SBBC (SBBC-AQS) exhibited optimal electron transfer properties and electrochemical activity. Thus, SBBC-AQS greatly promoted DIET-mediated CE, leading to 62% higher ca

Biosynthesis of poly(4-hydroxybutyric acid) by recombinant strains of Escherichia coli.

The aim of this study was the production of the homopolyester poly(4-hydroxybutyric acid) (poly(4HB)) with recombinant strains of Escherichia coli. Wild-type strains and other widely used non-recombinant strains of E. coli are not able to produce polyhydroxyalkanoic acids (PHA) as storage compounds and cannot utilize 4-hydroxybutyric acid as sole carbon source. Accordingly, hybrid plasmids of pBluescript vectors were constructed which harbored the Alcaligenes eutrophus PHA synthase gene (phaC) and the Clostridium kluyveri orfZ putatively encoding a 4-hydroxybutyric acid-coenzyme A transferase.

Fe3O4-Enhanced Continuous Production of Medium-chain Fatty Acids from Sludge: Metagenomic Perspective on Microbial Synergy.

Fe3O4 exhibits promising potential in enhancing chain elongation (CE) for medium-chain fatty acids (MCFAs) production from sludge, yet its application in continuous flow processes remains underexplored. In this study, MCFAs were continuously produced from sludge via a Fe3O4-enhanced CE process. Optimal performance was achieved with 20 g/L Fe3O4, 5.8 g COD/L/d ethanol loading rate (ELR), and 3-day hydraulic retention time (HRT), yielding 11.4 ± 0.8 g COD/L MCFAs and a production rate of 3.7 ± 0.1 g COD/L/d. n-Caproate and n-caprylate accounted for 40.9 % and 54.4 % of total MCFAs, respectively.
